Abstract
本发明涉及免疫细胞治疗技术领域,特别涉及一种淋巴细胞及其培养体系和培养方法。该培养体系包括灭活的基因工程细胞、IL‑2、FBS、抗CD3抗体和淋巴细胞基础培养基;基因工程细胞为表达IL‑21、IL‑12、CD80和CD206的细胞。将该培养体系与单核细胞进行共培养,可以得到大量的免疫细胞,其中NK细胞为主要成分。这些免疫细胞不但可以高效杀伤肿瘤细胞,而且可对CD206阳性的肿瘤相关巨噬细胞进行高效去除。

技术领域
本发明涉及免疫细胞治疗技术领域,特别涉及一种淋巴细胞及其培养体系和培养方法。
背景技术
NK细胞(自然杀伤细胞,Natural Killer Cell)行使着连接获得性免疫和固有免疫桥梁的作用。一方面,当机体发生感染及创伤时,NK细胞以防御者的身份通过非肽-MHC的识别方式快速、广泛、特异地识别抗原,及时地清除病原微生物、变异细胞,发挥固有免疫的作用。另一方面NK细胞被认为也部分参与了适应性免疫应答,能够影响αβT细胞和B细胞的效应功能。
在肿瘤的发生发展过程中,NK细胞既可以通过活化性受体直接识别肿瘤细胞并被活化,也可以被辅助细胞(单核细胞,巨噬细胞,树突状细胞等)活化。这些辅助细胞通过其模式识别受体来应答内外环境的变化,再通过分泌多种可溶性因子或直接接触的方式将信号传导给NK细胞。在人类,已经证实存在的可溶性因子有IL12、IL-18、typeI IFN、TNF-α等;直接接触的分子有GITRL/GITR、CD48/2B4、MICA、MICB、ULBP1-ULBP3/NKG2D、AICL/NKp80等。基于以上原理在体外扩增活化的NK细胞对肿瘤细胞显示出良好的杀伤活性,并应用于肿瘤生物治疗中。
目前制约NK细胞的临床应用的主要障碍之一就是难以得到足够数量的NK细胞。如何在体外实现NK细胞大规模扩增是目前NK细胞治疗的关键问题。外周血中NK细胞仅占很小的部分。而肿瘤病人的外周血中往往NK细胞的数量和活性有明显的下降。不同人NK细胞的性质有很大差异。寻找高效的个性化的NK细胞大规模扩增方法对NK细胞的临床应用起着关键性作用。
近年,人工抗原提呈细胞(使用基因工程技术制作的滋养层细胞)越来越多的用于NK细胞的体外扩增。比如把膜结合IL15和4-1BBL导入K562细胞,这种方法得到的人工抗原提呈细胞可以将NK细胞在21天平均扩增277倍。把MICA和4-1BBL导入K562细胞,并加以IL15刺激NK细胞可在21天平均扩增550倍。把mIL21、4-1BBL、CD64、CD86、tCD19导入K562细胞,这种方法得到的人工抗原提呈细胞可以将NK细胞3周平均扩增一万倍以上。
CN 103484429 A公开了一种NK细胞的高效制备方法,即通过联合细胞因子和饲养细胞的刺激作用,提高NK细胞的增殖速度和纯度。该发明将NCR3LG1和m IL-15同时转染到K562细胞,m IL-15可以调节NK细胞的活化和增殖,而NCR3LG1作为NK细胞表面主要的活化受体之一NKp30的配体,可以有效的刺激NK细胞活化,且两者有协同作用。再加上游离添加的IL-2和IL-21等因子的刺激,可以在21天的培养时间内,使PBMC细胞数量得到超过500倍的增殖,CD3-CD56+NK细胞的比例超过70%。
这些方法可以有效的扩增NK细胞,而且NK细胞的纯度也达到了非常高的水平。利用这些方法扩增的免疫细胞在体外试验中可以有效地杀伤肿瘤细胞,但是在动物体内试验中得到的效果与体外试验还是有一些差距。原因之一就是因为在体内肿瘤微环境的存在。仅仅提高免疫细胞的数量和对肿瘤的杀伤力,是不足以达到体内抗瘤的理想效果的。
发明内容
有鉴于此,本发明提供了一种淋巴细胞及其培养体系和培养方法。本发明通过在滋养细胞上表达TAM细胞特异性标志物,使体外扩增的免疫细胞同时具有更强的去除TAM细胞的能力,为免疫细胞治疗肿瘤提供新的可能性。
为了实现上述发明目的,本发明提供以下技术方案:
本发明提供了一种淋巴细胞的培养体系,该培养体系包括灭活的基因工程细胞、IL-2、FBS、抗CD3抗体和淋巴细胞基础培养基;
基因工程细胞为表达IL-21、IL-12、CD80和CD206的细胞。
肿瘤微环境是肿瘤细胞赖以生存的复杂环境,由多种不同的细胞外基质和细胞组成,它们通过多种途径限制免疫细胞的活性,促进肿瘤细胞的生长和转移。临床上利用治疗肿瘤的免疫细胞,只能杀伤肿瘤细胞是远远不够的,需要克服肿瘤微环境中的障碍才能获得更好的疗效。本发明提供一种体外激活扩增免疫细胞的方法,可以减少肿瘤微环境中的免疫抑制作用。
本申请发现饲养细胞上表达白细胞介素21、白细胞介素12和CD80,得到的抗原提呈细胞可将NK细胞在2周扩增几千倍。这些细胞可以对肿瘤细胞高效杀伤,但是对肿瘤相关细胞的作用比较弱。在此基础上,本申请制作了一种新的淋巴细胞扩增培养体系(包括白介素21、白细胞介素12、CD80、CD206、IL-2和抗CD3抗体的混合物)。采用本发明培养体系扩增的免疫细胞,NK细胞占大约72%,在第14天可以将细胞扩增约2000倍,对TAM的杀伤性明显增强,为免疫细胞过继免疫治疗的应用提供了新的路径。与原来的饲养细胞上表达白细胞介素21、白细胞介素12和CD80的扩增体系相比,扩增后的免疫细胞在体外实验中对肿瘤相关巨噬细胞的杀伤能力提高了接近2倍。这种方法扩增的免疫细胞有望在体内实验中打破肿瘤微环境的免疫抑制,得到更强的杀伤肿瘤细胞的效果。

