Abstract
本申请涉及一种水溶性Cu2‑XS纳米颗粒及其制备方法和应用,该纳米颗粒的Cu含量为25‑35wt%、S含量为15‑20wt%、磷酸化的糖的含量为45‑60wt%,水溶性Cu2‑XS纳米颗粒的尺寸小不易在生物器官中积累,可作为肿瘤靶向型光声成像和光热治疗的纳米颗粒,同时满足结构简单、光热转化效率高、光声效果好、生物安全性高、毒性小等性能要求,且Cu2‑XS含量可控、质量稳定,磷酸化的糖是人体所需供能小分子,安全性高且能特异性聚集在肿瘤部位,提高了治疗效果。同时该纳米颗粒采用生物矿化法合成,显示了良好的生物相容性和优越的肿瘤代谢靶向性。将合成思路中的Cu以放射性金属元素Cu64进行替换,将有望取代目前临床中广泛应用于正电子发射断层扫描(PET)类的放射性显像剂F18‑DG。

技术领域
本发明涉及一种水溶性Cu2-XS纳米颗粒及其制备方法和应用,属于生物医学纳米材料领域。
背景技术
手术和化疗是肿瘤治疗的常用手段,然而,手术仅能切除肉眼可见的瘤体,对不可见的亚临床病灶却难以清除;传统化疗往往存在靶向性差和剂量限制毒性的问题。光热治疗是一种继手术、化疗之后出现的新型治疗方法。光热治疗技术是利用光热转换材料把近红外光的能量转化为热能的一种微创技术,光热疗法在对癌症的治疗中可以实现局部升温进而杀伤癌细胞,而对正常的细胞没有损坏,是一种更加安全、高效的肿瘤治疗手段。
光能除了可以转化为热能外,还能转化为声能,用于光声成像(PAI)来诊断或监测疗效。在光声成像和光热治疗中,由于材料的吸收光谱位于近红外二区,故具有更深的穿透深度,可以达到更好的成像及治疗效果。同时以饥饿治疗和化学动力学治疗作为辅助疗法,可以全面提高材料在肿瘤治疗各阶段发挥的作用。
在光热治疗中,由于近红外光的组织穿透力较强,因此常用近红外光作为光热治疗的光源,许多具有较强的近红外光转换能力的纳米材料也常用于光热治疗研究。例如贵金属纳米材料、碳基纳米材料等无机纳米材料,以及共轭高分子纳米材料和近红外染料等有机纳米材料等。与有机光热耦联剂相比,无机纳米材料具有更好的光热稳定性,与常用的金基纳米材料相比,新型的半导体 Cu2-XS纳米光热材料不仅具有稳定的光热转换能力以及多模态成像功能。有望实现多模成像与光热治疗一体化平台的整合。
然而,纳米材料在体内的代谢以及毒性限制了其在生物体内的进一步应用。通常条件下合成的较大尺寸的纳米材料,容易在网状内皮系统器官中积累,造成严重的肝脾脏器的损伤。
发明内容
本发明的目的在于提供一种生物安全性高且相容性好、毒性小、光热转化效率高、且光声效果好的水溶性Cu2-XS纳米颗粒。
为达到上述目的,本发明提供如下技术方案:一种水溶性Cu2-XS纳米颗粒,所述纳米颗粒的Cu含量为25-35wt%、S含量为15-20wt%、磷酸化的糖的含量为45-60wt%。
进一步地,所述纳米颗粒的晶粒尺寸为10-20nm。
进一步地,所述磷酸化的糖为6-磷酸葡萄糖、6-磷酸果糖、3-磷酸甘油醛或 5-磷酸核糖。
本发明还提供一种水溶性Cu2-XS纳米颗粒的制备方法,用以制备如上所述的水溶性Cu2-XS纳米颗粒,所述制备方法包括:
S1、将含铜化合物溶解于分散介质中形成含铜化合物溶液,所述含铜化合物溶液的浓度为0.01-0.1mol/L,所述分散介质为去离子水;
S2、将所述含铜化合物溶液和磷酸化的糖溶液(浓度为0.01-0.1mol/L)混合搅拌20-40min,其中,所述含铜化合物溶液和所述磷酸化的糖溶液的体积比为1:1;
