技术领域
本申请涉及化妆品技术领域,更具体地说,它涉及一种抗炎抗敏止痒植物组合物及其制备方法和应用。
背景技术
皮肤敏感已成为影响人类健康的社会问题,皮肤容易过敏的皮肤成为敏感性皮肤,导致皮肤敏感现象的原因有很多,像饮食结构的变化、环境污染以及工作压力的增大、化妆品使用不当(包括长期使用劣质化妆品、含有激素类、抗生素类化妆品等)等。常见的皮肤过敏症状有皮肤紧绷、发红、发痒,甚至有灼烧感等。目前市场上大部分护肤品的成分复杂,很多原料都是由化学合成的,一般都添加了香精、香料、防腐剂等,加重了皮肤的负担,让敏感皮肤更加不堪重负,更易出现红肿、瘙痒等症状。
治疗皮肤过敏导致的瘙痒一方面要考药物,另一方面要注意对皮肤的调养,克制挠抓,挠抓会导致越抓越痒的恶性循环,目前市场上针刺敏感性皮肤使用的产品较多,随着植物护肤理念的流行,添加天然植物成分的化妆品受到消费者的青睐,天然植物的抗敏功效研究大多基于过敏介质理论,即有效的抑制过敏反应发生机制各个缓解中过敏性介质的产生和释放,植物中的黄铜类物质、多糖、生物碱、α-亚麻酸等成分具有抗炎、抗敏作用,且天然植物成分毒副作用小,一般不会产生药物依赖性,更易被患者接受。
针对上述中的相关技术,发明人认为虽然天然植物成分具有抗敏、抗炎作用,但天然植物成分的止痒效果起效慢,皮肤过敏者在临睡前使用时,难以快速止痒,无法轻易入睡。
发明内容
为了加快天然植物成分的止痒起效速度,本申请提供一种抗炎抗敏止痒植物组合物及其制备方法和应用。
第一方面,本申请提供一种抗炎抗敏止痒植物组合物,采用如下的技术方案:
一种抗炎抗敏止痒植物组合物,包括以下重量份的组分:马齿笕10-20份、苦参5-15份、忍冬花8-24份、艾叶10-40份、紫苏12-24份、侧柏8-30份、鼠尾草10-27份、木瓜7-14份、厚朴20-30份。
通过采用上述技术方案,由于采用忍冬花、马齿笕、鼠尾草等多种天然植物成分制备植物组合物,由于马齿笕中的主要活性成分是马齿笕皂甙和黄酮,能抑制透明质酸酶的活性,减少炎症因子白介素的分泌,马齿笕中的左旋去甲基肾上腺素、多巴胺等儿茶酚胺类物质,具有抗过敏特性,能收缩皮肤毛细血管,缓解面部红血丝,且马齿笕对长期使用激素类化妆品产生的皮肤过敏有明显舒缓作用,马齿笕中还含有丰富的ω-3脂肪酸,作为一种不饱和脂肪酸,ω-3脂肪酸在人体皮肤中是重要的抗炎成分,能收缩血管平滑肌,抑制干燥引起的皮肤瘙痒,因此,马齿笕能起到较强的抗炎、止痛、止痒的效果;鼠尾草具有促进细胞再生、修复皮肤、抑菌杀菌、清洁止痒的效果,还能收缩毛孔,紧实肌肤,鼠尾草和马齿笕配合使用,能在较快时间内起到舒缓肌肤,缓解面部紧绷,并起到快速止痒效果;忍冬花即金银花,具有清热解毒、杀菌止痒的效果,鼠尾草与忍冬花能产生协同作用,能快速杀菌面部细菌,从而达到止痒效果;木瓜中含有的主要活性物质为齐墩果酸,具有抗炎抑菌、快速愈合伤口的作用,艾叶性温,能消肿修复、抗敏抗炎、安神助眠,与木瓜配合使用,具有较好的止痒效果;紫苏具有抗炎、抗过敏、安心凝神和镇静的效果,将紫苏、木瓜和鼠尾草三者配合,能快速杀菌止痒;侧柏能杀菌止痒、抗炎、抗敏、舒缓刺激,厚朴能除湿止痒、生肌敛疮、清热解毒、镇痛抗炎,苦参具有清热燥湿、止痒抑菌的效果,将多种组分配合作用,能快速起到抗炎、抗敏的效果,缓解肌肤敏感,减少红肿,修复受损肌肤,并且止痒速度快,皮肤过敏者在临睡前使用时,能快速润肤止痒,使患者轻易入睡。
优选的,所述组分的重量份如下:马齿笕13-18份、苦参8-12份、忍冬花12-20份、艾叶20-30份、紫苏15-21份、侧柏13-25份、鼠尾草14-23份、9-11份木瓜、23-28份厚朴。
