Abstract
本发明涉及一种用于预防或治疗龋病和牙周病的牙套;牙套包括羧甲基壳聚糖、聚乙烯吡咯烷酮、甘油磷酸钠、海藻酸钠和铺料;羧甲基壳聚糖的脱乙酰度≥80%,羧甲基壳聚糖的羧甲基取代度为40%~65%;牙套的制备包括以下步骤:将羧甲基壳聚糖溶于去离子水,获得羧甲基壳聚糖溶液;将羧甲基壳聚糖溶液与海藻酸钠混合,再加入辅料搅拌,获得匀浆溶液;将匀浆溶液与聚乙烯吡咯烷酮混合,再加入甘油磷酸钠进行交联,形成水凝胶;将水凝胶加热熔融,导入牙套模具内,冷却至室温后进行干燥,去模,得到牙套。从而得到一种不会诱导耐药菌株的产生和引起菌群失调并且能够预防或治疗龋病和牙周病的牙套。

技术领域
本发明涉及龋病和牙周病预防和治疗技术领域,具体涉及一种用于预防或治疗龋病和牙周病的牙套。
背景技术
龋病和牙周病是人类口腔两大类最常见的慢性感染性疾病,不仅严重影响患者的口腔健康,同时也是全身多种疾病的危险因素。菌斑生物膜中的细菌是龋病和牙周病的始动因子。由于口腔独特的生理特点,有效清除菌斑生物膜、减少细菌的黏附与聚集,目前仍面临巨大的挑战,因此有关改善口腔抗菌材料、抗菌药物的抗菌性能的研究也一直没有停止过,各种新型抗菌措施也层出不穷。
牙齿抗菌,现有技术中一般采用牙齿封釉的技术来隔绝细菌对牙齿的浸蚀。虽可有效的抑制细菌的生成,防止龋齿、牙周炎、牙髓炎等的产生,但是操作复杂,成本高,作用时间短,不利于普及使用。抗菌牙套可有效抑制牙齿表面的致病菌,而且成本低,操作简单,便于普及,但其大多含有抗菌药物,容易诱导耐药菌产生和引起菌群失调,不利于牙周病的缓解。
发明内容
本发明旨在至少在一定程度上解决相关技术中的技术问题之一。为此,本发明的目的在于提供一种用于预防或治疗龋病和牙周病的牙套。
为此,在本发明的一方面,本发明提出了一种用于预防或治疗龋病和牙周病的牙套,其特征在于,所述牙套包括羧甲基壳聚糖、聚乙烯吡咯烷酮、甘油磷酸钠、海藻酸钠和铺料;所述羧甲基壳聚糖的脱乙酰度≥80%,所述羧甲基壳聚糖的羧甲基取代度为40%~65%;
所述牙套的制备包括以下步骤:
将所述羧甲基壳聚糖溶于去离子水,获得羧甲基壳聚糖溶液;
将所述羧甲基壳聚糖溶液与所述海藻酸钠混合,再加入所述辅料搅拌,获得匀浆溶液;
将所述匀浆溶液与所述聚乙烯吡咯烷酮混合,再加入甘油磷酸钠进行交联,形成水凝胶;
将所述水凝胶加热熔融,导入牙套模具内,冷却至室温后进行干燥,去模,得到所述牙套。
根据本发明实施例的用于预防或治疗龋病和牙周病的牙套,其通过羧甲基壳聚糖在高脱乙酰度、低羧甲基取代度的条件下,对致龋菌有较好地抑菌效果,能够有效清除菌斑生物膜、减少口腔中黏附的细菌,能够有效增加牙周新附着、促进牙周组织再生,有效地预防或治疗龋病和牙周病等口腔疾病,并且该牙套不存在西药成分,也没有使用抗菌剂,不会诱导耐药菌株的产生和引起菌群失调的问题。
另外,根据本发明上述实施例提出的用于预防或治疗龋病和牙周病的牙套,还可以具有如下附加的技术特征:
根据本发明的实施例,所述辅料包括维生素C、金银花、木糖醇和连翘。
根据本发明的实施例,去模后的牙套经过CaCl2溶液、NaOH溶液浸泡,再用蒸馏水洗涤,干燥。
根据本发明的实施例,干燥的条件为50℃~80℃,无菌。
根据本发明的实施例,所述羧甲基壳聚糖的分子量>23KD。

