Background

Selenium is a trace element necessary for human bodies and animal bodies, can effectively inhibit the proliferation of tumor cells, and can effectively enhance the disease resistance of human bodies under the synergistic action of vitamin E, protect the normal functions of human cells, thereby effectively reducing the risk of suffering from tumor diseases. Therefore, the vitamin A, the vitamin D, the vitamin E and the selenium are supplemented in a proper amount in a targeted manner, the cellular immunity can be regulated, the humoral immunity function can be enhanced, and the effect of auxiliary prevention and treatment of tumor diseases can be achieved.
Although many health care products combining various vitamins and trace elements exist in the market at present, most of the health care products play a role of a nutritional supplement, can be taken by various groups, and have very limited actual health care effect due to lack of pertinence and small dosage of a formula. Therefore, the present invention aims to develop a composition suitable for the middle-aged and elderly people to prevent tumors, select vitamins and trace elements with synergistic functions, and give a physiological sufficient amount to perform accurate nutrition to people with specific nutrient deficiency, thereby maximizing the health care function of the product.
Disclosure of Invention
The invention aims to provide a composition with the function of assisting in preventing tumor diseases of middle-aged and elderly people.
In order to achieve the purpose, the invention adopts the following technical scheme:
a composition for auxiliary prevention of tumor diseases comprises the following components:
(1) 160-800 μ g vitamin A;
(2) 2-15 μ g vitamin D;
(3) 5-150 μ g vitamin E;
(4) 10-100 μ g selenium;
(5) acceptable auxiliary materials.
In the composition, preferably, the content of vitamin A is 600-800 μ g, the content of vitamin D is 5-10 μ g, the content of vitamin E is 10-100 μ g, and the content of selenium is 20-100 μ g.
In the invention, the vitamin A source comprises vitamin A and beta-carotene of plant source.
In the present invention, the vitamin D includes vitamin D2 and vitamin D3.
In the present invention, the selenium includes inorganic selenium and organic selenium. Inorganic selenium includes but is not limited to sodium selenite, sodium selenate, organic selenium includes but is not limited to selenoprotein, selenium-rich yeast, selenate lipopolysaccharide, selenium-rich edible fungi, selenomethionine.
The composition can be prepared into oral preparations such as common tablets, chewable tablets, soft capsules, hard capsules, powder, granules and the like.
In the present invention, necessary and acceptable excipients are required to be added in order to prepare various suitable dosage forms. The adjuvants are selected from one or more of filler (dispersant), solubilizer, binder, disintegrating agent, lubricant, suspending agent, correctant, stabilizer, edible pigment, embedding substance, and coating premix.
The filler (dispersing agent) comprises one or more of starch, soluble starch, dextrin, maltodextrin, potato starch, corn starch, wheat starch, D-mannitol, calcium hydrophosphate, anhydrous calcium hydrophosphate, maltose, xylitol, sorbitol, microcrystalline cellulose, isomaltulose, milk powder, low-substituted hydroxypropyl cellulose, sucrose, glucose, lactose, pregelatinized starch, edible vegetable oil, polyethylene glycol and the like.
The suspending agent comprises one or more of beeswax, insect wax, lecithin, glyceryl monostearate, ethyl cellulose and the like, the wetting agent comprises purified water and ethanol, and the adhesive comprises one or more of hydroxypropyl methyl cellulose, povidone, hydroxypropyl cellulose, sodium carboxymethyl cellulose, starch, methyl cellulose, ethyl cellulose, gelatin, polyethylene glycol, pregelatinized starch, sucrose, glucose and the like.
The disintegrating agent comprises one or more of starch, carboxymethyl starch sodium, crospovidone, croscarmellose sodium, low-substituted hydroxypropyl cellulose, sodium bicarbonate, citric acid and the like.
The lubricant comprises one or more of magnesium stearate, stearic acid, talcum powder, silicon dioxide, hydrogenated vegetable oil, polyethylene glycol, sodium dodecyl sulfate, glyceryl behenate, sodium stearyl fumarate and the like.
