Abstract
本发明公开了一种保健食品,公开了一种抗衰老的皮肤修复的人参核桃肽组合物及其制备方法和应用,其人参核桃肽组合物由如下重量份数原料制成:人参肽20‑25份,核桃肽20‑30份,蜂王浆多肽15‑25份,枸杞糖肽7‑15份,胶原蛋白肽粉7‑10份、肉苁蓉提取物5‑9份,复合酵素6‑10份、食品级保鲜剂0.1‑0.5份。本发明以人参肽和核桃肽为主原料,配合蜂王浆多肽、枸杞糖肽、胶原蛋白肽粉和肉苁蓉提取物为辅助原料,且之间的配比合理,使得该人参核桃肽组合物的营养丰富、改善胃肠道功能、促进细胞生长的作用,共同作用可增强各成分的功效,起到调节免疫功能,延缓衰老,同时清除细胞中的脂褐质,清除自由基,延缓衰老之功效,并改善皮肤弹性的效果,对预防疲劳、亚健康症疗效显著。

技术领域
本发明涉及一种保健食品,尤其涉及一种抗衰老的皮肤修复的人参核桃肽组合物及其制备方法和应用。
背景技术
衰老是一个持续的变化过程,国际主流认为自由基损伤人体细胞,从而引起众多疾病,加速人体衰老。自由基从生成到淬灭,深刻影响着脂褐素的形成、线粒体DNA的突变、诱导细胞凋亡和蛋白质的合成,自由基与衰老间的关系,自由基可引起的疾病,从而提出了如何延缓衰老,为人类减缓自身衰老提供了一定的科学依据。抗衰老不是指让人增加寿命,而是对抗人体因为与外界的持续接触,包括呼吸(氧化反应)、外界污染、放射线照射等因素不断的在人体体内产生自由基所导致的折寿因素。
目前国内市场上销售的抗疲、抗衰老产品名目繁多,大致可以分为两类:一类是常规的功能性饮料,通过补充运动中所消耗的营养素来达到维持机体正常生理功能、缓解疲劳的目的,如红牛、力保健等属于此类,这类产品一般有效成分含量低,服用量大,时效性差;另一类是中草药制剂,即以中药材为主要原料制成的口服保健食品或药物,如专利号200410058060.5公开了一种抗疲劳保健食品胶囊,该专利将仙茅、淫羊藿、熟地黄、神骨脂、鹿鞭干燥粉碎后,混合复配,装入胶囊得到具有抗疲功效的保健食品胶囊;专利号201310279949.5公开了一种抗疲劳保健组合物及其制备方法,该专利以淫羊藿苷、黄芪甲苷、黄精提取物、西洋参提取物为原料,添加载体或稀释剂,制成具有改善疲劳,补气养阴,健脾润肺,提神醒脑作用的固体制剂或液体制剂。这些专利所涉及的产品配方和制作工艺各不相同,基本上都是以中草药材为主要原料,具有较好的抗疲劳保健作用,但是长期服用会产生一定的副作用,过多副作用会损害身体健康。因此,开发一种效果好、无副作用的药食同源产品,对内激活调节基因,对外降解生理毒垢,通过这种内外夹攻的根本性措施,使细胞充满活力,使生命健康幸福,达到抗衰老之效是社会的需求。
发明内容
本发明的目的是提供人参核桃肽组合物的制备方法及其在抗衰老方面的应用,充分利用人参和核桃中的有效成分,改良和优化现有的提取方法,提高提取率,利用人参核桃肽组合物的抗衰老活性将其应用到抗衰老保健食品中。
为了实现上述目的,本发明采用以下技术方案:
一种抗衰老的皮肤修复的人参核桃肽组合物,由如下重量份数原料制成:人参肽20-25份,核桃肽20-30份,蜂王浆多肽15-25份,枸杞糖肽7-15份,胶原蛋白肽粉7-10份、肉苁蓉提取物5-9份,复合酵素6-10份、食品级保鲜剂0.1-0.5份。
