本发明提供一种球孢白僵菌可分散油悬浮剂及高效使用方法,孢白僵菌Bb455可分散油悬浮剂中加入了独脚金内酯,能有效促进球孢白僵菌孢子萌发。球孢白僵菌Bb455专用助剂,可与孢白僵菌Bb455孢子完全相容,对孢子萌发无影响,同时提高孢白僵菌Bb455可分散油悬浮剂施用时在靶标上的润湿性能、附着性能,提高球孢白僵菌孢子Bb455对靶标害虫上的附着率和侵染率,从而提高杀虫速度和防治效果。

技术领域
本发明涉及生物农药杀虫剂领域,具体是一种球孢白僵菌菌株Bb455高效使用方法。
背景技术
球孢白僵菌(Beauveria bassiana)是一种广谱性的昆虫病原真菌。因为其致病力强且环境友好的特点,已经被广泛用于农、林及卫生害虫的生物防治,我国使用球孢白僵菌防治马尾松毛虫和玉米螟是国际上最大的害虫防治项目,取得良好的害虫持续控制效果。由于球孢白僵菌对害虫的防治效果较为良好且不会对环境和人体造成危害,因此越来越受到人们的重视。球孢白僵菌及其制剂是应用最广泛的生物制剂之一。目前国内登记的以球孢白僵菌为有效成分的农药品种有25个,其中制剂19个,应用于蛾类、蚜虫、棉铃虫、松毛虫等多种害虫的防治。
然而与化学农药相比,球孢白僵菌存在疏水作用强,剂型单一且生物活性易受环境影响,工作效率低等不足缺点。孢子在生产和保存过程中存在消解或使用时无法充分萌发等风险,从而导致产品球孢白僵菌防效低、效果不稳定等。球孢白僵菌主要通过接触侵染昆虫,孢子在虫体上成功附着后开始萌发、定殖、大量繁殖,同时分泌各种毒素物质,从而使害虫致死,这整个过程需要较长的时间,孢子有效附着及快速侵染是关键因素。筛选出生物活性高的球孢白僵菌菌株,加工成稳定的制剂产品,研制出一套有利于孢子附着及侵染,提高杀虫速度及防治效果的高效使用方法,是促进球孢白僵菌类生物农药大面积快速应用的有效手段,也是一项亟待解决的技术问题。
球孢白僵菌Bb455(保藏号为CGMCC NO.21472)由安徽省农业科学院植物保护与农产品质量安全研究所生物防治团队从宁国玉米种植区调查发现田间感染虫生真菌的草地贪夜蛾僵虫中分离鉴定获得。具体测序结果如下:

发明内容
本发明涉及一种球孢白僵菌可分散油悬浮剂及高效使用方法,本发明广泛的收集田间虫生真菌僵虫,经过室内毒力测定发现了对草地贪夜蛾、水稻二化螟等具有较高生物活性的球孢白僵菌菌株,命名为Bb455,保藏号为CGMCC NO.21472;针对筛选到的高效菌株,以提高该球孢白僵菌孢子萌发率、促进球孢白僵菌孢子快速有效附着定殖为基本目的,以独脚金内酯为球孢白僵菌孢子萌发促进物质,加入不影响球孢白僵菌孢子萌发的表面活性剂、油相载体及适宜的其它生物农药助剂,制各成稳定的球孢白僵菌Bb455可分散油悬浮剂;同时加入具有易润湿、易渗透、粘附作用的专用增效助剂以利于孢子附着及侵染,从而提高球孢白僵菌Bb455杀虫速度及防治效果,应用用于防治草地贪夜蛾、水稻螟虫等虫害。
具体技术方案如下:
一种孢白僵菌Bb455可分散油悬浮剂,各原料组分按质量百分比为:球孢白僵菌Bb455孢子粉(1000亿孢子/克)10%-50%、独脚金内酯0.1-0.2%、甲醇0.5-2%、脂肪醇与环氧乙烷缩合物1-10%、聚氧乙烯失水山梨醇脂肪酸酯1-10%、十二烷基苯磺酸钙1-10%、增稠剂黄原胶0.1-1%、硅酸镁铝1-5%、其余组分为菜籽油;所述孢白僵菌Bb455的保藏号为CGMCC NO.21472。
优选的,球孢白僵菌Bb455孢子粉(1000亿孢子/克)10%-50%、独脚金内酯0.16%、甲醇1%、脂肪醇与环氧乙烷缩合物7%、聚氧乙烯失水山梨醇脂肪酸酯3%、十二烷基苯磺酸钙5%、增稠剂黄原胶0.2%、硅酸镁铝1.5%-2.5%、其余组分为菜籽油。
本发明提供一种孢白僵菌Bb455可分散油悬浮剂,将独脚金内酯用甲醇溶解混入油相载体,将表面活性剂、独脚金内酯、增稠剂加入油相载体,将固形物研磨至粒径2-3微米的颗粒,在搅拌下加入球孢白僵菌孢子粉,充分搅拌后高速剪切30min,过滤去除残渣,得成品。
本发明提供一种孢白僵菌Bb455可分散油悬浮剂的使用方法,将球孢白僵菌Bb455可分散油悬浮剂用少量水稀释后,与专用助剂混合后,按一定用量稀释后采用低容量或超低容量技术喷雾;或采用常量喷雾,用于防治害虫;
所述的专用助剂为:各原料组分按质量百分比为:异辛醇24%、脂肪醇与环氧乙烷的缩合物12%、脂肪酸聚氧乙烯酯16%、成膜剂0.4%、沉降剂2%、增稠剂0.9%,其余的组分为油(黄油:油酸甲酯的质量比为1∶1);成膜剂为羟甲基纤维素、羟乙基纤维素、聚氨酯改性硝酸纤维、聚氨酯丙烯酸酯、聚乙烯丙烯酸酯改性丁二烯树脂中的一种或几种的混合。增稠剂为黄原胶、硅酸镁铝、聚丙烯酸钠、羟丙基甲酸纤维素、松香树脂、β-环状糊精中一种或几种的混合
所述的害虫为草地贪夜蛾、水稻螟虫。
本发明提供一种球孢白僵菌Bb455专用助剂,各原料组分按质量百分比为:异辛醇24%、脂肪醇与环氧乙烷的缩合物12%、脂肪酸聚氧乙烯酯16%、成膜剂0.4%、沉降剂2%、增稠剂0.9%,其余的组分为油(黄油∶油酸甲酯的质量比为1∶1);其制备方法:将异辛醇、水溶性表面活性剂、成膜剂、沉降剂、增稠剂在搅拌下引入油相,制成稳定油剂,充分搅拌后过滤去除残渣,得成品。
与现有技术相比,本发明具有如下有益效果:
1.本发明的孢白僵菌Bb455可分散油悬浮剂中加入了独脚金内酯,能有效促进球孢白僵菌孢子萌发,在附着虫体后可提高球孢白僵菌Bb455孢子快速萌发;以孢子萌发率为主要指标优选了表面活性剂、增稠剂等,有效保证了孢子的在有效期内的稳定贮存,加工成的制剂对多种害虫效果显著。
2.本发明的球孢白僵菌Bb455专用助剂,可与孢白僵菌Bb455孢子完全相容,对孢子萌发无影响,同时提高孢白僵菌Bb455可分散油悬浮剂施用时在靶标上的润湿性能、附着性能,提高球孢白僵菌孢子Bb455对靶标害虫上的附着率和侵染率,从而提高杀虫速度和防治效果。

生物测试实施例1
1.1球孢白僵菌高效菌株的筛选和鉴定
从采集的僵虫中分离白僵菌菌株,采用《农药室内生物测定试验准则 杀虫剂 第6部分:杀虫剂试验 浸虫法》(NYT-1154.6-2006)中的方法进行生物活性测定,筛选出高生物活性菌株Bb455,其生物活性测定结果如表1。

