The vegetable oil is purchased from Hecheng Natural perfume oil of Jiangxi Co.
The preparation method of the compound globefish liver oil composition comprises the following steps:
1) preparing globefish liver oil: extracting crude puffer liver oil from puffer liver by heating over water, saponifying with sodium hydroxide or other alkaline substances, centrifuging, washing with saline water, adsorbing with active carbon, fine filtering, detoxicating, deacidifying, deodorizing, degumming, decolorizing, and deodorizing to obtain refined puffer liver oil.
2) Mixing refined globefish liver oil and vegetable oil to obtain the composition.

In the present invention, the inventors have made a great technical contribution by making the following unexpected findings through creative efforts:
first, tetrodotoxin aqueous solution has no significant analgesic effect on mice, but has significant analgesic effect in oil phase preparations. Secondly, tetrodotoxin is mainly distributed in the liver of puffer fish, but not in the ovary. Thirdly, the tetrodotoxin is dissolved with the tetrodotoxin liver oil for a long time in the puffer fish liver to form rich tetrodotoxin lipidosome. And fourthly, the tetrodotoxin lipidation is very stable, has no toxicity of tetrodotoxin, cannot be destroyed by alkaline substances, and cannot cause harm to human bodies. The tetrodotoxin lipidate has similar efficacy to tetrodotoxin, can amplify the curative effect of the common medicament, and can inhibit the dependence and tolerance of the common medicament. Sixthly, the globefish liver oil can meet the food or medicine standard only through saponification treatment. And seventhly, the globefish liver oil is compatible with the vegetable oil phase containing effective components for resisting cancer, easing pain, removing addiction and the like, so that advantage complementation can be realized, and a more remarkable treatment effect can be generated.
The above findings, the present invention is confirmed by the following experiments and effect examples of the specific embodiments:
experiment I, comparison experiment of analgesic effect of tetrodotoxin in aqueous solution and in oil phase preparation
1. Experiment of analgesic effect of tetrodotoxin in aqueous solution on mice
A mouse hot plate method: mice were divided into 3 groups of 10 mice each, and injected with 0.3. mu.g/kg, 1.0. mu.g/kg, 3.0. mu.g/kg of tetrodotoxin aqueous solution (ip), respectively, and showed no significant difference (P > 0.05) compared to the administration before administration. ② mouse writhing method: the mice were divided into 3 groups of 10 mice each, and the abdominal cavity was injected with an aqueous acetic acid solution (0.4%/0.1 ml/10g), and then injected with an aqueous tetrodotoxin solution (ip) at 0.3. mu.g/kg, 1.0. mu.g/kg, and 3.0. mu.g/kg, respectively, which showed no significant difference (P > 0.05) from the administration level before the administration.
2. Experiment of analgesic effect of tetrodotoxin in oil phase preparation on mice
Four groups of 32 mice were divided in equal parts: low dose group, medium dose group, high dose group and blank group. Dissolving tetrodotoxin 1mg in soybean oil 1000ml, with tetrodotoxin content of 1 μ g/ml. The first three groups of mice were gavaged with 0.2ml, 0.3ml and 0.4ml of soybean oil, respectively, and the blank group was gavaged with soybean oil. The result shows that the tetrodotoxin oil phase preparation has very obvious analgesic effect (P is less than 0.001) on mice, and has no obvious dose-effect relationship. The tetrodotoxin has obvious analgesic effect in the oil phase preparation.
Experiment II, experiment that tetrodotoxin evolves into tetrodotoxin lipidation in grease
1. Dissolving tetrodotoxin 10mg in 1000ml soybean oil, with theoretical content of tetrodotoxin 10 μ g/ml. Within 48 hours, 40ml of the preparation is taken, 40ml of pure water is added, and the preparation is stirred, washed by water and finely filtered to obtain 40ml of water washing liquid. Mice (ip) were first injected with 0.1ml of aqueous wash solution and died, indicating that the aqueous wash solution contained monomeric tetrodotoxin. Then, the content of the tetrodotoxin in the water washing liquid is determined by a High Performance Liquid Chromatography (HPLC), and the result shows that the content of the tetrodotoxin is close to 10 mu g/ml.
