Abstract
本发明涉及卫生消毒技术领域,公开了无机纳米消毒物质在制备抑制和/或杀灭病原体的组合物中的应用,所述无机纳米消毒物质包括氧化锌,所述组合物优选为啫喱。上述组合物安全、无刺激、无污染,适合用于空气病原体灭活,特别是针对飞沫、气溶胶粒子为载体的病毒的灭活,净化空气。杀菌试验结果表明,上述组合物,作用时间仅5min,对金黄色葡萄球菌(ATCC6538)的杀菌率即可大于90%,符合GB15979‑2002《一次性使用卫生用品卫生标准》的要求。流感病毒H1N1灭活试验结果显示,上述组合物作用20.0min,对流感病毒H1N1的平均灭活对数值可达到3.20(即平均灭活率为99.94%)。

技术领域
本发明涉及卫生消毒技术领域,具体涉及一种抑制和/或杀灭病原体组合物,特别是一种抑制和/或杀灭病原体啫喱,及其制备方法和应用。
背景技术
人类患有的许多重要传染病都是由病原体造成的,包括狂犬病、天花、脊髓灰质炎、肝炎、肺炎、黄热病、免疫缺陷和各种脑炎病等等,它们中的许多是高度传染性的且会产生急性不适,且常常是致命的,其它的例如风疹和巨细胞病毒会造成先天畸形。
2019年12月以来,部分医疗机构陆续出现不明原因肺炎病人,持续开展流感及相关疾病监测,发现多起病毒性肺炎病例,均诊断为病毒性肺炎/肺部感染。2020年2月10日,国际病毒分类委员会的冠状病毒研究小组(CSG)在生命科学论文预印本平台上发布了关于新型冠状病毒命名的最新论文,正式将新型冠状病毒命名为“SARS-CoV-2"。此外,世界卫生组织将由该病毒引发的疾病正式命名为COVID-19(Corona Virus Disease 2019)。SARS-CoV-2 主要的传播途径是呼吸道飞沫传播和接触传播,气溶胶和粪-口等传播途径待进一步明确。COVID-19的临床表现主要有发热、咳嗽、气促、肌肉疼痛、精神错乱、头痛、咽喉痛、流涕、胸痛、腹泻、恶心呕吐等等,在较严重病例中,感染可导致肺炎、急性呼吸窘迫综合征、多器官衰竭,可在短时间内病情加重,甚至死亡。
作为社会基本细胞的单位、家庭和个人,做好疫情防控工作尤为重要,日常尽量避免到人群聚集、通风不良的公共场所,勤洗手,室内应做好空气消毒,高频接触的物体表面,如门把手、桌椅、玩具等应进行清洁处理,必要时进行消毒处理。室内消毒喷雾剂在此次疫情期间以及之后的日常生活中是非常必要的用品。
传统消毒杀菌喷雾剂,一般都是以醇、油、汀、水剂为主,添加化学消毒杀菌成分,进行空气及接触物杀菌消毒。优势是方便、价格便宜。缺点是安全性差、腐蚀性强、持久性差、携带不方便。特别是面对气溶胶传播的病毒,传统消毒制剂必须大量喷洒,以在气溶胶表面形成一定浓度的消毒液才能杀死和灭活病毒,对于一般家庭、单位和个人,这种浓度大剂量的喷雾消毒几乎是不可能。
发明内容
为克服现有技术的不足,本发明提供无机纳米消毒物质在制备抑制和/或杀灭病原体的组合物中的应用,所述无机纳米消毒物质包括氧化锌,所述组合物为喷雾剂、粉末剂、溶液剂、凝胶剂、膏剂或啫喱。
具体地,上述组合物包含如下重量份数的原料组分:无机纳米消毒物质 0.1-5份,成膜剂1-5份,稀释剂50-100份。
具体地,上述无机纳米消毒物质为氧化锌以及,选自氧化银、二氧化钛、二氧化硅等中的一种或多种的组合;在本发明的一个实施例中,上述无机纳米消毒物质包括氧化锌和二氧化硅。
具体地,上述组合物中,无机纳米消毒物质的重量份数可以为0.1、0.2、 0.3、0.4、0.5、0.6、0.7、0.8、0.9、1、1.2、1.4、1.6、1.8、2、2.2、2.4、2.6、 2.8、3、3.2、3.4、3.6、3.8、4、4.2、4.4、4.6、4.8、5份。
