Description
技术领域
本发明涉及能够促进小胶质细胞(microglia)向抗炎M2表型极性转化的极性转化促进剂。
背景技术
作为一种胶质细胞的小胶质细胞可认为是来源于中胚层的由卵黄囊分化而来的细胞。已知小胶质细胞具有作为受损细胞的去除、突触的维护检查等调整脑内外环境的免疫细胞的作用。例如,在保护大脑免受外伤性脑损伤的情况下,小胶质细胞活化并释放炎症细胞因子,从而使星形胶质细胞变化为反应性星形胶质细胞,发挥保护大脑的作用。另外,已知活化后的小胶质细胞诱发脑炎,促进神经退行性疾病。
另一方面,公开了将小胶质细胞作为靶点的神经退行性疾病的治疗方法。例如,提出了使用作为一种甘草次酸衍生物的间隙连接阻断剂的方法(非专利文献1、专利文献1)、使用异α酸的方法(非专利文献2)等。
已知小胶质细胞存在两种活化类型(非专利文献3)。即如图1所示,存在促炎M1表型小胶质细胞和抗炎M2表型小胶质细胞。促炎M1表型小胶质细胞产生白细胞介素-1β(IL-1β)、肿瘤坏死因子(TNF-α)、诱导型一氧化氮合酶(iNOS)等。IL-1β是效应因子,在IL-1β的存在下,小胶质细胞引起脑内炎症,释放出自由基活性氧等自由基。另外,在产生IL-1β的小胶质细胞中,吞噬老化(破坏的)细胞等代谢废物的能力降低。因此,促炎M1表型小胶质细胞对神经细胞产生伤害。而抗炎M2表型小胶质细胞产生精氨酸酶-1(ARG1)、白细胞介素-10(IL-10)、白细胞介素-4(IL-4)、转化生长因子(TGF)-β1等。ARG1是效应因子,通过释放ARG1,从而作用于保护神经细胞。抗炎M2表型小胶质细胞在抑制脑内炎症的同时,吞噬老化细胞等代谢废物的能力高,因此,有抗炎作用、即对神经细胞有保护作用。
然而,本发明人等在专利文献2中提供了新型的环状肽衍生物。虽然已知该环状肽衍生物是从作为一种冬草夏草的细脚拟青霉(Paecilomyces tenuipes)中提取的,对星形胶质表现出具有增殖活性,但尚不知其可作用于小胶质细胞并促进向抗炎M2表型小胶质细胞的极性转化。

本发明的目的在于提供新型的向对神经细胞有保护作用的抗炎M2表型小胶质细胞的极性转化促进剂。
用于解决问题的技术手段
本发明的实施方式涉及的向抗炎M2表型小胶质细胞转化的极性转化促进剂包含由下述通式(1)所示的坏状肽衍生物。

具体实施方式
本实施方式涉及的向抗炎M2表型小胶质细胞转化的极性转化促进剂(以下,也简称为“极性转化促进剂”)含有由以下通式(1)所示的环状肽衍生物作为有效成分。

式中,m为0~3的整数,n为1以上的整数。R1~R6分别独立地为氢原子或烃基。R7和R8分别独立地为羧基或其盐或者烷氧基羰基。R9为烃基、羟基、烷氧基或烷基羰氧基。R10和R11分别独立地为氢原子、烃基或烷基羰氧基。R12~R16分别独立地为氢原子或烃基。
在此,作为烃基,例如可举出:脂肪族烃基即直链状或支链状的饱和或不饱和的烃基、或者脂环式的烃基。烃基的碳原子数并无特别限定,优选碳原子数为1~6,更优选碳原子数为1~4。烷氧基、烷氧基羰基、烷基羰氧基中的烃部分也是同样的。作为优选的例子,烃基和烃部分分别为碳原子数1~4的烷基。
关于R7和R8,作为羧基的盐,例如可例示:碱金属盐、碱土金属盐等金属盐、乙酸铵盐等铵盐、二乙胺盐等胺盐等。
式(1)中,例如可以是:R1、R2、R3及R4分别独立地为烷基,n=2~4,R5和R6分别为氢原子,R7和R8分别独立地为羧基或其盐。
式(1)中,例如可举出以下的情况:R1、R2、R3及R4分别独立地为烷基,特别优选为甲基和乙基中的任一种,n=2~4,R5和R6分别为氢原子,R7和R8分别独立地为羧基或其盐,m=0,R10和R11分别为氢原子,R12和R13分别为氢原子,R14和R15分别独立地为氢原子或烷基(特别优选为甲基)中的任一种,R16为氢原子。
在一个实施方式中,作为由式(1)所示的环状肽衍生物,可以是由下述式(2)所示的化合物或其盐。