该菌株已保藏,保藏号为CGMCC NO..21472。
1.2独脚金内酯促进球孢白僵菌Bb455孢子萌发的效果测定
分别将含有独脚金内酯(浓度为0.16%)、不含独脚金内酯的不同含量球孢白僵菌Bb455可分散油悬浮剂,用无菌水均稀释成1.0×105孢子/mL悬浮液。加入到营养液中,定期调查孢子萌发率。从表2可以看出,独脚金内酯对球孢白僵菌Bb455孢子萌发具有明显的促进作用,各处理48h内孢子萌发率均达到80%以上,明显高于空白对照(萌发率70.2%)。

实施例11
一种球孢白僵菌可分散油悬浮剂高效使用方法,将球孢白僵菌Bb455可分散油悬浮剂用少量水稀释后,与专用助剂混合后,按一定用量稀释后采用低容量或超低容量技术喷雾;或采用常量喷雾,防治草地贪夜蛾、水稻螟虫等虫害。
所述的专用助剂,各原料组分按质量百分比为:异辛醇24%、脂肪醇与环氧乙烷的缩合物12%、脂肪酸聚氧乙烯酯16%、成膜剂羟甲基纤维素0.4%、沉降剂2%、增稠剂黄原胶0.9%,其余的组分为油(黄油∶油酸甲酯的质量比为1∶1);其制备方法:将异辛醇、水溶性表面活性剂、成膜剂、沉降剂、增稠剂在搅拌下引入油相,制成稳定油剂,充分搅拌后过滤去除残渣,得成品。
生物测试实施例2
2.1专用助剂与球孢白僵菌Bb455孢子相容性测定
分别将不同含量球孢白僵菌Bb455可分散油悬浮剂,用无菌水均稀释成1.0×107孢子/mL悬浮液,与不同浓度的专用助剂(按用水量)混合,然后加入到营养液中,48h后调查孢子萌发率。从表3可以看出,专用助剂对球孢白僵菌Bb455孢子萌发没有明显的影响,各处理浓度48h内孢子萌发率均达到80%以上,与空白对照无明显差异。

2.2不同浓度专用助剂对球孢白僵菌Bb455孢子侵染的影响
分别将不同含量球孢白僵菌Bb455可分散油悬浮剂,用无菌水均稀释成1.0×107孢子/mL悬浮液,与不同浓度的专用助剂混合(按用水量),采用《农药室内生物测定试验准则 杀虫剂 第6部分:杀虫剂试验 浸虫法》(NYT-1154.6-2006)中的方法,以草地贪夜蛾2龄幼虫为靶标,进行生物活性测定,7天后调查草地贪夜蛾幼虫侵染率。从表4可以看出,未添加专用助剂各孢子浓度处理7天内孢子萌发率最高为65.6%,而添加专用助剂后各孢子浓度处理7天内孢子萌发率均达到70%以上,与空白对照具有明显差异。试验结果表明,专用助剂对球孢白僵菌455孢子侵染具有明显促进作用,当浓度达到3%以上后侵染率无明显变化。

2.3专用助剂对球孢白僵菌Bb455杀虫活性的影响
将30亿孢子/克球孢白僵菌Bb455可分散油悬浮剂,用无菌水均稀释成1.0×107孢子/mL悬浮液,与3%浓度(按用水量)专用助剂混合,采用《农药室内生物测定试验准则杀虫剂第6部分:杀虫剂试验浸虫法》(NYT-1154.6-2006)中的方法,以草地贪夜蛾2龄幼虫为靶标,进行生物活性测定,7天后调查草地贪夜蛾幼虫死亡率。
从表5可以看出,草地贪夜蛾2龄幼虫处理后,添加专用助剂处理第4天开始死亡,累计死亡率为20.6%,而未添加专用助剂在第6天才开始死亡,专用助剂可以明显提高球孢白僵菌Bb455的杀虫速度。累计死亡率随着处理后时间的延长而增加,第12天以后增加趋势渐趋稳定,球孢白僵菌Bb455添加专用助剂处理对草地贪夜蛾2龄幼虫累计死亡率明显高于未添加助剂处理,其中添加专用助剂累计死亡率最高为87.8%,而未添加专用助剂累计死亡最高为76.5%,由此可知,专用助剂可明显提高球孢白僵菌Bb455的杀虫效果。