进一步的,所述蜂王浆多肽的制备步骤:
A、酶解:在蜂王浆中加入胰蛋白酶解液和0.9%的生理盐水,在35-40℃条件下酶解60min;
B、酶失活:将酶解后的液体在100℃沸水中煮沸2-3min,迅速降温至3-5℃;
C、离心:用去离子水与蜂王浆混合均匀后,在5000-8000rpm条件下,离心5min,得到蜂王浆上清液;
D、过滤:采用孔径为0.25-0.35μm的超滤膜过滤过滤;
E、冷冻干燥:采用真空冷冻干燥机进行处理,即可得到蜂王浆多肽。
本发明还提供了一种抗衰老的皮肤修复的人参核桃肽组合物的制备方法,所述制备方法包括以下步骤:
S1、按比例选取人参肽、核桃肽、肉苁蓉提取物,并将上述原料依次添加至混合研磨机内,对其进行混合研磨,待其混合搅拌15-25min后,即得到第一混合物;
S2、按比例将胶原蛋白肽粉置入搅拌器内,并在添加过程中进行混合搅拌,然后温度升高至36-44摄氏度,再次向其中添加枸杞糖肽,继续混合搅拌20-30min;
S3、在S2完成后,再次向搅拌器内添加蜂王浆多肽以及复合酵素,然后将温度升高至50-70摄氏度,再次混合搅拌6-10min后,继续向搅拌器内缓慢添加S1中得到的第一混合物,再次将温度降低至36-44摄氏度,缓慢搅拌90-120min,并在此期间按比例将食品级保鲜剂均匀添加至其中;
S4、上述完成后,取出,将其放置在45-55摄氏度的无菌环境中沸腾造粒干燥,获得所述的人参核桃肽组合物,即为具有抗衰老的皮肤修复的人参核桃肽组合物。
进一步的,在S1中的混合研磨的转速需始终保持在60-80转/min。
本发明还提供了一种抗衰老的皮肤修复的人参核桃肽组合物的应用,将抗衰老的皮肤修复的人参核桃肽组合物作为原料加入到抗衰老类化妆品、保健食品和保健饮品中。
本发明与现有技术相比,具有以下优点:(1)本发明以人参肽和核桃肽为主原料,配合蜂王浆多肽、枸杞糖肽、胶原蛋白肽粉和肉苁蓉提取物为辅助原料,且之间的配比合理,所使用的原料都是天然原料,使得该人参核桃肽组合物的营养丰富、改善胃肠道功能、美容养颜、促进细胞生长的作用,共同作用可增强各成分的功效,起到调节免疫功能,延缓衰老,同时清除细胞中的脂褐质,清除自由基,延缓衰老之功效,并改善皮肤弹性的效果,对治疗和预防疲劳、亚健康症疗效显著。
(2)本发明所采用的配方原料均为传统药食同源物品或新资源食品,不添加任何中草药、西药成分,无副作用,可长期服用,安全可靠。

为使本发明实现的技术手段、创作特征、达成目的与功效易于明白了解,下面结合具体实施方式,进一步阐述本发明。
实施例1
本发明公开了一种抗衰老的皮肤修复的人参核桃肽组合物,由如下重量份数原料制成:人参肽20份,核桃肽30份,蜂王浆多肽15份,枸杞糖肽7份,胶原蛋白肽粉7份、肉苁蓉提取物5份,复合酵素6份、食品级保鲜剂0.1份。
进一步的,所述蜂王浆多肽的制备步骤:
A、酶解:在蜂王浆中加入胰蛋白酶解液和0.9%的生理盐水,在35℃条件下酶解60min;
B、酶失活:将酶解后的液体在100℃沸水中煮沸2min,迅速降温至5℃;
C、离心:用去离子水与蜂王浆混合均匀后,在5000rpm条件下,离心5min,得到蜂王浆上清液;

S2、按比例将胶原蛋白肽粉置入搅拌器内,并在添加过程中进行混合搅拌,然后温度升高至36摄氏度,再次向其中添加枸杞糖肽,继续混合搅拌20min;