通过采用上述技术方案,进一步精确植物组合物中各原料的用量,使制成的植物组合物止痒效果更佳迅速,舒缓肌肤效果更好。
第二方面,本申请提供一种抗炎抗敏止痒植物组合物的制备方法,采用如下的技术方案:
一种抗炎抗敏止痒植物组合物的制备方法,包括以下步骤:
(1)将苦参、侧柏和厚朴粉碎,加水,超声处理后,用乙醇提取,得到第一提取液;
(2)将马齿笕、忍冬花、艾叶、紫苏和鼠尾草混合后粉碎,用超临界二氧化碳进行提取,提取压力为34-40MPa,温度为35-40℃,二氧化碳流量为20-30L/h,得第二提取液;
(3)将木瓜粉碎,加入发酵菌,混合均匀,加入3-5倍重量的去离子水,在25-45℃下发酵,将发酵液置于95-100℃下,加入乙醇,密封提取,制得木瓜发酵液;
(4)将第一提取液、第二提取液和木瓜发酵液混合均匀后,离心,浓缩,制得抗炎抗敏止痒植物组合物。
通过采用上述技术方案,将苦参。侧柏和厚朴粉碎后进行超声处理,超声能进一步使有效成分溶出,将马齿笕、忍冬花等组分用超临界二氧化碳进行提取,使提取的成分更易吸收,从而使止痒起效快,将木瓜用发酵菌进行发酵,从而制得含有木瓜酵素、具有去角质功效的果酸和蛋白酶、具有抗氧化功能的多酚、黄酮和SOD酶等成分,果酸能软化皮肤角质,蛋白酶能剥脱角质层,抗氧化的多酚、黄酮和SOD酶能清除自由基,减少皮肤损伤。
优选的,所述步骤(1)中向乙醇中加入吐温80,吐温80的加入量为乙醇用量的20-30%。
通过采用上述技术方案,向作为提取剂的乙醇中加入吐温80表面活性剂,能相互聚集形成胶束,增加乙醇对苦参、侧柏和厚朴的浸提能力,还能降低乙醇与苦参、侧柏和厚朴之间的界面张力,促进润湿,在胶束作用下有效成分易被分解、提取。
优选的,所述步骤(3)中发酵菌为酵母菌,加入量为木瓜用量的20-30%,发酵时间为24-72h。
通过采用上述技术方案,使用酵母菌对木瓜进行发酵,发酵速度快。
优选的,还包括水解蛋壳膜12-20重量份、白芷6-10重量份、烟酰胺10-20重量份和光甘草定15-25重量份;将白芷和光甘草定粉碎后,与苦参、侧柏和厚朴的粉碎物混合提取,得第一提取液;将水解蛋壳膜与第一提取液和第二提取液混合。
通过采用上述技术方案,当皮肤过敏好转后,由于机体过敏反应中各种复杂化学反应导致黑色素细胞在局部积累过多,引起黑色素沉着,在植物组合物中添加水解蛋壳膜、烟酰胺、白芷和光甘草定组分,由于水解蛋壳膜中的氨基酸及多肽中存在丰富的硫氢基,可以抑制酪氨酸酶的活性,阻止黑色素的生成,同时小分子肽中还含有生物抑制因子,通过俘获酪氨酸酶中的铜离子,进而抑制酪氨酸酶的活性,从而降低黑色素的形成,有效提升肌肤的亮度,改善暗沉,多肽能有效促进细胞新陈代谢,延缓细胞衰老,水解蛋壳膜中富含的多种活性物质协同作用,能极大程度起到增加肌肤弹性、提亮肤色的作用,白芷能对黑色素产生过程中的酪氨酸酶具有抑制作用,且白芷具有抗敏、抗炎和止痒的效果;烟酰胺能快速止痒,修复肌肤,缓解皮肤不适,具有抗敏止痒、滋润保湿的作用,而光甘草定具有舒缓抗敏的作用,其具有较强的美白作用,能消除肌底的黑色素,将水解蛋壳膜与白芷、烟酰胺和光甘草定配合作用,能有效改善过敏后引起的黑色素沉着,防止肌肤暗沉,且还能进一步改善植物组合物的止痒、抗敏和抗炎效果。
第三方面,本申请提供一种抗炎抗敏止痒植物组合物的应用,采用如下的技术方案:一种抗炎抗敏止痒植物组合物在化妆品制备中的应用。
通过采用上述技术方案,将该抗炎抗敏止痒植物组合物应用于化妆品中,能有效缓解、修复或治疗皮肤敏感瘙痒或抑制皮肤表面细菌、真菌。
