Abstract
本发明涉及一种粘附性药物微载体的制备方法,包括:配制载体溶液,并混合特定比例的听力保护性药物,制作液滴,并实现听力保护性药物的装载;固化制备的乳液液滴,并在固化过程中原位生成特定的表面微纳结构,得到药物微载体粗品;清洗,得到粘附性药物微载体。与现有技术相比,本发明通过对微载体表面理化性质和微纳结构的调控来增强其粘附性,应用于鼓室注射时,使该药物微载体粘附于圆窗膜及中耳粘膜,所负载的药物成份通过微载体的自然降解缓慢释放,药物微载体粘附于圆窗膜及中耳粘膜,能有效减少药物经咽鼓管丢失,增加药物缓释时间,提高进入内耳的药物浓度,具有药物缓释时间久、药物损失少、效果显著等优点。

技术领域
本发明涉及生物材料及转化医学领域,尤其是涉及一种粘附性药物微载体的制备方法。
背景技术
鼓室注射是获得性耳聋常用的治疗方法之一,药物主要通过圆窗膜运输至内耳。鼓室注射可提高内耳局部药物浓度,提高药物吸收率,减少地塞米松等保护性药物对全身的副作用,是一种安全有效的治疗方式。
药物微载体因为其能定向递送和持续释放,从而避免药物的迅速流失,提高药物利用率。因此,药物微载体鼓室注射治疗获得性耳聋,通过材料的自然降解,可在中耳腔中持续释放听力保护性药物,实现单次注射即达到持续治疗听力下降的目的,具有较高的临床运用价值。然而,目前药物微载体表面结构单一、理化性质简单、功能单一,容易从给药位点脱落甚至被快速降解或排泄流失,这使得药物作用时间过短,不能使药物微载体中的药物发挥最大化作用。
因此,用于鼓室注射治疗获得性耳聋的新型药物微载体有待开发。
发明内容
本发明的目的就是为了克服上述现有技术存在的缺陷而提供一种粘附性药物微载体的制备方法,通过对微载体表面理化性质和微纳结构的调控来增强其粘附性,可解决传统的药物微载体表面结构单一、理化性质简单、功能单一、对圆窗膜黏附性不够理想、作用于圆窗膜的药物少、难以快速跨越圆窗膜屏障、容易脱落经咽鼓管流失、在中耳腔停留时间短的缺点。
本发明的目的可以通过以下技术方案来实现:
本申请的目的是保护一种粘附性药物微载体的制备方法,包括以下步骤:
S1:配制载体溶液,并混合特定比例的听力保护性药物,制作液滴,并实现听力保护性药物的装载;
S2:固化S1中制备的乳液液滴,并在固化过程中原位生成特定的表面微纳结构,得到药物微载体粗品;
S3:对药物微载体粗品进行清洗,得到粘附性药物微载体。
进一步地,S1中通过搅拌、机械震荡、微流控中的一种方式制作乳液液滴。
进一步地,S1中通过原位包裹、多孔吸附、静电吸引、基团反应中的一种方式进行装载。
作为本技术方案的一种实施方式,S1中所述载体溶液为具粘附性的生物相容性材料,即通过自身具有粘附性的材料作为基底材料,制作具有粘附性的药物微载体。
进一步地,S1中可聚合高分子预聚物溶液为甲基丙烯酸酯明胶(Gel-MA)。
作为本技术方案的另一种实施方式,S2中所述载体溶液在固化过程中原位生成的表面微纳结构包括:花粉结构和/或花瓣结构,以此通过增加微载体表面的摩擦力和毛细作用、机械锁等增加药物微载体的粘附性。
进一步地,S2中通过溶剂蒸发/交换诱导高分子在液液界面发生相分离,使液滴在固化过程中生成不规则的凸起状花粉样或花瓣样表面微纳结构。
