背景技术
为了限制与人类食物的竞争,禽日粮(diet)中使用的饲料种类逐渐丰富,通过在日粮中使用谷物副产品、并引入更多本地资源,减少对进口的依赖,例如在欧洲对大豆的依赖。但是相比于传统的玉米和大豆日粮,这些替代性的饲料通常质量较低且不稳定。由此,鸡对这些非最优的新型日粮的消化效率(DE)正成为比以往更重要的饲料效率中的因素。提高消化效率以提高饲料效率因而是禽生产的经济效益和环境影响的关键因素之一。这加强了对消化效率的遗传选择的兴趣,消化效率是饲料效率的因素之一,其通过零氮贮留(zeronitrogen retention)校正的表观代谢能(AMEn)进行评估。
相比于以前的化学分析方法,近红外光谱(NIRS)技术的发展极大地提高了大规模测定消化效率的可行性。这也使得针对这项特性的基因实验成为可能,我们表明了消化效率是可遗传的,并且可以作为选择的标准(Mignon-Grasteau等人,2004)。然而制备来源于大量动物(禽)的排泄物样品仍然需要耗费大量时间。而且收集全部排泄物需要将动物(禽)在平衡试验期间单独分笼饲养,会损害动物的福利。

申请人在本发明中表明,血清在492nm的吸光度可以用作消化效率选择的间接标准。
发明内容
本发明的第一个目的在于提供一种评估禽(特别地为鸡)的消化效率(DE)的体外方法,通过测定所述禽血液的液体部分样品的颜色进行。
本发明还涉及一种根据本发明的体外方法用于选择禽的用途,所述禽具有改进的消化效率(DE),特别地所述禽为鸡。
本发明的另一个目的在于根据本发明的体外方法用于筛选饲料或日粮和/或用于生产饲料或日粮的用途,所述饲料或日粮适应于禽和/或为禽而优化的,特别地所述禽为鸡。

具体实施方式
本发明涉及一种评估禽(特别地为鸡)的消化效率(DE)的体外方法,其通过测定所述禽血液的液体部分样品的颜色进行。
根据本发明所述的“血液的液体部分”表示血液的非细胞部分。实际上,血液是人和其他动物的一种体液,由悬浮在血液的液体介质中的血细胞(红细胞、白细胞和血小板)组成。
本发明关注血液的液体介质,即血液的液体部分。
通过离心分离血液,可见三层:血浆(黄色上层)、血沉棕黄层(白色中间薄层)和红细胞层(红色底层)。
血浆相当于约55%的血液,是血液的液体介质,自身颜色为淡黄色。其本质上是含有92%水、8%血浆蛋白和痕量其他物质的水溶液。其还含有凝血因子(协助凝血)。
术语“血清”表示移除了凝血蛋白的血浆。
在本发明的一个具体的实施方案中,血液的液体部分样品为血浆样品或血清样品。
在一个优选的具体的实施方案中,血液的液体部分样品为血清样品。
在一个具体的实施方案中,禽选自:鸡、母鸡、火鸡、鹅、鸭、珠鸡、鹌鹑、和鸽。
在一个优选的具体的实施方案中,所述禽为鸡。
在本发明的一个优选的具体的实施方案中,血液的液体部分样品为鸡的血清样品。
根据本发明所述的“消化效率”(DE)是饲料效率的一个因素,通常使用零氮贮留校正的表观代谢能(AMEn)评估。
血液的液体部分样品、特别地为血清样品,可以在不同的时间(禽龄)采集。
在一个具体的实施方案中,血液的液体部分样品、特别地为鸡的血清样品,采集于第21至25天,优选地为第23至25天。实际上,根据发明人对鸡血清样品的研究,预计与DE关联的差异在该龄附近为最大。
在一个具体的实施方案中,血浆样品按下列步骤采集:
-采集血液样品,特别地为在3周时,在禽(特别地为鸡)的枕窦处采集,收集于含有抗凝剂的干燥的管内;
-收集的含有抗凝剂的样品在3000g离心10分钟;
-使用移液器取出上清(即血浆样品)。
作为抗凝剂的非限制性示例,可以提及例如EDTA、肝素盐、和柠檬酸钠。
在另一个优选的具体的实施方案中,血清样品按下列步骤采集:
-采集血液样品,特别地为在3周时,在禽(特别地为鸡)的枕窦处采集,收集于干燥的管内;
