Abstract

技术领域
本发明涉及土壤修复技术领域,具体涉及一种有机氯污染土壤的纳米零价铁-线虫协同修复技术。
背景技术
土壤有机氯污染是世界许多国家,尤其是发展中国家面临的重大环境问题之一。有机氯化物作为农药被广泛使用,多数化学性质稳定,不易降解,可通过食物链富集,产生严重的生态后果。有机氯污染物大多水溶性不高,使得土壤逐渐成为其重要的汇。例如五氯酚曾是全球使用最广泛的生物杀灭剂之一,它作为除草剂和防腐剂的功能逐渐被更加安全的新型农药所替代,但由于洪涝灾害及血吸虫病在传统流行病区域的反复爆发,五氯酚仍被大量用于防治血吸虫的中间寄主钉螺,在底泥中的浓度可高达48mg/kg。然而传统土壤修复技术如气相抽提、热处理、化学淋洗等不但需要消耗大量能源,而且容易破坏土壤原有结构,甚至引起二次污染。生物修复技术具有成本低、安全性高等优点,但由于受到生物代谢能力的限制,存在效率不高、周期长等不足。因此,亟需发展高效去除土壤中有机氯污染物的新技术。
近年来纳米零价铁在土壤污染修复方面展现出巨大潜力,具有比较面积大,反应活性高等优势,能够对有机氯污染物进行吸附、还原脱氯,已被越来越多地应用于有机氯污染土壤和地下水修复工程。但纳米零价铁在环境中不可避免地团聚、氧化失活,导致其在土壤修复中的实际利用率不高。在土壤及地下水修复应用中,往往需要将高达10g/L级别的纳米零价铁直接注入受污染区域。这不仅导致纳米零价铁土壤修复技术的成本高昂,而且进入环境的纳米材料会产生复杂的生物效应,尤其与目标污染物产生复合毒性,威胁生态安全和人类健康。因此,有必要寻求经济、高效的强化手段,在保障纳米零价铁土壤修复技术安全性的基础上降低成本。纳米-生物协同修复是一个新概念,旨在通过生物的生命活动强化纳米材料对污染物的降解效果,或通过纳米材料刺激生物对污染物的降解能力,有望形成低成本、高效率的联合修复技术。但由于对相关机理研究的匮乏,当前国内外仅存在极少数纳米-微生物或纳米-植物协同修复研究,纳米-土壤动物协同修复技术尚为空白。
线虫是一类典型的土壤动物,具有体型小、透明等优势,常作为指示生物被用于生态风险评估。更重要的是,它在有机质分解、植物营养矿化及养分循环过程中起重要作用,其生命活动能改变土壤中污染物的行为和归趋。已有证据显示线虫可通过取食、消化以及与细菌相互作用等活动改变土壤性质,促进土壤中菲的去除。最新研究还发现一定浓度的纳米材料不仅具有去除有机污染物的功能,还能诱导线虫抗氧化、解毒等生物学响应,增强线虫对有机污染物的抗性和降解。其中一些诱导产生的生物大分子如谷胱甘肽、半胱氨酸等不但可与铁/铁离子形成螯合作用,且因结构中具有巯基而呈还原性,已被证实能够在水介质中促进纳米零价铁对有机氯污染物的还原降解。因此,土壤中纳米零价铁与线虫的共存可能产生协同去除有机氯污染物的效果。考虑到线虫自身合成促降解化合物的能力较为有限,以人为添加促降解化合物为辅助,基于响应面法构建和优化一定组合和配比的纳米零价铁-线虫互作体系,有望在保证土壤生物安全的基础上协同去除土壤中有机污染物。这将填补纳米-动物协同修复技术的空白,为治理土壤污染提供绿色、经济、高效的新技术。
发明内容
本发明提供了一种有机氯污染土壤的纳米零价铁-线虫协同修复技术。与传统技术相比,该方法兼具较高的生态安全性以及优良的有机氯污染土壤修复效率;在筛选功能纳米材料的基础上,以人为添加促降解化合物为辅助,基于响应面法构建一定组合和配比的纳米零价铁-线虫互作体系,并优化参数以获得最优设计方案,可在保障修复效果的前提下精准、科学地减少纳米零价铁的用量,有效降低纳米土壤污染修复技术的成本。
