背景技术
介孔二氧化硅纳米颗粒由于其化学/热稳定性、大表面积、高负载能力、可调的表面性质和优异的生物相容性而被认为具有作为药物递送系统的巨大潜力。在各种二氧化硅纳米材料中,空心二氧化硅纳米球(HSN)的形态和特性与普通的介孔二氧化硅纳米颗粒的不同之处在于具有空心的内部空间和薄的多孔壳,使得它们能够包封大分子(诸如生物活性成分)并且由于内部空间大而表现出更高的负载能力;这样的目的可以例如通过调节壳的孔径来实现。当孔径小于大分子时,壳可以防止大分子在血液中的循环期间泄漏出去。HSN的形态和特性很大程度上取决于合成策略,而合成策略因应用而异。本发明探索了HSN作为载剂和佐剂以增强医学应用(例如,疫苗接种)的功效的潜力。
发明内容

在本发明中,如本文所用的术语“新抗原”是指具有较小尺寸的生物活性成分,诸如肽、DNA、RNA、核苷酸等。
空心二氧化硅纳米球(HSN)的医学应用
本发明人惊奇地发现,除了用作药物递送剂之外,空心二氧化硅纳米球(HSN)本身可表现出可用于医学应用,特别是免疫治疗的某些特性。
在一个方面,本公开提供了一种用于抑制有需要的受试者中的肿瘤生长的方法,其包含向所述受试者施用空心二氧化硅纳米球(HSN),从而增加肿瘤中的肿瘤浸润免疫细胞,其中所述HSN包含单层或多层二氧化硅壳,其中每个壳具有介孔并包封封闭空心空间,最内空心封闭空间任选地具有固体二氧化硅核,其中所述空间由任意两个二氧化硅壳或固体二氧化硅核之间的距离界定,并且其中通过动态光散射(DLS)测量的于介质中的HSN的流体动力学尺寸不大于150nm,其中所述介质在生物学上类似于或等同于磷酸盐缓冲盐水(PBS)。在一个实施方案中,肿瘤浸润免疫细胞包含,但不限于,T细胞、B细胞、自然杀手细胞、巨噬细胞、中性粒细胞、树突状细胞、肥大细胞、嗜酸性粒细胞、嗜碱性粒细胞等等。
在一些实施方案中,HSN的尺寸在30至150nm,优选40至100nm的范围内。
应当注意,HSN的尺寸,例如,流体动力学尺寸等对于确定它们是否适合于应用可能是至关重要的。透射电子显微术(TEM)是一种用于测量纳米颗粒的“原始”尺寸的常规手段,而流体动力学尺寸可更接近地反映存在于介质中的纳米颗粒的“表观”尺寸。特别地,HSN的流体动力学尺寸可直接确定它们是否可以应用于活体受试者。如果HSN的流体动力学尺寸太大,它们将容易在介质中聚集或生长。这种现象,即聚集,不仅妨碍了输送效率,而且还可能在医学应用中产生负面影响,例如循环系统堵塞、免疫系统快速清除等。流体动力学尺寸可以通过动态光散射(DLS)测量。

包封于HSN中的小生物活性成分/新抗原
本发明还提供了一种空心二氧化硅纳米球(HSN)缀合物,其包含HSN和包封于HSN中的小生物活性成分,其中HSN包含单层或多层二氧化硅壳,其中每个壳具有介孔并包封封闭空心空间,最内空心封闭空间任选地具有固体二氧化硅核,其中所述空间由任意两个二氧化硅壳或固体二氧化硅核之间的距离界定,并且其中通过动力学光散射(DLS)测量的于介质中的HSN的流体动力学尺寸不大于200nm,其中所述介质在生物学上类似于或等同于磷酸盐缓冲盐水(PBS)。
不受理论的约束,当HSN具有不大于100nm的尺寸(例如,通过TEM测量的)和不大于200nm,特别是不大于150nm的流体动力学尺寸(例如,通过DLS测量的)时,它们表现出优异的分散性和靶向淋巴结和肿瘤的特性。鉴于此,HSN适于携带生物活性成分,诸如抗原、新抗原等。特别地,生物活性成分被包封于孔内,使得它们在受试者中的递送期间不会泄漏出去。这确保了生物活性成分不会被存在于受试者中的蛋白酶降解并且到达免疫细胞存在的相同位置,从而增强免疫反应。所述结构还允许生物活性成分在每单位体积HSN中处于更高水平。
HSN的壳的孔径可以调节;当孔径小于生物活性成分(例如,大分子)的尺寸时,壳可以防止生物活性成分(例如,大分子)在血液中的循环期间泄漏出去。HSN的形态和特性很大程度上取决于合成策略,而合成策略因应用而异。因此,本发明人基于HSN可以作为载剂和佐剂施用的效果进行应用,从而增强疫苗接种的功效等。
本发明人还惊奇地发现,当使用微乳液方法生产包封某些生物活性成分(诸如肽)的HSN时,HSN负载的生物活性成分的水平可能不足或低于其它类型的生物活性成分。不受理论的约束,原因可能是在过程中形成反相微乳液,因为肽可能对在形成HSN的微乳液法中使用的表面活性剂具有亲和力。为了解决这一问题,本发明人发现,生物活性成分可以被修饰以在生物活性成分与表面活性剂和二氧化硅的亲和力之间产生差异,即更倾向于二氧化硅,使得生物活性成分可以更容易地被HSN包封。另一种方法是引入对生物活性成分具有高亲和力的分子。
因此,在一些实施方案中,小生物活性成分被修饰为具有如下结构:Y(n)-X-SBI-[X-Y(m)](r),其中Y为带正电荷的肽,X为酶可裂解的序列,SBI为小生物活性成分,且n、m和r中每一个均为整数,其中n和m×r中至少一个不为零。在一个实施方案中,n为不为零的整数且r为0。在一个实施方案中,m和r中每一个均为不为零的整数且n为0。在一个实施方案中,n、m和r中每一个均为不为零的整数。

