Description

Medicine for treating osteoporosis and treatment method thereof
Technical Field
The invention relates to the technical field of traditional Chinese medicines, in particular to a medicine for treating osteoporosis and a treatment method thereof.
Background
Osteoporosis is a systemic or systemic skeletal disease characterized by a decrease in bone mass, destruction of the microstructure of bone tissue, and an increase in bone fragility, which leads to an increase in the risk of fracture. With the aging world population, the disease has an increased incidence and is associated with serious complications, which place enormous physical and mental burdens on society and patients, and thus, the world health organization classifies it as one of four major elderly diseases.
Osteoporosis is caused by various factors, and the basic pathological mechanism of the osteoporosis is that the coupling of bone absorption and bone formation in the bone metabolic process is defective, so that the calcium and phosphorus metabolism in a human body is unbalanced, and the bone density is gradually reduced to cause clinical symptoms. The currently commonly used medicines for treating osteoporosis mainly comprise calcium agents, active vitamin D, estrogen replacement therapy, parathyroid hormone, diphosphonates and the like. These drugs suffer from the following disadvantages: the use of calcium agents alone has physiological absorption limits; active vitamin D is easy to cause hypercalcemia and hypercalcemia; the estrogen replacement therapy is a common means for preventing and treating postmenopausal osteoporosis, has strong effects of inhibiting bone resorption and promoting bone formation, and has side effects of inducing risks of breast cancer, endometrial cancer and the like; parathyroid hormone, although it promotes bone formation, has no established influence on endocrine system and clinical use; bisphosphonates are currently used in a wide range of applications, interfering with various biochemical functions of osteoclasts to inhibit bone resorption, but also interfering with the normal bone remodeling process of the body, which has many adverse effects over long-term use. Therefore, the development of the traditional Chinese medicine preparation for improving the osteoporosis and having small toxic and side effects is significant.

Further, in the step (1), the time for the first reflux extraction is 2 hours, and the time for the second reflux extraction is 1 hour.
In the scheme, the medicinal materials are always in a high-temperature environment, the extraction time is too long, the decomposition and the damage of the effective components can be caused, the treatment effect is reduced, the extraction time is too short, and the effective components cannot be fully dissolved out.
Further, in the step (1), the pH value of the solution is adjusted to 4.
Further, in the step (2), the volume ratio of the first ethanol addition to the filter residue is 1-3:1, and the volume ratio of the second ethanol addition to the filter residue is 1-2: 1.
In the above scheme, the volume of the solvent is too large, so that the waste of the solvent is caused, the time for subsequent concentration and the energy consumption are prolonged, the volume of the solvent is too small, the dissolution rate of the active ingredients in the medicinal materials is poor, the active ingredients are reduced, and the treatment effect is reduced.
Further, in the step (2), the time for the first reflux extraction is 1.5h, and the time for the second reflux extraction is 1 h.
In the scheme, the medicinal materials are always in a high-temperature environment, the extraction time is too long, the decomposition and the damage of the effective components can be caused, the treatment effect is reduced, the extraction time is too short, and the effective components cannot be fully dissolved out.
Further, the pH value of the solution is adjusted to 4 in the step (2).
Further, the volume concentration of ethanol in the step (2) is 75%.
In the scheme, the polarities of the ethanol solutions with different volume concentrations are different, and when the ethanol with the volume concentration of 75% is selected as a solvent, effective ingredients can be fully dissolved out, so that the treatment effect of the medicine is improved.

Test examples
Randomly finding 140 patients with osteoporosis at the age of 40-60 years, measuring and recording the bone density of each patient, then randomly dividing the patients into 7 groups of 20 patients, wherein the average age of each group is 48-49 years, each group has comparability, respectively taking the medicaments in the 7 groups of the patients according to the examples 1-3 and the comparative examples 1-4 three times a day and 20g each time for 2 months, measuring and recording the bone density of each patient, taking the average bone density of all the patients in each group as the final result of the group, and calculating and recording the effective rate of each group of the patients, wherein the specific result is shown in the table 1-2.
Treatment criteria were:
the effect is shown: the patient has increased bone density but does not reach the normal range;
the method has the following advantages: the bone density of the patient is recovered to a normal value;
and (4) invalidation: the bone density of the patients has no obvious change.
Effective rate is 100% of effective people/total people
Table 1: patient treatment for bone density conditions before and after treatment
Figure BDA0002966759670000101
As can be seen from the results in the above table, the drugs in examples 1-3 have a certain therapeutic effect on osteoporosis, and can significantly improve the bone density of patients, while the drugs in comparative examples 1-4 also have a certain therapeutic effect, but have poor therapeutic effects.
Table 2: statistics of effective rate
Total number of people (human) Number of effective people (human) Number of valid persons (human) Invalid number of people (human) Effective rate (%)
Example 1 20 20 18 0 90%
Example 2 20 20 18 0 90%
Example 3 20 20 20 0 100%
Comparative example 1 20 13 2 7 10%
Comparative example 2 20 14 3 6 15%
Comparative example 3 20 14 2 6 10%
Comparative example 4 20 15 4 5 20%
As can be seen from the data in the table above, the effective rates in examples 1-3 are all higher than those in comparative examples 1-4, which proves that the drugs in examples 1-3 have better therapeutic effect on osteoporosis.
Comparing comparative example 1 with example 3, it can be seen that the treatment effect of the formula is reduced after the addition of rhizoma cibotii, fructus cnidii and safflower in the formula is cancelled, which proves that the formula of the application is an integral body, the medicinal materials in the formula have interaction, and the integral effect of the formula is reduced after one or more of the medicinal materials are lacked.
Comparing the comparative example 2 with the example 3, the rhizoma cibotii is replaced by the cortex acanthopanacis with similar effect, the notopterygium root is replaced by the radix angelicae pubescentis, the fructus cnidii is replaced by the hippocampus, and the herba epimedii is replaced by the herba cistanches, although the replaced medicine has similar effect with the medicine before replacement, the whole effect of the formula is still poor, and the interaction between different medicinal materials in the application is proved.
Comparing the comparative example 3 with the example 3, the proportion relation in the formula is changed, although the components in the formula are not changed, the whole effect of the formula is reduced after the dosage is changed, and the application proves that the formula is a whole and any factor is changed to influence the treatment effect.
Comparing comparative example 4 with example 3, in comparative example 4, the extraction method of the medicinal materials is changed, resulting in the decrease of the therapeutic effect of the drug, and it is also confirmed that the preparation method in the present application is different from the conventional preparation method, which can increase the extraction amount of the effective ingredients.