该由式(2)所示的化合物是包含N-甲基-β-羟基多巴、缬氨酸、β-羟基亮氨酸、谷氨酸这4种氨基酸的肽为环状结构的化合物。
由式(1)所示的环状肽衍生物的制造方法没有特别限定,例如,如专利文献2(WO2016/047638)所记载,可以使用各种提取、分离方法从细脚拟青霉中(日语:ハナサナキタケ)采集,也可以通过将肽合成等各种公知的化学合成方法组合来制造。
由式(1)所示的环状肽衍生物具有促进使小胶质细胞从促炎M1表型极性转化为抗炎M2表型的作用,因此能够用作向抗炎M2表型小胶质细胞的极性转化促进剂。即,通过使由式(1)所示的环状肽衍生物作用于作为胶质细胞中的一种的小胶质细胞,从而能够使小胶质细胞极性转化(极化)为抗炎性的M2表型,通过成为M2表型,从而能够使小胶质细胞对神经细胞起保护作用。因此,本实施方式涉及的极性转化促进剂能够利用在用于治疗、预防伴随着脑部疾病或老化而产生的神经细胞伤害的医药品或食品中。
在一个实施方式中,上述极性转化促进剂可以作为医药组合物或食品组合物的有效成分而配合于其中,也可以提供用于促进向抗炎M2表型小胶质细胞的极性转化的医药组合物或食品组合物。这些医药组合物或食品组合物例如能够用于人或除人以外的哺乳动物(实验动物、宠物等)。
医药组合物可以使用将上述极性转化促进剂与作为医药品而允许的各种添加剂等或其他成分进行混合等公知技术来制造。医药组合物能够经口给药或非经口给药,作为其形态,若是经口给药用,则例如可举出:片剂、丸剂、粉剂、颗粒剂、胶囊剂、液状制剂(包含酏剂、糖浆剂、悬浮剂、溶液剂)等,若是非经口给药用,则例如可举出:注射剂、点滴剂等,也可以向脑内直接给药。
食品组合物能够使用将上述极性转化促进剂与作为食品而允许的各种添加剂等或其他成分进行混合等公知技术来制造。作为食品组合物,可举出:保健功能食品(营养功能食品、特定保健用食品及机能性食品)、膳食补充剂等。作为食品组合物的形态的例子,可举出:与经口给药用的医药组合物同样的形态,另外也可以是饮料、点心等形态。
作为能够配合在医药组合物或食品组合物中的添加剂,例如可举出:赋形剂、抗氧化剂,香料、调味料、甜味剂、着色剂、增稠稳定剂、发色剂、漂白剂、胶基、乳化剂、结合剂,稀释剂、防腐剂、稳定剂、凝固剂等。
医药组合物的每次给药或每天的有效成分量以及食品组合物的每餐或每天的有效成分量能够根据给药或摄取对象的年龄、体重、性别、所应用的疾病或状态等,或者基于非临床或临床的试验结果等,而适当进行设定。虽未特别限定,但作为上述极性转化促进剂的经口摄取量,以式(1)的环状肽衍生物的量计,例如,对于包括人在内的哺乳动物,每天可以为0.1μg/kg以上且50μg/kg以下,也可以为1μg/kg以上且25μg/kg以下。
另外,上述极性转化促进剂在体外(in vitro)的实验体系中即使是低浓度也可得到效果。体外的实验体系中的上述极性转化促进剂的浓度没有特别限定,以式(1)的环状肽衍生物的浓度计,例如可以是0.01μM以上且5μM以下(即1×10-8~5×10-6mol/L),可以是0.01μM以上且1μM以下,也可以是0.03μM以上且0.3μM以下。
实施例
1.MG6小胶质细胞的细胞培养
在使用了MG6小胶质细胞的细胞株(Riken Cell Bank)的以下试验中,MG6小胶质细胞的培养是使用Dulbecco′s Modified Eagle Medium(杜尔贝科改良伊格尔培养基)(DMEM,Thermo Fisher Scientific(赛默飞世尔科技))进行的。在DMEM中包含100μM的β-巯基乙醇、10μg/ml的胰岛素、10%胎牛血清(Gibco)、青霉素-链霉素(Gibco)、450mg/ml葡萄糖(Gibco)。
2.MG6小胶质细胞的细胞存活率的测定