2. Storing the oil phase preparation of tetrodotoxin for 40 days or more. Adding 40ml of pure water into 40ml of oil solution, stirring, washing, and finely filtering to obtain 40ml of water washing liquid. The water washing solution is first 0.1ml and injected into mouse (ip) without death, and this shows that the water washing solution contains no monomer tetrodotoxin. And then, measuring the content of the tetrodotoxin in the water washing liquid by using a High Performance Liquid Chromatography (HPLC), wherein the result shows that the content of the tetrodotoxin is 0.
The significance of this experiment is that tetrodotoxin, which has just begun to dissolve in soybean oil, still exists in a short time, and after 30 days or more, tetrodotoxin evolves into a new substance, which is called tetrodotoxin lipidate. The structure of this tetrodotoxin lipidate has not been clarified yet due to the limitations of the inventors' conditions.
Experiment III, experiment that tetrodotoxin is mainly distributed in liver of puffer fish but not ovary
Selecting one highly toxic maternal globefish, taking 100g of ovary and liver respectively, cutting up, soaking in 4 times of pure water respectively, stirring, standing for 4 hours, taking 10ml of supernatant respectively for later use. The liver soaking solution is diluted by 10 times of pure water, 0.1ml of injection mouse (ip) is taken, and the mouse dies within 15 min. The toxicity of the ovarian soak solution was tested in the same manner as above, and the mice did not die. Then, the experiment was continued by gradually decreasing the dilution ratio of the ovarian soak solution, and it was found that the mice died by diluting the ovarian soak solution with 5 times of pure water. The tetrodotoxin content in the liver is 2 times of the content of the ovarian tetrodotoxin. It can be seen that tetrodotoxin is mainly distributed in the liver, not in the ovary.
The significance of the experiment lies in that because tetrodotoxin is mainly accumulated in the liver of the puffer fish, a large amount of tetrodotoxin can be promoted to be dissolved with the puffer fish liver oil in the liver for a long time to form saturated tetrodotoxin lipidization. Tetrodotoxin extracted from globefish liver is just non-lipidated tetrodotoxin, but the lipidated tetrodotoxin cannot be separated.
Experiment four, the safety and the stability of the globefish liver oil (globefish toxin lipidation) are tested
1. And (3) safety test: taking 100ml unsaponified crude puffer liver oil, adding 50ml pure water, heating to 60 deg.C, and magnetically stirring for 1 hr to obtain 50ml water washing solution of crude puffer liver oil. The same method is used to prepare 50ml of saponified Fugu ocellatus liver oil water washing solution for use. Two groups of mice (ip) are injected with the two water washes, the first dose is 0.2ml, and the mice injected with crude oil water wash die in 5-15min, and the mice injected with saponified oil water wash do not die. The second injection of 0.4ml of non-dead mice resulted in no death. The crude puffer fish liver oil contains monomer tetrodotoxin, which can kill mouse, and the saponified puffer fish liver oil does not contain the toxicity of tetrodotoxin, and has reliable safety.
2. And (4) performing a stability test: the 24 mice were divided into 3 groups: crude puffer liver oil, saponified puffer liver oil, and blank group. The crude puffer fish liver oil and 0.3ml of saponified puffer fish liver oil are respectively taken to irrigate the stomachs of the first 2 groups of mice and irrigate the soybean oil in the blank group to carry out the licking test. The results show that both have substantially the same analgesic effect. The method shows that the saponified tetrodotoxin liver oil (tetrodotoxin lipidate) does not change the original characteristics and has good stability.
Fifth, the globefish liver oil after saponification can meet the food or drug standard
In the process of refining globefish liver oil, the invention adopts sodium hydroxide or other alkaline substances to saponify the crude globefish liver oil. Because the tetrodotoxin can disappear rapidly in sodium hydroxide or other alkaline substances, the invention not only can completely eliminate the tetrodotoxin in the puffer fish liver oil, but also can ensure that the acid value and other indexes of the puffer fish liver oil meet the relevant national regulations, and overcomes the defect that the puffer fish liver oil prepared by the prior art can not be used for food or medicines.
