Technical Field
The invention belongs to the field of medicine. In particular to the application of the longstamen onion bulb extract in preparing the medicine or the cosmetics for preventing or treating alopecia and promoting hair growth.
Background
Alopecia has a significant impact on the mental health and quality of life of an individual, and is a source of low self-esteem and anxiety in many patients, and serious people can cause depression. Normally shed hair is typically hair in the catagen and telogen phases, and the normal amount of hair is maintained because the hair entering the catagen phase is constantly in dynamic balance with the hair entering the catagen phase. Androgenetic alopecias (AGA), also known as seborrheic alopecia, belong to pathological alopecia, account for more than 90% of alopecia population, are mainly manifested by greasy scalp, pruritus, dandruff, alopecia and the like, are the most common clinical alopecia types, are caused by endocrine dyscrasia, are different from chemotherapy toxic and side effects, are mostly seen in young and old males between 20 and 30 years old, are characterized by progressive reduction of head and top hairs, mostly appear in adolescence, and aggravate with age. Androgenic alopecia is present in both males and females, but is more common in males. To date, the U.S. FDA has approved only two drugs for the treatment of androgenetic alopecia: the minoxidil tincture suitable for men and women and the oral finasteride tablet suitable for men only have obvious side effects, and the minoxidil tincture and the oral finasteride tablet are easy to relapse after stopping taking medicines.
Bulbus Allii Macrostemi (Alliiimacharostomunisbulb) is dried bulb of Bulbus Allii Macrostemi or Bulbus Allii Macrostemi of Liliaceae. The Bulbus Allii Macrostemi is a Chinese medicinal material with homology of medicine and food, and has effects of inhibiting blood platelet aggregation, reducing blood lipid, resisting atherosclerosis, inhibiting tumor, diminishing inflammation, resisting oxidation and inhibiting bacteria. The allium macrostemon has small toxic and side effects and is easy to plant, thus being a Chinese medicinal material with great development value.

The invention aims to provide a new application of a traditional Chinese medicine allium macrostemon extract.
Specifically, the invention provides an application of an allium macrostemon extract in preparing a medicine or a cosmetic for preventing and treating alopecia, wherein the allium macrostemon extract is prepared by the following method: pulverizing 500g of Bulbus Allii Macrostemi, extracting with 95% ethanol for 2 times by reflux extraction method, wherein the material-liquid ratio is 1:10 and 1:8 respectively; the extraction time is 2h and 1.5h respectively; the two filtrates are combined, concentrated and dried under reduced pressure.
In a preferred embodiment, the allium macrostemon extract is used as the only active ingredient for preparing the medicine or the cosmetic for preventing and treating the alopecia.

The invention arose in part from the unexpected discovery that: the allium macrostemon extract can promote the hair growth of a mouse model with androgenetic alopecia induced by testosterone. Therefore, the allium macrostemon extract can be used for preparing medicaments, health-care products, cosmetics, washing and caring products and the like for preventing and treating alopecia, particularly androgenetic alopecia.
Furthermore, the invention provides the application of the allium macrostemon extract in preparing the medicines or the cosmetics for preventing and treating the androgenetic alopecia.
The allium macrostemon extract can be prepared by the conventional method in the field, for example, the allium macrostemon Chinese medicinal material is crushed, the reflux extraction method is adopted to add 95 percent ethanol for extraction, the filtrate is combined, and the decompression, concentration and drying are carried out.
The medicinal carrier in the composition does not damage the pharmaceutical activity of the allium macrostemon extract, and the effective dosage of the allium macrostemon extract is nontoxic to human bodies when the dosage of the allium macrostemon extract plays a role of the medicinal carrier.
Such pharmaceutically acceptable carriers include, but are not limited to: lecithin, aluminum stearate, alumina, ion exchange materials, self-emulsifying drug delivery systems, tweens or other surfactants, serum proteins, buffer substances such as phosphates, glycine, sorbic acid, water, salts, electrolytes such as protamine sulfate, disodium hydrogen phosphate, potassium hydrogen phosphate, sodium chloride, zinc salts, magnesium silicate, mixtures of saturated fatty acid partial glycerides, and the like.
Other conventional pharmaceutical adjuvants such as binder (e.g. microcrystalline cellulose), filler (e.g. starch, glucose, anhydrous lactose and lactose beads), disintegrant (e.g. crosslinked PVP, croscarmellose sodium, low-substituted hydroxypropylcellulose), lubricant (e.g. magnesium stearate), and absorption enhancer, adsorption carrier, flavoring agent, sweetening agent, excipient, diluent, wetting agent, etc.
The allium macrostemon extract composition is prepared into medicines, cosmetics, washing care products and the like according to a conventional method in the field.
The cosmetic or hair care composition may be in the form of hair tonic, hair conditioner, hair essence, hair lotion, hair tonic lotion, shampoo, hair rinse, hair cream, hair tonic, hair massage cream, hair wax, hair gas, hair spray, hair pack, hair tonic, hair soap, hair cleansing foam, hair oil, hair drying agent, hair conditioner, hair dye, hair curling preparation, hair bleach, hair gel, hair glaze, hair dressing, hair tonic, hair moisturizer, hair mousse, and hair spray.