The flavoring agent comprises one or more of fruit powder, edible essence, high-intensity sweetener, sour agent and the like, wherein the fruit powder comprises one or more of orange powder, lemon powder, cherry powder, apple powder, strawberry powder, kiwi fruit powder, orange powder, seabuckthorn fruit powder, green plum fruit powder, mango fruit powder, mulberry fruit powder, hawthorn fruit powder, watermelon fruit powder, pineapple fruit powder, blueberry fruit powder, honey peach fruit powder, hami melon fruit powder, grape fruit powder, vanilla fruit powder, pomegranate fruit powder, rose fruit powder and the like.
The edible essence comprises one or more of vanilla essence, sweet orange essence, milk essence, apple essence, grape essence, strawberry essence, pineapple essence, honey peach essence, hami melon essence, lemon essence, cherry essence, sweet corn essence, banana essence, grape essence, rose essence, blackcurrant essence, green apple essence, mango essence, green mango essence, orange essence, fragrant orange essence, snow pear essence, strawberry essence, green plum essence, blueberry essence, waxberry essence, grapefruit essence, pineapple essence, guava essence, passion fruit essence, kiwi fruit essence, cherry essence, kiwi fruit essence, watermelon essence, papaya essence, hami melon essence, durian essence, lychee essence, hawthorn essence, olive essence, mulberry essence and the like.
The sweetening agent comprises sucralose, aspartame, mogroside, neotame and stevioside, the acidulant comprises citric acid, malic acid, lactic acid, tartaric acid and fumaric acid, the edible pigment comprises caramel color, gardenia yellow, curcumin, chlorophyll, capsicum orange, capsanthin, grape skin red, carmine and aluminum lake thereof, sunset yellow and aluminum lake thereof, brilliant blue and aluminum lake thereof, indigo blue and aluminum lake thereof, beet red, natural amaranth, gardenia blue, vegetable carbon black, iron oxide red, black bean red, quinoline yellow, lycopene, monascus red, yellow iron oxide, cocoa shell color, safflower yellow, monascus yellow and the like.
The stabilizer comprises one or more of dibutyl hydroxy toluene (BHT), Butyl Hydroxy Anisol (BHA), vitamin C, L-ascorbic acid, tricalcium phosphate, magnesium phosphate, calcium carbonate and the like.
The embedding substance comprises one or more of beta-cyclodextrin, alpha-cyclodextrin, gamma-cyclodextrin, dextrin, starch, edible modified starch, sucrose, maltodextrin, gum arabic and the like.
The composition provided by the invention screens vitamins and trace elements with synergistic functions, carries out scientific composition according to the investigation result of early large clinical epidemics, and synergistically plays a role in preventing tumor diseases of middle-aged and elderly people by supplementing the composition of vitamin A, vitamin D, vitamin E and selenium with optimal dose. Vitamins are essential nutrients for the human body, and a long-term deficiency of vitamins increases the risk of developing neoplastic diseases. Clinical test results suggest that the risk of cancer is increased due to the lack of vitamin D, the risk of lung cancer is increased by 3 times due to the lack of vitamin A, and the risk of oral cancer, esophageal cancer, gastric cancer and intestinal cancer is increased due to the lack of vitamin E. The above epidemiological investigations provide a basis for the present compositions. In actual product development, the combination of vitamin A, D, E and selenium in a specific dosage through specific design and a large number of experimental screens shows an unexpected tumor prevention effect in experiments. Compared with the products sold in the market, the composition is a beneficial product with better and more accurate effect, the action effect of the composition is obviously stronger than the single application effect of the composition, and the composition is suitable for being prepared into food or nutritional supplements and can also be added into compound medicines as an effective component.

Detailed Description
Example 1: tablet preparation (1000 tablets amount) of the composition of the invention
The formula is as follows:
the preparation method comprises the following steps: mixing vitamin A, D, E and sodium selenite with microcrystalline cellulose and sodium dodecylbenzenesulfonate, and sieving; adding lactose, polyvinylpolypyrrolidone and passion fruit essence and mixing; adding magnesium stearate, mixing, and tabletting. The coating premix is prepared into solution, and tablets of the composition are prepared by coating the tablets.