具体技术方案如下:
一种有机氯污染土壤的纳米零价铁-线虫协同修复技术,包括以下步骤:
(1)修复材料的筛选:
参考环境浓度配置五氯酚污染土壤,将一定数量的线虫添加至土壤中,测定添加不同粒径和表面改性的纳米零价铁对土壤中线虫存活率的影响,以评估其生态安全性;另一方面,比较有无线虫条件下不同粒径和表面改性的纳米零价铁对土壤五氯酚的降解效果;综合考虑以上两方面,选取线虫存活率高、五氯酚降解效果好的一种纳米零价铁材料用于构建纳米-动物协同修复体系;
(2)促降解化合物的选取:
在水介质中比较谷胱甘肽和半胱氨酸两种生物还原性分子对纳米零价铁降解五氯酚的促进效果;选取效果好的一种作为强化纳米零价铁-线虫协同修复体系的添加剂;
(3)单因素实验:
在合理范围内对纳米零价铁、促降解化合物及线虫的添加量进行多水平取值,获得各组分不同添加量下五氯酚降解率变化曲线;在单因素实验结果的基础上筛选合理控制效能指标下的取值范围;
(4)响应面实验与建模:
根据单因素实验确定的取值范围,利用Design-Expert 12软件在Box-BehnkenDesign中设计实验方案(三因素三水平共17组),按照设计将不同浓度的纳米零价铁、促降解化合物拌入土壤;再将含有不同数量线虫的溶液加入土壤;测定不同处理组土壤中五氯酚的降解率;将测定结果通过二次多项回归拟合,得到纳米零价铁-促降解化合物-线虫协同去除土壤中五氯酚的响应面方程;
(5)确定最优组合:
在Design-Expert 12软件中进行因素优先级排序,计算在尽可能减少纳米零价铁用量的前提下获得最佳五氯酚去除效率的纳米零价铁-促降解化合物-线虫配比,以此作为最优方案,并通过重复实验比较真实值与预测理论值的差异,验证优化结果的有效性。
有益效果
与现有技术相比,本发明具有以下有益效果:
本发明在筛选功能纳米材料的基础上,以人为添加促降解化合物为辅助,基于响应面法构建一定组合和配比的纳米零价铁-线虫互作体系,并优化参数以获得最优设计方案,最终优选出20nm零价铁作为修复材料,半胱氨酸作为促进剂,确定了最优化的修复组合:纳米零价铁为2g/kg,半胱氨酸为5g/kg,线虫为18百万条/kg,得到了拟合度高的模型方程,在保障修复效果和生态安全性的前提下科学地减少纳米零价铁的用量,降低技术成本,为土壤纳米修复技术指明了新的发展方向。

具体实施方式
下面结合具体实施例对本发明作进一步描述,以下列举的仅是本发明的具体实施例,但本发明的保护范围不仅限于此。
实施例1
本发明方法步骤具体如下:
(1)修复材料的筛选:
首先配置人工添加五氯酚(50mg/kg)的水稻农田污染土壤作为对照组,以原始土壤作为空白组,将粒径为20、50、100nm的零价铁颗粒、3μm片状零价铁以及羧甲基纤维素(CMC)表面改性的纳米零价铁分别与污染土壤混合均匀,得到含纳米零价铁(5g/kg)的五氯酚污染土壤。各组设置三个重复,均取2 g土壤置于六孔板中。每孔加入600μL双蒸水(调节土壤含水率至最大持水量的50%),加入1mL OD600为1.1-1.2的大肠杆菌OP50(作为线虫的食物)溶液,再添加1mL含有4万条秀丽隐杆线虫(Caenorhabditis elegans)的培养液,此时土壤为淹水状态。在20±1℃黑暗条件下培养72h,采用土-琼脂法分离线虫,在普通光学显微镜下计数,计算存活率(图1)。为验证线虫的促降解效果,另一部分处理组仅添加大肠杆菌和不含线虫的培养液。收集各处理组土壤,采用2,4,6-三溴苯酚作为内标,经超声提取、K2CO3溶液纯化和乙酸酐酯化后,通过GC-MS测定土壤样品中五氯酚及其降解产物的含量(图2)。