在本发明的一个实施例中,上述无机纳米消毒物质包括氧化锌0.1-2份,二氧化硅0.1-2份。
具体地,上述无机纳米消毒物质的粒径可以为1-100nm,具体如1、2、3、4、5、6、7、8、9、10、15、20、25、30、35、40、45、50、55、60、65、 70、75、80、85、90、95、100nm。
具体地,上述成膜剂可以为聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物。
具体地,上述组合物中,成膜剂的重量份数可以为1、1.2、1.4、1.6、1.8、 2、2.2、2.4、2.6、2.8、3、3.2、3.4、3.6、3.8、4、4.2、4.4、4.6、4.8、5份。
具体地,上述稀释剂可以为去离子水、乙醇等。
具体地,上述组合物中,稀释剂的重量份数可以为50、55、60、65、70、 75、80、85、90、95或100份。
在本发明的一个实施方式中,上述组合物还包含赋香剂。
具体地,上述赋香剂可以为香精、香料,特别是植物香精,包括但不限于,柠檬、薄荷、尤加利、茶树、檀香、玫瑰、茉莉等植物精油中的一种或多种。植物精油在无机纳米消毒物质吸附病毒病菌后,具有一定抗菌、抗微生物及抗病毒的辅助效果。
具体地,上述组合物中,赋香剂的重量份数可以为0.05-5份,具体如0.05、 0.1、0.2、0.3、0.4、0.5、0.6、0.8、1、1.2、1.4、1.6、1.8、2、2.2、2.4、2.6、 2.8、3、3.2、3.4、3.6、3.8、4、4.2、4.4、4.6、4.8或5份。
在本发明的一个实施例中,上述赋香剂包括薄荷精油0.1-0.5份,尤加利精油0.1-0.5份,柠檬精油0.1-0.5份,乳香精油0.05-0.3份。
在本发明的一个实施方式中,上述组合物还包含消毒剂,以强化消毒效果,例如,含氯消毒剂(包括无机氯化合物(如次氯酸钠、次氯酸钙、氯化磷酸三钠)、有机氯化合物(如二氯异氰尿酸钠、三氯异氰尿酸、氯铵T等))、含碘消毒剂(如碘伏、氯己定碘)、醇类消毒剂(如乙醇、异丙醇、正丙醇、戊二醇等)、酚类消毒剂、季铵盐类消毒剂(如单链季铵盐消毒剂(例如苯扎氯铵、苯扎溴铵、十二烷基二甲基苯氧乙基溴化铵)、双链季铵盐消毒剂(例如双羟乙基双羟丙基硬脂基氯化铵、双癸基二甲基氯化铵、双十烷基二甲基溴化铵、溴化(双十二烷基二甲基)乙撑二铵)、复合季铵盐消毒剂(例如 FMB1210、Bardac 208M)、聚季铵盐消毒剂)、双胍类消毒剂(如氯己定)、过氧化物类消毒剂(如过氧化氢、过氧乙酸等)、醛类消毒剂(如甲醛、戊二醛等)、中草药消毒剂等中的一种或多种;在本发明的一个实施例中,上述消毒剂包括醇类消毒剂和双链季铵盐消毒剂。
具体地,上述组合物中,上述消毒剂的重量份数可以为0.5-25份,如0.5、 1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、 20、21、22、23、24或25份。
在本发明的一个实施例中,上述消毒剂包括:乙醇3-15份,戊二醇0.5-1 份,双羟乙基双羟丙基硬脂基氯化铵1-5份。
具体地,上述组合物还可包含其他添加剂,如保湿剂、赋脂剂、增稠剂、 pH调节剂、肤感调节剂、稳定剂、缓冲剂或中草药等中的一种或多种。
具体地,上述保湿剂可选自甘油、1,3-丁二醇、1,3-丙二醇、己二酸二异丁酯、丁二酸二异丁酯、木糖醇、山梨糖醇或海藻糖等中的一种或多种。
