Abstract
本发明提供了一种发酵杜仲叶提取物及其在制备鸡饲料中的应用,所述的发酵杜仲叶提取物,所述的提取物由包括如下步骤的方法制成:(1)将杜仲叶烘干,然后放在沸水上蒸3‑10min,得到预处理物料;(2)将所述的预处理物料放入袋中,在真空40℃下进行发酵,当物料发黑后取出;(3)将发酵后的物料加水热提取2次,将提取液混合、浓缩后即得所述的发酵杜仲叶提取物。本发明所述的用于防治鸡输卵管炎的组合物中包含了发酵杜仲叶提取物、杨树花提取液、维生素A、维生素D、维生素E、黄芪多糖、使君子粉、川楝素、槟榔、南瓜子粉,各组分结合,有效的对鸡输卵管炎产生防治效果,组分为纯天然材质,无毒副作用。

技术领域
本发明属于发酵领域,尤其是涉及一种发酵杜仲叶提取物及其在制备鸡饲料中的应用。
背景技术
蛋鸡的输卵管炎有的是大肠杆菌、沙门氏菌等引起的,有的是由于产蛋高峰期,输卵管的粘膜功能部分丧失造成的,还有的是由于强烈的应激因素,如气温的变化、免疫力低下等造成的。另外,在产蛋高峰期最容易发生输卵管炎症,因为高产鸡群一般连产7天休产一天,蛋白的分泌、蛋壳的形成都需要输卵管的参与,高强度的机能兴奋状态很容易导致面膜层的代谢过快,造成老化和分泌机能的过早丧失也会导致输卵管炎症的发生。临床症状和解剖变化:病鸡精神不佳,体温升高,冠苍白,产蛋量下降,有的停止产蛋,并产软壳蛋,破蛋增多,排蛋后鸡有时卧地不起,排出乳白色蛋清样稀便或白绿色稀便,泄殖腔突出,疼痛不安,产出的蛋其蛋壳上往往带有血迹。输卵管内经常排出黄、白色脓样分泌物,污染肛门周围及其下面的羽毛,产蛋困难。随着病情的发展,病禽开始发热,而后热退,痛苦不安,呆立不动,两翅下垂,羽毛松乱,有的腹部靠地或昏睡,当炎症蔓延到腹腔时可引起腹膜炎,或输卵管破裂引起卵黄性腹膜炎。剖检可看到输卵管发生病变:输卵管黏膜充血,出血,水肿,内有大量黏液性白色分泌物,内积有异形蛋样渗出物,泄殖腔处有1个至2个形成完好的鸡蛋未能产出,有的患鸡腹腔处肠道粘连。
目前,在治疗和预防禽畜疾病方面,多采用化学合成药物,化学合成药物虽然剂量小、见效快,但往往治标不治本,存在治疗不彻底的弊端,且化学合成药副作用较大,在鸡的肌肉和鸡蛋里存在着化学药物残留,有害残留可能对人体造成危害;特别是抗生素的使用,容易使菌株产生抗药性而无效果;且肉类中药物残留严重,直接危害人体健康。病毒性感染在动物体上常常发生,一旦出现,会对养殖业造成严重损失,也会给人类造成危害。
发明内容
有鉴于此,本发明旨在克服现有技术中的缺陷,提出一种发酵杜仲叶提取物及其在制备鸡饲料中的应用。
为达到上述目的,本发明的技术方案是这样实现的:
一种发酵杜仲叶提取物,所述的提取物由包括如下步骤的方法制成:
(1)将杜仲叶烘干,然后放在沸水上蒸3-10min,得到预处理物料;
(2)将所述的预处理物料放入袋中,在真空40℃下进行发酵,当物料发黑后取出;
(3)将发酵后的物料加水热提取2次,将提取液混合、浓缩后即得所述的发酵杜仲叶提取物。
进一步,所述的步骤(1)中的烘干步骤的温度为100℃,时间为30-60min。

所述的用于防治鸡输卵管炎的组合物的制备方法,包括如下步骤:
(1)将使君子粉、川楝素、槟榔、南瓜子粉分别粉碎,并混合均匀,得到粉料;
(2)将所述的粉料与50-100倍量的水混合均匀,煮沸后蒸煮1-3小时,过滤残渣,将滤液浓缩后得到药液;
(3)将维生素A、维生素D、维生素E、杨树花提取液、发酵杜仲叶提取物、黄芪多糖溶于所述的药液中,混合均匀后浓缩,在得到的浓缩液中加入糜蛋白酶与α-半乳糖苷酶,混合均匀后即得所述的用于防治鸡输卵管炎的组合物。

