First, animal experiment
The animal model and the normal group of the xerophthalmia are observed by the treatment effect of the medicines of the examples and the control group.
Molding: female C57BL/6 mice, 20, were divided into 10 groups of two: model group, treatment group and control group; injecting scopolamine hydrobromide solution subcutaneously to the mouse to inhibit the tear secretion, placing the mouse in front of a room-temperature blower to accelerate the tear evaporation for 15 days, and establishing a mouse dry eye model.

The treatment period is 4 weeks, and the mice in the model group, the treatment group and the control group are detected for the amount of lacrimal secretion and the trend of staining of the sodium keratectin.
The treatment results are as follows: the detection result of the lacrimal secretion of the mouse shows that the lacrimal secretion and the conjunctival goblet cell number of the xerophthalmia mouse obtained by molding are obviously reduced compared with the normal group of mice. After drug treatment, the tear secretion amount of the dry eye mice is remarkably increased in the treatment groups of examples 1 to 5 of the invention compared with the control group and the treatment groups of examples 6 to 8, and the treatment groups of the invention are better than those of examples 1 to 3, particularly the treatment group of example 1 has the most remarkable effect.
Staining mouse eyeball tissue sections by using a PAS staining method for conjunctival goblet cells, and counting the change trend of the total number of the goblet cells in each group: the number of indexes of the embodiments 1 to 5 of the present invention is also significantly increased, and the effect is most remarkable particularly in the embodiment 1. The tear secretion amount of the dry eye mice in the treatment groups of examples 6 to 8 is significantly lower than that of the treatment groups of examples 1 to 5, and is lower than that of the control group of the commercially available eye drops, particularly as the tear secretion amount of example 7 is the smallest; the tear secretion amount of the dry eye mice in example 6 is higher than that of the mice in examples 7 and 8, but the number of goblet cells is reduced compared with that of the mice in the normal group, and the treatment group is considered to have limited drug effect, and eye inflammation is formed in the treatment process, eye surface lesion is generated, and then the goblet cells are damaged.
Results of change in corneal fluorescein sodium staining: the corneal fluorescein sodium staining of the mice in the normal group is 0 grade, and the corneal fluorescein sodium staining of the mice in the dry eye group is 4 grade. The corneal fluorescein sodium staining of mice in each treatment group of the embodiments 1-5 of the invention is reduced compared with that of a xerophthalmia group, and the corneal fluorescein sodium staining is classified into 1 grade, 2 grade, 1 grade, 2 grade and 3 grade; the control group was graded at 3, and the treatment groups of examples 6-8 were all graded at 4.
Therefore, the natural herbal eye lotion in examples 1 to 5 has significant improvement and treatment effects on dry eyes and dry eyes caused by experimental dry eye syndrome, wherein the combination of active ingredients has significant effects on simultaneously improving tear secretion and the number of goblet cells of a mouse with dry eye syndrome.
Second, clinical observations
The natural herbal eye lotion of example 1 of the present invention was selected for use with commercially available eye drops, sodium hyaluronate eye drops, for patients with dry eye and asthenopia, and the effects and the experience of use were observed and compared. The patient with eye fatigue aims at volunteer patients who work for a long time on a desk, do irregular work and rest and have eye dryness symptoms for at least one week in the morning; the xerophthalmia patient is a volunteer patient with mild clinical diagnosis of xerophthalmia; the patients in the above disease groups are 10 persons each, half a man and half a woman, and 18-60 years old.
2.1 therapeutic effect standard:
the improvement is remarkable: the symptoms of dry eyes and unsmooth eyes basically disappear, and the eyes do not have ache, foreign body sensation and burning sensation; in at least 1 week, eyes are not dry and uncomfortable after the morning of normal work and regular work and rest every day.