将灭活的基因工程细胞、IL-2、FBS、抗CD3抗体加入淋巴细胞基础培养基。
作为优选,灭活的方式为辐照灭活,辐照的剂量为100~500Gy,时间为10~60min。
优选地,辐照的剂量为100~300Gy,时间为20~40min。
优选地,抗CD3抗体在培养淋巴细胞第1天时添加。
本发明还提供了一种淋巴细胞的培养方法,包括如下步骤:
将单核细胞加入本发明培养体系,培养5~10天,离心;
在细胞中再次补加培养体系,再培养5~10天,离心。
作为优选,培养的条件为37℃、5%CO2;
作为优选,单核细胞为哺乳动物外周血单核细胞、脐带血单核细胞或胎盘单核细胞中的任意一种。
作为优选,培养体系中的基因工程细胞与单核细胞的比例为1:(0.25~5)。

本发明提供了一种淋巴细胞扩增培养体系(包括白介素21、白细胞介素12、CD80、CD206、IL-2和抗CD3抗体的混合物)。将上述培养体系与外周血单核细胞进行共培养,可以得到大量的免疫细胞,其中NK细胞为主要成分。这些免疫细胞不但可以高效杀伤肿瘤细胞,而且对CD206阳性的肿瘤相关巨噬细胞进行高效去除。