将MG6小胶质细胞在96孔板中以37℃培养24小时(5×103细胞/孔),接着,与各种浓度的环状肽衍生物一同以37℃培养48小时。作为环状肽衍生物,使用了通过专利文献2的实施例所记载的方法而得到的由上述式(2)所示的化合物的二乙胺盐(以下的各试验中相同)。作为环状肽衍生物的浓度,调整至培养液中的浓度为0μM、0.01μM、0.03μM、0.1μM、0.3μM、1μM。
细胞存活率的评价使用Cell-Counting Kit-8(CCK-8;Dojindo,Kumamoto,Japan(日本同仁化学研究所)),按照制造商的指示如下所述地进行。即,在用环状肽衍生物进行处理后,将10μL的CCK-8移送至96孔板,接着,以37℃培养2小时。根据该指示,使用酶标仪(日语:マイクロブレ一トリ一ダ一)以波长450nm读取光学浓度。细胞存活率使用以下的式子来计算。细胞存活率=(实验组的光学浓度/对照组的光学浓度)×100%
在环状肽衍生物给药48小时后,研究小胶质细胞的存活率,结果如图2所示,在环状肽衍生物的浓度为0.01~1μM的范围内没有确认到小胶质细胞的增殖,且其存活也未被抑制。于是,在以后的实验中,将环状肽衍生物的最佳浓度设定为1μM,用于功能性解析。另外,图2中的“ns”是指无统计学意义。
3.MG6小胶质细胞的极化试验
作为阿尔茨海默认知症的脑内的老年斑成分的嗜铬粒蛋白A(Chromogran in A,CGA)的神经毒性强于淀粉样蛋白β。另外,在蓄积有CGA的老年斑的周围确认到大量小胶质细胞,可认为原因在于小胶质细胞的活化(非专利文献4)。因此,作为接近脑内环境的条件,在存在CGA的病态条件下,进行基于环状肽衍生物的小胶质细胞的极化试验。
将MG6小胶质细胞在96孔板中以37℃培养24小时(5×103细胞/孔),接着,在添加CGA(Peptide Institute,Osaka)至浓度达到10nM的同时,添加环状肽衍生物达到浓度为1μM,以37℃进行培养。对于未添加环状肽衍生物而添加了CGA者以及未添加环状肽衍生物和CGA者(对照组),也同样进行了培养。
在添加处理24小时后和72小时后,进行实时定量PCR(RT-PCR)分析和统计分析,求出白细胞介素-1β(IL-1β)和精氨酸酶-1(ARG1)的基因表达量。已知受到刺激的小胶质细胞根据所产生的因子而极化为对神经细胞产生伤害的M1表型以及保护神经细胞的M2表型,M1表型小胶质细胞产生IL-1β,M2表型小胶质细胞产生ARG1(非专利文献3)。因此,根据IL-1β的基因表达来确定是M1表型,根据ARG1的基因表达来确定是M2表型。实时定量PCR(RT-PCR)分析和统计分析的方法如下所述。
[实时定量PCR(RT-PCR)分析]
从用环状肽衍生物处理后的MG6小胶质细胞的细胞株分离出mRNA,使用RNAisoPlus(Takara,Japan)根据制造者的指示提取总RNA。使用QuantiTect ReverseTranscription试剂盒(Qiagen,Japan),将合计800ng的提取出的RNA逆转录成cDNA。以95℃进行5分钟的最初的变性工序后,开始温度循环。各循环中包含:以95℃进行5秒钟的变性、以60℃进行10秒的复性和30秒的延伸。合计进行40次循环。使用利用了Corbett Rotor-Gene RG-3000AReal-Time PCR System的Rotor-Gene SYBR Green RT-PCR试剂盒(Qiagen,Japan),将cDNA进行2轮扩增。数据使用RG-3000A软件程序(版本Rotor-Gene 6.1.93、Corbett)进行评价。以下记载引物对(Invitrogen公司制,Custom primer)的序列。
IL-1β:5′-CAACCAACAAGTGATATTCTCCATG-3′(序列编号1)、以及5′-GATCCACACTCTCCAGCTGCA-3′(序列编号2);
ARG:5′-CGCCTTTCTCAAAAGGACAG-3′(序列编号3)、以及5′-CCAGCTCTTCATTGGCTTTC-3′(序列编号4)