作为本技术方案的另一种实施方式,S3中在清洗过程中对得到的药物微载体进行表面化学修饰,对微载体表面修饰具有组织黏附特性的物质。
进一步地,所述具有组织黏附特性的物质为聚多巴胺、卡波姆中的一种,使其通过氢键、亲疏水相互作用等发挥黏附效果。

与现有技术相比,本发明具有以下技术优势:

(2)本发明设计了一种通过制备具有微刺型花粉样微纳结构的粘附性药物微载体,增加了药物微载体的比表面积,增加了药物微载体对软湿界面的粘附性。
(3)本发明设计了一种通过对微载体表面包被具有粘附性的聚多巴胺,使其通过氢键、疏水相互作用等发挥黏附效果,制备具有粘附性的药物微载体用于鼓室注射治疗获得性耳聋。

具体实施方式
从本技术方案的构思上进行分析,本技术方案在传统药物微载体的基础上通过改变药物微载体的表面理化性质,增强软湿生物组织界面的粘附性,可增强通过鼓室注射治疗获得性耳聋的药物微载体的功能。
本技术方案中微载体的粘附性可以通过三个方面完成:使用生物相容性好的粘附性材料作为微载体的基底材料;通过诱导高分子的界面相分离使微载体表面呈现微刺型花粉样结构,可增加微载体与组织界面的摩擦力和机械锁;通过对微载体表面包被具有粘附性的聚多巴胺等成分,使得药物微载体具有更强的粘附性。本技术方案中药物装载的方法主要包括原位装载、多孔吸附、静电吸引或基团反应等方法。
本技术方案提供的一种粘附性药物微载体当其在鼓室注射中进行应用时,所述粘附性药物微载体的特征在于其能有效的粘附于圆窗膜及中耳粘膜上,延长药物微载体在中耳腔的停留时间,增加内耳中听力保护性药物的浓度,提高听力保护性药物的治疗作用,对后天性耳聋进行持续性治疗。
本技术方案中粘附性药物微载体通过鼓室注射治疗获得性听力损失的方法通过以下步骤:
(1)粘附性药物微载体的制备:
根据需求配制可聚合高分子预聚物溶液,混合一定比例的听力保护性药物;通过搅拌、机械震荡、微流控等技术制作乳液液滴;通过光照、溶剂挥发、温度诱导相变等过程固化乳液液滴并在固化过程中原位生成一定的表面微纳结构;固化完成后通过无水乙醇和超纯水等多次漂洗去除残留的溶剂,根据药物微载体的特性对其进一步进行表面化学修饰,浸泡于一定浓度的聚多巴胺溶液过夜,干燥后保存在容器中。
(2)粘附性药物微载体通过鼓室注射的步骤:
将干燥后的药物微载体与生理盐水按比例混合,超声震荡混匀,注入到注射器中,在耳内镜或显微镜的引导下于鼓膜前上象限刺破鼓膜,用针头稍扩大穿刺孔利于中耳腔气压与外界气压平衡,把药物微载体-生理盐水混合液缓慢注射到中耳腔内。
在一个较佳实例中,所述的一种粘附性药物微载体的制备方法,所述粘附性药物微载体选自生物相容性好、可降解的明胶甲基丙烯酸酯(GelMA),原位装载听力保护性药物地塞米松磷酸钠;并混合一定比例的HMPP。
在一个较佳实例中,所述的一种粘附性药物微载体的制备方法,所述的粘附性新型药物微载体的表面为花粉样表面微纳结构。
在一个较佳实例中,所述的一种粘附性药物微载体的制备方法,所述药物微载体表面覆盖了一层具有更强粘附性的聚多巴胺。
其中尤为重要的是,本技术方案中药物微载体的粘附性可通过以下几个方面实现:
(1)利用材料本身粘附性:利用自身具有粘附性的材料作为基底材料,制作具有粘附性的药物微载体;
(2)利用药物微载体表面的微纳结构:通过溶剂蒸发/交换诱导高分子在液液界面发生相分离使液滴在固化过程中生成不规则的凸起状花粉样或花瓣样表面微纳结构,从而具有粘附性。
