Description

Skin-care matrix with whitening, anti-allergy and anti-aging functions and preparation method and application thereof
Technical Field
The invention belongs to the technical field of cosmetics, relates to a skin-care matrix and a preparation method and application thereof, and particularly relates to a skin-care matrix with whitening, anti-allergy and anti-aging functions and a preparation method and application thereof.
Background
Skin aging is a vital stage accompanying human life activities and is influenced by various aspects such as genetic factors, environmental factors, complex morphological structures of the whole body, and the like. In recent years, with the rise of dietary culture and the change of living habits of people, a series of problems of exogenous accelerated aging, color spots, dark yellow and the like appear on the skins of a plurality of people, and characteristics of wrinkles, skin laxity and the like begin to be generated.
With the development of the science and technology of cosmetics, various skin care products have different quality, and a series of cosmetics with the functions of resisting aging, preserving moisture, whitening, resisting inflammation, repairing and the like are also endless. However, although some cosmetics have efficacy due to the addition of more active ingredients, many people are still sensitive to allergic phenomena such as erythema and swelling pain, and the safety is questioned. In addition, although the components such as vitamin C, hormone, polypeptide and the like which play an anti-aging effect in cosmetics at present can show obvious effects in a short time, most of the compounds have the defects of easy accumulation, difficult decomposition, low safety, poor stability and the like, and cause toxicity to human bodies.
On the other hand, because of the self-barrier function of human skin, general active ingredients are difficult to really enter the deep layer of the skin to play a role, and only can play a role in simply preserving moisture. In order to enhance the penetration and absorption of nutrient components in the skin care product, the external steam engine can be used for assisting in heating, so that a better beautifying effect is achieved. Products of many beauty salons achieve the effect of promoting absorption by means of the functions of introduction instruments (electrical conduction, ultrasonic introduction, radio frequency introduction, microcrystal introduction, micro-needle introduction and the like), but the long-term disordered use of the beauty instruments can cause certain damage to skin, and the use method is not simple and convenient, takes time, and has very limited effect of promoting absorption.

(2) Anti-allergic efficacy-Hyaluronidase in vitro inhibition test
Hyaluronic acid is a component that limits the diffusion of water and other extracellular substances in the tissue matrix, and after being hydrolyzed by hyaluronidase, cells become non-viscous, causing degranulation of cells and the exudation of new synthesized mediators, exerting biological effects, resulting in the occurrence of immediate type hypersensitivity. Hyaluronidase is a participant in allergic reactions, has strong allergic-related effects, and can relieve allergic phenomena and improve skin by inhibiting the activity of hyaluronidase.
The anti-allergic effects in vitro of examples 1 to 8 and comparative examples 1 to 3 were examined by the hyaluronidase inhibition method. Taking 0.1mL of CaCl2Reacting the solution (0.25mmol/L) and 0.5mL of hyaluronidase solution (100U/mL) in water bath at 37 ℃ for 20 min; adding 0.5mL of the extracting solution, and keeping the temperature for 20 min; then adding 0.5mL of sodium hyaluronate solution (0.5g/L), reacting for 30min, taking out and cooling for 5 min; adding 0.1mL of sodium hydroxide solution (0.4mol/L) and 0.5mL of acetylacetone solution (3.5mL of acetylacetone dissolved in 50mL of 1.0mol/L sodium carbonate solution), boiling in water bath for 15min, and immediately transferring to ice water bath for 5 min; 1mL of an Ellisib reagent (0.8g of p-dimethylaminobenzaldehyde dissolved in 15mL of concentrated hydrochloric acid and 15mL of absolute ethanol) was added dropwise, and diluted with 3mL of absolute ethanol, left at 25 ℃ for 20min for color development, and the absorbance was measured at 540nm with dipotassium glycyrrhizinate as a control. The hyaluronidase inhibition rate was calculated according to the following formula:
hyaluronidase inhibition { [ (A-B) - (C-D) ]/(A-B) } × 100%
In the formula: a-absorbance of control solution (the sample solution was replaced with acetic acid buffer solution); b-the absorbance of the control blank solution (the sample solution and the enzyme solution are replaced by acetic acid buffer solution); c-absorbance of the sample solution; d-absorbance of sample blank solution (acetic acid buffer solution was used instead of enzyme solution).
The inhibition rate of each group of samples on hyaluronidase was examined by using 5% dipotassium glycyrrhizinate as a positive control, and the results are shown in table 2:
TABLE 2

Respectively measuring the inhibition rate of the extracts with different concentrations, taking the concentration as an abscissa and the inhibition rate as an ordinate to make a straight line, obtaining a straight line equation, and calculating the concentration of the extract when the inhibition rate is 50 percent, namely the IC 50. The results are shown in Table 3:
TABLE 3
Figure BDA0002765415920000182