为了数据的标准化,评价内源性对照(肌动蛋白)以控制cDNA的输入,根据比较Ct法计算相对单位。将全部的实时RT-PCR实验重复3次,将结果表示为平均±SEM。
[统计分析]
数据表示为平均±SEM。通过使用利用了GraphPadPrism软件包的事后图基(posthoc Tukey)检测的一元或二元方差分析(ANOVA),进行统计分析。可以认为p<0.05的值具有统计学意义(GraphPad软件)。
结果是,对于添加了CGA和环状肽衍生物者(实施例)以及仅添加CGA者(比较例),以相对于对照组(没有添加环状肽衍生物和CGA)的mRNA的相对表达量的形式,示于图3。
在用CGA与环状肽衍生物一同进行处理的情况下,虽然在添加处理24小时后,IL-1β的基因表达比ARG1的基因表达高约5倍,小胶质细胞极化为促炎M1表型,但是在72小时后反转,变为ARG1的基因表达比IL-1β的基因表达高约9倍。该结果表明,作为小胶质细胞的极化,通过环状肽衍生物的长时间处理而极化为抗炎M2表型,对神经细胞有保护作用。另外,可以认为,通过暂时极化为促炎M1表型,能够短时间地提高小胶质细胞的活力,高效且快速地极化为抗炎M2表型。
另外,与仅用CGA进行处理的情况相比,在用CGA与环状肽衍生物一同进行处理的情况下,虽然在添加处理24小时后,IL-1β的基因表达量高达约4倍,促炎M1表型的活化效果高,但是在添加处理72小时后,促炎M1表型的活性降低至与仅用CGA处理的情况相比没有显著性差异的程度。另一方面,对于ARG1的基因表达量,在用CGA与环状肽衍生物一同进行处理的情况下,与仅用CGA进行处理的情况相比,添加处理24小时后显示高达约4倍的高活化效果,添加处理72小时后也显示2倍以上的高活化效果。由此可知,在接近脑内环境的存在CGA的环境(即,小胶质细胞活化的病态条件下),通过长期处理环状肽衍生物,向对神经细胞有保护作用的抗炎M2表型小胶质细胞的极性转化得以促进。
小胶质细胞是处于控制脑内环境和脑炎的脑内免疫第一线的细胞。M2表型小胶质细胞具有收集在脑内死亡的神经细胞或在脑内蓄积的淀粉样蛋白等代谢废物并排出到脑外的能力。进而,在脑炎时,M2表型小胶质细胞使脑炎迅速结束,保护神经细胞。因此,可以认为环状肽衍生物促进小胶质细胞向抗炎M2表型的极性转化,从而延缓脑老化,并且有助于外伤性脑损伤、脑中风、痴呆症等急性/慢性脑炎关联性疾病的预防、病态恢复。进而,可以认为有助于对包括自闭症、抑郁症在内的精神疾病等的预防和病态恢复。
4.环状肽衍生物的安全性试验
将MG6小胶质细胞的细胞在96孔板中以37℃培养24小时(5×103细胞/孔),接着,添加环状肽衍生物至浓度达1μM,以37℃进行培养。在添加处理24小时后和72小时后,进行与上述同样的实时定量PCR(RT-PCR)分析和统计分析,求出白细胞介素-1β(IL-1β)和精氨酸酶-1(ARG1)的基因表达量。
结果如图4所示,在环状肽衍生物的单独给药时,小胶质细胞在24小时后和72小时后均未活化,既未极化为促炎M1表型也未极化为抗炎M2表型。由此可知,环状肽衍生物不会被小胶质细胞识别为异物,在非病态条件下,不会将小胶质细胞活化,因此确认了环状肽衍生物对于小胶质细胞的安全性。
5.初代小胶质细胞培养
利用磁性细胞分选MACS(磁性活化细胞分选法)法,从成年小鼠的大脑(8周龄,雄性,Japan SLC,Inc.滨松,日本)分离出作为初代小胶质细胞的CD11b+细胞。详细而言,将大脑切割成小片,将得到的细胞悬浮液用应用了神经组织分离试剂盒(Miltenyi Biotec)的酶消化法以及Gentle MACS Dissociator(全自动组织单细胞分离器,Miltenyi Biotec)机械地解离,进一步将其移送至30mm的细胞筛,得到单细胞悬浮液。用CD11b微珠(MiltenyiBiotec)进行磁标记后,将细胞悬浮液注入到设置于磁性分离器(Miltenyi Biotec)的磁力(MACS)柱。用磷酸缓冲生理盐水(PBS)冲洗MACS柱后,按照非专利文献4所记载的方法收集CD11b阳性组分。在使用初代小胶质细胞的以下试验中,使用伊格尔MEM(NissuiPharmaceutical Co.,Ltd.)培养基进行初代小胶质细胞的培养。