S3、在S2完成后,再次向搅拌器内添加蜂王浆多肽以及复合酵素,然后将温度升高至50摄氏度,再次混合搅拌6min后,继续向搅拌器内缓慢添加S1中得到的第一混合物,再次将温度降低至36摄氏度,缓慢搅拌90min,并在此期间按比例将食品级保鲜剂均匀添加至其中;
S4、上述完成后,取出,将其放置在45摄氏度的无菌环境中沸腾造粒干燥,获得所述的人参核桃肽组合物,即为具有抗衰老的皮肤修复的人参核桃肽组合物。
本发明还提供了一种抗衰老的皮肤修复的人参核桃肽组合物的应用,将抗衰老的皮肤修复的人参核桃肽组合物作为原料加入到抗衰老类化妆品、保健食品和保健饮品中。
实施例2
本发明公开了一种抗衰老的皮肤修复的人参核桃肽组合物,由如下重量份数原料制成:人参肽21份,核桃肽29份,蜂王浆多肽16份,枸杞糖肽8份,胶原蛋白肽粉8份、肉苁蓉提取物6份,复合酵素7份、食品级保鲜剂0.2份。
进一步的,所述蜂王浆多肽的制备步骤:
A、酶解:在蜂王浆中加入胰蛋白酶解液和0.9%的生理盐水,在36℃条件下酶解60min;
B、酶失活:将酶解后的液体在100℃沸水中煮沸2min,迅速降温至3℃;
C、离心:用去离子水与蜂王浆混合均匀后,在5500rpm条件下,离心5min,得到蜂王浆上清液;
D、过滤:采用孔径为0.3μm的超滤膜过滤过滤;
E、冷冻干燥:采用真空冷冻干燥机进行处理,即可得到蜂王浆多肽。
本发明还提供了一种抗衰老的皮肤修复的人参核桃肽组合物的制备方法,所述制备方法包括以下步骤:
S1、按比例选取人参肽、核桃肽、肉苁蓉提取物,并将上述原料依次添加至混合研磨机内,在65转/min对其进行混合研磨,待其混合搅拌18min后,即得到第一混合物;
S2、按比例将胶原蛋白肽粉置入搅拌器内,并在添加过程中进行混合搅拌,然后温度升高至38摄氏度,再次向其中添加枸杞糖肽,继续混合搅拌25min;
S3、在S2完成后,再次向搅拌器内添加蜂王浆多肽以及复合酵素,然后将温度升高至55摄氏度,再次混合搅拌7min后,继续向搅拌器内缓慢添加S1中得到的第一混合物,再次将温度降低至38摄氏度,缓慢搅拌100min,并在此期间按比例将食品级保鲜剂均匀添加至其中;
S4、上述完成后,取出,将其放置在48摄氏度的无菌环境中沸腾造粒干燥,获得所述的人参核桃肽组合物,即为具有抗衰老的皮肤修复的人参核桃肽组合物。
本发明还提供了一种抗衰老的皮肤修复的人参核桃肽组合物的应用,将抗衰老的皮肤修复的人参核桃肽组合物作为原料加入到抗衰老类化妆品、保健食品和保健饮品中。
实施例3
本发明公开了一种抗衰老的皮肤修复的人参核桃肽组合物,由如下重量份数原料制成:人参肽23份,核桃肽27份,蜂王浆多肽20份,枸杞糖肽10份,胶原蛋白肽粉8份、肉苁蓉提取物7份,复合酵素6份、食品级保鲜剂0.3份。
进一步的,所述蜂王浆多肽的制备步骤:
A、酶解:在蜂王浆中加入胰蛋白酶解液和0.9%的生理盐水,在37℃条件下酶解60min;
B、酶失活:将酶解后的液体在100℃沸水中煮沸3min,迅速降温至5℃;
C、离心:用去离子水与蜂王浆混合均匀后,在7000rpm条件下,离心5min,得到蜂王浆上清液;
D、过滤:采用孔径为0.30μm的超滤膜过滤过滤;