在这种情况下,Y和Z可以提供对HSN的亲和力,HSN通常在表面X(酶可裂解序列)上带有微量负电荷,X,酶可裂解的序列,可被存在于施用HSN的受试者中的酶(诸如蛋白酶)裂解,使得SBI可以释放到环境中。这些基团的细节也在本公开的其它地方描述。
在一些实施方案中,小生物活性成分为新抗原。新抗原是先前未被免疫系统识别的新形成的抗原。免疫治疗中涉及基于肽新抗原的疫苗的新方法有望使治疗精度达到个体患者中个体肿瘤的水平。新抗原可以由肿瘤突变形成的改变的肿瘤蛋白或由病毒蛋白产生。新抗原的实例包括但不限于肿瘤特异性新抗原、肿瘤新表位、新抗原肽、新抗原DNA和新抗原RNA。
在一些实施方案中,小生物活性成分为衍生自病毒、细菌或微生物的抗原。
包含已知肿瘤特异性突变的肽以及突变多肽或肿瘤表位的片段。这些肽和多肽在本文中称为“新抗原肽”或“新抗原多肽”。
最近,已经报道了二氧化硅纳米颗粒作为疫苗中的潜在免疫佐剂。疫苗通常含有两种主要成分:抗原和佐剂。抗原可以衍生自被抗原特异性受体识别的致病生物的片段或癌细胞的表面蛋白。然而,当单独使用抗原时,用于疫苗的大多数抗原通常具有差的免疫原性、弱的免疫反应和差的免疫记忆。佐剂是诱导、增强、加速和延长针对抗原的特异性免疫反应的物质。对于疫苗,佐剂在产生针对抗原的强大且持久的适应性免疫反应中起关键作用。此外,理想的佐剂应同时充当抗原递送媒介物和免疫增强剂,因为单个颗粒中的抗原和佐剂应促进相同的抗原递呈细胞(APC)的摄取并导致更有效的免疫反应。
使用二氧化硅纳米颗粒作为疫苗接种的佐剂和载剂的优点包括:(1)保护抗原免于降解和变性;(2)有效靶向和激活抗原递呈细胞;(3)提高每体积抗原分子的浓度;以及(4)调节抗原递呈途径。二氧化硅纳米颗粒显示出固有的佐剂活性,并且可以有效地增强细胞和体液免疫。二氧化硅纳米颗粒的理化性质影响其与免疫系统的相互作用。因此,不同种类的二氧化硅纳米颗粒将诱导不同的免疫反应。用于增强免疫原性或免疫治疗功效的二氧化硅纳米颗粒必须完成两项关键工作:(1)有效地将抗原递送到APC或淋巴结,和(2)随后在APC内或附近释放抗原并激活免疫反应。二氧化硅纳米颗粒可以通过粒径和表面官能团修饰促进APC摄取和淋巴结靶向。与带负电的纳米颗粒相比,带正电或带中性电荷的纳米颗粒可以被树突细胞(DC)更有效地摄取。纳米颗粒以尺寸依赖性方式转运至淋巴结。当通过皮下注射施用大颗粒(>200nm)时,颗粒依赖于细胞运输通过DC迁移从皮肤传输至淋巴结,但认为这种途径效率较低。相反,小颗粒(<200nm)能够直接引流至淋巴结并且被淋巴结驻留细胞摄取。因此,小颗粒具有潜在的淋巴结靶向能力和更高的抗原递送效率。为了改善抗原特异性免疫反应的激活,由纳米颗粒递送的抗原应被保护免受蛋白酶的影响并且保持完整直到APC摄取。此外,当抗原被吸附、包封或掺入到纳米颗粒中时,与游离抗原相比,其产生更高的局部抗原浓度,并导致更强的免疫反应。纳米颗粒结合抗原的常见方式是通过共价键或非共价键(诸如疏水/亲水相互作用,范德华力,静电相互作用或氢键)将抗原附着在颗粒表面或颗粒的孔的内表面上。然而,附着在颗粒表面上的抗原可在原液或生理溶液中引起颗粒聚集,使得难以产生稳定悬浮液以进行应用。通过非共价键附着在颗粒上的抗原在注射入体内时可能泄漏;泄漏的抗原将被降解,而递送到APC的有效抗原的数量将减少。通过共价键将抗原与颗粒缀合可以解决抗原泄漏问题。然而,当抗原颗粒被APC摄取时,抗原不能被释放;颗粒将干扰抗原与APC抗原受体的相互作用,并降低诱导免疫反应的可能性。利用二氧化硅纳米颗粒作为抗原载剂和佐剂的能力,二氧化硅纳米颗粒具有解决传统疫苗开发中免疫原性差、免疫反应弱和免疫记忆差的问题的潜力。因此,本发明提供了一种服克上述问题并开发(空心)二氧化硅纳米球的方法,所述二氧化硅纳米球具有单分散粒径,并为疫苗应用提供抗原保护和良好的免疫原性和免疫治疗功效。
如上所述,新抗原可能未被HSN正确或有效包封。新抗原可以是肽、DNA、RNA等。在它们的序列中具有不超过200个氨基酸,优选不超过100个氨基酸,更优选不超过50个氨基酸的肽可被认为是新抗原。具有不超过1200个核碱基,优选不超过600个核碱基,且更优选不超过300个核碱基的DNA或RNA序列可被认为是新抗原。
包封于HSN中的小生物活性成分的应用
因此,在另一方面,本公开提供了一种包含本公开的HSN缀合物的疫苗组合物。
在另一方面,本公开提供了一种将小生物活性成分递送到受试者的方法,其包含向受试者施用本公开的HSN缀合物。
在另一方面,本公开提供了一种在免疫治疗中将新抗原递送到受试者的方法,其包含向受试者施用其中包封新抗原的HSN。
包封小生物活性成分的HSN的制备
本发明还提供了一种产生含有于其中的生物活性成分的空心二氧化硅纳米颗粒的方法:
(a)提供包含油相、表面活性剂、烷氧基硅烷和/或硅酸盐源、含有一或多种生物活性成分的水相以及任选的助表面活性剂的组合物,
(b)由步骤(a)所述的组合物形成油包水(W/O)微乳液;
(c)向(b)的W/O微乳液中加入引发剂,以形成封装生物活性成分的HSN;
(d)执行去稳定条件以使W/O微乳液不稳定并收集由所述微乳液如此形成的所得颗粒;以及