6.初代小胶质细胞细胞存活率的测定
将初代小胶质细胞的细胞接种于96孔板(104细胞/孔),与各种浓度的环状肽衍生物一同以37℃培养24小时、72小时。作为环状肽衍生物的浓度,调整至培养液中的浓度为0μM、0.01μM、0.1μM、1μM、5μM。细胞存活率的评价(24小时、72小时)如上述“2.”的MG6小胶质细胞存活率的测定所记载。
结果如图5所示,在环状肽衍生物的浓度为0.01~5μM的范围内没有确认到初代小胶质细胞的增殖,并且抑制存活的情况也几乎没有。另外,“*”表示P<0.05。
7.初代小胶质细胞的极化试验
作为大脑的常驻免疫细胞的小胶质细胞调节促炎性细胞因子和抗炎性细胞因子,从而对包括阿尔茨海默病在内的神经退行性疾病的神经炎症发挥重要的作用。已知白细胞介素-1β(IL-1β)为主要的炎症性细胞因子,转化生长因子(TGF)-β1为重要的抗炎性细胞因子(非专利文献5)。如上所述,对于作为神经分泌酸性糖蛋白的嗜铬粒蛋白A(CGA),作为小胶质细胞的活化候选因子而在阿尔茨海默病的老年斑中发现。因此,使用IL-1β和TGF-β1作为初代小胶质细胞的细胞和标志物,研究环状肽衍生物及CGA对炎症或抗炎的影响。
将初代小胶质细胞在96孔板中以37℃培养24小时(104细胞/孔),接着,在添加CGA(American Peptide Company、Anaspec)至浓度达10nM的同时,添加环状肽衍生物至浓度达1μM,以37℃进行培养。对于未添加环状肽衍生物而添加CGA者、未添加CGA而添加环状肽衍生物者以及未添加环状肽衍生物和CGA者(对照组),也同样地进行了培养。
在添加处理24小时后和72小时后进行实时定量PCR(RT-PCR)分析和统计分析,求出TGF-β1和IL-1β的基因表达量。实时定量PCR(RT-PCR)分析和统计分析的方法如上述“3.”的MG6小胶质细胞的极化试验所记载。其中,TGF-β1的引物对(Invitrogen公司制,Customprimer)为以下的序列。
TGF-β1:5′-TCAGACATTCGGGAAGCAGTG-3′(序列编号5)、以及5′-ATTCCGTCTCCTTGGTTCAGC-3′(序列编号6)
结果如图6所示。另外,图6中的“*”表示P<0.05,“##”表示P<0.01,“###”表示P<0.001。另外,“con”表示对照组,“肽”表示环状肽衍生物单独处理,“CGA”表示CGA单独处理,“肽+CGA”表示环状肽衍生物与CGA的组合处理。
如图6的(a)及(b)所示,在初代小胶质细胞中,与对照组相比,在CGA单独处理时,在24小时和72小时,使TGF-β1的表达多少有所增加。在CGA处理与环状肽衍生物的组合处理时,在24小时和72小时,使TGF-β1的表达大幅增加(平均3倍左右)。需要说明的是,与对照组相比,环状肽衍生物单独处理没有影响TGF-β1的表达。
另一方面,对于IL-1β的表达,如图6的(c)和(d)所示,在CGA单独处理时显著增加,在CGA处理与环状肽衍生物的组合处理时IL-1β的表达低至与对照同等或以下,与TGF-β1的表达具有相反的倾向。需要说明的是,与对照组相比,环状肽衍生物单独处理没有影响IL-1β的表达。
该结果表明,环状肽衍生物使TGF-β1增加,抑制IL-1β,由此使被CGA激活的小胶质细胞(促炎表型M1)转换为抗炎表型M2,即进行极性转化。若考虑到环状肽衍生物自身不会影响初代小胶质细胞中的TGF-β1或IL-1β的表达,则根据使用了初代小胶质细胞的本试验,也能够确认环状肽衍生物对于小胶质细胞的安全性,并且理解环状肽衍生物可有效地用于改善神经退行性疾病。
