Abstract
本发明公开了一种含有跨膜IL‑21蛋白激活的细胞因子诱导杀伤性细胞(cytokine‑induced killer cells,CIK细胞)分泌的外泌体(Exosome)及其内含物MicroRNA(miRNA),其均具有高效的抗菌性能,为治疗细菌感染提供了新的方法。此方法:(1)不同于抗生素治疗,外泌体具有较低的副作用;(2)外泌体可低温储存,miRNA可以人工合成,成本低,可以实现大规模的生产;(3)对多种细菌均有抑制效果,具有宽泛的抗菌谱。

技术领域
本发明涉及通过经跨膜IL-21激活的CIK细胞分泌的外泌体(Exosome)及其相关miRNA在抗菌中的应用。
背景技术
CIK细胞是一种新型的过继免疫治疗细胞,它是利用人外周血单个核细胞在体外通过多种细胞因子诱导培养一段时间后获得的一群免疫活性细胞,具有T淋巴细胞特性和非主要组织相容性复合体(major histocompatibility complex,MHC)限制性和强大的抗肿瘤活性,因其具备增殖速度快、杀瘤活性高、杀瘤谱广等优越性,常用于细胞免疫治疗。
CIK细胞是免疫细胞的一种,和NK细胞相比,其细胞类型完全不同。CIK细胞表面同时表达CD3和CD56两种膜蛋白分子,而NK细胞表面不表达CD3,只表达CD56分子。如图2所示,CIK外泌体同时表达CD3和CD56两种膜蛋白分子,而NK外泌体表面不表达CD3,只表达CD56分子,这与细胞表面表达一致。
外泌体,英文名又为Exosome,是一种脂质双分子膜包裹的细胞分泌出的小囊泡,大小在30-150nm左右,是纳米级的细胞外囊泡(extracellular vesicles,EVs)。EV包含具有不同大小(直径100-1000nm)的微泡和外体,其容易通过从细胞质膜出芽而产生。与EV不同,外泌体由多囊泡内体或多泡体产生,通常直径小于150nm。当内体或多泡体与质膜融合时,外泌体分泌被激活。最初被认为是无用的代谢“垃圾”,后来发现其中含有大量的蛋白质、RNA、细胞因子等多种生物活性物质。并且对于抗原提呈细胞(Antigen-presentingcell,APC)和肿瘤细胞来源的外泌体研究发现,外泌体在体内和体外均表现出了很强的免疫刺激活性,越来越受到研究人员的关注。
MicroRNA是非编码小RNA,通常长度为19-24个核苷酸,并且是编码RNA的调节因子。它们在转录后水平通过结合特定区域中的候选靶标“结合位点”来负向调节mRNA,从而导致蛋白质翻译的抑制。每个microRNA作用于多个mRNA靶标,而任何mRNA可能被不同的microRNA阻断。这个错综复杂的网络致力于保持对几种生物功能的微调。由于癌症显示miRNA表达异常表达,它们的改变在肿瘤进展中担当关键的功能。最近的研究已经暗示肿瘤外泌体microRNA在细胞间通信中并且赋予肿瘤侵袭性。在不同的细胞微环境刺激下,细胞能主动、特异地将miRNA包裹,并通过外泌体的形式分泌到细胞外环境以响应不同的刺激。
在过去数十年中,抗生素的广泛使用使得微生物耐药频率和其与相关的严重感染性疾病的以令人担忧的速度增加。在革兰氏阳性菌中,最重要的耐药性病原体是耐甲氧西林的金黄色葡萄球菌、耐β-内酰胺的肺炎球菌和多药耐药性肺炎球菌,以及耐万古霉素的肠球菌。肺炎克雷伯氏菌、大肠杆菌和奇异变形杆菌等革兰氏阴性菌中的β-内酰胺酶耐药性是造成革兰氏阴性菌耐药性的重要原因。因此临床治疗迫切需要新颖的抗菌剂,尤其具有新颖作用机制的抗菌剂。
本发明发现具有广泛的、强大的杀伤肿瘤细胞作用的CIK细胞,其外泌体不仅富含CIK细胞的胞浆成分,还富含大量的miRNA。这些miRNA在体外实验中可以显著的抑制细菌的生长,具有出色的抗菌功能。
发明内容
本发明的目的在于提供经跨膜IL-21激活的CIK细胞分泌的外泌体(Exosome)以及所含的miRNA在抗菌中的应用。
本发明的目的是通过以下技术方案实现的:一种经跨膜IL-21共同激活的CIK细胞分泌的外泌体。