本发明公开了一种淋巴细胞及其培养体系和培养方法,本领域技术人员可以借鉴本文内容,适当改进工艺参数实现。特别需要指出的是,所有类似的替换和改动对本领域技术人员来说是显而易见的,它们都被视为包括在本发明。本发明的方法及应用已经通过较佳实施例进行了描述,相关人员明显能在不脱离本发明内容、精神和范围内对本文所述的方法和应用进行改动或适当变更与组合,来实现和应用本发明技术。
术语解释:
肿瘤相关巨噬细胞(tumor associated macrophage,TAM)是浸润在肿瘤组织中的一种巨噬细胞。肿瘤相关巨噬细胞可以通过多种途径促进肿瘤的生长和转移。TAM在肿瘤的发生发展过程中发挥重要作用,可表现在多个环节。其中包括释放多种生长因子,促进肿瘤的生长;分泌多种蛋白酶,破坏肿瘤周边的基底膜,从而间接促进肿瘤向周边侵袭;分泌大量促血管生成的因子和调节血管生成的酶类,并可表达血管内皮生成因子VEGF,促进肿瘤血管和淋巴管的生成。TAM还可通过表达PD-L1以及释放IL-10等细胞因子,限制免疫细胞发挥效应。
CD206是M2型巨噬细胞标志物,具有高度特异性,与肿瘤细胞的增殖和转移密不可分。已有研究表明CD206与乳腺癌、卵巢癌、胰腺癌、前列腺癌等恶性肿瘤密切相关。
白细胞介素21(interleukin-21,IL-21)是一种细胞因子,与其受体结合后可以调节B细胞的增殖,促进T细胞、NK细胞的增殖、分化,并能提高NK细胞杀伤活性。
白细胞介素12(Interleukin-12,IL-12)是一种具有多种免疫调节功能的杂二聚体,它主要由活化的单核/巨噬细胞产生,其合成受多种因素的影响。并主要通过作用于T及NK细胞而发挥作用,它在感染免疫、肿瘤免疫及自身免疫性疾病中起着十分重要的作用。
白细胞介素-2(Interleukin-2,IL-2)是趋化因子家族的一种细胞因子。它是由多细胞来源(主要由活化T细胞产生),又具有多向性作用的细胞因子(主要促进淋巴细胞生长、增殖、分化);对机体的免疫应答和抗病毒感染等有重要作用,能刺激已被特异性抗原或致丝裂因数启动的T细胞增殖;能活化T细胞,促进细胞因子产生;刺激NK细胞增殖,增强NK杀伤活性及产生细胞因子,诱导LAK细胞产生;促进B细胞增殖和分泌抗体;激活巨噬细胞。
CD80和CD86是活化免疫细胞时的协同刺激因子,在自身免疫监控、体液免疫应答及移植反应中发挥重要作用。

第一方面,本发明提供一种体外扩增淋巴细胞的培养体系,其特征在于,所述培养体系包括:白细胞介素21(IL-21)、白细胞介素12(IL-12),CD80,CD206,IL-2和抗CD3抗体的混合物。
优选地,所述白细胞介素21、白细胞介素12和CD80,CD206,IL-2和抗CD3抗体的混合物的添加量为10-3000pmol,优选为100-800pmol,进一步优选为300-500pmol。

本发明K562细胞通过基因工程技术跨膜表达IL-21、IL-12、CD80,改造为刺激型工程细胞,使得IL-21、IL-12、CD80在细胞生物膜上正常表达,体外刺激NK生长,激发活性,抑制B细胞和T细胞的作用。
所述培养体系还包括白细胞介素2,所述白细胞介素2的添加量为50-400U/mL,优选为80-200U/mL。