(e)将步骤(d)中收集的颗粒分散在含水洗涤相中,以得到二氧化硅纳米颗粒。
在另一方面,所述方法包含以下特征中的至少一个:
(i)表面活性剂为离子型的;或者表面活性剂为非离子型的并且不存在氧化烯单元;

用于形成W/O微乳液的表面活性剂是本领域中常用的和公知的。优选地,在本发明中使用不具有氧乙烯单元的离子表面活性剂和非离子表面活性剂。不具有氧化烯单元的非离子表面活性剂的实例包括但不限于葡糖苷烷基醚、甘油烷基酯、椰油酰胺单乙醇胺(椰油酰胺MEA)、椰油酰胺二乙醇胺(椰油酰胺DEA)、月桂基二甲基氧化胺等。
如上所述,本发明人发现生物活性成分可以被修饰,使得生物活性成分可以更容易地被HSN包封,和/或对生物活性成分具有高亲和力的分子可以在制备HSN的微乳液法中引入水相中。
在一个实施方案中,小生物活性成分用氨基酸序列修饰。特别地,序列中的氨基酸是可以带正电荷的氨基酸。可以带正电荷的氨基酸的实例包括但不限于精氨酸(R)、赖氨酸(K)、组氨酸(H),具有可带正电荷的位置的非天然氨基酸等。

实施例
提供以下实施例以使本发明对本发明所属领域的普通技术人员而言更容易理解,但并不意图限制本发明的范围。
材料、方法和测试模型
透射电子显微镜法(TEM)
使用透射电子显微镜法(TEM)直接检查并验证二氧化硅纳米颗粒的外观。在75-100kV的加速电压下运行的Hitachi H-7100透射电子显微镜上拍摄TEM照片。将分散在乙醇或水中的样品滴在碳涂覆的铜网格上,并在空气中干燥以进行TEM观察。
动态光散射(DLS)
在马尔文纳米粒度电位仪(Malvern Zetasizer Nano SZS)(马尔文,英国)上,使用动态光散射(DLS)进行二氧化硅纳米颗粒在不同溶液环境中的尺寸测量。在室温下分析在不同溶液中形成的(溶剂化的)粒径:H2O,含有10%FBS的达尔伯克改良伊格尔培养基(Dulbecco's Modified Eagle Medium,DMEM),PBS缓冲液(pH 7.4)和5%葡萄糖。
OPSS-二氧化硅纳米颗粒
合成过程:在第一步,将85.2μL APTMS+253.2μL OPSS-接头(50mg OPSS-PEG-NHS,200/mL,在DMSO中)混合在一起,然后在37℃下搅拌过夜。为了分离APTMS-接头-OPSS,将ProElutTMC18管用3-5mL甲醇冲洗,然后用3-5mL去离子水冲洗。将混合物从100%DMSO稀释至10%DMSO溶液。将APTMS/APTMS-接头-OPSS混合物装载至管的顶部,然后用1mL 10%ACN洗涤以除去游离的APTMS试剂。用1.5mL甲醇洗脱样品,得到APTMS-接头-OPSS。最后,通过旋转蒸发浓缩APTMS-接头-OPSS溶液,并通过HPLC定量原液。
将20mL癸烷、3.5mL Igepal CO-520和1.1mL己醇混合在一起,然后加入1.2mL DI水。将混合物在20℃下搅拌20分钟,以形成反相微乳液体系。引入50μL稀释的APTMS(用DI水8X稀释)和200μL TEOS,然后搅拌20分钟。之后,向体系中加入500μL 28%NH4OH。将溶液在20℃下混合11分钟,然后加入65μL稀释的APTMS(用去离子水8X稀释)和260μL TEOS,然后在20℃下搅拌过夜。为了在颗粒表面修饰APTMS-接头-OPSS,向体系中加入15μL TEOS和ATMPS-接头-OPSS(4.2mg),并将体系搅拌过夜。将10mL乙醇引入溶液中以破坏微乳液体系,然后以14000rpm离心15分钟,得到OPSS-二氧化硅纳米颗粒。将沉淀用乙醇洗涤两次,然后离心以除去洗涤溶剂。用80mL去离子水在50℃下将OPSS-二氧化硅纳米颗粒洗涤1小时,以除去残留试剂并使颗粒变为空心。将沉淀用水洗涤两次,然后离心以除去洗涤溶剂。在上述合成过程之后,获得OPSS-二氧化硅纳米颗粒。
HSN中蛋白质的定量
通过两种方法定量HSN中的蛋白质的量:(1)酶活性或(2)荧光相关光谱法。
通过酶活性方法定量蛋白质@HSN:如果包封于HSN中的蛋白质为酶,HSN中的蛋白质的量可以从蛋白质@HSN的酶活性推导。
ASNase@HSN定量:通过纳氏(Nessler)试剂测定ASNase活性,所述试剂购自默克公司(Merck)。首先,将100μL的0.05M Tris-HCl(pH=8.6)和850μL的0.02M L-天冬酰胺混合,加入50μL 1.5M三氯乙酸(TCA)作为空白,并加入50μL D.I水作为样品。接着,将50μL的ASNase@HSN原液加入到混合物中,在37℃下温育50分钟。之后,取50μL温育的样品与100μL纳氏试剂混合,再加入2.5μL TCA用于测试样品以淬灭反应,并将混合物在室温下静置10分钟。最后,通过在480nm处的吸光度测定100μL样品的活性。