(3)利用药物微载体表面基团的粘附性:通过对微载体表面修饰具有组织黏附特性的物质。
本技术方案中制备的药物微载体能有效粘附于圆窗膜和中耳腔粘膜。通过鼓室注射使药物微载体进入中耳腔,利用药物微载体的粘附性,使得药物微载体粘附于圆窗膜和中耳粘膜表面,减少药物经咽鼓管丢失,提高中耳腔和进入内耳的药物浓度。随着材料的降解,所负载的药物成份通过粘附于圆窗膜和中耳腔粘膜的微载体的自然降解缓慢释放,促进保护性药物进入内耳对获得性耳聋进行治疗。
以下结合具体实施例对上述方案做进一步说明,参见图1与图2。应理解,这些实施例是用于说明本发明而不限于限制本发明的范围。凡根据本发明精神实质所做的等效变换或修饰,都应涵盖在本发明的保护范围之内。实施例中未注明的实施条件通常为常规实验中的条件。
实施例1
粘附性高分子微载体的制备及鼓室注射应用:
1.载地塞米松磷酸钠甲基丙烯酸甲酯明胶(Dexsp-GelMA)微载体的制备:按30mg地塞米松磷酸钠溶解于1ml 8%(w/v)GelMA和6%(v/v)HMPP配制高分子预聚物溶液,超声震荡混匀,制作单乳液模板,通过紫外光固化成球,通过无水乙醇和超纯水多次漂洗去除残留的溶剂,鼓风干燥机中30℃鼓风干燥后保存在容器中。
2.载地塞米松磷酸钠甲基丙烯酸甲酯明胶(Dexsp)微载体粘附性测试:
(1)选取豚鼠:分别选取16只听力正常的成年豚鼠,通过ABR测量其4、8、16、24、32kHz听阈。
(2)鼓室注射药物微载体:把已行ABR检测的16只豚鼠随机分为8组。分别称取约230mg Dexsp-GelMA药物微载体分别溶于100μl生理盐水中,注入1ml的注射器中,在显微镜下通过长约20mm的塑料针头经外耳道于豚鼠左耳鼓膜前上象限刺破鼓膜后缓慢注入豚鼠中耳腔。对照组鼓室注射100μl Dexsp。
(3)分别于注射后1-3-5-7-9-12-24-48小时处死豚鼠,取出听泡,在显微镜下打开听泡,观察药物微载体在中耳粘膜和圆窗膜的粘附情况。通过50倍镜观察圆窗膜及中耳粘膜,分别在圆窗膜和中耳粘膜上随机选择3个视野拍照,分别对圆窗膜和中耳粘膜上粘附的微球进行计数,取3个视野的微球计数值取平均值,比较不同时间粘附于圆窗膜和中耳粘膜上的微球数变化情况。
实施例2
基于界面不稳定性的粗糙粘附性微载体的制备及鼓室注射应用:
1.N-乙酰-L-半胱氨酸的聚苯乙烯微载体的制备:配制各相溶液,分散相由将N-乙酰-L-半胱氨酸溶解于10mg/ml聚苯乙烯/氯仿溶液、一定浓度的n-十六烷醇、氯仿溶液组成,连续相溶液由3mg/ml的SDS/甘油溶液组成。通过微流控技术形成单分散的单乳液液滴。因分散相中有机溶液扩散蒸发,液滴逐渐固化,表层形成褶皱。将悬浮的微球在去离子水和乙醇中透析以除去甘油、SDS、氯仿和n-十六烷醇。
2.载N-乙酰-L半胱氨酸的聚苯乙烯微载体在豚鼠中耳腔和圆窗膜的粘附性测试:步骤同实施例1中步骤2。
实施例3
基于表面化学修饰的粘附性微载体的制备及鼓室注射应用
1.修饰聚多巴胺的地塞米松磷酸钠甲基丙烯酸甲酯明胶(Dexsp-GelMA)微载体的制备:制备Dexsp-GelMA微载体,步骤同实施例1中步骤1;予以聚多巴胺溶液浸泡Dexsp-GelMA微载体过夜后,提取修饰聚多巴胺的Dexsp-GelMA微载体。
