Abstract
一种防疫用口罩及使用方法将纱布中的糖基通过过碘酸钠氧化法,将纱布中的纤维素的糖基氧化形成醛基,再与胰蛋白酶中的氨基进行共价偶联,通过加氢反应生成稳定的偶联物,从而制备固相胰蛋白酶纱布口罩,再将该纱布口罩与医用外科口罩组合使用。使用时该纱布固相酶是放在普通医用口罩的外面,使胰蛋白酶直接与最外面空气接触,通过小型的喷雾瓶给胰蛋白酶纱布口罩外表面进行喷雾pH7.2‑8.0的溶液,使胰酶充分与新冠病毒接触、并在最适pH条件下发挥酶的作用。这样将空气中的病毒外表蛋白降解,当人体这种酶处理过的吸入空气时将会减少其中的活病毒含量,达到防控疫病的目的。

技术领域
本发明涉及免疫学、免疫学技术中固相化酶技术、传染病领域内容,服务于防疫工作,通过阻断传染途径来,减少传染病的危害。
发明背景
由于新冠病毒是RNA病毒,很容易出现变异,生产的疫苗也不好百分之百确保有效。因而人们或许通过居家呆着、减少外出、出门戴口罩、保持社交距离等措施,隔断传染病的传播途径,努力减少染病的风险。
因而只能另想其他办法,留意到新冠病毒是一个不耐高温、对酒精敏感、对胰蛋白酶敏感。或许我们可以在传播途径上做一些努力,或许能阻断病毒的传播。考虑到英国、南非均出现新冠变异株,人类面对局面将更加不确定性,因而而设计了本发明,希望能对新冠病毒的控制有一定的帮助。
发明内容
一种防疫用口罩及使用方法,其内容是通过过碘酸钠氧化法,将纱布纤维素中的糖基上的羟基进行氧化形成含有醛基化合物,该糖基的化合物上的醛基在碱性条件与蛋白质中氨基进行共价偶联,再通过硼氢化钠溶液进行加氢反应,使形成的不饱和键通过加氢形成稳定的饱和键,从而制备稳定的固相化酶。
固相酶技术将胰蛋白酶固定到纱布口罩上,制备成固相化酶,通过小的喷瓶(例如6ml 塑料喷瓶,内装pH7.2-8.0范围NaOH稀溶液)给纱布的外表面进行喷汽雾,使酶处在湿润环境,并且将固相胰酶处在最适工作pH的范围(即pH7.2-8.0),再利用人们呼吸时的空气将胰酶工作的温度提升,提高酶的作用效果。胰酶在适宜的pH、温度等条件下将破坏新冠表面的蛋白的纤突等蛋白,从而使之不能与人体内的病毒受体结合。同时病毒保护层破坏后,其核酸也易被空气中或人们呼出的核酸酶降解,本口罩再与医用口罩联合使用,从而增加阻挡新冠病毒防控效果。
可行性分析:过碘酸钠氧化法是制备免疫学实验技术中酶标抗体制备常用的方法,就是利用辣根过氧化物酶中的糖基进行氧化形成席夫氏醛,该醛基再与抗体蛋白中的氨基进行共价偶联制备酶标抗体。纱布中的纤维素就β-1、6-萄葡糖的聚合产物,因而其中的糖基也可以进行过碘酸钠氧化,实现纱布上偶联胰蛋白酶,形成固相化胰蛋白酶,反应条件温和,酶的活性依然保持。
由于胰蛋白起作用的条件是碱性条件,并且在溶液中效果较好,然而人呼吸是不能在水溶液中进行,因而需要借助固相化酶技术,通过喷雾使固相酶处在适当的pH条件,至于温度主要是环境温度以及人们呼吸时的热汽给固相酶加温,使之达到较为适宜的温度。
使用时该纱布固相酶是放在普通医用口罩的最外面,使之直接与最外面空气接触,以便与新冠病毒接触,发挥作用,将空气中的病毒通过生物酶降解,实现空气中病毒含量降低的效果。由于纱布的空隙较大,因而纱布口罩不能单独使用,必需要与医用口罩联合使用,才能增加防疫效果。
医用口罩用在里面与人的口罩接触,避免纱布上固相酶与人的皮肤直接接触,也可以防止胰蛋白酶的残基脱落或胰蛋白酶的降解产物被人体直接呼吸进入人体内部,产生不必要的问题。
本发明的创新点在于:
1、利用病毒对胰蛋白的敏感性,建立固相化酶技术,使酶固定于载体--棉花纤维素的多糖基上,并且胰蛋白酶的酶活性依然保持。
2、通过喷雾瓶喷雾将固相酶的保持适宜环境在偏碱性条件,以便胰蛋白酶发挥较大的作用效果,因为胰酶在酸性条件下活性很低或几乎没有活性。并且微湿纱布对人们的呼吸影响不大,不会引起呼吸困难。
3、在固相酶使用的同时,增加使用医用外科口罩,避免胰酶常用时间较久后,酶失活后用户依然使用,同时避免胰酶与肌肤接触。虽然肌肤的pH不是胰酶的适合范围,也不能让胰酶直接与肌肤直接接触,减少意外状况发生。
4、本防疫口罩不仅对胰酶敏感的病毒有效果,对细菌也有降解效果,因而有一定的防疫其他呼吸道传染病效果。
具体实施方式
实施例1
将市售的二层纱布或三层纱布的普通口罩直接浸入0.05M NaIO4溶液中室温下反应0.5h,蒸馏水冲洗去掉多余的NaIO4,然后将该纱布口罩浸入含有1-5%的胰蛋白的0.05MpH9.6 NaHCO3-Na2CO3溶液中4℃过夜进行偶联反应,次日加入2-20mg/ml硼氢化钠,这时纱布口罩上出现许多气泡,即加氢反应。加氢反应0.5-2h后,用水冲洗终止反应。晾干即制备好固相化胰蛋白酶,这种偶联技术反应条件温和,酶的活性没有失去。
使用时将该胰蛋白处理的固相酶口罩(在外层)与医用外科口罩(在内层)组合使用,并结合喷瓶向固相酶的口罩外表面喷雾几下,维持胰蛋白的适宜pH条件。胰蛋白在碱性条件下,降解病毒的纤突蛋白,使病毒不能与宿主体内细胞表面受体结合,从而减少病毒的感染性,实现阻断病毒传播。
实施例2
将市售的普通三层纱布口罩直接浸入0.05M NaIO4溶液中室温下反应0.5h,蒸馏水冲洗去掉多余的过碘酸钠(NaIO4),然后将该纱布口罩浸入含有1%的胰蛋白的0.05MpH9.6 NaHCO3-Na2CO3溶液中4℃过夜进行偶联反应,次日加入2mg硼氢化钠,这时纱布口罩上出现许多气泡,即加氢反应。加氢反应0.5-2h后,用水冲洗终止反应。晾干即制备好固相化胰蛋白酶。
使用时将该胰蛋白处理的固相酶的纱布二层或三层剪切适当大小缝合在医用外科口罩的外层,使空气进入人体内首先与纱布上的胰蛋白接触,并结合喷瓶喷雾,维持胰蛋白的适宜pH条件,使胰蛋白在碱性条件下发挥作用最佳。胰蛋白在碱性条件下,降解病毒的纤突蛋白,使病毒不能与宿主体内细胞表面受体结合,从而减少病毒的感染性,实现阻断病毒传播。
Claims (2)
1.一种防疫用口罩及使用方法是通过过碘酸钠氧化法,将纱布纤维素中的糖基上羟基进行氧化形成多醛化合物,该糖基的化合物上的醛基在碱性条件与胰蛋白酶中氨基共价偶联,再通过硼氢化钠溶液进行加氢,使形成的不饱和键通过加氢形成稳定的饱和键,从而制备稳定的固相化胰蛋白酶。通过固相酶技术将胰蛋白酶固定到纱布口罩上,制备成固相化酶,通过小的喷瓶(例如6ml塑料喷瓶,内装pH7.2-8.0范围NaOH稀溶液)给纱布外表面进行喷汽雾,使酶处在湿润环境,并且将固相胰酶处在最适工作pH的范围(即pH7.2-8.0),再利用人们呼吸时的空气将胰酶工作的温度实现提升,提高酶的作用效果。胰酶在适宜的pH、适宜温度等条件下将降解病原的所有蛋白质。因而可以降解新冠表面的蛋白的纤突蛋白,从而使之不能与人体内的病毒受体结合能力。同时病毒保护层破坏后,其核酸也外露出来,易被空气中或人们呼出的核酸酶降解,再与医用口罩联合使用,从而增加阻挡病毒与人体细胞受体结合,从而实现防控疫病的效果。
2.依据1所述,二层或三层纱布口罩也可以直接用医用二层或三层纱布代替口罩,使用时添在在医用外科口罩的外面。