发酵杜仲叶提取物中:含有大量的杜仲多糖、杜仲黄酮,具有广谱抑菌、抗氧化抗应激、增强免疫力和改善肠道健康等作用。
杨树花提取液:含黄酮、有机酸、氨基酸、糖类和生物碱等物质,可防治家禽大肠杆菌,沙门氏菌,鸭巴氏杆菌、变形杆菌、葡萄球菌等多种细菌、引起的家禽输卵管炎感染,具有清热解毒、涩肠止泻、化湿止痢的功效,提高机体的抵抗力,对抗免疫制剂,调节机体的免疫功能,促进消化、促生长,缓解畜禽各种疾病发病期间的各种应激反应,补充氨基酸,可明显提高饲料转化率。
维生素A、维生素D与维生素E:如饲料中缺乏维生素A、D、E等均可导致炎。
黄芪多糖:对干扰素系统有明显的刺激作用,使动物机体产生内源性干扰素,抑制病毒的复制,从而达到病毒的目的;抗细菌作用:黄芪多糖对细菌及其病毒性产物有直接抑杀和解毒作用,更主要的是通过调动机体免疫防御功能而发挥扶正祛邪抑菌、杀菌的作用;增强机体免疫功能:黄芪多糖可特异性和非特异性的排除侵入肌体的异物,对特异免疫和非特异性免疫均有促进作用,提高机体的抗病能力。
使君子粉:具有抗皮肤真菌的作用,对许多病菌都有抑制作用。可以抗肠寄生虫和其他的一些虫病。
川楝素作:为一种从楝科和苦楝的树枝皮中分离而来的植物源杀虫剂,可有效驱除蛔虫,蛲虫和鞭中。
槟榔:具有“杀虫,破积,降气行滞,行水化湿”的功效,可用来治疗绦虫、钩虫、蛔虫、绕虫、姜片虫等寄生虫感染。
南瓜子粉:具有很好的杀灭血吸虫幼虫的作用,对于已经成熟的成虫,也能使其变性和虫数减少。同时,南瓜子也是有效的驱绦虫剂,且没有毒性和任何副作用,对蛲虫病、绦虫病、钩虫病等患者均有明显的效果。
糜蛋白酶:为胰腺分泌的一种蛋白水解酶,能迅速分解变性蛋白质,作用、用途与胰蛋白酶相似,比胰蛋白酶分解能力强、毒性低、不良反应小。糜蛋白酶用途广泛。可用于外科炎症、创伤、血肿及脓肿等,也可用于气管切开术。糜蛋白酶对痰多患者效果较好,使痰易于咳出。
α-半乳糖苷酶:催化α-半乳糖苷键的水解,可将饲料及豆制食品中的抗营养因子α-半乳糖苷类转化分解,改善其营养成分
相对于现有技术,本发明具有以下优势:
本发明所述的用于防治鸡输卵管炎的组合物中包含了发酵杜仲叶提取物、杨树花提取液、维生素A、维生素D、维生素E、黄芪多糖、使君子粉、川楝素、槟榔、南瓜子粉、糜蛋白酶与α-半乳糖苷酶,各组分结合,有效的对鸡输卵管炎产生防治效果,组分为纯天然材质,无毒副作用。
本发明所述的发酵杜仲叶提取物通过烘干、蒸制与发酵制成,烘干可将杜仲叶中的酶灭活生物活性物质易被酶分解,不利于有效成分的保存;蒸汽处理对鲜叶的穿透均匀,不会产生焦叶,叶子内部温度瞬时升高,使多酚类物质充分氧化,有利于各种活性成分的保留;采用发酵的方法进行制备,可有效的提高发酵杜仲叶提取物中的黄酮、桃叶珊瑚甙和绿原酸的含量,

实施例1
一种发酵杜仲叶提取物,所述的提取物由包括如下步骤的方法制成:
(1)将杜仲叶100℃烘干30min,然后放在沸水上蒸5min,得到预处理物料;
(2)将所述的预处理物料放入袋中,在真空40℃下进行发酵,当物料发黑后取出;
(3)将发酵后的物料加水热提取2次,将提取液混合、浓缩后即得所述的发酵杜仲叶提取物。

(3)将发酵后的物料加水热提取2次,将提取液混合、浓缩后即得所述的发酵杜仲叶提取物。

一种用于防治鸡输卵管炎的组合物,该组合物由包括如下重量份的原料制成:发酵杜仲叶提取物(仲能)3份,杨树花提取液5份,维生素A 0.02份,维生素D 0.02份,维生素E0.02份,黄芪多糖5份,使君子粉4份,川楝素4份,槟榔4份,南瓜子粉4份。所述的组合物还包括糜蛋白酶与α-半乳糖苷酶,所述的糜蛋白酶与α-半乳糖苷酶的质量比为1:1;所述的糜蛋白酶的添加量为0.002份。所述的糜蛋白酶的酶活为20万U/g,所述的α-半乳糖苷酶的酶活为80万U/g。