修饰聚多巴胺的Dexsp-GelMA微载体在豚鼠中耳腔和圆窗膜的粘附性测试:步骤同实施例1中步骤2。
实验结果表明,与直接注射药物或利用传统药物微载体相比,本实施例方案使用粘附性的新型药物微载体进行鼓室注射,可使药物微载体粘附于圆窗膜和中耳粘膜,减少药物和药物微载体经咽鼓管的丢失量,延长药物作用时间,增加圆窗膜和中耳粘膜局部药物浓度。通过增加药物微载体的粘附性,可延长药物微载体的作用时间,同时提高进入内耳的药物浓度,从而优化治疗效果。
可见,本技术方案通过原位包裹、多孔吸附、静电吸引使保护性药物装载在载体中。通过利用材料本身粘附性;药物微载体表面的微纳结构;药物微载体表面基团的粘附性制备具有粘附性的新型药物微载体,通过鼓室注射使药物微载体进入中耳,使药物微载体粘附于圆窗膜和中耳粘膜。该微载体能粘附于圆窗膜及中耳腔粘膜,提高圆窗膜局部药物浓度,延长药物微载体作用于圆窗膜的时间。
上述的对实施例的描述是为便于该技术领域的普通技术人员能理解和使用发明。熟悉本领域技术的人员显然可以容易地对这些实施例做出各种修改,并把在此说明的一般原理应用到其他实施例中而不必经过创造性的劳动。因此,本发明不限于上述实施例,本领域技术人员根据本发明的揭示,不脱离本发明范畴所做出的改进和修改都应该在本发明的保护范围之内。
Claims (10)
1.一种粘附性药物微载体的制备方法,其特征在于,包括以下步骤:
S1:配制载体溶液,并混合特定比例的听力保护性药物,制作液滴,并实现听力保护性药物的装载;
S2:固化S1中制备的乳液液滴,并在固化过程中原位生成特定的表面微纳结构,得到药物微载体粗品;
S3:对药物微载体粗品进行清洗,得到粘附性药物微载体。
2.根据权利要求1所述的一种粘附性药物微载体的制备方法,其特征在于,S1中通过搅拌、机械震荡、微流控中的一种方式制作乳液液滴。
3.根据权利要求1所述的一种粘附性药物微载体的制备方法,其特征在于,S1中通过原位包裹、多孔吸附、静电吸引、基团反应中的一种方式进行装载。
4.根据权利要求1所述的一种粘附性药物微载体的制备方法,其特征在于,S1中所述载体溶液为具粘附性的生物相容性材料。
5.根据权利要求4所述的一种粘附性药物微载体的制备方法,其特征在于,S1中可聚合高分子预聚物溶液为甲基丙烯酸酯明胶。
6.根据权利要求1所述的一种粘附性药物微载体的制备方法,其特征在于,S2中所述载体溶液在固化过程中原位生成的表面微纳结构包括:花粉结构和/或花瓣结构,以此通过增加微载体表面的摩擦力和毛细作用、机械锁等增加药物微载体的粘附性。
7.根据权利要求1所述的一种粘附性药物微载体的制备方法,其特征在于,S2中通过溶剂蒸发/交换诱导高分子在液液界面发生相分离,使液滴在固化过程中生成不规则的凸起状花粉样或花瓣样表面微纳结构。
8.根据权利要求1所述的一种粘附性药物微载体的制备方法,其特征在于,S3中在清洗过程中对得到的药物微载体进行表面化学修饰,对微载体表面修饰具有组织黏附特性的物质。
9.根据权利要求8所述的一种粘附性药物微载体的制备方法,其特征在于,所述具有组织黏附特性的物质为聚多巴胺、卡波姆中的一种。
10.根据权利要求9所述的一种粘附性药物微载体的制备方法,其特征在于,所述具有组织黏附特性的物质为聚多巴胺。