Example 2: preparation of Soft capsules of the composition of the invention (1000 capsules)
The formula is as follows:
the preparation method comprises the following steps: adding Cera flava into soybean oil, heating at 50 deg.C to dissolve Cera flava, standing to room temperature, adding vitamin A, vitamin D, vitamin E, sodium selenite, and L-ascorbic acid into soybean oil to obtain suspension. Preparing gelatin solution from gelatin, purified water, grape skin red and p-hydroxybenzoate, pelleting the obtained suspension by a pelleting machine to obtain soft capsules, and carrying out shaping, washing and drying treatment to obtain the soft capsules of the composition.
Example 3: preparation of hard capsules (1000 granules) of the composition of the invention
The formula is as follows:
the preparation method comprises the following steps: mixing vitamin A, vitamin E, sodium selenate, stearic acid and 50% microcrystalline cellulose, sieving, adding lactose and magnesium stearate, mixing, filling by using a full-automatic capsule filling machine, and polishing to obtain the hard capsule of the composition.
In order to prove that the health-care product has the effect of preventing tumor diseases of middle-aged and elderly people, the following experiment is designed to mainly analyze the relationship between the levels of vitamin A, vitamin D and vitamin E in blood plasma and the tumor disease risk of middle-aged and elderly people with hypertension.
Example 4: preparation of granules of the composition according to the invention (1000 parts)
The formula is as follows:
the preparation method comprises the following steps: crushing sucrose and sieving with a 80-mesh sieve for later use; preparing binder from polyvidone K90 with 50% ethanol solution, adding vitamin A, vitamin E, and sodium selenate into the binder, and stirring. Adding sugar powder, maltodextrin, sodium carboxymethyl starch, Hami melon fruit powder and Hami melon essence, mixing, adding prepared adhesive containing vitamin A, vitamin E and sodium selenate to prepare soft material, granulating with 24-mesh sieve, drying at 50 deg.C for 2 hr, and grading with 24-mesh sieve to obtain the granule of the composition.
Example 5: chewable tablet preparation (1000 tablets amount) of the composition of the invention
The formula is as follows:
the preparation method comprises the following steps: preparing hydroxypropyl methylcellulose into adhesive with purified water, adding vitamin A, E into the adhesive, and stirring. Mixing sodium selenite with microcrystalline cellulose, sieving, adding stevioside, pineapple essence, milk powder and xylitol, mixing, adding a prepared binding agent containing vitamin A to prepare a soft material, granulating by using a 24-mesh sieve, drying for 2 hours at 55 ℃, granulating by using the 24-mesh sieve, adding magnesium stearate, mixing, and tabletting to obtain the chewable tablet of the composition.
Example 6: influence of vitamin A, vitamin D, vitamin E and trace element selenium deficiency on primary liver cancer rats
Wistar rats, male, 80, 150-170 g. Feeding conditions are as follows: the room temperature is 21-25 ℃, the relative humidity is 40-60%, and the light and the dark alternate between 12h and 12 h. 4 per cage, free food intake, water, change bedding every other day.
After 1 week of adaptive feeding, rats were randomly divided into a normal group (normal feed feeding), a deficient group (feed deficient in vitamin a, vitamin D, vitamin E, trace element selenium), a composition group (feed deficient in vitamin a, vitamin D, vitamin E, trace element selenium) and 20 animals per group. The formula of each group of feed is as follows:

The method adopts intraperitoneal injection of diethyl nitrosamine to model primary liver cancer of a rat, and specifically comprises the following steps: except for the normal group, each group was intraperitoneally injected with diethylnitrosamine (40mg/Kg) 1 time/week for 12 consecutive weeks. Normal group injected with normal saline of the same volume for 1 time/week for 12 weeks; composition groups the composition of example 1 (VA + VD + VE + Se) (80+1.5+15+10) ug/kg was administered by gavage for 12 consecutive weeks during the entire molding cycle (80+1.5+15+ 10).