本发明实施例中组合物的使用方法:将组合物加入到饲料中,组合物与饲料的质量比为1-3:100,其他按照常规喂养即可。
临床实验:
选择1000只患有鸡输卵管炎的病鸡作为实验对象,分为5组,每组200只,分别对应实施例1-4与对比例1。
病鸡症状为:病鸡冠厚,鲜红,腹部下垂;蛋壳上往往带有血迹,血斑,内经常排出黄、白色脓样分泌物,污染肛门周围及羽毛,困难;随着病情的发展,病禽开始发热,厌食,痛苦不安,呆立不动,两翅下垂,羽毛松乱。
对比例2
一种发酵杜仲叶提取物,所述的提取物由包括如下步骤的方法制成:
(1)将杜仲叶100℃烘干30min,然后放在沸水上蒸5min,得到预处理物料;
(2)将所述的预处理物料放入袋中,在真空40℃下进行发酵,当物料发黑后取出;
(3)将发酵后的物料加水热提取2次,将提取液混合、浓缩后即得所述的发酵杜仲叶提取物。

将实施例1-4与对比例1-2得到的组合物混入饲料中,每日投喂1次,其他管理采用常规管理,测试结果如表1所示。

由表1中数据可知,实施例1-4中的组合物均对鸡输卵管炎有一定的治愈效果,各组分均有一定的作用效果,而没有糜蛋白酶与α-半乳糖苷酶的对比例2的结果均不理想,说明加入糜蛋白酶与α-半乳糖苷酶,与其他组分产生了协同效果,对鸡输卵管炎有明显的治愈效果,并且,发酵杜仲叶提取物采用的本发明所述的烘干、蒸煮与发酵的制备方法,也对组合物起到进一步的提升效果。
以上所述仅为本发明的较佳实施例而已,并不用以限制本发明,凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (10)
1.一种发酵杜仲叶提取物,其特征在于:所述的提取物由包括如下步骤的方法制成:
(1)将杜仲叶烘干,然后放在沸水上蒸3-10min,得到预处理物料;
(2)将所述的预处理物料放入袋中,在真空40℃下进行发酵,当物料发黑后取出;
(3)将发酵后的物料加水热提取2次,将提取液混合、浓缩后即得所述的发酵杜仲叶提取物。
2.根据权利要求1所述的发酵杜仲叶提取物,其特征在于:所述的步骤(1)中的烘干步骤的温度为100℃,时间为30-60min。
5.根据权利要求4所述的用于防治鸡输卵管炎的组合物,其特征在于:所述的组合物还包括糜蛋白酶与α-半乳糖苷酶,所述的糜蛋白酶与α-半乳糖苷酶的质量比为1:1;所述的糜蛋白酶的添加量为0.001-0.01份。
6.根据权利要求4所述的用于防治鸡输卵管炎的组合物,其特征在于:所述的糜蛋白酶的酶活为20万U/g,所述的α-半乳糖苷酶的酶活为80万U/g。
7.根据权利要求4所述的用于防治鸡输卵管炎的组合物,其特征在于:所述的杨树花提取物由包括如下步骤的方法制成:将杨树花置于水中煎煮2次,每次煎煮1-2小时,将煎煮液混合、浓缩,然后在浓缩液中加入乙醇,2-5℃条件下沉淀2次,每次6-12小时,取出沉淀后将乙醇挥发完成后得到杨树花提取液。
8.权利要求3-7中任一项所述的用于防治鸡输卵管炎的组合物的制备方法,其特征在于:包括如下步骤:
(1)将使君子粉、川楝素、槟榔、南瓜子粉分别粉碎,并混合均匀,得到粉料;
(2)将所述的粉料与50-100倍量的水混合均匀,煮沸后蒸煮1-3小时,过滤残渣,将滤液浓缩后得到药液;
(3)将维生素A、维生素D、维生素E、杨树花提取液、发酵杜仲叶提取物、黄芪多糖溶于所述的药液中,混合均匀后浓缩,在得到的浓缩液中加入糜蛋白酶与α-半乳糖苷酶,混合均匀后即得所述的用于防治鸡输卵管炎的组合物。
9.一种用于防治鸡输卵管炎的组合物的在制备鸡饲料中的应用。
10.一种用于防治鸡输卵管炎的组合物的在制备治疗鸡输卵管炎药物中的应用。