S3、向反应体系中加入pH调节剂调整pH至10;
S4、向反应体系中加入0.1ml含硫化合物溶液(1mol/L),搅拌至反应体系颜色呈棕褐色;
S5、将整个反应体系转移至水浴加热并继续搅拌反应8min,得到墨绿色澄清透明均一的纳米颗粒悬浊液;
S6、将所述悬浊液离心分离,弃去下方液体,得到水溶性Cu2-XS纳米颗粒。
进一步地,所述含铜化合物为氯化铜、硝酸铜或硫酸铜。
进一步地,所述pH调节剂为氨水或氢氧化钠溶液。
进一步地,所述含硫化合物为硫代乙酰胺。
进一步地,所述水浴加热的温度为100℃。
进一步地,将所述悬浊液离心分离的具体步骤为:用100K的超滤管进行离心,转速为3000rpm、时长为5min,并且使用去离子水重复离心分离。
本发明还提供一种如上所述的水溶性Cu2-XS纳米颗粒在肿瘤靶向型光声成像和光热治疗的应用。
本发明的有益效果在于:水溶性Cu2-XS纳米颗粒的尺寸小不易在生物器官中积累,可作为肿瘤靶向型光声成像和光热治疗的纳米颗粒,同时满足结构简单、光热转化效率高、光声效果好、生物安全性高、毒性小等性能要求,且Cu2-XS 含量可控、质量稳定,磷酸化的糖是人体所需供能小分子,安全性高且能特异性聚集在肿瘤部位,提高了治疗效果。同时该纳米颗粒采用生物矿化法合成,显示了良好的生物相容性和优越的肿瘤代谢靶向性。决定了其未来在核医学方向也有广阔的应用前景,若将该材料的合成思路中的Cu以放射性金属元素Cu64进行替换,将有望取代目前临床中广泛应用于正电子发射断层扫描(PET)类的放射性显像剂F18-DG。
上述说明仅是本发明技术方案的概述,为了能够更清楚了解本发明的技术手段,并可依照说明书的内容予以实施,以下以本发明的较佳实施例并配合附图详细说明如后。

具体实施方式
下面将结合附图对本发明的技术方案进行清楚、完整地描述,显然,所描述的实施例是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
此外,下面所描述的本发明不同实施方式中所涉及的技术特征只要彼此之间未构成冲突就可以相互结合。
本申请提供一种水溶性Cu2-XS纳米颗粒,该纳米颗粒的Cu含量为25-35wt%、 S含量为15-20wt%、磷酸化的糖的含量为45-60wt%,其中x为0-1。Cu2-XS纳米颗粒的外周具有磷酸化的糖,故具有水溶性。
请参见图1,该纳米颗粒的电镜图,可以得到纳米颗粒的晶粒尺寸为10-20nm。纳米颗粒的晶粒尺寸较小,降低了其在生物体内的器官的积累。磷酸化的糖为 6-磷酸葡萄糖、6-磷酸果糖、3-磷酸甘油醛或5-磷酸核糖,但不仅限于此,磷酸化的糖还可以为其他化合物,在此不一一列举。
其中,水溶性Cu2-XS纳米颗粒具有良好的PA/MRI/PET/TI多模态成像功能以及PTT和IRT协同治疗效果,有望实现精准诊断下的高效光热治疗,达到诊疗一体化,具有广阔的应用前景,其可作为肿瘤靶向型光声成像和光热治疗的纳米制剂。磷酸化的糖作为Cu2-XS纳米颗粒的稳定剂,其是人体所需供能小分子,安全性高且能特异性聚集在肿瘤部位,从而实现制剂的靶向性,提高了治疗肿瘤的效果,降低了在生物体内的器官的积累,避免对器官造成损伤。
请参见图2至图4,在λ=808nm,2W·cm-2的近红外光照下,不同浓度下的水溶性Cu2-XS纳米颗粒具有良好的光热转化效率,并且具有良好的稳定性。经过计算水溶性Cu2-XS纳米颗粒的光热转化效率在近红外光波长808nm时为70%,光热转化效率在光波长1064nm时为84%。
