Abstract
本发明提供了一种多孔植物多糖凝胶膜体及其制备方法,属于面膜制作技术领域。本发明采用卡拉胶0.20‑30.0%,魔芋胶0.30‑30.0%、羟乙基纤维素0.10‑15.0%,甘油0.5‑30.0%,β‑环状糊精1.0‑3.0%,光甘草定0.05‑1.0%,纯化水为原料,制备了一种多孔植物多糖凝胶膜体。本发明提供的多孔植物多糖凝胶膜体具有稳定性高、不易被氧化降解,功效更持久,保质期长等优点。

技术领域
本发明涉及面膜制作技术领域,尤其涉及一种多孔植物多糖凝胶膜体及其制备方法。
背景技术
光甘草定是一种黄酮类物质,是从光果甘草中提取、分离出来的一种甘草黄酮,是一种脂溶性化合物,具有很强的抗自由基氧化作用,能有效清除体内新陈代谢过程中所产生的自由基、使对氧化敏感的生物大分子(低密度脂蛋白LDL,DNA)和细胞壁等免受自由基氧化损伤。从而可以防止与自由基氧化有关的某些病理变化,如动脉粥样硬化,细胞衰老等。由于光甘草定具有出色的抗氧化和抗黑色素形成的作用,能增强细胞能量,提高肌肤机能,抑制色斑、皱纹等生成。提高肌肤的抗氧化力,避免紫外线伤害,提高肌肤弹力及张力,因而光甘草定可以用于制作各类护肤品(如面霜、面膜等),通常是将光甘草定的有效成分粘附在无纺布或蚕丝膜上,但是由于光甘草定对光和热稳定性差,遇光和受热后易分解、变色、结晶析出,使其功效下降,因此,本申请的目的是优化含有光甘草定化妆品的生产工艺,提高其功效的稳定性。
β-环状糊精分子具有略呈锥形的中空圆筒立体环状结构,具有亲水性。β-环状糊精作为新型药用辅料主要用于增加药物的稳定性,防止药物氧化与分解,用于提高药物的溶解和生物利用度,是良好的稳定剂,但在化妆品行业应用较少。
专利号CN 106619163 A公开了一种光甘草定纳米混悬剂及其制备方法,该专利以光甘草定、稳定剂和水为原料,结合超声预分散和高压均质法制备光甘草定纳米混悬剂。虽然该发明的光甘草定纳米混悬剂可以改善光甘草定的溶解性,提高使用效率,但是该专利制备的光甘草定纳米混悬剂存在对光和热的稳定性差,遇光和受热后易分解,功效降低的缺陷。
专利号CN 108815012A公开了一种水溶性光甘草定的制备方法,该专利通过将光甘草定溶液滴加到α-环糊精溶液中进行微囊包裹、干燥、粉碎,得到光甘草定粉末,具有可长期保存和水溶性较好的特点,但是该专利制备的水溶性光甘草定的功效稳定性和持久性较差。
发明内容
有鉴于此,本发明提供了一种多孔植物多糖凝胶膜体,由如下质量百分数的组分构成:卡拉胶0.20-30.0%,魔芋胶0.30-30.0%、羟乙基纤维素0.10-15.0%,甘油0.5-30.0%,β-环状糊精1.0-3.0%,光甘草定0.05-1.0%,余量为纯化水。
一种权利要求1所述多孔植物多糖凝胶膜体的制备方法,包括以下步骤:
(1)按比例称量β-环状糊精和光甘草定,加入容器中,然后加入纯化水,采用均质机进行均质后得到溶液A;
(2)按比例称取卡拉胶、魔芋胶、羟乙基纤维素、甘油,加入纯化水得到混合物,然后升温搅拌均匀,得到溶液B;
(3)将溶液B降温后倒入溶液A中,搅拌均匀、灌装、冷却后即可包装。
优选的,步骤(1)中所述均质机的转速为18000rpm。
优选的,步骤(1)中所述均质的温度为15-20℃。
优选的,步骤(1)中所述均质的次数为3次,每次均质5min,均质后将混合溶液保温20℃备用。
优选的,步骤(2)中所述搅拌时将混合物的温度升至95℃,以60rpm的转速搅拌15min,直至所有物质完全溶解。
优选的,步骤(3)中所述溶液B降温至80℃。
优选的,步骤(3)将降温后的溶液B倒入溶液A后,以60rpm的转速搅拌0.5-1.0小时。