-收集的样品在室温下静置15分钟,直至凝血;
-凝集的血液在3000g离心10分钟;
-使用移液器取出上清(即血清样品)。
根据本发明所述的“样品颜色”是禽的一项具体特征,在本发明提供的背景下,受到食物(日粮)中所含色素和禽的消化效率(DE)的影响。
特别地,颜色与上述样品通过分光光度法测定的吸光度有关。
原理如下:样品(溶液)在白光照射下,如吸收了白光的一部分光辐射,则会显示出颜色。样品(溶液)的颜色是透过的光辐射的颜色,即吸收的颜色的互补色。根据样品(溶液)的性质,有色光的光谱呈现暗的吸收带。对于每次波长为λ的光辐射,样品(溶液)吸收的光的部分由其吸光度A(λ)测定。因此吸光度是一项反映样品(溶液)吸收光辐射的能力的物理量。
吸光度主要由两个参数决定:样品(溶液)的浓度,及透过样品(溶液)的光的波长。
因此吸光度是一项与波长为λ的光在样品(溶液)中的吸收强度相关的量。存在使吸光度达到最大值的波长。因此样品(溶液)的颜色对应于互补色。
可见光光谱的波长为400至700nm。
在一个具体的实施方案中,根据本发明所述血液的液体部分样品的颜色、特别是血清样品的颜色、优选地为鸡的血清样品的颜色,由400至520nm的吸光光谱测定,优选地为430至516nm,更优选地为460至500nm,进一步优选地为492nm波长。
申请人表明,特别是对于鸡的血清样品,发现特别地在492nm处差异最显著,与类胡萝卜素的吸收区域相对应,尤其是叶黄素和玉米黄素,这两者是谷类、特别是小麦中主要的类胡萝卜素。而且,由于叶黄素和玉米黄素比其他在此光谱区域中的类胡萝卜素更易被鸡吸收,如γ-胡萝卜素、虾青素和隐黄素,可以预测该492nm处的吸收峰很有可能与叶黄素和玉米黄素相对应。
在一个具体的实施方案中,对于大规模评估或筛选,禽的多个样品可以置于多孔板中,其吸光度由自动分光光度计测定。因此本发明还涉及一种评估和/或选择DE的体外方法,其通过测定血液的液体部分样品的颜色进行,其特征在于所述血液的液体部分样品、特别地为血清样品、优选地为鸡的血清样品,置于多孔板中(每孔一个样品),(每个样品的)吸收光谱使用分光光度计测定。
在一个具体的实施方案中,血液的液体部分样品、特别地为血清样品的颜色的测定由如下步骤实现:
-根据预先定义的方案,将200μL血液的液体部分样品、特别地为血清样品,置于板的孔中(每孔一个样品);
-使用分光光度计获取300至600nm的吸收光谱(2nm步进);
-所述样品的吸光度测定(每个样品一个吸光度)在400至520nm进行,优选地为430至516nm,更优选地为460至500nm,进一步优选地为492nm波长,依照分光光度计的指示,优选地为进行一式三份;且
-保存分光光度结果,以备进一步差异分析。
在一个优选的实施方案中,板为透明的96孔板。
在一个优选的具体的实施方案中,血液的液体部分样品为血清样品,优选地为鸡的血清样品,且吸收光谱在492nm波长处测定。
血液的液体部分样品、特别地为血清样品、优选地为鸡的血清样品,其吸收峰、特别是在492nm波长处,推测应与更高的禽吸收的类胡萝卜素浓度相对应。
本发明还涉及根据本发明的体外方法用于筛选具有改进消化效率(DE)的禽的用途,特别地所述禽为鸡。
这意味着选育在血液的液体部分样品(特别是血清样品)中特别是在492nm波长处具有更高吸光度值的禽,可以达到更高的消化效率值(由AMEn评估)。
在一个具体的实施方案中,本发明所述的体外方法包括选择禽(特别地为鸡)的步骤,所述禽的血浆或血清样品(优选地为血清样品)在492nm波长处具有更高的吸光度值。
本发明还涉及根据本发明的体外方法用于筛选饲料或日粮和/或用于生产饲料或日粮的用途,所述饲料或日粮为适应于禽(特别地为鸡)和/或为禽(特别地为鸡)而优化的饲料或日粮。