优选的,所述抗炎抗敏止痒的植物组合物在化妆品中的添加量为1-10%。
通过采用上述技术方案,抗炎抗敏止痒植物组合物在化妆品中添加量适量,具有较好的缓解肌肤,抗敏、抗炎和快速止痒的效果。
优选的,所述化妆品为修复乳,所述修复乳由包含以下重量份的组分制成:1.5-2%E-102乳化剂、2-3%鲸蜡硬脂醇、1.5-3%硬脂酸、2-7%矿油、1.5-2.5%葵酸甘油三脂、1.5-2.5%聚二甲基硅氧烷;B相7-9%甘油、0.2-0.3%黄原胶、0.05-0.06%EDTA二钠、1-10%抗炎抗敏止痒植物组合物、3-7%细辛挥发油、15-25%β-环糊精、0.6-1%改性玉米淀粉、1-2%淀粉、余量为去离子水。
通过采用上述技术方案,在晚上临睡前进行护肤时,因修复乳吸收较慢,若在床上侧身躺时,修复乳易粘附在枕头上,因此使用细辛挥发油、β-环糊精、玉米改性淀粉和淀粉作为渗透促进剂,加快修复乳的渗透速率,在临睡前使用时,能快速吸收,不易粘附在枕头上。
优选的,所述修复乳的制备方法如下:(1)将黄原胶分散在甘油中,升温至85℃,加入EDTA二钠,搅拌均匀;(2)将E-102乳化剂、鲸蜡硬脂醇、硬脂酸、矿油、葵酸甘油三脂和聚二甲基硅氧烷混合,搅拌均匀;(3)将步骤(1)和步骤(2)所得物搅拌均匀,冷却至50℃后,加入抗炎抗敏止痒植物组合物,调节pH至7;(4)将改性玉米淀粉、淀粉和去离子水混合均匀,糊化,加入细辛挥发油、β-环糊精和步骤(3)所得物,混合均匀后注入超临界二氧化碳,超临界二氧化碳注入流速为20L/h,在45℃和25MPa下混合均匀,制得修复乳。
通过采用上述技术方案,将各组分混合后,使用改性玉米淀粉、淀粉、细辛挥发油和β-环糊精的混合物将植物组合物、黄原胶等其余成分进行包裹,因改性玉米淀粉具有凝胶和增稠作用,与淀粉协同能生成弹性凝胶体,细辛挥发油中主要成分为甲基丁香油酚,具有解热、抗炎的作用,具有促进渗透的作用,在细辛挥发油和β-环糊精的作用下,该弹性凝胶体能快速携带植物组分物、E-102乳化剂等成分进入角质层,加快了有效成分的渗透系数,使修复乳具有快速止痒的效果,且能快速渗透。
综上所述,本申请具有以下有益效果:
1、由于本申请采用马齿笕、鼠尾草、木瓜、忍冬花等成分制备植物组合物,由于马齿笕具有抗敏特性,还能舒缓肌肤、抑制瘙痒、宁心安神,鼠尾草能促进细胞再生、抑菌杀菌、紧致皮肤,忍冬花、艾叶、厚朴、侧柏等能清热解毒抗炎、抗敏、舒缓刺激、止痒抑菌,马齿笕与鼠尾草协同作用,木瓜与艾叶协同作用,紫苏、鼠尾草和木瓜协同作用,均能有效加快植物组合物的止痒起效时间,使过敏者在临睡前使用时,较快起到止痒效果,防止过敏者因皮肤瘙痒而反复挠抓皮肤而难以入睡。
2、本发明的方法中优选向提取苦参、厚朴和侧柏的乙醇溶液中加入吐温80,能有效增加乙醇对苦参、侧柏和厚朴的浸提能力,降低乙醇与苦参、厚朴和侧柏之间的界面张力,促进润湿,增大提取率。
3、本申请中优选向植物组合物中加入水解蛋壳膜、白芷、光甘草定和烟酰胺,由于水解蛋壳膜具有阻止黑色素形成,提高肌肤亮度的效果,白芷具有抗菌止痒、抑制黑色素产生过程中酪氨酸酶活性的作用,烟酰胺能快速止痒、抑制黑色素合成,光甘草定具有较强的美白效果,能有效防止机体过敏后局部黑色素积累过多,引起黑色素沉着,肌肤色泽不均匀。
4、本申请中优选将植物组合物制备成修复乳,在修复乳中加入细辛挥发油、β-环糊精、改性玉米淀粉和淀粉,将植物组合物等成分在超临界二氧化碳的作用下,被改性玉米淀粉和淀粉形成的凝胶体包裹,凝胶体上含有细辛挥发油和β-环糊精,在β-环糊精喝细辛挥发油的作用下,凝胶体能快速被肌肤吸收,从而使修复乳快速渗透,在临睡前使用时,不易粘附在枕头上。