与现有技术相比,本发明具有以下有益效果:本发明利用β-环状糊精的结构特点,通过β-环状糊精的包埋技术,将光甘草定包埋在β-环状糊精的空洞内,起到保护和增溶作用,提高光甘草定的稳定性,有效防止光甘草定被氧化降解,使制作出的凝胶膜体在使用时发挥更高效率,使用过程中缓慢释放,发挥功效作用更持久,同时由于稳定性的提高,也可延长产品保质期。

为使本发明的目的、技术方案和优点更加清楚,下面将对本发明的技术方案进行清楚、完整的描述。显然,所描述的实施例仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动的前提下所得到的所有其它实施例,都属于本发明所保护的范围。
下述实施例中所述试验方法或测试方法,如无特殊说明,均为常规方法;所述原料和助剂,如无特殊说明,均从常规商业途径获得,或以常规方法制备。
实施例1
一种多孔植物多糖凝胶膜体,由如下质量百分数的组分构成:卡拉胶8.0%,魔芋胶5.0%、羟乙基纤维素4.0%,甘油3.0%,β-环状糊精2.0%,光甘草定0.5%,余量为纯化水。
一种权利要求1所述多孔植物多糖凝胶膜体的制备方法,包括以下步骤:
(1)按比例称量β-环状糊精和光甘草定,加入容器中,然后加入适量纯化水,将环境温度控制在15-20℃,采用均质机在18000rpm的转速下进行高速均质3次,每次5min,后得到溶液A,在20℃保温备用;
(2)按比例称取卡拉胶、魔芋胶、羟乙基纤维素、甘油,倒入混合罐,加入适量纯化水,将温度升至95℃,在60rpm的转速下搅拌15min,直至所有物质溶解完全,得到溶液B;
(3)将溶液B降温至80℃后倒入溶液A中,在60rpm的转速下搅拌0.5-1.0小时,然后将混合液趁热倒入灌装机中灌装进面膜托,待冷却后即可包装。
实施例2
一种多孔植物多糖凝胶膜体,由如下质量百分数的组分构成:卡拉胶8.0%,魔芋胶5.0%、羟乙基纤维素4.0%,甘油3.0%,β-环状糊精3.0%,光甘草定1.0%,余量为纯化水。
制备方法同实施例1。
对比例1
与实施例1的区别在于原料组成中不包括β-环状糊精,制备方法同实施例1。
对实施例1-2和对比例1进行性能检测结果如下:
(1)产品稳定性试验对比
将实施例1-2和对比例1所制备的产品在常规储存条件20-30℃保存,每隔3天进行取样检测,比较凝胶面膜产品的抗氧化性能,评价产品功效的稳定性。抗氧化活性的检测按总氧自由基清除能力检测法测定(DPPH法),具体检测方法如下:
1)样品A测定:取一块凝胶膜体,加入适量95%的乙醇,研磨,然后再加入95%的乙醇将上述磨碎后的凝胶膜体稀释至光甘草定的浓度为0.030mg/ml,用滤纸过滤,取1ml过滤液加入2mlDPPH,混合均匀,静置30分钟,波长520nm,测吸光值。
2)空白对照A0测定:取DPPH溶液2ml加入试管,加95%乙醇1ml,混合均匀,静置30分钟,波长520nm,测吸光值。
计算:自由基清除率=(A-A0)/A0×100%
实施例1-2和对比例1的产品稳定性试验结果如下:
从上表试验结果可知,含β-环状糊精的凝胶膜体的自由基清除率明显高于无β-环状糊精凝胶膜体,说明β-环状糊精对光甘草定分子结构稳定具有明显保护作用。
(2)应用效果试验
受试者年龄18-58岁,测试部位肤色ITA°值在20°-40°,健康女性或男性,受试部位至少有一个和周围邻近皮肤的ITA°差值大于10°的明显色斑,且直径不小于3mm。入选和排除标准选择合格的受试者,并按随机表分为试验组(32人)和对照组(30人),在受试部位左右两侧色斑对称的情况下,可分为试验产品组和对照产品组,确保最终完成有效例数不少于30人/组。
