Abstract

本发明公开了化合物Xerucitrinin A在制备防治香蕉枯萎病病原菌药物中的应用。本发明的主要目的是解决有效抑制香蕉枯萎病病原菌问题,以现有的杀菌剂,未能有效对香蕉枯萎病病原菌的抑制而不伤害香蕉植株。通过采用我们所发明的杀菌剂处理方法,可以在限制香蕉枯萎病病原菌生长的同时,而不对香蕉植株造成不良影响。

Description

化合物Xerucitrinin A在制备防治香蕉枯萎病病原菌药物中 的应用
技术领域:
本发明属于香蕉病虫害防治领域,具体涉及化合物Xerucitrinin A在制备防治香蕉枯萎病病原菌药物中的应用。
背景技术:
尖孢镰刀菌是引起香蕉枯萎病的主要原因。其中1号生理小种与4号生理小种是严重威胁着全球香蕉产量(Siddhesh,International Journal of Pest Management,2015,61(3):250-263)。其土传特性使得控制甚至消灭尖孢镰刀菌变得十分困难(Carvalhais,Frontier in plant science,2019,10)。随着病情的发展,植株叶片开始变黄,并逐渐枯萎。伴随着病情的进一步蔓延,叶片会掉落,围着假茎形成一圈。新生叶片会出现边缘皱褶(Berg,Molecular Plant Pathology,2010,8(3))。一旦香蕉植株被尖孢镰刀菌感染,目前来说还未有有效的控制措施。这种困难是由于尖孢镰刀菌可以产生大量孢子,这些孢子可以在土壤中存活30年之久(Ploetz,Plant Health Prog,2000,10:1-7)。现有的杀菌剂与熏蒸方法不能有效的控制尖孢镰刀菌孢子蔓延(Ploetz,Plant Health Progress,2005)。
目前大多数杀菌剂存在一种较为常见的问题,即其抑菌浓度与对植物生长的限制作用存在一种矛盾的关系。往往在抑制了病原菌生长的同时,对植物的生长也造成一定的负面影响。需要在抑制病原菌生长与植物生长中间寻找平衡。进一步筛选出对病原菌有效作用的杀菌剂。
Salendra等从桔青霉菌(Penicillium citrinum SCSIO 41017)中首次分离出天然化合物Xerucitrinin A(Salendra,Natural Product Research,2019)。本课题组于2020年,首次从链霉菌Sm4-1986中分离出化合物Xerucitrinin A(Wu,Natural ProductResearch,2020),结构式如下所示:

发明内容:
本发明的主要目的是解决有效抑制香蕉枯萎病病原菌问题,以现有的杀菌剂,未能有效对香蕉枯萎病病原菌的抑制而不伤害香蕉植株。通过采用我们所发明的杀菌剂处理方法,可以在限制香蕉枯萎病病原菌生长的同时,而不对香蕉植株造成不良影响。因此本发明的第一个目的是提供化合物Xerucitrinin A在制备防治香蕉枯萎病病原菌药物中的应用。
优选,所述的防治香蕉枯萎病病原菌药物还包括农药学上可接受的助剂。
优选,所述的防治香蕉枯萎病病原菌药物还包括其它的防治香蕉枯萎病病原菌的药物。
优选,所述的防治香蕉枯萎病病原菌药物的剂型为可湿性粉剂、水分散粒剂、悬浮剂或乳油。
优选,所述的防治香蕉枯萎病病原菌药物中化合物Xerucitrinin A的浓度为3mM。
本发明与现有技术相比,具有以下效果:
1.化合物Xerucitrinin A具有抑制香蕉枯萎病病原菌生长的作用。
2.在化合物Xerucitrinin A完全抑制香蕉枯萎病病原菌生长的浓度下对香蕉植株无不良影响。
因此可以将化合物Xerucitrinin A用于制备防治香蕉枯萎病病原菌的药物。

