Abstract

一种测序文库的构建方法和得到的测序文库及测序方法，构建方法包括：将线性核酸分子环化形成环状核酸分子，滚环扩增得到多拷贝的长片段核酸分子，然后合成互补链得到双链的长片段核酸分子；与转座复合体混合孵育形成带有转座复合体的长片段核酸分子，与带有分子标签序列的固相载体混合孵育，使得分子标签序列与转座复合体上的转座子序列连接；将转座复合体上的转座酶从长片段核酸分子上释放出来，同时长片段核酸分子被打断成多个连接有转座子序列和分子标签序列的短片段核酸分子；对短片段核酸分子进行聚合酶链式扩增，得到测序文库。