过氧化氢酶@HSN定量:通过H2O2测定法测定过氧化氢酶活性。将约40μg CAT@HSN分散在50μL D.I水中并与50μL的25μM H2O2混合。将它们在黑暗中在37℃温育12分钟以进行反应。之后,将混合物离心以收集上清液,并与由5μL AmplexRed试剂、10μL 10单位/mL HRP和485μL 50mM磷酸盐缓冲液(pH=7.4)构成的100μL稀释的试剂(A22188,英杰公司(Invitrogen))在室温下混合10分钟,以检测剩余的H2O2。然后通过在530nm激发后在585nm的荧光发射来测量样品。根据已知的H2O2浓度,用标准曲线估算过氧化氢酶@HSN活性。
辣根过氧化物酶(HRP)@HSN定量:通过过氧化物酶测定法测定HRP活性。在室温下将50μL HRP@HSN与700μL磷酸钠缓冲液(SPB)(0.05M,pH=7.8)和750μL邻苯二胺二盐酸盐(OPD)溶液(50mL SPB中的20mgOPD,与167μL H2O2,pH=7.8)混合1小时。然后,将100μL样品与1M磷酸充分混合以终止反应,并通过在490nm处的吸光度测量混合物溶液以用于活性测定。
通过荧光相关光谱法(FCS)的蛋白质@HSN定量:利用具有543nm绿激光的共焦激光扫描显微镜(PicoQuant Microtime 200),还通过FCS测定HSN中包封的蛋白质的数量。荧光染料缀合蛋白质用于蛋白质@HSN的合成和随后的检测。将体积为100μL的样品溶液置于载玻片上,然后用543nm激光激发荧光染料,并通过光电二极管检测荧光计数率。测量过程可以分为三个阶段。首先,测量FCS激光的焦体积是拟合相关函数的一个因素。在此,使用具有已知扩散系数(共焦体积中的平均停留时间)的游离染料R6G来测量计数率,然后测定543nm激光的焦体积。基于以上内容,检测RITC的计数率以知晓RITC被激发时释放的光子数。其次,通过测量不同浓度的RITC缀合蛋白质,推导出RITC缀合蛋白质的计数率,以确定浓度与计数率之间的相关性。其应该是线性相关的。最后,通过543nm激光对RITC蛋白质@HSN进行了测量,得到计数率,从而估算HSN中包封了多少蛋白质。
蛋白酶耐受性测定
将400μL 2x10-3mg/mL蛋白酶混合物溶解在10mM NaOAc和5mM CaOAc(pH=7.5)中。然后,将蛋白质@HSN溶解在1mL H2O中并与蛋白酶溶液混合。在30下温育30分钟后,取反应溶液用于通过酶活性测定法测定溶液中剩余蛋白质的量。
通过ELISA测定抗原@HSN诱导的IgG抗体
采用酶联免疫吸附测定法(ELISA)检测抗原@HSN诱导的抗原特异性抗体。将96孔板在4℃下用0.05M碳酸盐/碳酸氢盐缓冲液(pH=9.5)中的100μL 10μg/mL抗原溶液包被20小时。接下来,将孔中的溶液排出,向孔中加入300μL含有0.1%牛血清白蛋白(BSA)的0.1MPBS(pH=7.2),并将96孔板在室温下在定轨摇床上振摇1.5小时以洗去未吸附的抗原。之后,将100μL在0.05%吐温-20PBS中的1600倍稀释的小鼠血浆加入板中,并在室温下温育1小时或在4℃温育过夜。温育后,将孔用PBS洗涤两次,然后加入100μL稀释的第二抗体(1:10000[HRP缀合的第二抗体]或1:200[荧光染料缀合的第二抗体])并在室温下温育1小时。如果使用荧光染料缀合的第二抗体进行检测,则通过在560nm激发下在660nm处的荧光发射来测量样品。如果用HRP缀合的第二抗体进行检测,则将样品与100μL新鲜底物溶液混合,所述新鲜底物溶液含有在0.1M柠檬酸盐缓冲液(pH=6.0)和167μL H2O2中的20mg OPD,并在室温下温育30分钟,之后,加入100μL 1M磷酸以终止反应,并取100μL样品用于通过分光光度计在490nm下测量吸光度。

通过慢病毒颗粒感染,利用luc_4T1细胞构建表达Her2的4T1细胞。将慢病毒载体CHL0042(其对博来霉素(zeocin)和具有EcoRI-AmCyan-XhoI的pcDNA3.1(+)_NheI-HER2-HindIII具有抗性)消化,并通过限制性内切酶和连接酶将其与慢病毒载体连接,产物命名为plenti-zeo-HER2。按照标准磷酸钙转染方案进行病毒包装:将10μg plenti-zeo-HER2质粒、9μg pCMV-Δ8.91质粒和2.5μg水疱性口炎病毒G蛋白(VSVG)质粒与含有2.5M CaCl2的HEBS(HEPES,NaCl,右旋糖,KCl和Na2HPO4)混合,在室温下温育20分钟,并在10cm培养皿中逐滴加入制备的2x106 HEK293T细胞中以制备慢病毒颗粒。20小时后用新鲜培养基替换培养基,转染72小时后收集含有病毒的上清液。接下来,收集病毒溶液并将其加入到制备的靶细胞(luc-4T1)中,在8ng/ml聚凝胺的存在下温育48小时以感染靶细胞。最后,将细胞与100μg/mL博来霉素温育14天以选择Her2-luc转染的4T1细胞系。以抗Her2/ERBB2(cat.10004-RP04,义翘神州(Sino Biological))作为第一抗体和Alex fluor 488山羊抗小鼠IgG作为第二抗体的,通过IFC染色确认Her2蛋白表达。
Her2_ECD蛋白表达
