Description

一种纯中药制剂及其在治疗高尿酸血症与痛风药物中的应用
技术领域
本发明涉及医学药剂领域，特别涉及一种纯中药制剂及其在治疗高尿酸血症与痛风药物中的应用。
背景技术
高尿酸血症与痛风是因多种基础性疾病引起，发病后又可导致多组织、多器官异源性病变的嘌呤代谢紊乱性疾病。该疾病既有DNA遗传或严重肝脏疾病所致的黄嘌呤氧化酶(HGRT)的缺陷，又有内源性嘌呤生成过多或外源性嘌呤的过度摄入，还有肾脏排泄障碍与尿酸倒吸收等因素，致使血清尿酸升高。在治疗药物的研究中，容易误将治疗的靶向集中与痛风止痛，补救性或介入性控制尿酸水平，忽略或遗漏了引起嘌呤代谢紊乱的病根。从曾有和现有的高尿酸血症与痛风治疗产品中，如苯溴马隆、非布司他和白漂醇等可明显看出补救性控制尿酸是短期或暂时的，无法实现血清尿酸正常、自然、持续稳定。
根据传统中医与现代医学理论，研究表明，肝脾肾功能异常是导致嘌呤代谢紊乱的主要根源，嘌呤代谢紊乱又是导致高尿酸血症的主要根源，高尿酸血症又是引起痛风的主要根源。
在曾有和现有众多技术中，以非布司他、苯溴马隆为列，以补充嘌呤氧化酶或介入性控制尿酸素降低尿酸。首先是脱离了“矫正嘌呤代谢紊乱”这一病根，再是药物对肝肾损伤，导致恶性循环。
为了矫正嘌呤代谢紊乱，达到尿酸的生成与排泄平衡，使血清尿酸长期保持在正常阈值，从而避免尿酸盐在体内的沉积，导致痛风等各种异源性疾病的产生，特作以下创新发明。
发明内容
本发明意在提供一种纯中药制剂及其在治疗高尿酸血症与痛风药物中的应用，以解决嘌呤代谢紊乱的问题。
本方案中的一种纯中药制剂，包括的原料及其重量份数为：白梅花50～60g，龙胆草50～60g，肝炎草60～70g，柴胡50～60g，五味子50～60g，岩黄连50～60g，决明子50～60g，虎杖50～60g，肾炎草70～80g，蒲公英50～60g，猫须草50～60g，益母草50～60g，女贞子70～80g,百合50～60g，酒大黄20～30g，熟地黄60～70g，盐泽泻30～40g，山药90～100g，黄芪90～100g，猪苓50～60g，厚朴50～60g，炒仓木20～30g，炒白术20～30g，大枣20～30g，炒鳖甲50～60g，葛根80～90g，炒硝石50～60g，伸筋草90～100g，淡竹叶40～50g，桑叶40～50g，车前子80～90g，薏苡仁80～90g，天葵子80～90g，毛冬青50～60g，桃仁50～60g，当归50～60g，丹参30～40g，威灵仙90～100g，灯芯草50～60g，露露通50～60g，生地黄50～60g，泽泻40～50g，山茱萸20～30g，茯苓20～30g，石斛50～60g，炒黄柏20～30g，秦艽80～90g，扛板归50～60g。
进一步，本申请中的纯中药制剂在制备治疗高尿酸血症与痛风药物或药物组合物中的应用。
本方案的有益效果：按照传统中医理论：肝藏血，主疏泄，调畅气机，促进脾胃运化，是人体新陈代谢之枢纽。若肝脏受病邪侵扰，就会阻遏静脉，致使肝郁气滞而引起脾胃运化和新陈代谢无力，二便便秘，造成尿酸排泄障碍、若肝气郁结，气郁则生火，火热又耗损肝肾之阴，加剧体内阴阳失衡。由此可见，肝病不但直接导致黄嘌呤氧化酶缺乏，引起尿酸生成过多，而且还会累及脾胃和肾脏，造成代谢功能紊乱加剧。因此，根据高尿酸血症与痛风疾病病理，调治肝脏是重中之重。本方案中用白梅花用于疏肝，龙胆草和肝炎草利胆退黄，柴胡解郁，五味子益气养阴，岩黄连、决明子和虎杖益肝排毒；