The invention overcomes the defects of the prior art by the following technologies:
the invention utilizes the characteristics of the globefish liver oil (globefish toxin lipidation) to carry out compatibility with trace plant essential oil containing various anti-cancer substances, analgesic substances and/or addiction-removing substances, thereby not only amplifying the treatment effect of the plant essential oil, but also inhibiting the possible side effect of the plant essential oil.
The composition of the invention has the technical advantages compared with tetrodotoxin: the composition of the invention has no toxicity of tetrodotoxin and no harm to human body; but can also amplify the curative effect of the common medicine and inhibit the dependence and tolerance of the common medicine.
The composition of the invention has technical advantages over individual puffer liver oil or individual vegetable oil: the composition of the invention amplifies the anticancer, analgesic and addiction-removing effects of the volatile oil and inhibits the possible side effects of the volatile oil.
The invention has the following technical effects:
first, the present invention utilizes globefish liver oil to solve the common problems of cancer cell division, proliferation, metastasis and implantation. The cancers produced by the human body are various, but all cancer cells have its commonness. The common property refers to that cancer cells have the characteristics of loose structure, easy fragmentation, easy movement and easy formation of new lesions. The globefish liver oil can directly kill cancer cells by enhancing the phagocytic power of immune cells, inhibit the growth and diffusion of the cancer cells, and inhibit the aggregation of platelets by promoting blood circulation to remove blood stasis, thereby preventing the cancer cells from aggregating, proliferating, nidating and forming a metastasis in blood vessels.
Secondly, the invention solves the individual problem of cancer cells by applying the plant volatile oil with various varieties and low content. All cancer cells have their individuality, which refers to the diversity of cancer cells and the variability that appears after the formation of new lesions. The invention aims at different cancer cells and selects volatile oil containing a plurality of different anti-cancer substances. The volatile oil has strong penetrability and can reach tumor parts in a short time, so that different anticancer substances can be brought into cancer cells to kill different cancer cells at multiple targets. Fifthly, adding 1.5kg of active carbon into the puffer liver oil clear liquid, controlling the temperature to be 60-80 ℃, stirring and adsorbing for 60 minutes, pouring into a fine filter paper bag, and performing fine filtration twice to obtain about 10kg of refined puffer liver oil for later use.
Example 2 preparation of Compound Fugu liver oil composition for treating cancer
Taking 8kg of refined globefish liver oil, adding 1kg of pressed oil of magnolia sieboldii seeds, and uniformly stirring for later use.

③ preparing 2 thousands of soft capsules from 10kg of the composition, wherein each capsule contains 0.5g of composition net weight, and each capsule contains 5 volatile oils, 2% of each volatile oil, such as magnolia flower oil, asarum oil, angelica dahurica oil, garlic oil and safflower oil, and 10% of the total volatile oil; peppermint oil, clove oil, sage oil, burreed tuber oil, zanthoxylum oil, ginger oil, lithospermum oil, perilla leaf oil and other 8 volatile oils, each content being 1.25%, and the total content being 10%; the content of refined globefish liver oil is 80%.

collecting 100g of allium macrostemon oil, citrus oil, curcuma oil, zedoary turmeric oil, ginger oil, geranium oil, garlic oil, blumea oil, vitex oil and perilla leaf oil respectively, adding 1000g (volatile oil) in total, mixing with the above-mentioned first step, and uniformly stirring so as to obtain 10kg of swellfish liver oil and vegetable oil composition;
③ preparing 2 million capsules from 10kg of the composition, wherein each capsule contains 0.5g of the composition by net weight, 1 percent of volatile oil, 10 percent of dracaena odorata seed pressed oil and 80 percent of refined globefish liver oil;

taking 7kg of refined globefish liver oil, adding 0.9kg of brucea javanica squeezed oil, and uniformly stirring for later use;