从金斯瑞(Genescript)购买Her2表达载体pET24-hHER2 ECD op的胞外结构域,并将其转化至BL21细胞中以产生Her2_ECD蛋白。在37℃用0.5mM的IPTG诱导大肠杆菌(E.coli)(BL21)中Her2_ECD蛋白过表达4小时。温育后,通过离心收集BL21。接下来,将细胞在10%甘油PBS中在4℃下进行超声处理以破碎细胞,并将溶液离心以收集沉淀,沉淀中富含Her2_ECD蛋白包涵体。向沉淀中加入1.5%肌氨酸以溶解Her2_ECD蛋白,并在4℃下振荡过夜,然后通过离心收集富含蛋白质的上清液。将蛋白质溶液过滤并利用His-tag柱(HisTrap FF,通用电气(GE))通过FPLC(AKTA纯,通用电气)用200mM咪唑磷酸盐缓冲液(0.05M NaH2PO4,0.3M NaCl,pH=8)在4℃下纯化。纯化的蛋白质通过SDS-PAGE和使用抗Her2/ERBB2抗体(10004-RP04,义翘神州)的蛋白质印迹进行表征,并通过纳米光度计(N60,IMPLEN)进行定量。
CD8+T细胞分析方法中的IFN-γ表达
在第1天、第8天和第15天,用相同量的新抗原(NeoAg)或NeoAg@HSN免疫雌性C57BL/6小鼠(6-8周)。在不同的天数从经过疫苗接种的小鼠收集约300μL的外周血。外周血单个核细胞(PBMC)通过1083分离并转移至96孔板中的200μL T细胞培养基(补充有10%FBS、100UmL-1青霉素/链霉素(penn/strep)、55μMβ-巯基乙醇、1x MEM非必需氨基酸溶液和1mM丙酮酸钠的RPMI 1640)。为了进行肽再刺激,将PBMC用各种抗原表位(20μg/mL)处理2小时。随后,通过向培养液中加入GolgePlug来终止细胞因子分泌,再温育4小时。将细胞沉淀并用抗CD16/CD32在室温下染色10分钟,然后将细胞用抗CD8-FITC或抗CD4-FITC在室温下染色20分钟。洗涤细胞,随后使用50μL 2%甲醛溶液固定过夜。第二天,将细胞洗涤并在室温下在50μL 0.5%皂苷中渗透15分钟,然后将细胞洗涤并用抗IFNγ-APC染色20分钟。最后,将染色的细胞沉淀并重悬于200μL缓冲液中,并使用流式细胞术(贝登(BD)FACSCanto II)进行分析。
合成实施例1
蛋白质@HSN的合成
天冬酰胺酶(ASNase)@HSN的合成:将作为油的20mL癸烷、作为表面活性剂的3.5mLCO-520、作为助表面活性剂的1.1mL己醇和2mg ASNase溶解在700μL 1.43mg/mL NaF和500μL 10mg/mL NaF中。将上述所有物质混合以产生反相微乳液体系。然后,将一部分二氧化硅源、50μL用乙醇稀释8倍的APTMS和200μL TEOS加入到体系中,将其在20℃连续搅拌。1小时后,将另一部分二氧化硅源、50μL稀释的APTMS和200μL TEOS加入到体系中,在20℃下连续搅拌20小时。搅拌过夜后,将500μL氢氧化铵(aq)(2.24-2.4重量%)缓慢加入到混合物中,同时搅拌20分钟。接下来,向混合物中逐滴加入50μL TEOS,将其搅拌4小时,然后加入250μLPEG-硅烷和25μL TEOS,并在20℃下搅拌20小时以修饰颗粒表面。之后,加入95%乙醇以使微乳液体系不稳定,并通过在14000rpm下离心20分钟收集颗粒。将颗粒用95%乙醇洗涤一次,并用D.I水洗涤两次,并转移到80mL D.I水中。然后,将溶液保持在40℃下并搅拌1小时以获得ASNase@HSN。最后,通过离心收集ASNase@HSN,用D.I水洗涤两次,并储存在4℃的D.I水中。
辣根过氧化物酶(HRP)@HSN合成:HRP@HSN的合成过程与ASNase@HSN的描述相同,但是用2.67mg HRP代替酶2mg ASNase。
过氧化氢酶@HSN合成:将作为油的20mL癸烷、作为表面活性剂的3.5mL CO-520、作为助表面活性剂的1.1mL己醇以及50000单位过氧化氢酶溶解在1200μL 10mg/mL NaF中。将上述所有物质混合以产生反相微乳液体系。然后,将一部分二氧化硅源、50μL用D.I水稀释8倍的APTMS和200μL TEOS加入体系中,在20℃下连续搅拌。2小时后,加入250μL 8.5mM氢氧化铵(aq)并搅拌20分钟;然后,将另一部分二氧化硅源、50μL稀释的APTMS和200μL TEOS加入体系中,将其在20℃下搅拌20小时。搅拌过夜后,将100μL氢氧化铵(aq)(5.6-6重量%)缓慢地引入到混合物中,同时搅拌10分钟。接下来,向混合物中逐滴加入50μL TEOS并搅拌2小时,然后加入250μL PEG-硅烷和25μL TEOS并在20℃下搅拌20小时以修饰颗粒表面。之后,加入95%乙醇以使微乳液体系不稳定,并通过在14000rpm下离心20分钟收集颗粒。将颗粒用95%乙醇洗涤一次,并用D.I水洗涤两次,并转移到80mL D.I水中。然后,将溶液保持在40℃并搅拌1小时以获得过氧化氢酶@HSN。最后,通过离心收集过氧化氢酶@HSN,用D.I水洗涤两次,并储存在4℃的D.I水中。
TEM和DLS测量