特别地,该评估禽消化效率(DE)的体外方法,可有助于筛选适应于和/或为提高消化效率(DE)而优化的饲料或日粮,其中所述禽饲喂了待测的饲料或日粮。
“适应于禽和/或为禽而优化”表示饲料或日粮的成分适应于禽的营养,和/或甚至经过优化,表示按预测可以被禽更好地消化。
本发明的另一个目的在于血液的液体部分样品的颜色用作筛选禽消化效率(DE)的间接标准的用途,特别地所述样品为血浆样品或血清样品,优选地为禽的血清样品,特别地所述消化效率为鸡的消化效率(DE)。
下面通过非限制性实施例解释本发明。
实施例
分别按对小麦日粮消化效率高或消化效率低选择两个品系的鸡,并比较其血清颜色。颜色通过血清在300至578nm的吸光度评估。两个品系在430至516nm的颜色不同,与类胡萝卜素(比如叶黄素和玉米黄素)的吸收区域相对应。
再评估血清颜色测定的遗传力,及其与消化效率的遗传相关性。
材料与方法
动物(禽)
在其余的实施例描述中,术语“动物”、“禽(poultry)”、“禽(bird)”或“鸡”可互换使用。
本文报道的所有动物管理与实验程序符合法国和欧洲关于动物实验的规定。饲养禽(鸡)的实验单位已在农业部作用于动物实验的登记,执照号码为C-37-175-1和A-17-661。实验中用于禽(鸡)的所有程序均包括在协议00885.02、2015121516442084-3202和2015040111006849中,经伦理委员会C2EA-19和C2EA-84批准,由法国研究部签发。
本研究所用的数据来源于两个连续实验,使用按高(D+)或低(D-)消化效率选择的不同的鸡的品系。初始种群为市售的中等生长程度的仔鸡,7周龄时体重达2kg。在最初8代,给禽饲喂一种难以消化的日粮,含有55%里亚托(Rialto)小麦(Mignon-Grasteau等人,2004),为一种坚硬、黏稠的小麦品种。使用第3周时平衡试验测定的AMEn,依照消化效率选择繁殖组。接下来的9代,繁殖两个品系,但不进行AMEn选择。在第20代,重新开始选择过程,采用相同的选择标准。
实验1.在第20代,无法获取更多新的里亚托小麦。进行预实验,将其换成含有其他品种小麦和黑麦的日粮,以模拟里亚托小麦的效应。两种日粮的成分在表1中给出。

禽(鸡)平养16天,再移入单独的笼中。23至25日时,每个品系、每种日粮取20只禽进行平衡试验。第23和25天(23和25日)时对禽称重。按Bourdillon等(1990)建议,在此期间收集全部排泄物。排泄物经冻干处理,如Bastianelli等(2010)所述使用NIRS进行分析,获取AMEn值。
在平衡试验结束时(25日),对于饲喂里亚托日粮的动物,从枕窦处取血样,测定血清颜色。将血液在室温下放置15分钟直至凝血,再离心(3000g,10分钟)制备血清。将200μL来自禽的血清样品转移到透明96孔板中(奥地利,克雷姆斯明斯特,Greiner Bio-One),使用Infinite M200分光光度计(瑞士,曼尼多夫,Tecan)测定其300至572nm的吸光光谱(2nm步进)。
实验2.连续孵化两次产生第21代的禽(鸡)(N=192D+,192D-)。给禽饲喂实验1(表1)测验过的替代日粮。孵化后平养13天(13日),再转移到单独的笼中进行平衡试验。平衡试验在第21至23天进行(21至23日),第24天(24日)采集动物血样。第21和23天时对禽称重。平衡试验、排泄物处理、血样处理及血清颜色测定的程序与实验1中相同。测定342至572nm间每2nm的血清吸光度。
表型与遗传分析
在300至578nm间每2nm的动物血清颜色数据上,使用Welch检验(Welch,1947)测试D+和D-之间的吸光度差异,其中所述动物饲喂有实验1中的里亚托小麦日粮。