And (5) observing and recording the death condition of the rat in the experimental process, and calculating the survival rate. After the 12 week-end dosing, fasting was performed for 12 hours, and abdominal aorta was opened to collect blood and the rats were sacrificed after pentobarbital sodium anesthesia. And storing blood samples in an EDTA anticoagulation tube and an inert separation gel procoagulant tube respectively for subsequent determination. Picking up spleen and weighing and recording; the general condition of the liver of each group of rats is visually observed, and whether cancer nodules appear or not.
The experimental results are as follows: the rats in the normal group have good growth condition, obvious weight gain, glossy hair and normal drinking water. The ADESe deficient rats began to eat significantly less at week 3 than the normal rats, had dull hair, was slow to lose, or gain weight, and died first at week 5. Rats in the cancer model group started to eat significantly less, lazy and dull hair at week 5 and died at week 1 at week 6. The rats are extremely thin before death, have hair loss and lassitude, cannot eat and drink at the later stage, are compressed, and show a cachexia state. The composition group had less food intake than the normal group, lacked luster, lost motion, lost weight or increased slowly, and the composition group had the first death at week 10.

The survival rate of each group is found by statistical analysis, the survival rate of the ADESe-deficient group is lower than that of the cancer model group, and the composition has better anti-cancer effect and higher survival rate.

The statistics of the tumor incidence of each group shows that except that the incidence of the cancer of the normal group is 0, the incidence of the liver cancer of the cancer model group and the ADESe group is 90-100%, and the incidence of the liver cancer is obviously reduced to 50% by supplementing vitamins and Se.
The serum AFP, AFP-L3 levels in surviving rats were measured by ELISA and the results are shown in Table 4:

Note: in comparison to the cancer model group,aP<0.01; in comparison with the normal group,bP<0.01; in comparison with the ADESe-deficient group,cP<0.01。
compared with the cancer model group, the AFP level of the ADESe-deficient group is obviously increased (P <0.01), and the AFP value of the composition group for surviving rats is obviously lower than that of the ADESe-deficient group and the model group, thereby showing better anti-cancer effect (P < 0.01).
AFP (alpha-fetoprotein) is a protein synthesized by embryonic liver cells, is closely related to abnormal rising of AFP of adults and carcinogenesis, and is clinically commonly used for screening and early diagnosis of liver cancer. But due to the poor specificity of AFP, it is often used in combination with the detection of AFP-L3 (alpha-fetoprotein heteroplasmon) to reduce false positive events. The data analysis shows that the deficiency of vitamin A, vitamin D, vitamin E and trace element selenium can accelerate and aggravate the cancer progression of the primary liver cancer rat, and has obvious promotion effects on the cancer progression and the cancer death rate; and the incidence of cancer can be reduced by properly supplementing deficient vitamin A, vitamin D, vitamin E and trace element selenium.
Claims (7)
1. A composition for assisting in preventing tumors of middle-aged and elderly people comprises 160-800 mu g of vitamin A, 2-15 mu g of vitamin D, 5-150 mu g of vitamin E, 10-100 mu g of selenium and acceptable auxiliary materials.
2. The composition of claim 1, wherein: the content of the vitamin A is 600-800 mu g; the content of the vitamin D is 5-10 mu g; the content of the vitamin E is 10-100 mu g; the selenium content is 20-100 mug.
3. The composition of claim 1, wherein: the vitamin A comprises vitamin A and beta-carotene of plant origin.
4. The composition of claim 1, wherein: the vitamin D comprises vitamin D2 and vitamin D3.
5. The composition of claim 1, wherein: the selenium comprises inorganic selenium and organic selenium, the inorganic selenium comprises sodium selenite and sodium selenate, and the organic selenium comprises selenoprotein, selenium-enriched yeast, selenate lipopolysaccharide, selenium-enriched edible fungi, selenocysteine and selenomethionine.
6. The composition of claim 1, wherein: the composition can be prepared into oral preparations such as common tablets, chewable tablets, soft capsules, hard capsules, powder, granules and the like.
7. Use of the multivitamin and trace element composition of any one of claims 1 to 6 in the preparation of a product for the adjuvant prevention of tumors in the middle-aged and elderly people.