具体实施方式
水解蛋壳膜的制备例1
制备例1:将10kg鸡蛋壳粉碎,加入240L浓度为水,再加入24L浓度为36%的醋酸,在45℃下保温1.5h后常温静置24h,过滤,水洗,在60℃下干燥1.5h,切碎,加入10L浓度为20%的硫酸,加热至80℃回流提取5h,加入3kg碳酸钙,过滤,喷雾干燥,粉碎至1um,制得水解蛋壳膜。

(1)将苦参、侧柏和厚朴粉碎至400目,加三者总重3倍的水,超声处理后,用浓度为60%的乙醇提取,得到第一提取液,超声频率为25000Hz,时间为30min,乙醇的用量为苦参、侧柏和厚朴三者总重的3倍,向乙醇中加入吐温80,吐温80的加入量为乙醇用量的20%;
(2)将马齿笕、忍冬花、艾叶、紫苏、鼠尾草和木瓜混合后粉碎,用超临界二氧化碳进行提取,提取压力为34MPa,温度为40℃,二氧化碳流量为20L/h,提取10h,得第二提取液;
(3)将木瓜粉碎,加入发酵菌,混合均匀,加入5倍重量的去离子水,在25℃下发酵,发酵时间为72h,将发酵液置于95℃下,加入木瓜用量2倍的乙醇,密封提取,制得木瓜发酵液,发酵菌为酵母菌,加入量为木瓜用量的20%;
(4)将第一提取液、第二提取液和木瓜发酵液混合均匀后,离心,浓缩,制得抗炎抗敏止痒植物组合物。

实施例6:一种抗炎抗敏止痒植物组合物,其原料用量如表1所示,该抗炎抗敏止痒植物组合物的制备方法,包括以下步骤:
(1)将苦参、侧柏和厚朴粉碎至600目,加三者总重3.5倍的水,超声处理后,用浓度为60%的乙醇提取,得到第一提取液,超声频率为28000Hz,时间为25min,乙醇的用量为苦参、侧柏和厚朴三者总重的3倍,向乙醇中加入吐温80,吐温80的加入量为乙醇用量的25%;
(2)将马齿笕、忍冬花、艾叶、紫苏、鼠尾草和木瓜混合后粉碎,用超临界二氧化碳进行提取,提取压力为37MPa,温度为38℃,二氧化碳流量为25L/h,提取8h,得第二提取液;
(3)将木瓜粉碎,加入发酵菌,混合均匀,加入4倍重量的去离子水,在35℃下发酵,发酵时间为36h,将发酵液置于100℃下,加入木瓜用量2.5倍的乙醇,密封提取,制得木瓜发酵液,发酵菌为酵母菌,加入量为木瓜用量的25%;
(4)将第一提取液、第二提取液和木瓜发酵液混合均匀后,离心,浓缩,制得抗炎抗敏止痒植物组合物。
实施例7:一种抗炎抗敏止痒植物组合物,其原料用量如表1所示,该抗炎抗敏止痒植物组合物的制备方法,包括以下步骤:
(1)将苦参、侧柏和厚朴粉碎至800目,加三者总重4倍的水,超声处理后,用溶剂提取,得到第一提取液,超声频率为30000Hz,时间为20min,溶剂浓度为60%的乙醇,乙醇的用量为苦参、侧柏和厚朴三者总重的3倍,向乙醇中加入吐温80,吐温80的加入量为乙醇用量的30%;
(2)将马齿笕、忍冬花、艾叶、紫苏、鼠尾草和木瓜混合后粉碎,用超临界二氧化碳进行提取,提取压力为40MPa,温度为40℃,二氧化碳流量为30L/h,提取7h,得第二提取液;
(3)将木瓜粉碎,加入发酵菌,混合均匀,加入3倍重量的去离子水,在45℃下发酵,发酵时间为24h,将发酵液置于100℃下,加入木瓜用量3倍的乙醇,密封提取,制得木瓜发酵液,发酵菌为酵母菌,加入量为木瓜用量的30%;
(4)将第一提取液、第二提取液和木瓜发酵液混合均匀后,离心,浓缩,制得抗炎抗敏止痒植物组合物。
实施例8:一种抗炎抗敏止痒植物组合物,与实施例1的区别在于,还包括6kg水解蛋壳膜、4kg白芷、5kg烟酰胺和3kg光甘草定,水解蛋壳膜选自水解蛋壳膜的制备例1,该抗炎抗敏止痒植物组合物的制备方法包括以下步骤:
(1)将苦参、侧柏和厚朴粉碎至400目,将光甘草定和白芷粉碎,与苦参、侧柏和厚朴粉碎物混合,加混合物总重3倍的水,超声处理后,用溶剂提取,得到第一提取液,超声频率为25000Hz,时间为30min,溶剂为乙醇,溶剂的用量为苦参、侧柏、厚朴、白芷和光甘草定总重的3倍;
(2)将马齿笕、忍冬花、艾叶、紫苏、鼠尾草和木瓜混合后粉碎,用超临界二氧化碳进行提取,提取压力为34MPa,温度为40℃,二氧化碳流量为200L/h,提取10h,得第二提取液;(3)将木瓜粉碎,加入发酵菌,混合均匀,加入5倍重量的去离子水,在25℃下发酵,发酵时间为72h,将发酵液置于95℃下,加入木瓜用量2倍的乙醇,密封提取,制得木瓜发酵液,发酵菌为酵母菌,加入量为木瓜用量的20%;
(4)将第一提取液、第二提取液、水解蛋壳膜和木瓜发酵液混合均匀后,离心,浓缩,制得抗炎抗敏止痒植物组合物。
实施例9:一种抗炎抗敏止痒植物组合物,与实施例8的区别在于,水解蛋壳膜的用量为9kg,白芷的用量为7kg,烟酰胺的用量为8kg、光甘草定的用量为5kg。
实施例10:一种抗炎抗敏止痒植物组合物,与实施例8的区别在于,水解蛋壳膜的用量为12kg,白芷的用量为10kg,烟酰胺的用量为10kg、光甘草定的用量为8kg。

对比例2:一种抗炎抗敏止痒植物组合物,与实施例1的区别在于,未添加鼠尾草和忍冬花。
对比例3:一种抗炎抗敏止痒植物组合物,与实施例1的区别在于,未添加木瓜和艾叶。
对比例4:一种抗炎抗敏止痒植物组合物,与实施例1的区别在于,未添加紫苏、鼠尾草和木瓜。
对比例5:一种抗敏止痒植物组合物的制备方法,包括以下步骤:称取粉防己300g、铁冬青100g、黄芩100g、虎杖100g、光果甘草100g,混合、粉碎、过20目筛,加入12倍量浓度为60%的乙醇,回流提取3次,每次40min,合并滤液,减压浓缩,真空干燥,得提取物A;称取绿茶50g、七星剑50g、互叶白千层50g、母菊50g、积雪草50g、迷迭香50g,混合、粉碎、过20目筛,置于挥发油提取器中,加15倍水,常规方法提取3小时,放出精油,得提取物B;提取精油后,在绿茶、其详见、互叶白千层、母菊、积雪草和迷迭香提取器中加入乙醇,使提取液的乙醇浓度为50%,继续回流提取1h,过滤,减压浓缩,真空干燥,得提取物C,用吐温80/水(V/V=5:95)溶解提取物A、提取物B和提取物C,得一种抗敏止痒的植物组合物。
对比例6:一种可抗敏止痒修复的组合物,包括以重量百分比的物质:15%燕麦β-葡聚糖、4%膜夹黄芪提取物、2%防风根提取物、1.5%金盏花提取物、0.8%合欢花提取物、0.8%天麻根提取物、0.8%粉防己提取物、1.5%芽孢杆菌/大豆发酵提取物、4%1,2-己二醇,余量为丁二醇。
应用例
以实施例1制备的抗炎抗敏止痒植物组合物制备修复乳,修复乳的原料用量如表2所示,修复乳的制备方法如下:(1)将黄原胶分散在甘油中,升温至85℃,加入EDTA二钠,搅拌均匀;(2)将E-102乳化剂、鲸蜡硬脂醇、硬脂酸、矿油、葵酸甘油三脂和聚二甲基硅氧烷混合,升温至80℃,搅拌均匀;(3)将步骤(1)和步骤(2)所得物搅拌均匀,冷却至50℃后,加入抗炎抗敏止痒植物组合物,调节pH至7;(4)将改性玉米淀粉、淀粉和去离子水混合均匀,在90℃下糊化1h,加入细辛挥发油、β-环糊精和步骤(3)所得物,混合均匀后注入超临界二氧化碳,超临界二氧化碳注入流速为20L/h,在45℃和25MPa下混合均匀,制得修复乳。