具体地,上述赋脂剂可选自十四酸异丙酯、透明质酸(优选低分子透明质酸)或PEG-7甘油椰油酸酯等中的一种或多种。
具体地,上述增稠剂可选自卡波姆940、卡波姆941、卡波姆Ultrez 20、羟甲基纤维素、羟丙基甲基纤维素、羟乙基纤维素、黄原胶或海藻酸钠等中的一种或多种。
具体地,上述pH调节剂可选自精氨酸、三乙醇胺或氢氧化钠等中的一种或多种。
具体地,上述肤感调节剂可选自硅石、聚甲基硅倍半氧烷、甲基丙烯酸甲酯交联聚合物、乙烯基聚二甲基硅氧烷/聚甲基硅氧烷硅倍半氧烷交联聚合物中的一种或多种。
本领域技术人员可根据实际情况选择上述添加剂的种类及其在组合物中的量。
在本发明的一个实施方式中,上述组合物包含以下重量份数的原料组分:无机纳米消毒物质0.1-5份,成膜剂1-5份,稀释剂50-100份,赋香剂0.05-5 份,消毒剂0.5-25份,赋脂剂0.01-0.05份。
在本发明的一个实施例中,上述组合物包含以下重量份数的原料组分:去离子水70-90份、乙醇3-15份、戊二醇0.5-1份,薄荷精油0.1-0.5份、尤加利精油0.1-0.5份、柠檬精油0.1-0.5份、乳香精油0.05-0.3份、透明质酸 0.01-0.05份、聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物1-5份、双羟乙基双羟丙基硬脂基氯化铵1-5份、氧化锌0.1-2份、二氧化硅0.1-2份。
在本发明的具体实施方式中,上述组合物包含:去离子水900g、乙醇30g、戊二醇10g,薄荷精油1g、尤加利精油1g、柠檬精油5g、乳香精油3g、透明质酸0.1g、聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物10g、双羟乙基双羟丙基硬脂基氯化铵1g、氧化锌1g、二氧化硅2g。
在本发明的具体实施方式中,上述组合物包含:去离子水700g、乙醇150g、戊二醇5g,薄荷精油5g、尤加利精油5g、柠檬精油5g、乳香精油0.5g、聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物50g、双羟乙基双羟丙基硬脂基氯化铵50g、氧化锌20g、二氧化硅20g。
在本发明的具体实施方式中,上述组合物包含:去离子水800g、乙醇100g、戊二醇10g,薄荷精油5g、尤加利精油3g、柠檬精油3g、乳香精油1g、透明质酸0.5g、聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物20g、双羟乙基双羟丙基硬脂基氯化铵20g、氧化锌10g、二氧化硅10g。
具体地,上述组合物可以为医学药物制剂涉及的喷雾剂、粉末剂、溶液剂、凝胶剂、膏剂、啫喱等任何形式,也可以被制成洗液(如洗手液、洗衣液)、棉签、湿纸巾、口罩、牙膏、隐形眼镜护理液等形式。
在本发明的一个实施方式中,上述组合物为啫喱。将其置于带泵头的容器中,其可通过泵头喷出,使具有多孔结构的多囊结构悬浮于空气中,吸附空气中的病毒、细菌和重金属的纳米颗粒,净化空气;也可喷洒于皮肤、毛发、衣物、家居以及医院、办公室等场所,实现抑制和杀灭病原体。
本发明所述的组合物可以为消毒产品,例如可以是医用消毒产品,也可以为日常家用消毒产品。
具体地,上述组合物可用于皮肤、毛发、衣物、家居等。
本发明还提供了一种抑制和/或杀灭病原体的组合物,所述的组合物包含如下重量份数的原料组分:无机纳米消毒物质0.1-5份,成膜剂1-5份,稀释剂50-100份,所述无机纳米消毒物质包括氧化锌。
本发明还提供一种上述组合物的制备方法,其包括将上述原料混合的步骤。