请参见图5和图6,图5为图1所示的水溶性Cu2-XS纳米颗粒的浓度在0、 1μg/ml、2μg/ml、4μg/ml、6μg/ml、8μg/ml的溶液分别在λ=780nm、808nm、 900nm、980nm近红外照射下的光声效果的线性拟合曲线。图6为图1所示的水溶性Cu2-XS纳米颗粒的浓度在0、1μg/ml、2μg/ml、4μg/ml、6μg/ml、8μg/ml、 10μg/ml、12μg/ml的溶液在λ=808nm近红外照射下的光声效果。在λ=808nm, 2W·cm-2的光照下,水溶性Cu2-XS纳米颗粒能够同时满足光热转化效率高、光声效果好、生物安全性高、毒性小等性能要求。
请参见图7,图7为图1所示的水溶性Cu2-XS纳米颗粒的浓度在0、1μg/ml、 2μg/ml、4μg/ml、6μg/ml、8μg/ml的溶液的紫外吸收光谱。该纳米颗粒在波长 1000nm左右有特异性吸收峰,说明在此波长附近的近红外光照射下有良好的光热和光声效果。
本发明还提供一种水溶性Cu2-XS纳米颗粒的制备方法,用以制备上述所示的水溶性Cu2-XS纳米颗粒,该制备方法包括:
S1、将含铜化合物溶解于分散介质中形成含铜化合物溶液,含铜化合物溶液的浓度为0.01-0.1mol/L,分散介质为去离子水;
S2、将含铜化合物溶液和磷酸化的糖溶液(浓度为0.01-0.1mol/L)混合搅拌20-40min,其中,含铜化合物溶液和磷酸化的糖溶液的体积比为1:1;
S3、向反应体系中加入pH调节剂调整pH至10;
S4、向反应体系中加入0.1ml含硫化合物溶液(1mol/L),搅拌至反应体系颜色呈棕褐色;
S5、将整个反应体系转移至水浴加热并继续搅拌反应8min,得到墨绿色澄清透明均一的纳米颗粒悬浊液;
S6、将悬浊液离心分离,弃去下方液体,得到水溶性Cu2-XS纳米颗粒。
其中,含铜化合物为氯化铜、硝酸铜或硫酸铜,含硫化合物为硫代乙酰胺。关于含铜化合物和含硫化合物的原料不仅限于此,含铜化合物和含硫化合物还可以为其他原料,比如,含硫化合物可以为硫化钠、硫代硫酸钠或硫脲等,在此不一一列举。在本实施例中,含铜化合物为氯化铜,含硫化合物为硫代乙酰胺。
pH调节剂为氨水或氢氧化钠溶液,在一较佳实施例中,使用氢氧化钠作为 pH调节剂,且氢氧化钠的浓度为2mol/L。
搅拌可使用磁力搅拌,在不仅限于此,还可以使用其他搅拌方式,在此不一一列举。
在步骤S5中,水浴加热的温度为100℃。
在步骤S6中,将悬浊液离心分离的具体步骤为:用100K的超滤管进行离心,转速为3000rpm、时长为5min,并且使用去离子水重复离心分离。
该制备方法绿色、简单易行、用时短、易大规模生产、且成分简单、安全性高、毒性小、产物性能稳定。通过合理控制体系中铜离子与磷酸化的糖的摩尔比,本实施例中,在常温条件下,氯化铜和磷酸化的糖的摩尔比为1:1,在磁力搅拌反应生成葡萄糖-6磷酸铜,稍微过量的硫代乙酰胺在pH为10的环境下缓慢地释放硫离子,将反应速率降低,使得生成的纳米颗粒形状更完好。
下面以具体实施例对上述制备方法进行详细说明:
步骤一、将CuCl2配成0.1mol/L的水溶液;6-磷酸葡萄糖溶液配成0.1mol/L 的水溶液;
步骤二、常温条件下,分别取0.4ml配好的CuCl2溶液和0.4ml的6-磷酸葡萄糖溶液加入9ml去离子水中磁力搅拌30min,可观察到反应体系呈现淡蓝色稍混浊状态;