第二方面,本发明提供一种淋巴细胞的扩增方法,利用如第一方面所述的培养体系,以外周血单核细胞为原料,进行培养。

优选地,所述扩增方法,包括以下步骤:
(1)培养:采集哺乳动物外周血、脐带血或胎盘单核细胞中的任意一种单核细胞;
(2)培养体系制备:将所述K562细胞进行辐照灭活,再加入培养液中,再向培养液中加入白细胞介素2;再向培养液中加入OKT3;
(3)扩增:将步骤(1)得到的单核细胞加入步骤(2)所述的培养体系中,培养5-10天,离心获得单核细胞,再次补加培养体系,再培养5-10天;
(4)收获:培养结束后离心收集获得的淋巴细胞。
本发明中优选地,所述K562工程细胞与所述单核细胞的比例为1:(0.25-5),优选为1:(0.25-4),进一步优选为1:(0.5-1.5)。
优选地,步骤(2)所述辐照的剂量为100-500Gy,优选为100-300Gy。
优选地,步骤(3)所述的培养的天数为7天。
第三方面,本发明提供一种如第二方面所述的扩增方法扩增的淋巴细胞。
本发明中,所述淋巴细胞可以用来进行自体移植和异体移植,在受捐助人(成年人或者未成年人)的组织相容性抗原以及血液抗原与捐助人(成年人或者未成年人)的组织相容性抗原以及血液抗原相匹配的前提下,捐助人被激活和扩增的淋巴细胞可以通过静脉滴注的方式注入到被捐助人的血管中。
第四方面,本发明提供一种如第二方面所述的扩增方法扩增的淋巴细胞用于制备治疗肿瘤性疾病和/或传染性疾病的药物。
优选地,所述肿瘤性疾病选自但不限于急性髓细胞性白血病、胶质细胞瘤、前列腺肿瘤、恶性黑色素瘤、肾细胞恶性肿瘤、乳腺癌、肺癌或肝癌中的任意一种或至少两种的组合;

