Abstract

本发明公开了一种肉桂醛纳米微球悬浮剂的制备方法,属于抗菌剂制备领域。该方法具体为将油溶性的肉桂醛与改性的玉米淀粉溶于有机溶剂中,得到有机相溶液,再将有机相溶液缓慢滴入到聚乙烯醇水相溶液混合搅拌均匀后得到初乳液,接着对初乳液进行超声处理后常温下搅拌至有机溶剂完全挥发得到粒径为100‑300nm肉桂醛纳米微球。本发明方法所采用的原材料生物相容性好,对生产者安全,并且制备工艺简单,可大规模工业化生产,产品质量稳定可靠,可以有效地提高肉桂醛的稳定性,具有非常广阔的产业化应用前景。

Description

一种肉桂醛纳米微球悬浮剂的制备方法
【技术领域】
本发明属于抗菌剂制备领域,具体涉及一种肉桂醛纳米微球悬浮剂的制备方法。
【背景技术】
肉桂醛又名桂皮醛、苯丙烯醛,是一种醛类有机化合物,广泛存在于肉桂、清化桂、天竺桂、川桂、银叶桂等植株中,是美国食品药品监督管理局分类为GRAS(通常公认安全)的香精油。国内外学者通过大量研究表明肉桂醛具有多种药理活性,对抗糖尿病及抗肥胖具有较好疗效,同时还具有防治循环系统疾病、抗菌及抗肿瘤等作用,并且被认为是较为安全的药物,其给药剂量也往往比较大;此外,还作为香料、显色剂和食品添加剂被广泛使用。研究表明含有肉桂醛的薄膜,颗粒和纳米纤维已显示出在多种用途中的优势,包括药物,食品和抗菌包装。比如,包含肉桂醛的活性包装可以延长新鲜蘑菇的货架期。另外,肉桂醛的亚微米乳剂因其抗肿瘤活性和药代动力学特性而可用于临床。然而,肉桂醛结构中含有苯环等疏水性结构,难溶于水,且具有高挥发性,低稳定性以及对氧气,光和热的高敏感性,因此肉桂醛存在难以长时间保存的问题,另外直接使用肉桂醛生物利用度非常低,这些缺点严重地限制了其在医药、化工和食品等行业的使用。
【发明内容】
为了克服肉桂醛的上述缺点,本发明选用可食性的玉米淀粉作为载体材料,负载肉桂醛,用载体包裹肉桂醛制备成纳米颗粒,可有效地改善肉桂醛的水溶性和易挥发的问题。
本发明的目的是通过以下方式来实现的:
本发明提供了一种肉桂醛纳米微球悬浮剂的制备方法,具体包括如下步骤:
(1)将肉桂醛与改性的玉米淀粉按质量比2:1-10溶于有机溶剂中,得到有机相溶液;
(2)将聚乙烯醇溶于水中,配制重量浓度为0.5-2.0%的水相溶液;
(3)按有机相溶液与水相溶液的体积比为1:4-10,将有机相溶液缓慢滴入水相溶液中,混合均匀后得到初乳液;
(4)对初乳液进行超声乳化后,在常温下搅拌至有机溶剂完全挥发,即得到肉桂醛纳米微球悬浮剂。
进一步,在步骤(1)中所述的有机溶剂为二氯甲烷或三氯甲烷。
进一步,在步骤(1)中所述改性的玉米淀粉是通过如下方法获得的:将玉米淀粉均匀分散在冰醋酸中,加入冷却的乙酸酐,15分钟后向反应液中加入对甲苯磺酸,置于80℃下搅拌3小时,接着用去离子水洗涤混合物得到固体产物,最后将固体产物在40℃烘箱中干燥过夜得到改性的玉米淀粉。
进一步,在步骤(4)中所述的超声处理具体为在功率300-500W的条件下超声处理3-5min。
本发明还提供了一种如上所述的肉桂醛纳米微球悬浮剂的制备方法得到的肉桂醛纳米微球。
优选地,所述的肉桂醛纳米微球平均粒径为100-300nm。
上述原材料均可以通过市购的方式获得,其中,肉桂醛购自北京百灵威科技有限公司;玉米淀粉购自长春大成实业有限公司。
本发明的特点和有益效果如下:
1)本发明制备的肉桂醛纳米微球经改性的玉米粉包裹后,与原液相比,可以提高其稳定性,也便于应用和贮存;
2)本发明制备的肉桂醛纳米微球可以实现超长时间的缓控释作用,提高有效成分的疗效、降低其不良反应和延缓给药周期。
3)采用本发明制备的肉桂醛纳米微球,其平均粒径小于300nm,载药量可以达到30%以上;
4)本发明方法工艺简单,成本低廉,不产生污染物,产品质量稳定可靠;
5)本发明获得的肉桂醛纳米微球悬浮剂及其制备方法具有非常广阔的应用前景。