应用例8:与应用例1的区别在于,修复乳的制备方法为:(1)将黄原胶分散在甘油中,升温至85℃,加入EDTA二钠,搅拌均匀;(2)将E-102乳化剂、鲸蜡硬脂醇、硬脂酸、矿油、葵酸甘油三脂和聚二甲基硅氧烷混合,升温至80℃,搅拌均匀;(3)将步骤(1)和步骤(2)所得物搅拌均匀,冷却至50℃后,加入抗炎抗敏止痒植物组合物,调节pH至7;(4)将改性玉米淀粉、淀粉和去离子水混合均匀,在90℃下糊化1h,加入细辛挥发油、β-环糊精和步骤(3)所得物,混合均匀,制得修复乳。
应用例9:与应用例1的区别在于,使用丁二醇替代细辛挥发油,使用聚乙二醇替代β-环糊精。
性能检测方法
一、抗炎抗敏止痒植物组合物的性能检测
将各实施例和各对比例制备的植物组合物按照以下方法进行检测,将检测结果记录于表3中:1、止痒效果:取230只体重为20g的健康小鼠,随机分为23组,每组雌雄各半,用1%硫化钠将小鼠背毛脱除,脱毛皮肤区面积为1.5cm×1.5cm,2天后用于实验;实验时22组动物背部脱毛区对应涂抹实施例1-16和对比例1-6制备的植物组合物,第23组作为对照组,涂抹蒸馏水,涂抹量为1mL/kg,每天一次,连续涂抹5天后,在末次涂抹后30min,对小鼠尾静脉注射右旋糖酐0.95mg/kg,以小鼠前爪搔头部、后抓搔躯干、嘴咬全身各部位作为瘙痒指征,记录30分钟内小鼠瘙痒次数。
2、止痒起效时间:取230只体重为300g的白色豚鼠,通脱毛剂将右后足脱毛,然后随机分为23组,用细砂纸轻轻摩擦豚鼠的右后足脱毛皮肤使之发红,但不出血,1-22组豚鼠对应涂抹实施例1-16和对比例1-6制备的植物组合物,第23组涂抹醋酸曲安奈德尿素软膏,每组涂抹1mL/kg,当豚鼠出现瘙痒反应后,再向脱毛区涂抹植物组合物0.2mL,记录豚鼠从涂抹植物组合物至停止舔咬右后足创面部位的时间,此时间为植物组合物抗敏止痒起效时间;3、透明质酸酶抑制率:将实施例1-16和对比例1-6制备的植物组合物分别配制成浓度为50mg/mL的水溶液作为样品液,取0.1mL浓度为0.25mmoL/L的氯化钙溶液和0.5mL透明质酸酶液,37℃保温培养20min,加入样品液0.5mL,继续37℃保温培养20min,加入0,5mL透明质酸钠液,37℃保温30min,常温放置5min,加入0.1mL浓度为0.4moL/L的氢氧化钠溶液和0.5mL乙酰丙酮溶液,置于沸水浴中加热15min后立即用冰水冷却5min,加入埃尔利希试剂1mL,并用3mL无水乙醇进行稀释,放置20min显色,用分光光度计测定其吸光度值,根据以下公式计算样品对透明质酸酶的抑制率:透明质酸酶抑制率(%)=[(A-B)-(C-D)]/(A-B),式中A-对照溶液ABS值(用醋酸缓冲液替代样品溶液)、B-对照空白溶液ABS值(用醋酸缓冲液替代样品溶液及酶液)、C-试样溶液ABS值、D-试样空白溶液ABS值(用醋酸缓冲液替代酶液)。
4、美白功效检测:选取110名年龄在22-54岁具有敏感肌肤的志愿者,随机分为22组,每组5人,22组志愿者对应涂抹实施例和对比例制备的植物组合物,每天洁面后涂抹,每日早晚各1次,使用前1天和使用后28天使用MEXAMETER MX18皮肤色素仪测试皮肤黑红色素含量,连续测试3次,取平均值。

由表3中数据可以看出,实施例1-7制备的植物组合物具有较强的止痒效果和较快的止痒速度,且透明质酸酶的抑制率高,从而减少透明质酸的分解,减少组胺及炎症因子的释放量,从而起到抗敏、抗炎的效果。
实施例8-10中添加了水解蛋壳膜、白芷、光甘草定和烟酰胺,与实施例1-7相比,实施例8-10制备的植物组合物的止痒效果和止痒起效时间得到进一步的改善,且涂抹实施例8-10制备的植物组合物后,黑色素含量降低,能有效抑制黑色素的合成和沉积,从而防止过敏后皮肤黑色素沉降而导致局部皮肤变黑。