对实施例1中合成的蛋白质@HSN纳米颗粒进行TEM测量,结果表明,所有蛋白质@HSN具有约50至95nm的平均粒径和小的粒度标准偏差,这反映了颗粒的均匀性。通过动态光散射(DLS)测量了蛋白质@HSN在不同溶液环境中的粒径。DLS结果显示,蛋白质@HSN在水、PBS和含血清的培养基中在约60至约100nm的范围内良好分散。
蛋白质@HSN的定量
包封于二氧化硅纳米颗粒中的蛋白质的量为约1%-7%(重量百分比)。使用上述方法学,定量结果源自酶活性或荧光相关光谱法。
蛋白酶耐受性测定
为了确定本文公开的HSN对包封于其中的ASNase免受蛋白酶降解的保护作用,对游离的ASNase或包封于HSN(NTT3_39)中的ASNase进行蛋白酶消化,并通过ASNase活性测定法来确定剩余的ASNase活性。因此,进行蛋白酶耐受性测试以评估HSN对包封于其中的ASNase的保护作用。
将游离ASNase和包封于HSN(NTT3_39)中的ASNase(均含有相同量的ASNase)离心并分散在1mL PBS缓冲液(pH7.5)中,与400μL蛋白酶溶液(2×10-3mg蛋白酶/mL,在10mMNaOAc+5mM CaOAc(pH7.5)中)混合,并在37℃下进行蛋白酶消化30分钟。在消化后,通过ASNase活性测定法测定样品中的ASNase活性。可以清楚地观察到,在用蛋白酶混合物降解30分钟后,游离ASNase的活性降低至低于原始活性的20%。然而,包封ASNase的NTT3_39表现出优异的保护作用。结果表明,一旦悬浮于溶液中,蛋白质(抗原)将被蛋白酶快速降解。因此,如果蛋白质通过非共价键附着于纳米颗粒,一旦颗粒被注射到体内,蛋白质将开始从颗粒泄漏出来,导致降解。HSN内包封的抗原能够提供优异的蛋白酶保护。
HSN淋巴结靶向
诱导适应性免疫的效率在很大程度上取决于免疫系统特别是淋巴结的适当靶向。淋巴结是抗原特异性T细胞被激活和致敏的部位。使用纳米颗粒将抗原直接递送到淋巴结将改善抗原特异性免疫反应的功效和安全性。对于开发具有淋巴结靶向能力的HSN,HSN的大小和在溶液中的悬浮很重要。将近红外荧光发射染料(cy5.5)缀合的HSN(尺寸为约50-100nm(通过TEM检测),并且在水和缓冲液中单分散)皮下注射到小鼠的后足垫中,并通过IVIS在不同时间点检测HSN的生物分布以探索HSN的淋巴结靶向能力。Cys5.5-HSN首先集中在注射部位,并在淋巴结中持续长达7天。在给药后第7天,将小鼠解剖,并且在腹股沟淋巴结、腘窝淋巴结、髂淋巴结和肾淋巴结处存在显著的荧光信号(图1)。这一结果表明,HSN的较小尺寸和良好的悬浮表现出淋巴结靶向能力,这对于疫苗的抗原递送媒介物是有利的。
蛋白质@HSN诱导免疫反应
用天冬酰胺酶包封的HSN(ASNase@HSN)(尺寸较小(约65nm)且在溶液中悬浮良好)来测试抗原特异性免疫反应。小鼠分别静脉注射、皮下注射或肌肉注射20μg ASNase或具有相同蛋白量的ASNase@HSN的200μL PBS溶液,每周一次,共3周;在第17天收获75-100μL血液,并通过ELISA测定法检测血清的ASNase特异性抗体水平。ASNase免疫小鼠的血清未显示ASNase特异性抗体信号。即使ASNase剂量是四倍时,也仅检测到少量抗体。相反,与ASNase相比,ASNase@HSN诱导显著量的ASNase特异性抗体(图2)。不同的ASNase@HSN给药途径显示相似水平的ASNase特异性抗体诱导。此外,与仅有抗原的组相比,用其它蛋白质诸如过氧化氢酶和辣根过氧化物酶作为抗原代替ASNase仍然显示出强烈的免疫反应。因此,被HSN包封的抗原显著增强了抗原的免疫原性,并允许各种给药途径。
Her2_4T1@HSN和HSN免疫治疗
为了证明抗原@HSN作为癌症疫苗的潜力,选择Her2胞外结构域蛋白作为抗原并包封于HSN中。为了证明Her2_ECD@HSN可以诱导免疫反应并抑制Her2相关的肿瘤细胞增殖,将Her2过表达的乳腺癌细胞皮下(s.c.)植入免疫活性BALB/c小鼠的胁腹中来构建动物模型。通过用编码人Her2 cDNA的逆转录病毒载体转导乳腺癌细胞系4T1,产生过表达Her2的乳腺癌细胞系(Her2_4T1)。BALB/c小鼠每隔一周静脉内接种Her2_ECD@HSN三次。在第三次疫苗接种后一天,将Her2_4T1细胞皮下植入小鼠的胁腹,并且每周两次监测肿瘤大小。与对照组相比,接种Her2_ECD@HSN的小鼠显示出对肿瘤生长的显著抑制(图3),且血浆中抗Her2_ECD抗体的量也高于对照组。这一结果表明,Her2_ECD@HSN可以成功诱导Her2特异性适应性免疫反应,以抑制肿瘤增殖。这证实了HSN包封的抗原可以提高免疫原性和免疫治疗功效。