肾是人体先天之本，是人体新陈代谢与维护人体酸碱平衡的稳定；是人体水分、营养物质、电解质的调节器；是清除体内废物、毒素的血筛子。人体代谢中产生的废物与其他毒物，除小部分由肠胃排泄外，绝大多数是由肾脏排出体外。例如人体每天产生尿酸约700毫克，其中500毫克经小便排泄，200毫克经大便排泄。若肾脏受到药物损伤或肌酐高胱抑素c，尿素氮增高，都会引起肾炎，肾孟肾炎，肾间质性肾炎，肾病综合症。无论是急性或者慢性肾功能不全皆能导致尿路排泄障碍或受阻，产生尿酸盐结石或对尿酸的重吸收，引起高尿酸血症。据《中华医学杂志》2012年2月28日第92卷第八期N08专家论坛载：“血尿酸每升高60umo/l，肾脏疾病风险增加71％，肾功能恶化风险增加3倍”。由此可见肾病与高尿酸血症不但可能因果相互转化、相互自身，而且形成恶性循环，逐步加重疾病。因此治疗高尿酸血症与通风，必须调治好肝肾。本方案中用肾炎草、蒲公英、猫须草和益母草清热解毒，消炎利尿，女贞子滋肾益阴，百合润燥补脾健胃，酒大黄排泄肌酐，熟地和盐泽泻滋肾阴、泻肾经虚火。以确保尿酸排泄。
脾脏是人体最大的免疫器官，是机体体液免疫和细胞免疫的主要器官。在传统中医力，脾主运化、升清、统血。是输布水谷精微于全身，营养五脏六腑及各组织，是供给生命动力、气血生化之源。若脾失健运，不但消化吸收，营养物质的传输市场，免疫率下降，而且还会造成运化清浊不分，致使嘌呤代谢功能紊乱，水液与嘌呤物在体内停滞，形成痰湿。由于痰湿淤阻脉络、关节，引起关节中章，皮下结节。由此可见，脾失健运后，不但会引起嘌呤代谢紊乱，而且还是痛风关节炎的引发病灶。因此，健脾燥湿必不可缺。本方案中用薏苡仁健脾燥湿、利尿消肿，用猪苓参湿泄熟，用厚朴燥湿消淤，用苍术、白术大枣补虚。
散结，化浊，这是排除体内沉积尿酸，疏通脉络，消除高尿酸血症及其所致痛风等异源性疾病的关键技术。因高尿酸血症会导致大量的尿酸盐在体内沉积。当尿酸盐沉积时，关节受到挤压，造成关节畸形，活动功能障碍或关节坏死。若急性关节炎发作，常会在踝关节等处产生撕裂样或刀割样或咬噬样或烧灼样剧烈疼痛，这就是痛风。当尿酸盐沉积于血管壁时，造成血管粥样硬化和血管狭窄，引起高血压或心肌梗塞或脑梗等疾病，同时增加并发糖尿病、冠心病等代谢综合症的风险。当尿酸盐沉积于肾脏时，会引起尿路结石、慢性肾水肿、肾绞痛、尿血、肾炎，甚至尿毒症。若不软坚散结，溶酸化浊，沉积的尿酸盐就无法排出，淤阻的脉络也就无法疏通。本方案用威灵仙和炒鳖甲软坚，用天葵子和炒硝石化石。
排酸降算、疏通脉络。尿酸的生成和排降是一个动态过程，其生成与排降都与DNA先天因素或肝脾肾病理因素所致的嘌呤代谢紊乱相关联，是指尿酸生成与排降的自然平衡。本方案阐述了条例肝脾肾，矫正嘌呤代谢紊乱对治疗高尿酸血症与痛风的关键性。排酸降酸与溶酸化浊一脉相承。目的是对沉积于体内尿酸融排，达到疏通脉络，消除异源性疾病，为治愈高尿酸血症和痛风的其他异源性疾病消除病灶，从根本上解决痛风反复发作。
调整营养，补起不足，泻其有余，恢复阴阳的相对平衡是治疗疾病的基础，也是恢复元气，巩固根本，调节人体抗病能力的重要保障技术。按照传统中医理论，高尿酸血症与痛风内生风热，湿热相并。只有滋阴泻失，消痰排毒，方能恢复阴阳的相对平衡，本方案才去“三泻联动”，确保阴阳平衡，同时助力于“嘌呤代谢紊乱”得到矫正。

将实施例1中的纯中药制剂应用于制备治疗高尿酸血症与痛风药物或药物组合物中。，得出如下数据：

结论：将本方案制得的药物或药物组合物进行上千使用炎症，跟踪回访120余人，经统计显示，有效率98.7％，治愈率95.2％。除肌酐、尿素氮严重身高，影响治愈外，个体差异性并不明显。服用本药物或药物组合物治疗痛风，每月降低血尿酸最少58，最高146.最早治愈的患者已时隔近5年，至今未发现治愈后又反弹的先例。因此本方案中的纯中药制剂可矫正嘌呤代谢紊乱，达到尿酸的生成与排泄平衡，使血清尿酸长期保持在正常阈值，从而避免尿酸盐在体内的沉积，避免痛风等各种异源性疾病的产生。
