Description
技术领域
本发明涉及一种急性热应激细菌感染动物模型的构建方法及其应用。
背景技术
随着全球温度升高和高密度集约化养殖,热应激对畜禽健康和生产性能的负面影响是制约家禽生产的一个重要因素。尤其位处热带和亚热带地区的家禽养殖业,热应激事件愈加频繁,被认为是家禽生产中面临的突出问题[1],给畜牧业带来严重的经济损失。据报道,美国的家禽业每年因热应激造成的经济损失平均为1.28亿美元[2]。
家禽4周龄以后最适宜的生长温度是16-25℃,超过32℃时就会出现明显的热应激症状。研究表明,热应激严重影响着家禽的生产性能[3],造成生长和代谢紊乱,免疫抑制及肉品质下降[4]等。大量研究表明热应激可造成肉鸡采食量和日增重明显减少,并降低饲料利用率[5]。Liu等[6]检测热应激下猪空肠组织的基因表达谱,发现热应激影响猪免疫、内分泌、细胞损伤修复、信号转导相关基因的表达。在肉鸡中,热应激除了降低生长性能和高发病率和死亡率外,能抑制肠道营养转运蛋白对肠道活力和免疫力产生负面影响[7]。鸡大肠杆菌病是鸡的常见多发病,可导致感染鸡只发病、死亡和生产性能下降。目前,单独热应激或大肠杆菌感染对肉鸡肠道影响的报道较多,但是急性热应激后感染大肠杆菌鸡的肠道免疫功能研究目前还没有报道。建立一个急性热应激条件下鸡感染大肠杆菌病模型是非常有必要的。

本发明在热应激后采感染大肠杆菌的方式,通过双因素共同作用从而得到急性热应激大肠杆菌感染鸡动物模型,可用于急性热应激对鸡肠道免疫损伤及大肠杆菌病的发病机制,为急性热应激情况下鸡肠道免疫类药物的研发和生产提供科学依据。
本发明在诱导鸡急性热应激后,恢复7天,此时鸡免疫力最为低下,随后感染大肠杆菌造模,更贴合鸡的养殖中所处的环境,模拟效果更好,且更能契合生产需求。

本发明提供一种急性热应激细菌感染动物模型的构建方法。
在一些实施例中,所述构建方法包括以下步骤:
1)诱导急性热应激;
2)恢复;
3)感染细菌。
在有道急性热应激之前,动物于正常温度环境中进行适应性饲养。
本发明实施例中先热应激,在此基础上感染细菌造模更贴合动物的养殖中所处的环境,模拟效果更好,且更能契合生产需求。
本发明实施例中的恢复是指:动物诱导急性热应激后,在适应其生长的环境中养殖一段时间。在热应激的基础上恢复一段时间,此时动物整体免疫功能低,随后感染细菌模拟动物在热环境中更容易感染细菌或病毒性疾病的状态,更贴合实际养殖时动物的状态。

实施例1
一种急性热应激大肠杆菌感染肉鸡模型(heat stress+E.coli)的构建方法
设置正常对照组、大肠杆菌组(E.coli)和造模组,每组10只肉鸡。将肉鸡放在温度可控(25℃)的房间中,进行12小时的明暗循环(从7:00到19:00点亮)。鸡可以自由食用标准的实验饮食和饮用水。进行急性热应激造模时,将鸡放进一个大型恒温培养箱中进行热应激实验,鸡于41℃条件下热应激12小时后恢复7天,随后造模组和大肠杆菌组(E.coli)分别灌胃1mL E.coli O157:H7(1×109cfu/mL)。
E.coli感染后第4天,处死所有动物并分离小肠组织,测定血清中DAO、IgA、盲肠中大肠杆菌数量,TNF-α、IL-1β蛋白表达。

根据上述测定结果可知,模型组与正常对照组和大肠杆菌组的血清中DAO、IgA、盲肠中大肠杆菌数量,TNF-α、IL-1β蛋白表达均存在差异,本发明动物模型可用于急性热应激对鸡肠道免疫损伤及大肠杆菌病的发病机制,为急性热应激情况下鸡肠道免疫类药物的研发和生产提供科学依据。

Claims (2)
1.一种急性热应激细菌感染动物模型的构建方法,其特征在于,包括以下步骤:
1)诱导急性热应激;
2)恢复;
3)感染细菌;
所述诱导急性热应激包括:将动物于40-42℃饲养10-14h;
所述动物为鸡;
所述细菌为大肠杆菌;
所述恢复步骤包括:将急性热应激的动物于16-25℃饲养5~9d;
通过灌胃0.5-1.5mL细菌悬浮液感染细菌,所述细菌悬浮液中含有1×109-1.5×109cfu/mL的细菌;
感染后第4天,处死动物。
2.通过权利要求1所述的急性热应激细菌感染动物模型的构建方法构建的动物模型在药物筛选中的应用,所述药物用于治疗动物急性热应激情况下的细菌感染。