结合以下实施例对本发明的原理和特征进行描述,所举实施例只用于解释本发明,并非用于限定本发明的范围。
实施例1:改性的玉米淀粉的制备
将玉米淀粉均匀分散在冰醋酸中,加入冷却的乙酸酐,15分钟后向反应液中加入对甲苯磺酸,置于80℃下搅拌3小时,接着用去离子水洗涤混合物得到固体产物,最后将固体产物在40℃烘箱中干燥过夜得到改性的玉米淀粉。
实施例2:肉桂醛纳米微球悬浮剂的制备
将肉桂醛与改性的玉米淀粉按质量比1:1溶于二氯甲烷中;然后,按体积比1:4将其缓慢滴入1.0%的聚乙烯醇水溶液中,得到初乳液;然后以300w功率超声乳化4min,常温下搅拌至有机溶剂完全挥发,即可得到平均粒径为100nm的肉桂醛纳米微球悬浮剂。
实施例3:肉桂醛纳米微球悬浮剂的制备
将肉桂醛与改性的玉米淀粉按质量比1:3溶于三氯甲烷中;然后,按体积比1:5将其缓慢滴入2.0%的聚乙烯醇水溶液中,得到初乳液;然后以500w功率超声乳化5min,常温下搅拌至有机溶剂完全挥发,即可得到平均粒径为150nm的肉桂醛纳米微球悬浮剂。
实施例4:肉桂醛纳米微球悬浮剂的制备
将肉桂醛与改性的玉米淀粉按质量比1:5溶于二氯甲烷中;然后,按体积比1:4将其缓慢滴入1.0%的聚乙烯醇水溶液中,得到初乳液;然后以400w功率超声乳化5min,常温下搅拌至有机溶剂完全挥发,即可得到平均粒径为200nm的肉桂醛纳米微球悬浮剂。
下面将描述本发明肉桂醛纳米微球的性能试验。
实验例1:肉桂醛纳米微球的粒径分布和形貌表征
将实施例2中制得的肉桂醛纳米微球悬浮剂用去离子水稀释至0.5mg/mL,吸取1.5mL样品溶液加入比色皿中,利用激光粒度分析仪测定该纳米微球的水合粒径为301.8nm;另取10μL的样品溶液,铺展到洁净光滑的硅片表面,待自然晾干后,用扫描电子显微镜和透射电子显微镜对纳米微球进行形貌观察;最后,用Nano Measurer 1.2软件统计实际粒径分布情况,该结果列于附图1。
实验例2:肉桂醛纳米微球的释放行为
取5ml实施例2中的样品溶液密封于透析袋中,放入装有95mL,5%甲醇溶液到棕色反应瓶中;在25℃恒温摇床中震荡透析24h,分别于第0.5h、1h、2h、4h、7h、10h和24h从反应瓶中取出3mL透析液,同时补回等量的介质溶液。利用紫外分光光度仪,测定溶液中肉桂醛的浓度,计算各个取样时间点肉桂醛的累积释放百分率,该结果列于附图2。从附图2可知,肉桂醛原液释放得非常地迅速,1h即可完全释放,经载体包埋后的肉桂醛纳米微球,其释放速率得到了一定的减缓。
实验例3:肉桂醛纳米微球的热稳定性测试
吸取20ml实施例2中的样品放入洁净的安培瓶,密封后分别放置在0℃冰箱、25℃室温和54℃恒温箱中进行稳定性实验,14d取出样品,按上述方法进行制样,用扫描电子显微镜对样品进行形貌观察,该结果列于附图3。基于该实验结果,可以证明本专利制备的肉桂醛纳米微球在低温(0℃)、常温(25℃)和高温(54℃)环境中性能稳定,可以长期储藏。
实验例4:肉桂醛纳米微球的光稳定性测试
称取市售肉桂醛和肉桂醛纳米微球样品,用二甲苯溶液配置浓度250mg/L的母液,使用移液枪吸取1mL置于直径为3.4cm的石英培养皿中。在室温黑暗条件下,待甲苯挥发形成药膜后,将石英培养皿转入氙弧灯光照培养箱中25℃、250W条件下连续光照,分别在1h、4h、8h、12h、24h、48h时间点取样,用4mL甲醇将药膜充分溶解转移到样品瓶中,利用高效液相色谱测定样品的含药量,计算肉桂醛的分解率。抗光解分解率实验结果如附图4所示,经过48小时的紫外光照,肉桂醛剩余有效成分为33.1%,经改性后的肉桂醛纳米微球的抗光分解能力显著性的增强,48小时紫外光照后剩余有效成分约为56.4%。
实验例5:肉桂醛纳米微球的抗菌性能测试
人们对使用自然灭菌以防止由大肠杆菌或某些其他与食物相关的不良微生物引起的可能感染的需求日益增加,而不是使用化学防腐剂。源自植物精油作为抑制食源性病原体的天然抗菌剂已受到广泛关注。在植物精油中,已有报道肉桂醛是抗大肠杆菌的有效精油之一。我们测试了肉桂醛和肉桂醛淀粉纳米微球对大肠杆菌(E.coli)的抗菌活性。测试方法如下,将大肠杆菌制成150μL含有约108cfu mL-1的菌悬液,然后生长在Luria-Bertani琼脂上,将肉桂醛和肉桂醛淀粉纳米微球切成直径为1cm且重量相同的圆形片。为了比较,还测定了没有加肉桂醛样品抗菌活性。每个待测组分样品分别放置在铺有大肠杆菌的琼脂平板上,在将96孔板放在37℃恒温培养箱中培养24h前后,用酶标仪分别测定其OD值。能明显抑制大肠杆菌生长的浓度即为该抑菌剂的最小抑菌浓度(MIC)。从抗菌结果来看,经过淀粉包裹后的肉桂醛纳米微球的抗菌活性仍然保持(表1)。

Claims (2)

1.一种肉桂醛纳米微球悬浮剂的制备方法,其特征在于该方法包括如下步骤:
(1)将肉桂醛与改性的玉米淀粉按质量比2:1-10溶于有机溶剂中,得到有机相溶液;
(2)将聚乙烯醇溶于水中,配制重量浓度为0.5-2.0%的水相溶液;
(3)按有机相溶液与水相溶液的体积比为1:4-10,将有机相溶液缓慢滴入水相溶液中,混合均匀后得到初乳液;
(4)对初乳液进行超声乳化后,在常温下搅拌至有机溶剂完全挥发,即得到肉桂醛纳米微球悬浮剂;
其中,在步骤(1)中所述的有机溶剂为二氯甲烷或三氯甲烷;