E、冷冻干燥:采用真空冷冻干燥机进行处理,即可得到蜂王浆多肽。
本发明还提供了一种抗衰老的皮肤修复的人参核桃肽组合物的制备方法,所述制备方法包括以下步骤:
S1、按比例选取人参肽、核桃肽、肉苁蓉提取物,并将上述原料依次添加至混合研磨机内,在70转/min对其进行混合研磨,待其混合搅拌22min后,即得到第一混合物;
S2、按比例将胶原蛋白肽粉置入搅拌器内,并在添加过程中进行混合搅拌,然后温度升高至40摄氏度,再次向其中添加枸杞糖肽,继续混合搅拌25min;
S3、在S2完成后,再次向搅拌器内添加蜂王浆多肽以及复合酵素,然后将温度升高至65摄氏度,再次混合搅拌10min后,继续向搅拌器内缓慢添加S1中得到的第一混合物,再次将温度降低至36-44摄氏度,缓慢搅拌110min,并在此期间按比例将食品级保鲜剂均匀添加至其中;
S4、上述完成后,取出,将其放置在50摄氏度的无菌环境中沸腾造粒干燥,获得所述的人参核桃肽组合物,即为具有抗衰老的皮肤修复的人参核桃肽组合物。
本发明还提供了一种抗衰老的皮肤修复的人参核桃肽组合物的应用,将抗衰老的皮肤修复的人参核桃肽组合物作为原料加入到抗衰老类化妆品、保健食品和保健饮品中。
实施例4
本发明公开了一种抗衰老的皮肤修复的人参核桃肽组合物,由如下重量份数原料制成:人参肽25份,核桃肽20份,蜂王浆多肽25份,枸杞糖肽15份,胶原蛋白肽粉10份、肉苁蓉提取物9份,复合酵素6份、食品级保鲜剂0.1份。
进一步的,所述蜂王浆多肽的制备步骤:
A、酶解:在蜂王浆中加入胰蛋白酶解液和0.9%的生理盐水,在40℃条件下酶解60min;
B、酶失活:将酶解后的液体在100℃沸水中煮沸3min,迅速降温至5℃;
C、离心:用去离子水与蜂王浆混合均匀后,在8000rpm条件下,离心5min,得到蜂王浆上清液;
D、过滤:采用孔径为0.35μm的超滤膜过滤过滤;
E、冷冻干燥:采用真空冷冻干燥机进行处理,即可得到蜂王浆多肽。
本发明还提供了一种抗衰老的皮肤修复的人参核桃肽组合物的制备方法,所述制备方法包括以下步骤:
S1、按比例选取人参肽、核桃肽、肉苁蓉提取物,并将上述原料依次添加至混合研磨机内,对其进行混合研磨,待其混合搅拌25min后,即得到第一混合物;
S2、按比例将胶原蛋白肽粉置入搅拌器内,在80转/min并在添加过程中进行混合搅拌,然后温度升高至44摄氏度,再次向其中添加枸杞糖肽,继续混合搅拌30min;
S3、在S2完成后,再次向搅拌器内添加蜂王浆多肽以及复合酵素,然后将温度升高至70摄氏度,再次混合搅拌10min后,继续向搅拌器内缓慢添加S1中得到的第一混合物,再次将温度降低至44摄氏度,缓慢搅拌120min,并在此期间按比例将食品级保鲜剂均匀添加至其中;
S4、上述完成后,取出,将其放置在55摄氏度的无菌环境中沸腾造粒干燥,获得所述的人参核桃肽组合物,即为具有抗衰老的皮肤修复的人参核桃肽组合物。
本发明还提供了一种抗衰老的皮肤修复的人参核桃肽组合物的应用,将抗衰老的皮肤修复的人参核桃肽组合物作为原料加入到抗衰老类化妆品、保健食品和保健饮品中。
应用试验:小鼠口服本发明胶囊对体内抗衰老指标影响的试验:
以实施例1所得的抗衰老的皮肤修复的人参核桃肽组合物(以下简称组合物)为受试药物。