步骤三、在反应体系中加入0.1ml的NaOH溶液(2mol/L)调节PH至10,观察到反应体系蓝色加深,液体由浑浊变透明;
步骤四、向反应体系中加入0.1ml硫代乙酰胺溶液(1mol/L),磁力搅拌下观察到反应体系颜色呈棕褐色;
步骤五、当观察到反应体系的棕褐色不再加深,将整个反应体系转移至100℃水浴加热并继续搅拌反应8min,最终得到透明的墨绿色液体。
步骤六、将得到透明的墨绿色液体使用100K的超滤管离心分离,转速为 3000rpm、时长为5min,弃去下方无色透明液体,加入去离子水进行重复离心清洗,重复1-3次洗去杂质,得到水溶性Cu2-XS纳米颗粒。
本发明还提供一种如上所示的水溶性Cu2-XS纳米颗粒在肿瘤靶向型光声成像和光热治疗的应用。
综上,水溶性Cu2-XS纳米颗粒的尺寸小不易在生物器官中积累,可作为肿瘤靶向型光声成像和光热治疗的纳米颗粒,同时满足结构简单、光热转化效率高、光声效果好、生物安全性高、毒性小等性能要求,且Cu2-XS含量可控、质量稳定,磷酸化的糖是人体所需供能小分子,安全性高且能特异性聚集在肿瘤部位,提高了治疗效果。同时该纳米颗粒采用生物矿化法合成,显示了良好的生物相容性和优越的肿瘤代谢靶向性。决定了其未来在核医学方向也有广阔的应用前景,若将该材料的合成思路中的Cu以放射性金属元素Cu64进行替换,将有望取代目前临床中广泛应用于正电子发射断层扫描(PET)类的放射性显像剂F18-DG。

以上实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。
Claims (7)
1.一种水溶性Cu2-XS纳米颗粒的制备方法,其特征在于,所述制备方法包括:
S1、将含铜化合物溶解于分散介质中形成含铜化合物溶液,所述含铜化合物溶液的浓度为0.01-0.1mol/L,所述分散介质为去离子水;
S2、将所述含铜化合物溶液和浓度为0.01-0.1mol/L的磷酸化的糖溶液混合搅拌20-40min,其中,所述含铜化合物溶液和所述磷酸化的糖溶液的体积比为1:1;
S3、向反应体系中加入pH调节剂调整pH至10;
S4、向反应体系中加入0.1ml的浓度为1mol/L含硫化合物溶液,搅拌至反应体系颜色呈棕褐色;
S5、将整个反应体系转移至水浴加热并继续搅拌反应8min,得到墨绿色澄清透明均一的纳米颗粒悬浊液;
S6、将所述悬浊液离心分离,弃去下方液体,得到水溶性Cu2-XS纳米颗粒,其中,所述纳米颗粒的Cu含量为25-35wt%、S含量为 15-20wt%、磷酸化的糖的含量为45-60wt%;所述纳米颗粒的晶粒尺寸为10-20nm。
2.如权利要求1所述的水溶性Cu2-XS纳米颗粒的制备方法,其特征在于,所述磷酸化的糖为6-磷酸葡萄糖、6-磷酸果糖、3-磷酸甘油醛或5-磷酸核糖。
3.如权利要求1所述的水溶性Cu2-XS纳米颗粒的制备方法,其特征在于,所述含铜化合物为氯化铜、硝酸铜或硫酸铜。
4.如权利要求1所述的水溶性Cu2-XS纳米颗粒的制备方法,其特征在于,所述pH调节剂为氨水或氢氧化钠溶液。
5.如权利要求1所述的水溶性Cu2-XS纳米颗粒的制备方法,其特征在于,所述含硫化合物为硫代乙酰胺。
6.如权利要求1所述的水溶性Cu2-XS纳米颗粒的制备方法,其特征在于,所述水浴加热的温度为100℃。
7.如权利要求1所述的水溶性Cu2-XS纳米颗粒的制备方法,其特征在于,将所述悬浊液离心分离的具体步骤为:用100K的超滤管进行离心,转速为3000rpm、时长为5min,并且使用去离子水重复离心分离。