对于遗传分析,使用了每代经选择的D+和D-品系的所有可用数据,即分别为4626、5068和417只动物的AMEn、23日时的体重和血清颜色。对于血清颜色,我们采用实验1和实验2中收集的吸光度数据,342至572nm间每2nm进行采集。使用VCE6.0进行遗传分析(Neumaier和Groeneveld,1998;Groeneveld等人,2010)。我们采用了包含孵化(N=28)、饲养室(N=8)和性别的固定效应的动物模型。对于血清颜色,也考虑了板的固定效应(N=6)。动物系谱总共包括6376只动物。考虑到特征数目大,而相邻波长的吸光度变量之间高度相关,我们进行了包含各AMEn、23日时的体重和2个不同波长下血清的吸光度的一系列分析。对2个不同波长下的所有可能的因素组合均进行测试,共进行276项分析。下列结果为每个分别的分析所得的遗传参数和其标准误的平均值。
结果与讨论
实验1表型结果
实验1所得数据分析显示,两种日粮饲喂的D+和D-品系间AMEn的差异在同一范围内(里亚托及替代日粮分别为424及501kcal.kg-1DM)。新日粮与初始日粮同样难消化,AMEn值比初始日粮低4.7%(表2)。

简要来说,D+和D-血清颜色在430至516nm之间有显著差异,D+品系血清颜色更深(图1)。“血清颜色更深”表示血清的着色程度更强烈(深黄色)。发现492nm处差异最显著,与类胡萝卜素的吸收区域相关,尤其是叶黄素和玉米黄素(Rodriguez-Amaya,2001),它们是谷物、尤其是小麦中主要存在的类胡萝卜素(Abrar等人,2015)。而且,由于叶黄素和玉米黄素比其他在此光谱区域中的类胡萝卜素更易被鸡吸收,如γ-胡萝卜素、虾青素和隐黄素(Surai等人,2001),我们可以预测该492nm处的吸收峰很有可能与叶黄素和玉米黄素相对应。
血清颜色的遗传参数
AMEn和体重(BW)的遗传力估计值为中至高(分别为0.36±0.02和0.50±0.04)。AMEn与BW之间的遗传相关性不显著(0.09±0.06)。这些估计值与这些品系过去所得的估计值相一致(Mignon-Grasteau等人,2004)。
血清颜色的遗传力估计值(表3)很低,且在最低(342至432nm)和最高(512至572nm)波长处不显著。462至502nm之间的估计值中等(0.24至0.31),与0有显著差异,492nm处遗传力最大。

这些估计值与过去研究(Stone等人,1971;Washburn和Ruff,1978)中8周龄仔鸡的类胡萝卜素浓度的遗传力相近(0.20至0.32)。在波长392至502nm之间,AMEn和血清颜色之间的遗传相关性最高(0.76至0.97)。相比之下,在同一区域内,血清颜色和体重的遗传相关性则很低且不显著。考虑遗传力和遗传相关性的估计值,血清在492nm的吸光度应是最有效的选择AMEn的间接标准,产生的间接响应估计会达到AMEn对直接选择响应的78%(图2)。
类胡萝卜素的血清浓度高度取决于动物从日粮中吸收的能力。类胡萝卜素的吸收受到肠道pH值和日粮纤维含量的影响(Faure等人,1999)。在本研究的情况下,两个品系之间的肠道pH值不同(de Verdal等人,2010),我们使用的日粮纤维含量也很高,可能影响类胡萝卜素的吸收。而且,消化效率和类胡萝卜素含量之间的联系也可以解释为消化效率选择的间接后果。
我们对这些不同品系的实验表明,该选择不仅影响消化,也影响肠道结构和免疫系统。实际上,类胡萝卜素影响肠道上皮细胞之间的缝隙连接,从而影响肠道结构(Faure等人,1999),且增强细胞介导的免疫应答和体液免疫应答,从而影响免疫系统(Chew和Park,2004)。
考虑血清颜色测定的简易性、其遗传力水平和其与消化效率的遗传相关性,血清颜色似是选择禽消化效率(DE)的有潜力的间接标准,可以用在提高消化效率的选择方案中。