结果显示,无线虫存在时,不同粒径和表面改性的纳米零价铁对土壤中五氯酚的去除率较为接近,例如其中5g/kg 20nm零价铁对五氯酚的去除率约为36.2%,降解产物包括多种四氯酚、三氯酚、氯酚及四氯氰醌;而有线虫存在时,小粒径纳米零价铁的安全性和修复效果较好,其中20nm零价铁显著提高五氯酚污染土壤中线虫的存活率,并且线虫的存在促进了20nm零价铁对五氯酚的还原脱氯,去除率为74.4%。因此,本案例以20nm零价铁作为优选修复材料用于后续实验。
(2)促降解化合物的选取:
本案例设置5个处理组:配置10mg/L的五氯酚水溶液置于10mL棕色小瓶,分别向小瓶中加入50 mg谷胱甘肽、50mg半胱氨酸、50mg 20nm零价铁、50 mg谷胱甘肽+50mg 20nm零价铁、50mg半胱氨酸+50mg 20nm零价铁;反应24h后,以2,4,6-三溴苯酚作为内标,经超声提取、K2CO3溶液纯化和乙酸酐酯化后,采用高效气相色谱-质谱法测定溶液样品中五氯酚及其降解产物的浓度。结果表明半胱氨酸的促降解效果显著优于谷胱甘肽(图3),因此本案例选取半胱氨酸用于强化纳米零价铁-线虫协同修复体系。
(3)单因素实验:
本案例参考环境工程应用及前期实验参数设计单因素实验,确定纳米零价铁、半胱氨酸及线虫添加量的取值范围分别为0.1~5g/kg、0.1~5g/kg、1~20百万条/kg,测定各组分多水平下的五氯酚去除率(图4)。
(4)响应面实验与建模:
根据单因素实验结果,选择合适的纳米零价铁、半胱氨酸及线虫添加量,分别为0.5~5g/kg、1~5g/kg、10~20百万条/kg;以上述三个因素为自变量,以五氯酚去除率为响应值,采用三因素三水平实验进行五氯酚去除率的优化研究,利用Design-Expert 12软件在Box-Behnken中设计17组实验方案。按照设计将20nm零价铁、半胱氨酸拌入土壤;再将含有不同数量线虫的溶液加入土壤;测定72h后不同处理组土壤中五氯酚的降解率。

对表1中的数据进行二次多项回归拟合,最佳拟合模型为Quadratic model,响应面方程为:五氯酚去除率=79.54+29.3A+4.38B+1.99C-5.47AB+1.97AC+1.13BC-25.69A2-1.61B2-1.76C2。

表2模拟项Model的P值小于0.01,表明拟合的回归方程是极显著的;AB的决定系数R2为0.86,说明Quadratic model模型与真实值拟合程度较好,实验误差小。响应面图如图5所示:图5A为纳米零价铁和半胱氨酸的响应面分析,随着纳米零价铁和半胱氨酸添加量的增大,五氯酚的去除率提升,之后逐渐平缓;从响应面三维曲面坡度的缓急程度可以看出纳米零价铁对五氯酚去除率的影响要比半胱氨酸的显著;AB的P值小于0.05,表明纳米零价铁与半胱氨酸对五氯酚的降解存在显著的交互作用,其类型为协同。图5B为纳米零价铁和线虫的响应面分析,随着纳米零价铁和线虫添加量的增大,五氯酚的去除率提升,之后趋于平缓;从响应面三维曲面坡度的缓急程度可以看出纳米零价铁对五氯酚去除率的影响要比线虫的显著。图5C为半胱氨酸和线虫的响应面分析,随着半胱氨酸和线虫添加量的增大,五氯酚的去除率变化不显著。因此,从响应面的三维曲面可以分析出,三种组分对五氯酚去除率的影响顺序为纳米零价铁>半胱氨酸>线虫。
(5)确定最优组合:
由于纳米零价铁的市场价格远高于半胱氨酸和线虫,其用量是决定该土壤修复技术成本的最重要因素。通过Design-Expert 12软件对组合进行优化,将纳米零价铁用量设为最小化,五氯酚去除率设为最大化,半胱氨酸和线虫的添加量在实验范围内取值,得出最少纳米零价铁添加量下使五氯酚去除率最大化的纳米零价铁-半胱氨酸-线虫组合方案,即纳米零价铁为2g/kg,半胱氨酸为5g/kg,线虫为18百万条/kg时五氯酚去除率预测值为71.7%;在该实验条件下进行三组平行实验,得到此方案五氯酚去除率的模拟平均值是70.2%,与预测值非常接近,相对误差为1.5%,说明该二次多元回归模型较为可靠,优化结果具有实际指导意义,故确定为最优组合。与步骤(1)中结果对比可知,相同修复周期内,优化后的协同修复体系对土壤中五氯酚去除率约为单一使用纳米零价铁(5g/kg)的1.9倍,而该体系中纳米零价铁的用量仅为单一修复体系的40%。
以上实施例仅用以说明本发明的技术方案而非限制,本领域技术人员可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的宗旨和范围,其均应涵盖在本发明的权利要求范围当中,都属于本发明保护的范围。
Claims (4)
1.一种有机氯污染土壤的纳米零价铁-线虫协同修复技术,其特征在于,在筛选功能纳米材料的基础上,以人为添加促降解化合物为辅助,构建并优化纳米零价铁-线虫互作体系,具体包括以下步骤:
(1)修复材料的筛选:
参考环境浓度配置五氯酚污染土壤,将一定数量的线虫添加至土壤中,测定添加不同粒径和表面改性的纳米零价铁对土壤中线虫存活率的影响,以评估其生态安全性;另一方面,比较有无线虫条件下不同粒径和表面改性的纳米零价铁对土壤五氯酚的降解效果;综合考虑以上两方面,选取线虫存活率高、五氯酚降解效果好的一种纳米零价铁材料用于构建纳米-动物协同修复体系;
(2)促降解化合物的选取:
在水介质中比较谷胱甘肽和半胱氨酸两种生物还原性分子对纳米零价铁降解五氯酚的促进效果;选取效果好的一种作为强化纳米零价铁-线虫协同修复体系的添加剂;
(3)单因素实验:
在合理范围内对纳米零价铁、促降解化合物及线虫的添加量进行多水平取值,获得各组分不同添加量下五氯酚降解率变化曲线;在单因素实验结果的基础上筛选合理控制效能指标下的取值范围;
(4)响应面实验与建模:
根据单因素实验确定的取值范围,利用Design-Expert 12软件在Box-Behnken Design中设计实验方案(三因素三水平共17组),按照设计将不同浓度的纳米零价铁、促降解化合物拌入土壤;再将含有不同数量线虫的溶液加入土壤;测定不同处理组土壤中五氯酚的降解率;将测定结果通过二次多项回归拟合,得到纳米零价铁-促降解化合物-线虫协同去除土壤中五氯酚的响应面方程;
(5)确定最优组合:
在Design-Expert12软件中进行因素优先级排序,计算在尽可能减少纳米零价铁用量的前提下获得最佳五氯酚去除效率的纳米零价铁-促降解化合物-线虫配比,以此作为最优方案,并通过重复实验比较真实值与预测理论值的差异,验证优化结果的有效性。
2.根据权利要求1所述的一种有机氯污染土壤的纳米零价铁-线虫协同修复技术,其特征在于,步骤1中小粒径纳米零价铁的安全性和修复效果较好,优选20nm零价铁作为修复材料。
3.根据权利要求1所述的一种有机氯污染土壤的纳米零价铁-线虫协同修复技术,其特征在于,步骤2中选取半胱氨酸用于强化纳米零价铁-线虫协同修复体系。
4.根据权利要求1所述的一种有机氯污染土壤的纳米零价铁-线虫协同修复技术,其特征在于,步骤5中纳米零价铁-半胱氨酸-线虫最优组合方案为纳米零价铁为2g/kg,半胱氨酸为5g/kg,线虫为18百万条/kg。