在试验期间受试部位必须使用本发明对比例1和实施例1的试验产品,对比例1为不加环状糊精作为对照产品,试验期间每天敷试验面膜一次,不能使用其他任何具有祛斑美白功效或者可能对测试结果产生影响的产品,不能有暴晒情况,并应做好试验部位的防晒工作。
检测环境条件:试验检测过程中仪器测试环境应在温度为20-24℃、相对湿度为RH50-60%,自然光线,避免阳光直射的环境下进行。
连续使用试验面膜8周,在试验前和在使用面膜后2周、4周、8周对面部皮肤进行仪器检测,记录实验组和对照组所有人检测数据,分别将两试验组数据平均、汇总。
1.皮肤色度测量
本实验用分光测色仪(美能达CM-25d肤色测量仪)分别测量试验及对照组试验人员面部皮肤。
评价方法依据国际照明委员会(CIE)规定的L*a*b*色度系统,分别测量试验区及对照区域各个观测时点的L*、a*、b*数据,每个区域测试三次,并记录。计算肤色个体类型角(individule type angle,ITA°)的值:计算公式:
在化妆品美白功效评价检验方法中,用ITA°值来表征人体皮肤颜色的参数。ITA°值越大,肤色越浅,反之肤色越深。
2.皮肤黑素检测
用皮肤黑素测试仪(主机CKMPA580探头MexameterMX18,分析软件为CK公司“MultiProbesAdapter MPA5/9”),分别测量试验组及对照组试验部位色斑区皮肤的MI值,每个测试区测试三次,并记录;测试区MI值越小,肤色越浅,反之越深。
皮肤色度检测结果如下所示:
结论:含β-环状糊精的实施例1比对比例1有较好的抗氧化性能,ITA°平均值较使用前提高28.5%,比对照组提高10.8%,皮肤色值下降显著。
皮肤黑色素检测结果如下所示:
结论:含β-环状糊精的实施例1比对比例1有较好的抗氧化性能,MI平均值下降比对比例1高4.2%,抑制色素产生效果明显。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (8)
1.一种多孔植物多糖凝胶膜体,其特征在于,由如下质量百分数的组分构成:卡拉胶0.20-30.0%,魔芋胶0.30-30.0%、羟乙基纤维素0.10-15.0%,甘油0.5-30.0%,β-环状糊精1.0-3.0%,光甘草定0.05-1.0%,余量为纯化水。
2.一种权利要求1所述多孔植物多糖凝胶膜体的制备方法,其特征在于,包括以下步骤:
(1)按比例称量β-环状糊精和光甘草定,加入容器中,然后加入纯化水,采用均质机进行均质后得到溶液A;
(2)按比例称取卡拉胶、魔芋胶、羟乙基纤维素、甘油,加入纯化水得到混合物,然后升温搅拌均匀,得到溶液B;
(3)将溶液B降温后倒入溶液A中,搅拌均匀、灌装、冷却后即可包装。
3.根据权利要求2所述的多孔植物多糖凝胶膜体的制备方法,其特征在于,步骤(1)中所述均质机的转速为18000rpm。
4.根据权利要求2所述的多孔植物多糖凝胶膜体的制备方法,其特征在于,步骤(1)中所述均质的温度为15-20℃。
5.根据权利要求2所述的多孔植物多糖凝胶膜体的制备方法,其特征在于,步骤(1)中所述均质的次数为3次,每次均质5min,均质后将混合溶液保温20℃备用。
6.根据权利要求2所述的多孔植物多糖凝胶膜体的制备方法,其特征在于,步骤(2)是将混合物升温至95℃,搅拌15min。
7.根据权利要求2所述的多孔植物多糖凝胶膜体的制备方法,其特征在于,步骤(3)溶液B降温至80℃。
8.根据权利要求2所述的多孔植物多糖凝胶膜体的制备方法,其特征在于,步骤(3)将降温的溶液B倒入溶液A搅拌0.5-1.0小时。