制备过程:(1)将5g壳聚糖均匀分散在异丙醇或乙醇溶液中,然后加入35%的NaOH溶液进行碱化反应2.5h,之后再加入氯乙酸,在75℃恒温水浴中反应2~2.5h,同时开启冷凝装置和搅拌器,反应结束后待温度降至室温并用冰醋酸调PH至中性。将反应混合物过滤,滤饼用75%乙醇溶液洗涤多次,再用无水乙醇洗涤,真空干燥,得到羧甲基壳聚糖产品;(2)将羧甲基壳聚糖溶于去离子水,得到羧甲基壳聚糖溶液;(3)将羧甲基壳聚糖溶液与海藻酸钠水溶液混合,加入维生素C、金银花、木糖醇,连翘,充分搅拌,得到匀浆溶液;(4)将匀浆溶液与聚乙烯吡咯烷酮溶液混合,再加入甘油磷酸钠进行交联、中和,形成水凝胶;(5)将水凝胶进行加热熔融,倒入牙套模具内,使凝胶均匀分布,冷却至室温后进行第一次恒温干燥,去模,得到牙套;(6)经第一次恒温干燥后去模得到的牙套依次经过CaCl2溶液、NaOH溶液浸泡,用蒸馏水洗涤,对洗涤后的牙套进行第二次恒温干燥(50℃~80℃,无菌)。
本实施例中提供的配方能够有效增加牙周新附着、促进牙周组织再生。以牙周膜细胞为研究对象,研究不同羧甲基取代度(A=20%、B=40%、C=65%、D=95%)和不同脱乙酰度(a=25%、b=55%、c=85%)的不同浓度(300~10ug/ml)羧甲基壳聚糖(CMCS)对牙周膜细胞增殖的影响。根据四唑盐比色实验(MTT)结果,在大致相同的脱乙酰度的条件下,如图1可知当取代度过高或过低,都不利于羧甲基壳聚糖对牙周膜细胞的增殖作用,当取代度处于40%~65%之间,有利于促进对牙周膜细胞的增殖作用,且随着取代度的升高,增殖作用逐渐降低;在取代度差别不大的条件下,如图2所示,所有的样品均对牙周膜细胞的增殖有促进作用,且脱乙酰度越高,作用越明显。结合图1和图2可知,随着羧甲基壳聚糖浓度的升高,细胞的增殖作用越明显,但是由于考虑成本因素,羧甲基壳聚糖浓度的最佳范围建议在100~200ug/ml之间。
实施例2不同浓度的羧甲基壳聚糖对变形链球菌表面疏水性的影响
实验采用微生物黏着碳氢化合物法(MATH)来研究羧甲基壳聚糖对变形链球菌表面疏水性的影响。如图3可知,随着羧甲基壳聚糖浓度的升高,变形链球菌表面疏水率逐渐降低,当浓度为140ug/ml时,其疏水率的下降幅度逐渐趋于平缓,表明羧甲基壳聚糖溶液浓度为140~190ug/ml时作用于变形链球菌表面,其表面疏水性明显降低,同时还可抑制变形链球菌的黏附作用。它的作用机制是在羧甲基壳聚糖溶液pH为6.0的条件下,羧甲基壳聚糖的氨基质子化形成聚阳离子,和细胞表面带负电荷的基团作用,使羧甲基壳聚糖吸附于细胞表面,降低细菌的疏水性,进而降低细菌的粘附能力。
实施例3不同浓度的羧甲基壳聚糖对变形链球菌脱落的影响
实验采用死菌/活菌荧光染色和激光共聚焦扫描显微镜技术相结合的方法来研究羧甲基壳聚糖对变形链球菌生物膜黏附和脱落的影响。如图4所示,随着羧甲基壳聚糖浓度的增加细菌数量逐渐减少,且与空白组(羧甲基壳聚糖浓度为0时)有显著性差异,结果证明150ug/ml至200ug/ml的羧甲基壳聚糖溶液可以有效促进玻片表面形成的菌斑脱落,为羧甲基壳聚糖在口腔疾病预防方面的应用提供了理论支持。
综上,根据本发明实施例的用于预防或治疗龋病和牙周病的牙套,其通过羧甲基壳聚糖在高脱乙酰度、低羧甲基取代度的条件下,对致龋菌有较好地抑菌效果,能够有效清除菌斑生物膜、减少口腔中黏附的细菌,能够有效增加牙周新附着、促进牙周组织再生,有效地预防或治疗龋病和牙周病等口腔疾病,并且该牙套不存在西药成分,也没有使用抗菌剂,不会诱导耐药菌株的产生和引起菌群失调的问题。

Claims (5)
1.一种用于预防或治疗龋病和牙周病的牙套,其特征在于,所述牙套包括羧甲基壳聚糖、聚乙烯吡咯烷酮、甘油磷酸钠、海藻酸钠和铺料;所述羧甲基壳聚糖的脱乙酰度≥80%,所述羧甲基壳聚糖的羧甲基取代度为40%~65%;
所述牙套的制备包括以下步骤:
将所述羧甲基壳聚糖溶于去离子水,获得羧甲基壳聚糖溶液;
将所述羧甲基壳聚糖溶液与所述海藻酸钠混合,再加入所述辅料搅拌,获得匀浆溶液;
将所述匀浆溶液与所述聚乙烯吡咯烷酮混合,再加入甘油磷酸钠进行交联,形成水凝胶;
将所述水凝胶加热熔融,导入牙套模具内,冷却至室温后进行干燥,去模,得到所述牙套。
2.如权利要求1所述的用于预防或治疗龋病和牙周病的牙套,其特征在于,所述辅料包括维生素C、金银花、木糖醇和连翘。
3.如权利要求1所述的用于预防或治疗龋病和牙周病的牙套,其特征在于,去模后的牙套经过CaCl2溶液、NaOH溶液浸泡,再用蒸馏水洗涤,干燥。
4.如权利要求1所述的用于预防或治疗龋病和牙周病的牙套,其特征在于,干燥的条件为50℃~80℃,无菌。
5.如权利要求1所述的用于预防或治疗龋病和牙周病的牙套,其特征在于,所述羧甲基壳聚糖的分子量>23KD。