实施例11与实施例8-10相比,未添加水解蛋壳膜,与实施例8-10相比,止痒效果和止痒起效时间相差不大,但透明质酸酶的抑制率明显增大,黑色素含量与使用前相比明显降低,说明水解蛋壳膜能明显增大植物组合物的抗敏、抗炎效果,并提高植物组合物抑制黑色素的能力。
实施例12中未添加白芷,与实施例8-10相比,对小鼠的止痒效果好,且起效时间快,能有效降低黑色素的含量,具有美白效果。
实施例13中未添加白芷和水解蛋壳膜,与实施例8-10相比,止痒效果下降,止痒起效时间变慢,且黑色素含量使用前与使用后变化无实施例8-10明显,抑制黑色素效果不及实施例8-10。
实施例14中未添加光甘草定和烟酰胺,实施例14制备的植物组合物与实施例8-10相比,止痒速度下降,且止痒效果减弱,黑色素含量下降率与实施例8-10相比有所降低,对于肌肤的美白效果减弱。
实施例15中未添加水解蛋壳膜和烟酰胺,由表3中数据可知,实施例15制备的植物组合物的止痒效果下降,止痒起效时间长,使用后黑色素含量与使用前相比下降较少,抑制黑色素合成的效果减弱。
实施例16中未添加水解蛋壳膜和光甘草定,表3中显示,植物组合物的止痒效果和止痒时间与实施例8-10相比无较大变化,但黑色素含量在使用前后的改变量较小,黑色素抑制效果减弱。
对比例1-4制备的植物组合物止痒效果下降,止痒起效时间长,且抑制黑色素的效果较弱。
对比例5和对比例6为现有技术制备的植物组合物,止痒起效时间较长,止痒效果差,且对肌肤中黑色素含量抑制效果不及本申请实施例8-10。
二、应用例1-9制备的修复乳的性能检测
将应用例1-9制备的修复乳按照以下方法进行检测,将检测结果记录于表4中:
渗透性能检测:取200g的雄性大鼠,将大鼠断颈处死后,用理发器剃去腹部的鼠毛,剪下腹部皮肤,出去皮下脂肪层、血管和残留物,用生理盐水洗净,并检测其完整性,用生理盐水漂洗后,置于-10℃冰箱中冷冻保存备用;采用卧式Franz扩散装置进行体外透皮试验,将大鼠皮肤夹在供给室和接受室之间,皮肤角质层面向供给室,有效渗透面积为1.14cm2,供给室内加入4.5mL样品(各实施例和对比例制备的植物组合物),接受室内加入4.5mL磷酸盐缓冲液(PBS),且供给室和接收室的液面完全没过皮肤的最上缘,在恒温32℃下保持恒速300r/min,不断搅拌,于1min、2min和5min用移液器吸取接收室的溶液0.1ml作为测试液,用紫外分光光度计测试水溶性活性物含量,每个人样品的透皮试验重复3次,累积透皮量的计算公式如下:式中,Q:累积渗透量;S:有效扩散面积;V:接收室内接收液体积;Ci:第1次至上次取样时接收液中样品溶度;Cn:该次取样时接收液中样品浓度。
表4应用例1-9制备的修复乳渗透量测试
结合表2和表4中数据,以及应用例8-9可知,使用细辛挥发油、β-环糊精、改性玉米淀粉、淀粉等组分制成的修复乳应用例1-3能快速渗透至角质层,使面部在涂抹修复乳后快速恢复干爽,不粘腻。
而应用例4中未添加细辛挥发油,应用例5中未添加β-环糊精,对比例6中未添加细辛挥发油和β-环糊精,由检测结果可知,应用例6修复乳的渗透效果对应用例4-5差,而应用例4-5与应用例1-3相比,渗透速度慢,说明细辛挥发油和β-环糊精具有较好的协同作用,能加速有效组分的渗透,使脸部保持干爽。
应用例7中未添加玉米淀粉和淀粉,难以形成渗透包裹物,渗透速度下降,应用例8中未使用超临界二氧化碳超进行包裹制作,对比例9中使用丁二醇替代细辛挥发油,使用聚乙二醇替代β-环糊精,对比例8-9制备的修复乳渗透速度慢。