下面结合实施例,进一步阐述本发明:
实施例1基因工程细胞的制备
基因工程细胞的制备方法如下:首先构建能稳定表达跨膜白介素21、白介素IL12A、跨膜白介素12B和CD80、CD206的载体。各自的载体上分别含有病毒启动子和选择标记基因。跨膜白介素21或跨膜白介素12B是通过CD4的跨膜部分连接到细胞膜上,白介素IL12A和CD80为跨膜蛋白。用这些载体对K562细胞进行转染,根据标记基因用相应抗生素进行阳性筛选,用流式细胞仪挑选表达阳性的细胞,制作基因工程细胞,如图1。
实施例2淋巴细胞扩增培养体系的配制
将实施例1制备的基因工程细胞用于淋巴细胞的扩增,包括以下步骤:
(1)灭活基因工程细胞:通过100Gy放射线照射30分钟,获得灭活的K562工程细胞;
(2)配制培养基:取淋巴细胞培养液RPMI1640和10%胎牛血清,再向混合培养基加入步骤(1)灭活的K562工程细胞,添加量为1×106/mL,再添加IL-2,IL-2的添加量为50U/mL;在扩增淋巴细胞第1天添加抗CD3抗体50ng/mL。
实施例3健康志愿者淋巴细胞的扩增
(1)培养:第0天采集新鲜健康志愿者血液,经离心收集血清储存,经淋巴分离液分离获得人外周血单个核细胞(PBMC),接种在所述培养基中,密度为1×106/mL,添加所述灭活基因工程细胞与所述单核细胞的比例为1:1,接种到体外扩增淋巴细胞的培养体系中;第1天添加抗CD3抗体OKT3,OKT3的添加量为50ng/mL。
(2)扩增:37℃、5%CO2条件下培养到7天,收集细胞,离心,去除上清,加入新鲜培养基,再加入所述灭活基因工程细胞,再培养7天;
(3)收获:培养结束后离心收集获得NK细胞。
采用的稳定表达跨膜白介素21、白介素IL12A、跨膜白介素12B和CD80的K562工程细胞与PBMC细胞共培养得到的NK细胞作为对照,(工程细胞在第0天和第7天添加);
采用自动细胞计数仪对扩增的NK细胞进行计数,并绘制曲线。可以看出NK细胞在以上条件下扩增后,PBMC细胞数量显著增加,NK细胞数量在7天时进入对数生长期,14天时约增长了余约5000倍,NK细胞占比70%以上,如图2。
实施例4将单核细胞诱导为肿瘤相关巨噬细胞
分离健康志愿者单核细胞,将细胞按5×106/ml接种于培养瓶中,37℃、5%CO2培养箱培养2-4h后,去除未贴壁细胞。将贴壁细胞经EDTA消化后离心重悬,对照组为普通培养,实验组用20ug/ml IL-4和IL-13进行刺激5天,收集细胞。用标记的抗人CD206单克隆抗体进行染色,用流式细胞仪进行检测,可见实验组细胞与对照组相比,高表达CD206,如图3。ELISA检测实验组细胞的培养液中的IL-10,可见IL-10分泌明显增多,如图4。说明肿瘤相关巨噬细胞的诱导是成功的。
实施例5免疫细胞对肿瘤细胞的杀伤作用
实验组:人外周血单核细胞在稳定表达跨膜白介素21、白介素IL12A、跨膜白介素12B和CD80、CD206的基因工程细胞和IL-2、OKT3的共同作用下,培养14天后得到的免疫细胞对K562细胞的杀伤作用。
对照组:人外周血单核细胞在稳定表达跨膜白介素21、白介素IL12A、跨膜白介素12B和CD80的基因工程细胞和IL-2的共同作用下,培养14天后得到的免疫细胞对K562细胞的杀伤作用作为对照。
结果显示,在实验组的免疫细胞对肿瘤细胞的杀伤作用略低于对照组。在效靶比高的时候,比如4:1的时候,二者对肿瘤细胞的杀伤作用几乎相同。如图5。
实施例6免疫细胞对肿瘤相关巨噬细胞的杀伤作用
实验组:人外周血单核细胞在稳定表达跨膜白介素21、白介素IL12A、跨膜白介素12B和CD80、CD206的基因工程细胞和IL-2、OKT3的共同作用下,培养14天后得到的免疫细胞对肿瘤相关巨噬细胞的杀伤作用。
对照组:人外周血单核细胞在稳定表达跨膜白介素21、白介素IL12A、跨膜白介素12B的基因工程细胞和IL-2的共同作用下,培养14天后得到的免疫细胞对肿瘤相关巨噬细胞的杀伤作用作为对照。
结果显示,实验组的免疫细胞对肿瘤相关巨噬细胞的杀伤作用几乎是对照组的2倍。如图6。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (10)
1.一种淋巴细胞的培养体系,其特征在于,所述培养体系包括灭活的基因工程细胞、IL-2、FBS、抗CD3抗体和淋巴细胞基础培养基;
所述基因工程细胞为表达IL-21、IL-12、CD80和CD206的细胞。
3.根据权利要求1所述的培养体系,其特征在于,所述IL-21为跨膜IL-21;所述IL-12由IL-12A和IL-12B组成,所述IL-12B为跨膜IL-12B。
4.根据权利要求1至3中任一项所述的培养体系,其特征在于,所述基因工程细胞的宿主细胞为K562细胞;所述抗CD3抗体为OKT3;所述淋巴细胞基础培养基为RPMI 1640。
5.权利要求1至4中任一项所述培养体系的制备方法,其特征在于,包括如下步骤:
构建表达IL-21、IL-12、CD80和CD206的细胞,经灭活,得到灭活的基因工程细胞;
将灭活的基因工程细胞、IL-2、FBS、抗CD3抗体加入淋巴细胞基础培养基。
6.根据权利要求5所述的制备方法,其特征在于,所述灭活的方式为辐照灭活,所述辐照的剂量为100~500Gy,时间为10~60min。
7.一种淋巴细胞的培养方法,其特征在于,包括如下步骤:
将单核细胞加入权利要求1至4中任一项所述培养体系,培养5~10天,离心;
在细胞中再次补加培养体系,再培养5~10天,离心。
8.根据权利要求7所述的培养方法,其特征在于,所述培养的条件为37℃、5%CO2;
所述培养体系中的基因工程细胞与所述单核细胞的比例为1:(0.25~5)。
9.权利要求7或8所述培养方法获得的淋巴细胞。
10.权利要求9所述淋巴细胞在制备治疗肿瘤性疾病和/或传染性疾病的药物中的应用。