具体地,上述制备方法还包括将混合的原料进行均质处理的步骤。
具体地,上述制备方法还包括将混合的原料进行去气泡处理的步骤。
在本发明的一个实施方式中,上述制备方法包括如下步骤:
(1)将稀释剂与消毒剂、赋香剂混合,得到溶液;
(2)将成膜剂与无机纳米消毒物质混合,加入步骤(1)所得溶液中,搅拌,均质;
(3)将步骤(2)所得混合物进行真空去气泡处理。
具体地,上述制备方法还包括灌装的步骤。
本发明还提供上述组合物在制备抑制和/或杀灭病原体的产品中的应用。
在本发明的一个实施例中,上述病原体为病毒,例如流感病毒,特别是甲型流感病毒,如流感病毒H1N1。
在本发明的一个实施例中,上述病原体为细菌,特别是革兰氏阳性菌,例如金黄色葡萄球菌。
本发明的发明人以纳米微粒为载体,得到一种抑制和/或杀灭病原体组合物,特别是抑制和/或杀灭病原体啫喱。该组合物安全、无刺激、无污染,特别适合用于空气病原体灭活,特别是针对飞沫、气溶胶粒子为载体的病原体的灭活。该组合物可用于皮肤和毛发上,附着于其表面,形成保护膜,可有效预防接触性病毒,防阻细菌、病毒通过身体表面携带,例如,可以用于头发、鼻腔和眼角,能够有效预防病毒从眼睛(佩戴护目镜)和鼻腔(和口罩结合)侵入,并且不让病毒不在人体表面残留,还可以改善皮肤状况,净化毛孔和预防痤疮。该组合物也可用于外部衣物上,避免衣物因为静电效应吸附大量细菌和病毒,形成天然保护层。该组合物还可用于医院、公共场所、办公室、家居等易接触感染源地方,可以长时间抑菌杀毒(例如,可长达4 小时以上),比传统杀菌消毒有效时间长很多,其可能够有效提升公共卫生安全保护。该抑制和/或杀灭病毒组合物可制成啫喱,其可通过泵头喷出,使具有多孔结构的多囊结构悬浮于空气中,吸附空气中的病毒、细菌和重金属的纳米颗粒,净化空气。杀菌试验结果表明,本发明制备的组合物,作用时间仅5min,对金黄色葡萄球菌(ATCC6538)的杀菌率即可大于90%,符合 GB 15979-2002《一次性使用卫生用品卫生标准》的要求。流感病毒H1N1灭活试验结果显示,本发明制备的组合物作用20.0min,对流感病毒H1N1的平均灭活对数值可达到3.20(即平均灭活率为99.94%)。

下面将结合本发明实施例,对本发明的技术方案进行清楚、完整地描述,显然,所描述的实施例仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1
配方组成:
去离子水900g、乙醇30g、戊二醇10g,薄荷精油1g、尤加利精油1g、柠檬精油5g、乳香精油3g、透明质酸0.1g、聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物10g、双羟乙基双羟丙基硬脂基氯化铵1g、氧化锌1g、二氧化硅2g。
制备工艺如下:
(1)将去离子水加入消毒原料乙醇、戊二醇和双羟乙基双羟丙基硬脂基氯化铵、香精油薄荷精油、尤加利精油、柠檬精油和乳香精油,搅拌至完全溶解,得到A溶液;
(2)将透明质酸、聚合物材料聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物和纳米材料氧化锌、二氧化硅混合,加入A溶液中,搅拌、做均质;
(3)在化妆品反应锅中做真空去气泡处理;
(4)灌装于喷雾瓶中。
实施例2
配方组成:
去离子水700g、乙醇150g、戊二醇5g,薄荷精油5g、尤加利精油5g、柠檬精油5g、乳香精油0.5g、聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物50g、双羟乙基双羟丙基硬脂基氯化铵50g、氧化锌20g、二氧化硅20g。