根据上述免疫治疗结果,抗原@HSN可以显著增强免疫反应,因为HSN可以同时为载剂和自身佐剂。本发明中HSN的较小尺寸和良好悬浮给予诸如淋巴结靶向、肿瘤靶向(EPR作用)和自身免疫原性(自身佐剂)之类的特性。这些特性允许HSN潜在地用于另一种抗肿瘤生长的治疗方法中,其中将HSN颗粒(无抗原包封)静脉内施用到体内;颗粒能够在淋巴结和肿瘤中积累。之后,因为具有自身佐剂性质的HSN在肿瘤中积累,HSN将局部诱导免疫反应,并同时调节肿瘤微环境。增强免疫反应可激发抗肿瘤免疫,以抑制肿瘤生长。为了证明这一概念,在肿瘤植入后的第3、10、17天,在胁腹给小鼠皮下植入Her2_4T1细胞,并通过静脉内施用HSN进行治疗。与对照组相比,用HSN治疗的小鼠肿瘤体积较小。还测试了HSN在4T1肿瘤动物模型中的抗肿瘤功效,其显示HSN可抑制4T1肿瘤生长(图4)。相反,根据先前的实验,在仅用MSN(无药物)治疗的小鼠中肿瘤生长未被抑制。这些结果表明,HSN具有不同于其它二氧化硅纳米颗粒的一些特殊且独特的特性,使得HSN表现出更高的免疫原性和免疫治疗功效。
HSN在肿瘤中诱导局部免疫反应
携带4T1肿瘤的小鼠接种不同种类的HSN,每隔一周接种一次,接种三次,最后一次接种后一天,收集脾、淋巴结和肿瘤并消化用于后续的免疫细胞染色。接种HSN的小鼠脾和淋巴结中的免疫表型与对照组小鼠相似,相反,肿瘤中的CD8a+细胞、F4/80细胞和CD11c细胞数量增加,这说明细胞毒性T细胞、巨噬细胞、树突状细胞在肿瘤周围募集(图5)。这些结果表明,HSN的较小尺寸和良好的悬浮可以通过EPR效应在肿瘤中积累并募集肿瘤周围的免疫细胞,从而增强肿瘤部位的肿瘤浸润免疫细胞并抑制肿瘤生长,而无全身性不良反应。
新抗原@HSN的合成和提高包封肽的二氧化硅纳米颗粒的负载能力的方法
新抗原是一类由肿瘤特异性突变产生的HLA结合肽。它们可用作癌细胞从正常细胞分化的生物标志物。因此,新抗原肽是开发肿瘤免疫治疗抗原@HSN的良好抗原。用上述合成蛋白质@HSN的方法并将蛋白质替换为新抗原肽,来合成NeoAg@HSN。然而,出人意料地发现,肽不能有效地包封于HSN中。在微乳液去稳定化过程中,大多数新抗原肽悬浮在上清液中,而在HSN中几乎检测不到。这种出人意料的结果可能是由于以下事实:肽通常由疏水性和亲水性氨基酸残基组成;肽的两亲性可导致肽与含有聚乙二醇的表面活性剂(诸如CO520,Triton X-100和吐温20)之间发生强烈的相互作用。这些类型的非离子表面活性剂通常用于反相微乳液体系中。为了解决这一问题,提出了三种提高二氧化硅纳米颗粒的肽负载能力的方法:(1)将反相微乳液中使用的含聚乙二醇的表面活性剂替换为不含氧化烯单元的表面活性剂或离子型表面活性剂(诸如磺基琥珀酸二辛酯钠盐(AOT)、十六烷基三甲基溴化铵(CTAB));(2)设计所述肽是为了降低肽的两亲性,并增加肽与二氧化硅分子之间的相互作用,以使肽更易于被二氧化硅分子包围,并易于被二氧化硅纳米颗粒包裹。通过在肽序列之前、之后或两侧添加多电荷基序,并在多电荷基序与肽序列之间插入酶可裂解序列来设计肽;(3)将对新抗原肽具有亲和力的分子或颗粒加入微乳液的水相中,以提高肽的包封效率,诸如1、带正电荷的新抗原与带负电荷的分子之间的静电相互作用;2、疏水性新抗原与带电荷较少或不带电荷的分子之间的范德华相互作用;以及3、在新抗原和分子之间的可裂解共价键,包括二硫键、酶可裂解序列、酸可裂解部分。在加入到微乳液的水相中之前,分子和新抗原可以是单独的或预先混合的。分子可以是硅烷、聚合物、树枝状大分子或二氧化硅纳米颗粒。利用设计的新抗原肽可以成功合成NeoAg@HSN(在一个实施方案中,在原始新抗原肽的N末端添加聚精氨酸序列和酶可裂解序列。在一个实施方案中,在原始新抗原肽的N末端添加巯基氨基酸)且NeoAg@HSN中肽的量是可检测的。
正电荷修饰的新抗原@HSN的合成
合成过程:将20mL癸烷、3.5mL CO-520和1.1mL己醇混合,然后将溶解于350μL D.I水、250μL 10mg/mL NaF和25μL稀释的APTMS中的1-2mg正电荷部分修饰的新抗原的水相溶液加入油相,同时搅拌30分钟,得到微乳液体系;然后向体系中加入100μL TEOS并搅拌1小时。之后,加入25μL稀释的APTMS和100μL TEOS并在20℃下搅拌18小时。第二天,加入500μL28-30重量%NH3(aq)和100μL TEOS并在20℃下搅拌4小时,然后,加入250μL PEG-硅烷、25μLTEOS并在20℃下搅拌16-18小时。第三天,通过加入2倍体积的95%乙醇使微乳液体系不稳定并在14000rpm下离心20分钟来收集颗粒。将颗粒用95%乙醇洗涤两次,并用D.I水洗涤一次,并转移至80mL D.I水中。然后,将溶液保持在40℃下并搅拌1小时以除去多余的残余物。最后离心收集新抗原@HSN并用D.I水洗涤两次。新抗原@HSN中新抗原的负载量高于1%重量百分比,并且通过TE和MDLS测量的粒径示于表1中。

巯基部分修饰抗原@HSN的合成
合成方法有两个步骤。1、巯基部分修饰的抗原与含有邻二硫吡啶(OPSS)的二氧化硅纳米颗粒混合,以在抗原和二氧化硅颗粒之间产生二硫键。2、将Ag-二氧化硅纳米颗粒溶液作为水相引入微乳液体系中合成抗原@HSN。
1.抗原和OPSS-二氧化硅纳米颗粒的混合物
