(1) Adding 25% DMSO and 500U/mL hyaluronidase into 0.1mol/L acetate buffer solution, adding 0.25mL of compound essential oil with different concentrations, 1g of compound essential oil nanocapsule and 2.5mmol/L CaCl2Incubate at 37 ℃ for 20 min.

as can be seen from Table 4, each of the compositions exhibited anti-allergic effects, and composition 5 exhibited the highest anti-allergic activity. Compared with the compositions 1, 2 and 3 respectively, the anti-allergic activity of the composition 4 is obviously improved, which shows that the anti-allergic activity of the combination of 3 natural plant essential oils is improved, and the activity of the combination is reduced if any one of the natural plant essential oils is absent. The composition 4 is prepared into nanocapsules, the hyaluronic acid inhibition rate of the nanocapsules is obviously higher than that of the composition 4, the essential oil in the compositions 1-4 is volatile and has short action time, and the prepared nanocapsules can overcome the defect and improve the anti-allergy activity of the nanocapsules.
3. And (4) conclusion: the compound essential oil and the nanocapsules thereof have high hyaluronic acid inhibition rate, show high anti-allergy activity and play a role in quickly resisting allergy and maintaining stability of damaged skin.
Test example 2
The compound essential oil of the invention has in vitro inhibitory activity on staphylococcus and propionibacterium acnes:
1. materials and methods
1.1 test materials
The preparation method of each composition was the same as in test example 1.
1.2 test methods
(1) Preparing strain liquid: 2 strains of staphylococcus and propionibacterium acnes are taken, are respectively cultured for about 24 hours at 37 ℃ on a broth agar inclined plane, and are shaken and stirred uniformly to be used as strain liquid.
(2) Preparing a culture medium plate containing bacteria: cooling the BS culture medium sterilized by high-pressure steam to 45-50 ℃, and respectively adding 105-106And (3) fully shaking and uniformly mixing the CFU/mL staphylococcus and propionibacterium acnes, pouring the mixture into a sterilized culture dish, and cooling to obtain the bacterium-containing culture medium plate.
(3) And (3) measuring the inhibition zone: after the BS culture medium containing staphylococcus and propionibacterium acnes is solidified, a hole puncher with the diameter of 7mm is used for punching, four holes can be punched in each culture dish, 20 mu L of compound essential oil and 80 mu g of compound essential oil nanocapsules are added into each hole, and the holes are cultured in an incubator at 37 ℃ for 24 hours, and then the diameter of the inhibition zone is measured. Each composition was tested 10 times and the mean and standard deviation calculated.
2. Results and analysis
The bacteriostatic activity of each composition against staphylococci and propionibacterium acnes is shown in table 5.

as can be seen from Table 5, each of the compositions had inhibitory effects against Staphylococcus and Propionibacterium acnes, and composition 5 had the best bacteriostatic effect. Compared with the compositions 1, 2 and 3, the bacteriostatic activity of the composition 4 is obviously improved, which shows that the bacteriostatic activity of the combination of the 3 natural plant essential oils is improved, and the activity of the combination is reduced if any one of the natural plant essential oils is absent. The composition 1-4 cannot play the best antibacterial effect because the essential oil is volatile, and the compound essential oil is prepared into the nanocapsule, so that the exertion of the essential oil is reduced, and the antibacterial effect can be exerted durably and stably.
3. And (4) conclusion: the compound essential oil nanocapsule can effectively inhibit staphylococcus and propionibacterium acnes, so that the effect of removing acnes for a long time is achieved.
Test example 3
The compound essential oil of the invention has the following inhibiting effect on melanin pigmentation:
1. materials and methods
1.1 test materials
The preparation method of each composition was the same as in test example 1.

(1) Culturing of B16 cells: b16 cells at 95% humidity, CO2The culture was carried out in air at a concentration of 5% in DMEM containing 10% FBS at 37 ℃.
(2) Determination of melanin content: b16 cells at 5X 10 per well4The density of the seed is inoculated inAfter culturing for 24h on a 24-pore plate, removing the culture medium on the upper layer after the cells are completely attached to the wall, adding 500 mu L of each composition containing medicine for culturing, setting a normal group and a positive control group, continuously culturing for 48h, removing the supernatant, washing for 5 times by PBS (phosphate buffer solution), digesting the cells by trypsin, collecting the cells into a centrifuge tube, centrifuging for 10min at 10000r/min, removing the supernatant, dissolving melanin by 150 mu L of 2mol/L NaOH of 10% DMSO (sodium hydroxide), preserving the melanin in water bath at 80 ℃ for 2h, finally sucking 100 mu L of the dissolved solution, transferring the dissolved solution into a 96-pore plate, measuring the content of the melanin by the absorbance of the supernatant of each pore at the wavelength of 405nm, wherein the smaller the content is represented by the percentage relative to the absorbance of a blank control, and the stronger the inhibition effect of the sample on melanin pigmentation is proved.
2. Results and analysis
The inhibitory effect of each composition on melanin pigmentation is shown in table 6.

as can be seen from table 6, each composition had an inhibitory effect on melanin pigmentation, and No. 5 had the best effect on the inhibition of pigmentation. The lowest melanin content of composition 4 compared to compositions 1, 2, and 3, respectively, indicates that the melanin content is significantly reduced in the combination of 3 natural plant essential oils, and the inhibition is reduced in the absence of any one. The composition 1-4 has higher melanin content than composition 5 due to volatile essential oil, and can reduce volatilization of essential oil and inhibit melanin pigmentation.