1.2 Experimental animals
C57BL/6 male mice weighing 20 ± 2g (8 weeks), provided by the experimental animals center of the university of medicine in shanghai, animal certification No.: SYXK (Shanghai) 2009-0069. Placing in conventional breeding environment, and freely taking food and drinking water.
2. Experimental methods
2.1 preparation of Allium macrostemon extract
(1) Preparing the allium macrostemon extract powder: pulverizing 500g of Bulbus Allii Macrostemi, extracting with 95% ethanol for 2 times by reflux extraction method, wherein the material-liquid ratio is 1:10 and 1:8 respectively; the extraction time is 2h and 1.5h respectively; mixing the two filtrates, concentrating under reduced pressure, drying to obtain dry extract, pulverizing, and sieving to obtain 50g of Bulbus Allii Macrostemi extract powder.
(2) Adding 3g of Bulbus Allii Macrostemi extract powder into 100ml of 1, 2-propylene glycol, stirring and mixing to obtain 3% Bulbus Allii Macrostemi extract solution.
2.2 identification of Chinese medicine of longstamen onion bulb
The identification of the allium macrostemon traditional Chinese medicinal material is carried out according to the method of China pharmacopoeia of 2020, and allium macrostemon medicinal material powder and allium macrostemon extract powder are taken, 20ml of normal hexane is added, ultrasonic treatment is carried out for 20min, filtration is carried out, the filtrate is volatilized to dryness, and 1ml of normal hexane is added to residues for dissolution, so that the residues are used as a test solution. Preparing reference medicinal material of Bulbus Allii Macrostemi 4g, and making reference medicinal solution in the same way. Performing thin layer chromatography (general rule 0502) test, sucking 10ul of the above three solutions, respectively dropping on the same silica gel G thin layer plate, developing with n-hexane-ethyl acetate (10:1) as developing agent, taking out, air drying, spraying 10% sulphuric acid ethanol solution, heating at 105 deg.C until the spots are clearly developed, and inspecting under ultraviolet lamp (365 nm).
2.3 establishment and group sampling of androgenetic alopecia mouse model

A C57BL/6 male mouse (20 +/-2 g) with uniform weight is selected to be adaptively raised for 1 week, is mixed with rosin and paraffin, is heated, dissolved and cooled, is coated on the back of the mouse to remove hair, and induces the hair follicle of the mouse to enter a growth phase from a resting phase, wherein the hair follicle is not different from the spontaneously formed hair follicle in the growth phase in morphology, and the skin is pink. The experimental area on the back of each group of mice on the next day of hair removal was divided into groups and smeared according to the following method:
blank control group: smearing the solvent for 30min, and smearing blank matrix;
model group: coating 0.1% testosterone for 30min, and coating blank matrix;
minoxidil group: applying 0.1% testosterone for 30min, and applying 3% minoxidil solution (prepared with 1,2 propylene glycol);
allium macrostemon extract group: applying 0.1% testosterone for 30min, and applying 3% Bulbus Allii Macrostemi extract solution (preparation method is shown in 2.1);

2.4.1 Hair Length measurement
The hair was plucked out before, during and after the depilatory area at 15d, 18d and 21d, respectively, and the lengths thereof were measured and averaged.
2.4.2 gross weight measurement
After day 21, the mice were sacrificed and 3 x 3cm area of hair was cut along the hair roots in the depilated area, weighed on an analytical balance and averaged.
2.5 statistical analysis
All experimental data were repeated 3 times, the results were expressed as mean ± standard deviation, and One-way analysis of variance (One-way ANOVA) and LSD test were performed on the experimental data using SPSS 16.0 statistical software, with P <0.05 being statistically significantly different.
3. Results
3.1 identification of Chinese medicine of longstamen onion bulb
FIG. 1 shows that in the chromatogram of the test sample, the Chinese medicinal material of Bulbus Allii Macrostemi and the contrast of Bulbus Allii Macrostemi have fluorescent spots with the same color at the corresponding positions of the chromatogram; the allium macrostemon extract, the allium macrostemon traditional Chinese medicinal material and the allium macrostemon reference substance also have similar fluorescent spots on the corresponding positions of the chromatogram, but the fluorescence intensity is slightly low, thereby proving that the traditional Chinese medicinal material is the allium macrostemon.
3.2 therapeutic Effect of Allium macrostemon extract on mice with androgen alopecia induced by Testosterone
3.2.1 Effect of Allium macrostemon extract on Hair growth
Table 1 shows the effect of allium macrostemon extract on hair length. The hair lengths of the modeling days 15, 18 and 21 are selected for analysis, the hair growth of the blank control group is more, and the difference is significant (P is less than 0.001) compared with that of the alopecia model group, which shows that testosterone can inhibit the hair growth, and the model is successfully established; the minoxidil and the allium macrostemon extract can promote the hair growth, and the hair growth is remarkably different from the hair loss model group on 15 th, 18 th and 21 th days of the model building (P is less than 0.05 or P is less than 0.01 or P is less than 0.001). The above shows that the allium macrostemon extract can stimulate the growth of hair.

3.2.2 Effect of Allium macrostemon extract on Hair weight
Table 2 shows the effect of allium macrostemon extract on the gross weight of mice. The mice are sacrificed on the 21 st day, a depilation area is selected, hairs with the area of 2 x 2cm are cut off, the blank control group is weighed by an analytical balance, the hair weight of the hair loss model group is 10.08 +/-3.64 mg, the minoxidil group is 63.35 +/-7.42 mg, and the allium macrostemon extract group is 36.2 +/-5.31 mg, so that the hair growth can be promoted by the minoxidil and the allium macrostemon extract. Compared with the hair loss model group, the hair weights of the minoxidil group and the allium macrostemon extract have significant difference (P is less than 0.01 or P is less than 0.001). The above shows that the allium macrostemon extract can stimulate the growth of hair.
