Claims (2)
- 標的核酸を増幅する方法であって、
A.クランプオリゴヌクレオチドおよび標的核酸を含む第一混合物において、第一および第二標的結合領域を標的核酸にアニールする工程であって、前記クランプオリゴヌクレオチドは3'−末端に前記第一標的結合領域を、かつ5'−末端に前記第二標的結合領域を、そして前記第一および第二標的結合領域の間にユーザー定義のテザー領域を含み、前記テザー領域が1つまたは複数の捕捉エレメントを含み、前記クランプオリゴヌクレオチドを前記標的核酸に沿って伸長し、第一核酸と前記標的核酸を含む第一二本鎖を生成する工程;
B.前記第一核酸の末端をライゲーションし環状核酸を生成し、任意には前記環状核酸を前記標的核酸から除去する工程;
C.前記テザー領域の少なくとも1つのプライマー結合部位を結合可能なプライマーを含む第二混合物において、前記環状核酸を前記テザー領域の前記捕捉エレメントを結合する捕捉手段を含む固体支持体上に捕捉し、そして前記プライマーを伸長して第二核酸と前記第一核酸を含む第二二本鎖核酸を生成する工程;および
D.前記第二核酸を前記第一核酸から解離し、それによって前記標的核酸を増幅する工程
を含む方法。 - 前記テザー領域がさらに1つまたは複数のプライマー部位、1つまたは複数のアンカー部位、1つまたは複数のバーコード部位、1つまたは複数のアダプター部位、1つまたは複数の検出部位、1つまたは複数の配列決定プライマー結合部位、またはそれらの組み合わせを含む請求項1記載の方法。