制备工艺如下:
(1)将去离子水加入消毒原料乙醇、戊二醇和双羟乙基双羟丙基硬脂基氯化铵、香精油薄荷精油、尤加利精油、柠檬精油和乳香精油和溶剂,搅拌至完全溶解,得到A溶液;
(2)将聚合物材料聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物和纳米材料氧化锌、二氧化硅混合,加入A溶液中,搅拌、做均质;
(3)在化妆品反应锅中做真空去气泡处理;
(4)灌装于喷雾瓶中。
实施例3
配方组成:
去离子水800g、乙醇100g、戊二醇10g,薄荷精油5g、尤加利精油3g、柠檬精油3g、乳香精油1g、透明质酸0.5g、聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物20g、双羟乙基双羟丙基硬脂基氯化铵20g、氧化锌10g、二氧化硅10g。
制备工艺如下:
(1)将去离子水加入消毒原料乙醇、戊二醇和双羟乙基双羟丙基硬脂基氯化铵、香精油薄荷精油、尤加利精油、柠檬精油和乳香精油,搅拌至完全溶解,得到A溶液;
(2)将透明质酸、聚合物材料聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物和纳米材料氧化锌、二氧化硅混合,加入A溶液中,搅拌、做均质;
(3)在化妆品反应锅中做真空去气泡处理;
(4)灌装于喷雾瓶中。
实施例4:杀菌率检测
1、材料
(1)样品:实施例1所制备产品;
(2)试验菌株:金黄色葡萄球菌(ATCC 6538)(第5代),菌株来源于广东省微生物研究所;
(3)仪器设备:培养箱(GDCTC-139);
(4)培养基(批号):NA(1087361)。
2、方法
(1)中和剂鉴定试验:GB 15979-2002《一次性使用卫生用品卫生标准》附录C产品杀菌性能、抑菌性能与稳定性测试方法;
(2)杀菌试验:GB 15979-2002《一次性使用卫生用品卫生标准》附录C 产品杀菌性能、抑菌性能与稳定性测试方法,用该样品原液,作用时间5分钟,试验重复3次;
(3)中和剂:双倍D/E肉汤;
(4)检测环境:温度22℃,相对湿度52%;培养条件:37℃,48h;试验温度:20℃±1℃。

上述试验结果表明,用本发明上述实施例1制备的样品原样,作用时间 5min,对金黄色葡萄球菌(ATCC 6538)的杀菌率≥90%,符合GB 15979-2002 《一次性使用卫生用品卫生标准》的要求。
实施例5:流感病毒H1N1灭活试验
一、材料
1.试验用病毒株:流感病毒疫苗株H1N1(Influenza A virus H1N1, VR-1469)。
2.9d-11d龄白壳鸡胚,SPF级。
3.实施例1制备的样品原样。
4.中和剂:D/E中和肉汤。
5.有机干扰物质:3%牛血清白蛋白(BSA)。
6.培养箱(加湿)、lml无菌注射器、生物安全柜、照蛋器、石蜡及无菌器材等。
二、方法
1.检测依据:《消毒技术规范》(2002年版)第2.1.1.10项。
2.病毒悬液制备:用血凝滴度≥1:640的流感病毒H1N1悬液与3%牛血清白蛋白有机干扰物对倍稀释,备用。
3.中和剂鉴定试验:试验用实施例1制备的样品原样,作用20.0min,用生理盐水适宜稀释,各稀释度分别取样液0.2mL接种4枚鸡胚尿囊腔。试验温度为20℃恒温。试验重复3次。
4.病毒灭活试验:试验用实施例1制备的样品原样,作用20.0min,用生理盐水适宜稀释,各稀释度分别取样液0.2mL接种4枚鸡胚尿囊腔。试验温度为20℃恒温。试验重复3次。