进一步地,通过如下方法激活得到:配制CIK培养液,培养CIK细胞。其中CIK培养液中,含有IL-2(500U/ml),CD3(50ng/ml),IFN-γ(1000U/ml)和跨膜IL-21(100U/ml),至少培养24天后,从CIK细胞培养液上清中分离得到CIK外泌体。
进一步地,所述外泌体包含一条或一条以上选自SEQ ID NO.1~SEQ ID NO.1079所示,或下表中1080~2246号所示的miRNA。
本发明还提供上述外泌体在抗菌中的应用。
本发明还提供具有SEQ ID NO.1~SEQ ID NO.1079所示的任一序列的miRNA。
本发明还提供SEQ ID NO.1~SEQ ID NO.1079所示序列的任一条miRNA,或1080~2246号miRNA中任一序列的miRNA在抗菌中的应用。
进一步地,上述应用为:用于制备抗菌制剂。
进一步地,针对的细菌选自肺结核杆菌、嗜碱芽孢杆菌、解淀粉芽孢杆菌、短芽孢杆菌、环状芽孢杆菌、克劳氏芽孢杆菌、凝结芽孢杆菌、灿烂芽孢杆菌、迟缓芽孢杆菌、地衣芽孢杆菌、巨大芽孢杆菌、嗜热脂肪芽孢杆菌、枯草芽孢杆菌、苏云金芽孢杆菌、棒状杆菌属、变铅青链霉菌、鼠灰链霉菌、酿脓链球菌、间擦疹链球菌、海豚链球菌、肺炎链球菌、金黄色葡萄球菌、中间葡萄球菌、伪中间葡萄球菌、表皮葡萄球菌、屎肠球菌、伤寒沙门氏菌、大肠杆菌、霍乱弧菌、脑膜炎奈瑟氏菌、淋病奈瑟氏菌和绿脓杆菌中的一种或多种。
本发明的有益效果在于:本发明发现对CIK细胞分泌的外泌体包含的序列为SEQID NO.1~SEQ ID NO.1079的全新miRNA和1080~2246号的已知miRNA进行测试,发现具有抗菌能力,可用于抗菌,该抗菌手段:(1)不同于抗生素治疗,外泌体具有较低的副作用;(2)可通过联合使用增强治疗效果;(3)外泌体可低温储存,miRNA可以人工合成,成本低,可以实现大规模的生产;(4)对多种细菌均有抑制效果,具有宽泛的抗菌谱。

具体实施方式
下面结合实施例对本发明作进一步说明。
实施例1:免疫细胞中的单核细胞外泌体不具备抗菌性能
为了确定免疫细胞中的单核细胞外泌体是否也能抑制细菌生长,我们选取单核细胞外泌体作为研究对象,同时将CIK外泌体和NK外泌体作为阳性对照。通过实施例2的方法将三者外泌体按照不同比例和上述四种细菌混合,通过测量光密度(OD)量化细菌的数量。结果所示,CIK细胞和NK细胞外泌体针对伤寒沙门氏菌(CT18)具备抗菌性能,如图4A所示;针对金黄色葡萄球菌(LAC)具备抗菌性能,如图4B所示;针对柠檬酸杆菌(C.R.)具备抗菌性能,如图4C所示;针对大肠杆菌(E.COLI)具备抗菌性能,如图4D所示。而单核细胞外泌体针对伤寒沙门氏菌(CT18)没有抗菌功能,如图4A所示;针对金黄色葡萄球菌(LAC)没有抗菌功能,如图4B所示;针对柠檬酸杆菌(C.R.)没有抗菌功能,如图4C所示;针对大肠杆菌(E.COLI)没有抗菌功能,如图4D所示。
由此可见,根据NK细胞外泌体具有抗菌性能,不能预见任意的包括CIK在内的免疫细胞都具有抗菌性能。
实施例2:CIK细胞培养、外泌体的获取及其结构特征
配制A和B两种CIK培养液,用两种培养液分别培养出CIK细胞。其中CIK培养液A中,含有IL-2(500U/ml),CD3(50ng/ml),IFN-γ(1000U/ml);其中CIK培养液B中,含有IL-2(600U/ml),CD3(50ng/ml),IFN-γ(1000U/ml)和跨膜IL-21(100U/ml)。经过24天培养后,从这两种CIK细胞培养液上清中分离得到CIK外泌体-A和CIK外泌体-B。
用CIK培养液A培养CIK细胞的步骤如下:
第1天,从正常人外周血中提取2×10^7的PBMC,细胞计数,加20毫升培养液A进行培养;
第3天,补充CIK培养液A;
第5天,补充CIK培养液A;
第8天,提纯出CD3+CD56+双阳细胞,加CIK培养液A;
第24天,离心法收集细胞培养液上清。300g离心,10分钟,4℃,保留上清。
用CIK培养液B培养CIK细胞的步骤如下:
第1天,从正常人外周血中提取2×10^7的PBMC,细胞计数,加20毫升培养液B进行培养;
第3天,补充CIK培养液B;