将50mg OPSS-二氧化硅纳米颗粒分散在6.3-7mL DI水中,然后向溶液中加入375μL乙酸盐缓冲液(100mM,pH4.2)酸性缓冲液或375μL NaH2PO4(100mM,pH6.5)和750μL NaOH(25mM)碱性缓冲液。将溶液搅拌至均匀,并加入75-150μL抗原溶液(10-20mg/mL)。在4℃或室温下保持搅拌1-3天。之后,用含有0.025%TFA和水的20%ACN/DMSO洗涤颗粒。将溶液离心以获得颗粒,并通过HPLC分析上清液以测定抗原负载量。
2.巯基部分修饰抗原@HSN的合成
将37.5mL癸烷、6.56mL Igepal CO-520和2.06mL己醇混合在一起,并在20℃下搅拌。向混合物中加入2250μL抗原-二氧化硅纳米颗粒(50mg/2250μL)作为水相,然后在20℃下搅拌10分钟。将37.6μL 8倍稀释的APTMS、150μL TEOS和83.34μL 28%NH4OH引入混合物中,在20℃℃搅拌10分钟。之后,向混合物中加入37.6μL 8倍稀释的APTMS、150μL TEOS和83.34μL 28%NH4OH,在20℃下搅拌4小时。然后,向混合物中加入37.6μL TEOS和375.2μLPEG-硅烷,并在20℃下搅拌过夜。向溶液中引入20mL乙醇以破坏微乳液体系,并在14000rpm下离心15分钟以获得抗原@HSN颗粒。将颗粒用乙醇和水洗涤两次,并储存在水中。粒径(TEM)小于100nm,并且在PBS中通过DLS测量的颗粒的流体动力学尺寸小于150nm。
NeoAg@HSN免疫原性(在CD8+T细胞中的IFN-γ表达)和免疫治疗
在第1天、第8天和第15天,分别用NeoAg溶液(含有50μg MC38_mS或MC38_mL新抗原肽,有或没有50μg聚IC)和NeoAg@HSN溶液(MC38_mS@HSN或MC38mL@HSN溶液,其含有50μg新抗原肽,有或没有50μg聚IC)免疫雌性C57BL/6小鼠(6-8周)。在第28天和第35天,从经过疫苗接种的小鼠收集约300μL外周血。检测CD8 T细胞中IFN-γ表达的方法如上所述。与用MC38_mS或MC38_mL肽免疫的小鼠相比,用MC38_mS@HSN或MC38_mL@HSN免疫的小鼠在CD8 T细胞中显示出显著更高的IFN-γ表达(图6)。这一结果表明,HSN可以作为载剂和自身佐剂,以增强新抗原肽的免疫原性。免疫接种结果显示,每隔一周用NeoAg@HSN免疫,免疫三次的小鼠中抑制了MC38肿瘤的生长,而用NeoAg肽+聚IC(佐剂)免疫的小鼠中肿瘤大小与对照组相近。

12.根据权利要求8所述的HSN缀合物,其中所述小生物活性成分被修饰以具有结构Y(n)-X-SBI-[X-Y(m)](r),其中Y为带正电荷的肽,X为酶可裂解序列,SBI为小生物活性成分,并且m、n和r中的每一个均为整数,其中n和m×r中至少一个不为零。
13.根据权利要求8所述的HSN缀合物,其中所述小生物活性成分被修饰以具有结构Y(n)-X(a)-SBI-X(b)-Y(m),其中Y为带正电荷的肽,X为酶可裂解序列,SBI为小生物活性成分,并且n、m和r中的每一个均为整数,其中n和m中至少一个不为零。
14.根据权利要求8所述的HSN缀合物,其中所述小生物活性成分被修饰以具有结构Z(c)-Y(n)-X(a)-SBI-X(b)-Y(m)-Z(d),其中Z为含有巯基的分子,Y为带正电荷的肽,X为酶可裂解序列,SBI为小生物活性成分,并且a、b、c、d、m和n中的每一个均为整数,其中c、d、n、m中至少一个不为零。
15.一种疫苗组合物,其包含根据权利要求8所述的HSN缀合物。
16.一种将小生物活性成分递送到受试者的方法,其包含向所述受试者施用根据权利要求8所述的HSN缀合物。
17.一种在免疫治疗中将抗原或新抗原递送到受试者的方法,其包含向受试者施用根据权利要求8所述的HSN缀合物,其中抗原或新抗原被包封于所述HSN中。
18.根据权利要求17所述的方法,其中所述新抗原为肿瘤特异性新抗原、肿瘤新表位、肿瘤特异性新抗原、肿瘤新表位、新抗原肽、新抗原DNA或新抗原RNA,其中所述抗原为病毒抗原、细菌抗原或微生物抗原。
19.一种用于产生含有于其中的一或多种生物活性成分的空心二氧化硅纳米颗粒的方法,其包含以下步骤:
(a)提供包含油相、表面活性剂、烷氧基硅烷和/或硅酸盐源、含有一或多种生物活性成分的水相以及任选的助表面活性剂的组合物,
(b)由步骤(a)中所描述的组合物形成油包水W/O微乳液;
(c)向(b)的所述W/O微乳液中加入引发剂,以形成封装所述一或多种生物活性成分的HSN;
(d)执行去稳定条件以使所述W/O微乳液不稳定并收集由所述微乳液如此形成的所得颗粒;以及
(e)将步骤(d)中所收集的所述颗粒分散于含水洗涤相中,得到所述二氧化硅纳米颗粒。
20.根据权利要求19所述的方法,其进一步包含以下特征中的至少一个:
(i)所述表面活性剂为离子型的;或者所述表面活性剂为非离子型的并且不存在氧化烯单元;
(ii)所述一或多种生物活性成分在使用前被氨基酸序列修饰,其中所述序列中的氨基酸为可以带正电荷或含有巯基的氨基酸;以及
(iii)对所述一或多种生物活性成分具有亲和力的物质被引入步骤(a)中的所述水相中。
21.一种根据权利要求19或20所述的方法产生的空心二氧化硅纳米颗粒。
