背景技术
柠檬尤加利精油是从一种有柠檬香味的蓝桉树植物中提炼出来的。柠檬桉树油富含清洁香茅醛,是清洁表面和皮肤的理想精油。除了它的局部清洁的好处,柠檬桉树可以用来清洁和刷新空气。柠檬尤加利精油,是柠檬桉树的提取物,主要用于驱赶害虫,这些害虫包括蚊子、蜱虫和其他咬人的虫子。提取的柠檬尤加利精油经过精制,增加了其活性成分PMD的含量。市售的O柠檬尤加利精油E产品通常含有30%的OLE和20%的PMD。合成PMD是在实验室中制造的,它也被用作驱虫剂;市售的合成PMD产品的PMD浓度比商用柠檬尤加利精油产品低,含合成PMD的产品的PMD浓度约为10%。
脂质体(Liposomes)是由卵磷脂和神经酰胺等制得的脂质体(空心),具有的双分子层结构与皮肤细胞膜结构相同,对皮肤有优良的保湿作用,尤其是包敷了保湿物质如透明质酸、聚葡糖苷等的脂质体是更优秀的保湿性物质;纳米脂质体作为一种新产品,已经开始在多个领域被开发利用;纳米脂质体技术在化妆品、食品行业领域已取得了良好的效果。纳米脂质体是一种由两性类脂质形成的具有类似生物膜双分子层的微型囊泡,纳米脂质体具有良好的生物相容性,并且纳米脂质体不会对柠檬桉精油的主要活性成分造成破坏;纳米脂质体能够有效地保护包裹在内的柠檬桉精油,避免了精油的挥发。
为了提高制备柠檬尤加利精油纳米脂质体的稳定性,改善溶解性,为此我们提出一种柠檬尤加利精油纳米脂质体及制作工艺。

本发明的目的在于提供一种柠檬尤加利精油纳米脂质体及制作工艺,柠檬尤加利精油纳米脂质体的稳定性,改善溶解性。
为实现上述目的,本发明提供如下技术方案:一种柠檬尤加利精油纳米脂质体,由柠檬桉精油、大豆卵磷脂、胆固醇、乳化剂、表面活性剂、薄荷、薰衣草、稳定剂、抗氧化剂组成,其组分按重量百分比计为:柠檬桉精油5-25%、大豆卵磷脂20-40%、胆固醇10-20%、乳化剂4-12%、表面活性剂4-12%、薄荷2-4%、薰衣草2-4%、稳定剂2-4%、余量为抗氧化剂。

步骤二:有机相的制备:将柠檬桉精油、大豆卵磷脂、胆固醇、薄荷、薰衣草、抗氧化剂加入到去离子水中,充分溶解;
步骤三:水相的制备:将表面活性剂、稳定剂和乳化剂充分溶解在PBS缓冲液中;
步骤四:柠檬尤加利精油纳米脂质体悬浮液的制备:恒温状态下,用磁力搅拌器将步骤二中制备的有机相不断搅拌,然后用注射器按水相注入有机相中并继续搅拌,得到柠檬尤加利精油纳米脂质体悬浮液;

作为本发明的一种优选的技术方案,所述步骤二中,将柠檬桉精油、大豆卵磷脂、胆固醇、薄荷、薰衣草、抗氧化剂加入到36-50℃的去离子水中,充分溶解。
与现有技术相比,本发明的有益效果是:
(1)纳米脂质体的磷脂层将柠檬桉精油的活性物质包封,与外界环境隔开,有效地保护了柠檬桉精油中的活性物质,增加了制备柠檬尤加利精油纳米脂质体的稳定性;
(2)本发明还改善了制备柠檬尤加利精油纳米脂质体的溶解性;
(3)薄荷的加入进一步增加了抗菌效果。