第5天,补充CIK培养液B;
第8天,提纯出CD3+CD56+双阳细胞,加CIK培养液B;
第24天,离心法收集细胞培养液上清。300g离心,10分钟,4℃,保留上清。
采用离心法对收集的细胞培养液上清中的外泌体进行纯化收集,具体步骤如下:
1.2.1300g离心10min,4℃,取上清
1.2.22000g离心10min,4℃,取上清
1.2.310,000g离心30min,4℃,取上清
1.2.4100,000g离心70min,4℃,取沉淀。
1.2.5加入到PBS中,加入DTT(二硫苏糖醇,终浓度5-50mM/L);清洗;100,000g离心70min,4℃,取沉淀。
1.3外泌体-A和外泌体-B粒径分布图
将获取的外泌体-A和外泌体-B分别用PBS稀释100倍,利用Zetasizer Nano ZSE系统检测外泌体的粒径分布,并用Dispersion Technology Software对检测结果进行分析。如图1A和图1B所示,外泌体-A和外泌体-B分布在80-210nm。
另外,跨膜IL-21可以直接加入,也可以通过滋养细胞(K562)来引入,通常与培养的CIK细胞按照1:1培养。
实施例3:外泌体-A和外泌体-B抗菌性能
为了比较外泌体-A和外泌体-B抑制细菌生长的差异性,我们选取了伤寒沙门氏菌(CT18),金黄色葡萄球菌(LAC),柠檬酸杆菌(C.R.),大肠杆菌(E.COLI)作为检测对象。将实施例2获得的两种CIK外泌体分别按照不同比例和上述四种细菌混合,通过测量光密度(OD)量化细菌的数量。
结果如图3A所示,可以看出,外泌体-B比外泌体-A抑制的效果显著增强。另外,通过大量实验证明,在对CIK外泌体的扩增激活过程中,培养液中的组分对于外泌体的抗菌效果具有重要影响。培养液中缺失或增加组分,或者改变本发明的培养液中的组分含量均不利于获得高抗菌效能的外泌体。
另外,随着浓度的增加,外泌体-B显著抑制伤寒沙门氏菌(CT18),金黄色葡萄球菌(LAC),柠檬酸杆菌(C.R.),大肠杆菌(E.COLI)的生长,如图3B所示。
实施例4:CIK外泌体miRNA测序
根据实施例2获得的CIK外泌体-B,委托南京世和基因生物技术有限公司对CIK外泌体中miRNA进行测序。采用NEBNext Multiplex Small RNA Library Prep Set forIllumina(NEB,USA)进行建库,并对待测序样本添加测序接头。采用Illumina X-ten PE150平台对样本进行测序,每个样本至少300M reads。为了减少假阳性,信噪比大于100的独立序列纳入miRDeep log-odds score的计算,并设置miRDeep log-odds score的范围为-10~10.为了发掘CIK外泌体中全新的miRNA,以miRDeep score=0作为cut-off值,所有超过cut-off值的miRNA作为全新的miRNA。在测序中发现了1167个已知的miRNA(1080~2246号)。除此之外,全新序列的miRNA有1079个(SEQ ID NO.1~SEQ ID NO.1079)。
实施例5:CIK外泌体中已知序列的miRNA抗菌性能
为了确定外泌体miRNA是否可以抑制细菌生长,选取柠檬酸杆菌(C.R.),大肠杆菌(E.COLI),伤寒沙门氏菌(CT18),金黄色葡萄球菌(LAC)作为检测对象。
将实施例4筛选得到的已知序列的miRNA随机分成8组,每一组中随机选出30个样本进行抗菌测试,结果表明,全部具有抗菌性能(金黄色葡萄球菌、大肠杆菌、柠檬酸杆菌),可以确定实施例4筛选得到的已知序列的miRNA均具有抗菌性能。下面以这240个样本中的hsa-miR-376c-3p(1095号)、hsa-miR-103a-2-5p(1114号)、hsa-miR-15a-5p(1083号)、hsa-miR-106a-3p(1085号)、hsa-miR-548e-3p(1093号)、hsa-miR-598-3p(1082号)、hsa-miR-196b-5p(1084号)、hsa-miR-548d-5p(1089号)、hsa-miR-130a-3p(1087号)、hsa-miR-374a-3p(1088号)为例,人工合成这些miRNA类似物,进行抗菌试验如下:
(1)将上述10种类似物按照2.5μM的浓度和四种细菌共孵育,通过检测OD600计算细菌生长抑制率,发现hsa-miR-376c-3p、hsa-miR-598-3p、hsa-miR-196b-5p、hsa-miR-548d-5p、hsa-miR-130a-3p、hsa-miR-374a-3p这6种miRNA具有抑菌效果。测试结果如表1所示。
(2)根据上面结果,挑选hsa-miR-376c-3p类似物以2.5μM浓度和三种细菌共孵育,通过检测OD600量化细菌的数量。测试结果如图5,hsa-miR-376c-3p针对金黄色葡萄球菌(LAC)具备抗菌性能,如图5A所示;针对柠檬酸杆菌(C.R.)具备抗菌性能,如图5B所示;针对大肠杆菌(E.COLI)具备抗菌性能,如图5C所示。
通过与阴性对照PBS组和NC组对比,可以证明,这随机挑选的hsa-miR-376c-3p能抑制细菌的生长,对三种菌都有较为明显的抑制效果。

实施6:CIK外泌体全新序列的miRNA的抗菌性能
将实施例4筛选得到的全新序列的miRNA随机分成10组,每一组中随机选出30个样本进行抗菌测试,结果表明,全部具有抗菌性能,可以确定实施例4筛选得到的全新序列的miRNA均具有抗菌性能。下面以这300个样本中的chr13_30450(SEQ ID NO.75)、chr13_30113(SEQ ID NO.88)、chr10_20084(SEQ ID NO.80)、chr9_18346(SEQ ID NO.83)、chr8_19806(SEQ ID NO.94)、chrX_42223(SEQ ID NO.81)、chr18_31118(SEQ ID NO.87)、chr5_10926(SEQ ID NO.79)、chr21_33580(SEQ ID NO.86)、chr9_18349(SEQ ID NO.89)为例,人工合成这些miRNA类似物,进行抗菌试验如下:
(1)将上述10种类似物按照2.5μM和四种细菌共孵育,通过检测OD600计算细菌生长抑制率,发现chr8_19806、chr18_31118、chr5_10926、chr21_33580、chr9_18349这5种miRNA具有抑菌效果。测试结果如表1所示。
(2)根据上述结果,挑选chr18_31118类似物以2.5μM浓度和三种细菌共孵育,通过检测OD600量化细菌的数量。测试结果如图5;针对金黄色葡萄球菌(LAC)具备抗菌性能,如图5A所示;针对柠檬酸杆菌(C.R.)具备抗菌性能,如图5B所示;针对大肠杆菌(E.COLI)具备抗菌性能,如图5C所示。
通过与阴性对照PBS组和NC组对比,可以证明,挑选的chr18_31118能显著的抑制细菌的生长,并且chr18_31118对三种菌生长的抑制都存在效果。

Claims (6)
1.一种经跨膜IL-21共同激活的CIK细胞分泌的外泌体,通过如下方法激活得到:配制CIK培养液,培养CIK细胞;其中CIK培养液中,含有500U/ml的IL-2,50ng/ml的CD3,1000U/ml的IFN-γ和100U/ml跨膜IL-21,至少培养24天后,从CIK细胞培养液上清中分离得到CIK外泌体。
3.如权利要求1所述的外泌体在抗菌中的应用;针对的细菌选自伤寒沙门氏菌、金黄色葡萄球菌、柠檬酸杆菌、大肠杆菌。
4.hsa-miR-598-3p在抗菌中的应用,所述hsa-miR-598-3p的序列为:UACGUCAUCGUUGUCAUCGUCA;针对的细菌选自伤寒沙门氏菌、金黄色葡萄球菌、柠檬酸杆菌、大肠杆菌。
5.权利要求3所述的外泌体在制备抗菌制剂中的应用,针对的细菌选自伤寒沙门氏菌、金黄色葡萄球菌、柠檬酸杆菌、大肠杆菌。
6.权利要求4所述的hsa-miR-598-3p在制备抗菌制剂中的应用,针对的细菌选自伤寒沙门氏菌、金黄色葡萄球菌、柠檬酸杆菌、大肠杆菌。
