Description

Composite agilawood leaf concentrated powder for relaxing bowel and preparation method thereof
Technical Field
The invention relates to the technical field of food processing, in particular to compound agilawood leaf concentrated powder for relaxing bowel and a preparation method thereof.
Background
The agilawood is a low-yield traditional Chinese medicine in Lingnan, has the effects of promoting qi circulation, relieving pain, warming middle energizer, stopping vomiting and absorbing qi and relieving asthma as a traditional Chinese medicine in China, and is used for treating chest and abdomen swelling pain, stomach cold, vomiting and hiccup, kidney deficiency and qi regurgitation and asthma. The agilawood leaves are non-medicinal parts of agilawood, and related researches are relatively few, and experiments show that the agilawood leaf water extract has the functions of relaxing bowels, expelling toxin, promoting sleep, reducing fat, tonifying qi and the like. However, the single eaglewood leaf water extract has limited effect on intestinal tract regulation and is not easy to store. The concentrated powder prepared from the single aquilaria sinensis leaf water extract has the defects of poor solubility, low mouthfeel, insufficient flavor, obvious astringency, limited regulating function effect on the gastrointestinal system and the like.

2. Composite agilawood leaf concentrated powder bowel relaxing function experiment
(1) Establishment of Slow Transit Constipation (STC) animal model
Adult healthy male mice were selected, weighing 20 mice (20. + -.2) g.
The test pieces were divided into model control group and blank control group, each group containing 10 animals. The model control group was gavage with 5mg/kg.BW of loperamide hydrochloride every day, and the blank control group was fed normally for 7 days. On the 7 th day, after the model control group is filled with loperamide hydrochloride for 30min, the model control group and the blank control group are filled with stomach ink (the stomach filling amount of the ink is 0.2 ml/piece), and compared with the blank control group, the model control group obviously has the phenomena of prolonged defecation time and reduced defecation grain number; and (3) killing the mice after the ink is infused for 25-30 min, comparing the ink propulsion rates of the two groups of mice, and judging that the model building is successful if the small intestine ink propulsion rate of the model control group of mice is obviously reduced.
(2) Influence of compound agilawood leaf concentrated powder on gastrointestinal driving force of STC mice
Adult healthy male mice were selected, weighing (20. + -.2) g, and divided into 5 groups of 10 mice each.

The method comprises the following specific steps:
2 experimental groups at both high and low doses: dissolving 0.5g of compound agilawood leaf concentrated powder (prepared in example 3) in 100ml of distilled water to serve as a low-dose liquid medicine; dissolving 2.5g of compound agilawood leaf concentrated powder in 100ml of distilled water to be used as high-dose liquid medicine; on the basis of normal feeding, per stomach is irrigated with 0.6ml of the low-dose liquid medicine/high-dose liquid medicine and 5mg/kg of loperamide hydrochloride for 14 days;
positive control group: dissolving 2g fructus Cannabis pill in 100ml distilled water as positive control liquid medicine; on the basis of normal feeding, performing intragastric administration on 0.6ml of the positive control liquid medicine and 5mg/kg of loperamide hydrochloride for 14 days every day;
model control group: the feed is normally fed, and the stomach is drenched with 0.6 ml/pig of distilled water and 5mg/kg.BW of loperamide hydrochloride every day for 14 days.
Blank control group: feeding normally, and feeding with 0.6 ml/per stomach distilled water every day for 14 days;
during the period, daily records:
weight change, food intake and water intake of the mice;
② observing the health condition and mental state of the mouse and various physiological activities. Stool color and shape;
③ the number of the defecation grains, the water content of the excrement and the dry weight of the excrement.
On day 14, the gavage was performed 30min after gavage with loperamide hydrochloride. After a further 30min (i.e. the ink was gavage for 30min) the mice were sacrificed.
Opening the intestinal cavity to separate mesentery, cutting the intestinal canal from pylorus, lower end to ileocecal part, placing on a tray, slightly drawing the small intestine into a straight line, measuring the length of the intestinal canal as the total length of the small intestine, and measuring the length of the intestinal canal from the pylorus to the front edge of ink as the advancing length of the ink.
The ink propulsion rate was calculated as follows:
the ink propulsion rate (%) — the ink propulsion length (cm)/total small intestine length (cm) × 100%. (ii) a