5、检测环境温度:21.5℃-22.3℃;相对湿度:48%-49%。
三、结果
1.中和剂鉴定试验
经3次重复试验,在20℃恒温试验条件下,实施例1制备的样品原液中和剂鉴定结果为:第1组病毒平均滴度值为2.56,第2组病毒平均滴度值为 3.26。第3组、第4组和第5组病毒平均滴度值分别为6.39、6.44和6.44,第 6组为阴性对照组。具体结果如下表所示。

2.流感病毒灭活效果
经3次重复试验,在20℃恒温试验条件下,应用实施例1制备的样品原液,作用20.0min,对流感病毒H1N1的平均灭活对数值为3.20。具体结果如下表所示。

注:阴性对照收获尿囊液的血凝滴度为阴性;灭活率=[10[对照组]-10[试验组]]/10[对照组]×100%
四、结论
1.经3次重复试验,在20℃恒温试验条件下,所用D/E中和肉汤的中和剂溶液可有效中和实施例1制备的样品原液对流感病毒H1N1的残留作用,且中和剂和中和产物对流感病毒H1N1及鸡胚的生长基本无影响。
2.经3次重复试验,在20℃恒温试验条件下,应用实施例1制备的样品原液,作用20.0min,对流感病毒H1N1的平均灭活对数值为3.20(即平均灭活率为99.94%)。

Claims (10)
1.无机纳米消毒物质在制备抑制和/或杀灭病原体的组合物中的应用,所述无机纳米消毒物质包括氧化锌,所述组合物为喷雾剂、粉末剂、溶液剂、凝胶剂、膏剂或啫喱。
2.如权利要求1所述的应用,其特征在于,所述无机纳米消毒物质的粒径为1-100nm。
3.如权利要求1所述的应用,其特征在于,所述的组合物包含如下重量份数的原料组分:无机纳米消毒物质0.1-5份,成膜剂1-5份,稀释剂50-100份。
4.如权利要求3所述的应用,其特征在于,所述成膜剂为聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物。
5.如权利要求3所述的应用,其特征在于,所述组合物还包含赋香剂,所述赋香剂的重量份数为0.05-5份;
所述赋香剂优选为植物香精;
更优选地,所述赋香剂包括:薄荷精油0.1-0.5份,尤加利精油0.1-0.5份,柠檬精油0.1-0.5份,乳香精油0.05-0.3份。
6.如权利要求3所述的应用,其特征在于,所述组合物还包含消毒剂,所述消毒剂的重量份数为0.5-25份;
优选地,所述消毒剂包括醇类消毒剂和双链季铵盐消毒剂;
更优选地,所述消毒剂包括:乙醇3-15份,戊二醇0.5-1份,双羟乙基双羟丙基硬脂基氯化铵1-5份。
7.如权利要求3所述的应用,其特征在于,所述组合物还包含其他添加剂;
优选地,所述其他添加剂选自:保湿剂、赋脂剂、增稠剂、pH调节剂、肤感调节剂、稳定剂、缓冲剂和中草药中的一种或多种。
8.如权利要求1所述的应用,其特征在于,所述组合物包含以下重量份数的原料组分:去离子水70-90份、乙醇3-15份、戊二醇0.5-1份,薄荷精油0.1-0.5份、尤加利精油0.1-0.5份、柠檬精油0.1-0.5份、乳香精油0.05-0.3份、透明质酸0.01-0.05份、聚烯酸(酯)类/C10-30烷基丙烯酸酯交联聚合物1-5份、双羟乙基双羟丙基硬脂基氯化铵1-5份、氧化锌0.1-2份、二氧化硅0.1-2份。
9.如权利要求1-9任意一项所述的应用,其特征在于,所述的组合物用于皮肤、毛发、衣物或家居。
10.一种抑制和/或杀灭病原体的组合物,其特征在于,所述的组合物包含如下重量份数的原料组分:无机纳米消毒物质0.1-5份,成膜剂1-5份,稀释剂50-100份,所述无机纳米消毒物质包括氧化锌。