具体实施方式
为了具体阐明本发明所采取的手段及实施效果,以下结合实施例及附图对本发明作进一步说明。
实施例1
铬天青(CAS)检测平板,即铁载体筛选培养基:1mL 20%葡萄糖溶液,3mL 10%酪素,100μL 1mmoL/L CaCl2,2mL 1mmoL/L MgSO4,100mL H2O,1.8g琼脂,加热,约60℃时缓慢加入CAS染液和磷酸盐缓冲液各5mL,即得CAS蓝色检测培养基。
CAS染液:1mmol/L铬天青,4mmol/L十六烷基三甲基溴化铵,0.1mmol/L FeCl3
磷酸盐缓冲液(pH 6.8):2.427g Na2HPO4·12H2O,0.5905g NaH2PO4·2H2O,0.075gKH2PO4,0.250g NH4Cl,0.125g NaCl,100mL H20。所有上述溶液均用去离子水配制。
将无菌牛津杯放置在CAS检测平板中间,用无菌枪头吸取0.2mM化合物溶液200μL加入到牛津杯中,静置12小时,观察牛津杯周围有无嗜铁圈出现。
结果显示,化合物Xerucitrinin A具有结合Fe的功能,运用CAS平板检测Xerucitrinin A的Fe结合能力,如图1,在牛津杯中加入Xerucitrinin A,放置3个小时后,可在牛津杯周围出现嗜铁圈,表明Xerucitrinin A具有Fe结合能力。通过结合铁而抑制香蕉枯萎病病原菌的生长。
实施例2
在高氏一号培养液中加入终浓度为3mM的XcA(即化合物Xerucitrinin A),并且加入过滤下来的尖孢镰刀菌孢子悬浮液,孢子终浓度为1×107spores/mL。对照处理则用同体积的无菌水处理。孢子悬浮液在28℃黑暗条件下培养24h。收集尖孢镰刀菌孢子,在4℃、2.5%(v/v)戊二醛溶液中过夜固定。孢子固定后用0.1mol/L磷酸盐缓冲液(PBS,pH 7.4)漂洗2次后,用30%、50%、70%、80%、90%、95%和100%的乙醇溶液梯度脱水20min,然后进行干燥与喷金处理。用扫描电镜观察尖孢镰刀菌(TR4)孢子形态。
结果显示,在高氏一号培养液中,添加3m M浓度的Xerucitrinin A可以改变尖孢镰刀菌孢子形态,使尖孢镰刀菌孢子表面发生明显邹缩,并影响尖孢镰刀菌孢子的萌发与生长(图2)。
实施例3
XcA在乙醇中溶解。为了研究XcA对巴西蕉生长的影响,在高氏一号培养液中添加3mM的XcA。在对照中加入与化合物浓度相等的乙醇体积(CK)。设置阴性对照,在高氏一号培养液中添加200μM 8羟基喹啉(8HQ),然后将巴西蕉幼苗栽种到含有上述处理培养液的培养板中,每个处理6个重复(6株巴西蕉幼苗)。将含巴西蕉幼苗的培养板置于生长箱中生长10天,以观察对植物生长影响。各处理持续生长2个月,期间补充等体积无菌水,测定各处理株高、根重与苗重指标。
结果显示,在高氏一号培养基中分别添加无菌水,3mM化合物Xerucitrinin A(XcA)与200μM 8HQ。与对照相比,3mM化合物Xerucitrinin A对香蕉幼苗生长无不良影响(图3)。
化合物Xerucitrinin A可以作为防治香蕉枯萎病病原菌菌剂中的有效添加成份,具有良好的应用价值。

1.化合物Xerucitrinin A在制备防治香蕉枯萎病病原菌药物中的应用。
2.根据权利要求1所述的应用,其特征在于,所述的防治香蕉枯萎病病原菌药物还包括农药学上可接受的助剂。
3.根据权利要求1所述的应用,其特征在于,所述的防治香蕉枯萎病病原菌药物还包括其它的防治香蕉枯萎病病原菌的药物。
4.根据权利要求1所述的应用,其特征在于,所述的防治香蕉枯萎病病原菌药物的剂型为可湿性粉剂、水分散粒剂、悬浮剂或乳油。
5.根据权利要求1所述的应用,其特征在于,所述的防治香蕉枯萎病病原菌药物中化合物Xerucitrinin A的浓度为3mM。