选用雌性昆明小鼠50只,体重18-22克,随机分为5组,分别为空白对照组,模型对照组,受试药物高、中、低3个剂量组,每组10 只。高、中、低组分别按1.0g/kg、0.5g/kg、0.25g/kg的剂量灌胃给药(相当于人体每日用量3g的20倍、10倍和5倍,以60公斤体重计),每日1次,空白对照组和模型对照组给予等重量的生理盐水,连续给药(或生理盐水)30天。
给药的同时,按照文献方法建立衰老动物模型,模型对照组和给药组每天均于颈背部皮下注射1%D-半乳糖注射液120mg/kg,空白对照组每天在颈背部皮下注射等量的生理盐水,连续注射30天。
于停药后第2天,眼眶取血,分离血清,备用。采用南京建成生物工程研究所生产的试剂盒分别检测血清总超氧化物歧化酶(SOD)活力(羟胺法)、谷胱甘肽过氧化物酶(GSH-Px)含量(DTNB法)和丙二醛(MDA)含量(TBA法)。

从表1可以看出,D-半乳糖所致衰老小鼠模型对照组血清总SOD含量下降,明显低于空白对照组,给药各剂量组与模型对照组比较,给药各剂量组血清总SOD含量均增高,且有一定量效关系。说明本发明组合物具有提高D-半乳糖衰老小鼠血清总SOD酶活力的作用。

从表2可以看出,D-半乳糖所致衰老小鼠模型对照组血清MDA含量明显高于空白对照组,给药各剂量组的MDA含量与模型对照组比较,血清MDA含量均降低。说明本发明组合物能降低血清MDA含量。

从表3可以看出,D-半乳糖所致衰老小鼠模型对照组血清GSH-Px活性降低,明显低于空白对照组,给药各剂量组与模型对照组比较,中、高剂量组血清GSH-Px活性显著增高(P<0.05)。说明本发明组合物有显著提高D-半乳糖衰老小鼠血清GSH-Px活性的作用。
试验结果说明,本发明的抗衰老的皮肤修复的人参核桃肽组合物可以提高衰老小鼠血清中SOD活性和GSH-Px活性、降低衰老小鼠血清中MDA的含量,具有抗氧化、抗衰老的作用。
此外,应当理解,虽然本说明书按照实施方式加以描述,但并非每个实施方式仅包含一个独立的技术方案,说明书的这种叙述方式仅仅是为清楚起见,本领域技术人员应当将说明书作为一个整体,各实施例中的技术方案也可以经适当组合,形成本领域技术人员可以理解的其他实施方式。

1.一种抗衰老的皮肤修复的人参核桃肽组合物,其特征在于,由如下重量份数原料制成:人参肽20-25份,核桃肽20-30份,蜂王浆多肽15-25份,枸杞糖肽7-15份,胶原蛋白肽粉7-10份、肉苁蓉提取物5-9份,复合酵素6-10份、食品级保鲜剂0.1-0.5份。
2.根据权利要求1所述的一种抗衰老的皮肤修复的人参核桃肽组合物,其特征在于,所述蜂王浆多肽的制备步骤:
A、酶解:在蜂王浆中加入胰蛋白酶解液和0.9%的生理盐水,在35-40℃条件下酶解60min;
B、酶失活:将酶解后的液体在100℃沸水中煮沸2-3min,迅速降温至3-5℃;
C、离心:用去离子水与蜂王浆混合均匀后,在5000-8000rpm条件下,离心5min,得到蜂王浆上清液;

S3、在S2完成后,再次向搅拌器内添加蜂王浆多肽以及复合酵素,然后将温度升高至50-70摄氏度,再次混合搅拌6-10min后,继续向搅拌器内缓慢添加S1中得到的第一混合物,再次将温度降低至36-44摄氏度,缓慢搅拌90-120min,并在此期间按比例将食品级保鲜剂均匀添加至其中;