Claims (10)
1.一种抗炎抗敏止痒植物组合物,其特征在于,包括以下重量份的组分:马齿笕10-20份、苦参5-15份、忍冬花8-24份、艾叶10-40份、紫苏12-24份、侧柏8-30份、鼠尾草10-27份、木瓜7-14份、厚朴20-30份。
2.根据权利要求1所述的抗炎抗敏止痒的植物组合物,其特征在于:所述组分的重量份如下:马齿笕13-18份、苦参8-12份、忍冬花12-20份、艾叶20-30份、紫苏15-21份、侧柏13-25份、鼠尾草14-23份、9-11份木瓜、23-28份厚朴。
3.权利要求1-2任一项所述的抗炎抗敏止痒植物组合物的制备方法,其特征在于,包括以下步骤:
(1)将苦参、侧柏和厚朴粉碎,加水,超声处理后,用乙醇提取,得到第一提取液;
(2)将马齿笕、忍冬花、艾叶、紫苏和鼠尾草混合后粉碎,用超临界二氧化碳进行提取,提取压力为34-40MPa,温度为35-40℃,二氧化碳流量为20-30L/h,得第二提取液;
(3)将木瓜粉碎,加入发酵菌,混合均匀,加入3-5倍重量的去离子水,在25-45℃下发酵,将发酵液置于95-100℃下,加入乙醇,密封提取,制得木瓜发酵液;
(4)将第一提取液、第二提取液和木瓜发酵液混合均匀后,离心,浓缩,制得抗炎抗敏止痒植物组合物。
4.根据权利要求3所述的抗炎抗敏止痒植物组合物的制备方法,其特征在于,所述步骤(1)中向乙醇中加入吐温80,吐温80的加入量为乙醇用量的20-30%。
5.根据权利要求3所述的抗炎抗敏止痒植物组合物的制备方法,其特征在于,所述步骤(3)中发酵菌为酵母菌,加入量为木瓜用量的20-30%,发酵时间为24-72h。
6.根据权利要求3所述的抗炎抗敏止痒植物组合物的制备方法,其特征在于,还包括水解蛋壳膜12-20重量份、白芷6-10重量份、烟酰胺10-20重量份和光甘草定15-25重量份;将白芷和光甘草定粉碎后,与苦参、侧柏和厚朴的粉碎物混合提取,得第一提取液;将水解蛋壳膜与第一提取液和第二提取液混合。
7.权利要求1-6任一项所述的抗炎抗敏止痒植物组合物在化妆品制备中的应用。
8.根据权利要求7所述的抗炎抗敏止痒的植物组合物的应用,其特征在于,所述抗炎抗敏止痒的植物组合物在化妆品中的添加量为1-10%。
9.根据权利要求8所述的抗炎抗敏止痒的植物组合物的应用,其特征在于,所述化妆品为修复乳,所述修复乳由包含以下重量份的组分制成: 1.5-2% E-102乳化剂、2-3%鲸蜡硬脂醇、1.5-3%硬脂酸、2-7%矿油、1.5-2.5%葵酸甘油三脂、1.5-2.5%聚二甲基硅氧烷;B相7-9%甘油、0.2-0.3%黄原胶、0.05-0.06% EDTA二钠、1-10%抗炎抗敏止痒植物组合物、3-7%细辛挥发油、15-25%β-环糊精、0.6-1%改性玉米淀粉、1-2%淀粉、余量为去离子水。
10.根据权利要求9所述的抗炎抗敏止痒的植物组合物的应用,其特征在于,所述修复乳的制备方法如下:(1)将黄原胶分散在甘油中,升温至85℃,加入EDTA二钠,搅拌均匀;(2)将E-102乳化剂、鲸蜡硬脂醇、硬脂酸、矿油、葵酸甘油三脂和聚二甲基硅氧烷混合,搅拌均匀;(3)将步骤(1)和步骤(2)所得物搅拌均匀,冷却至50℃后,加入抗炎抗敏止痒植物组合物,调节pH至7;(4)将改性玉米淀粉、淀粉和去离子水混合均匀,糊化,加入细辛挥发油、β-环糊精和步骤(3)所得物,混合均匀后注入超临界二氧化碳,超临界二氧化碳注入流速为20L/h,在45℃和25MPa下混合均匀,制得修复乳。
