(1) the invention provides a preparation method of convenient and instant cubilose, which is characterized in that cubilose filaments are treated by glutamine transaminase, so that the crosslinking degree of protein molecules in the cubilose filaments is improved, the pH value of a system is adjusted to 0-5, the internal structure of the cubilose filaments is further more compact, the cubilose filaments and the glutamin transaminase are cooperated, the thermal stability of the cubilose filaments during high-temperature sterilization is improved, the cubilose filaments are not dissolved during high-temperature sterilization and later storage, and the problem of water melting of the cubilose filaments is avoided. Meanwhile, through the combination of glutamine transaminase treatment and acid treatment, sialic acid loss is reduced under the synergistic effect, and the nutritional value of the cubilose is improved.
(2) The production process is scientific and reasonable, simple in process and strong in operability, and industrial and automatic continuous production is easy to realize in factories.

In order to make the aforementioned objects, features and advantages of the present invention more comprehensible, specific embodiments thereof are described in detail below with reference to examples of the specification.
In the following description, numerous specific details are set forth in order to provide a thorough understanding of the present invention, but the present invention may be practiced in other ways than those specifically described and will be readily apparent to those of ordinary skill in the art without departing from the spirit of the present invention, and therefore the present invention is not limited to the specific embodiments disclosed below.
Furthermore, reference herein to "one embodiment" or "an embodiment" means that a particular feature, structure, or characteristic described in connection with the embodiment is included in at least one implementation of the invention. The dry cubilose used in the invention comprises cubilose, cubilose strips, cubilose corners and cubilose pieces which are generally sold in the market; the glutamine transaminase is generally sold in the market, and the enzyme activity is more than or equal to 100U/g.
Example 1
A. Soaking to remove impurities: weighing 50g of dry edible bird's nest, soaking in 10L of normal temperature deionized water for 5h, removing bird's hair, cleaning for 3 times, taking out and draining;
B. internal crosslinking: adding the drained bird's nest silks into 10L of 2% glutamine transaminase aqueous solution, crosslinking for 6h at 40 ℃, taking out and draining;
C. and (3) shrinking the structure: immersing the drained bird's nest silk in 10L of water, adding 0.05mol/L of sulfuric acid to adjust the pH value of the system to 2, soaking for 40min at 60 ℃, taking out and draining.
D. Cleaning and drying: soaking and cleaning the above nidus Collocaliae filaments at normal temperature for several times, taking out the nidus Collocaliae filaments, draining, and drying at 40 deg.C.
E. Subpackaging and filling liquid: 2g of dried bird's nest silk is put into a glass bottle, 100ml of water is added, and the bottle is sealed.
F. High-temperature sterilization: sterilizing the sealed nidus Collocaliae at 121 deg.C for 20min, cooling, inspecting, and packaging.
Example 2

C. Cleaning and drying: soaking and cleaning the above nidus Collocaliae filaments at normal temperature for several times, taking out the nidus Collocaliae filaments, draining, and drying at 40 deg.C;
D. Subpackaging and filling liquid: 2g of dried bird's nest silk is put into a glass bottle, 100ml of water is added, and the bottle is sealed.
E. High-temperature sterilization: sterilizing the sealed nidus Collocaliae at 121 deg.C for 20min, cooling, inspecting, and packaging.
Example 3
A. Soaking to remove impurities: weighing 50g of dry edible bird's nest, soaking in 10L of normal temperature deionized water for 5h, removing bird's hair, cleaning for 3 times, taking out and draining;
B. internal crosslinking: adding the drained bird's nest silks into 10L of 1.5% glutamine transaminase aqueous solution, crosslinking for 6h at 40 ℃, taking out and draining;
C. cleaning and drying: soaking and cleaning the above nidus Collocaliae filaments at normal temperature for several times, taking out the nidus Collocaliae filaments, draining, and drying at 40 deg.C;
D. subpackaging and filling liquid: 2g of dried bird's nest silk is put into a glass bottle, 100ml of water is added, and the bottle is sealed.
E. High-temperature sterilization: sterilizing the sealed nidus Collocaliae at 121 deg.C for 20min, cooling, inspecting, and packaging.
Example 4
A. Weighing, filling and sealing: 2g of common bird's nest silk is put into a glass bottle, 100ml of water is added, and the bottle is sealed.
B. High-temperature sterilization: sterilizing the sealed nidus Collocaliae at 121 deg.C for 20min, cooling, inspecting, and packaging.
Example 5
A. Weighing and filling: and sealing: 2g of common bird's nest silk is put into a glass bottle, 100ml of water is added, and the bottle is sealed.
B. High-temperature sterilization: by adopting the sterilization process disclosed in the invention patent CN105325797A, the sealed cubilose is heated to 105 ℃ and is kept warm for 5min, then is heated to 118 ℃ for sterilization for 16min, and back pressure is applied to the whole process for sterilization.
The method for measuring the solid content comprises the following steps: QB 1007 and 1990 can food net weight and solid content determination.
The solid content of the newly-prepared bird's nest cans in examples and comparative examples was measured, and the solid content of the newly-prepared bird's nest cans in examples and comparative examples was measured after placing the cans in a 37 ℃ incubator for 6 months in an accelerated test, and the results are shown in table 1.

As can be seen from table 1, when the bird's nest silk is directly sterilized at high temperature without treatment, the solid content obtained after sterilization is low, the water-melting phenomenon occurs during storage, and the solid content is completely melted after 6 months of storage, so that the bird's nest silk disappears. After the bird's nest silk is subjected to the denaturation treatment of glutamine transaminase and acid, the bird's nest silk and the acid transaminase are subjected to synergistic action, so that the internal structure of the bird's nest silk is tighter, and the thermal stability during high-temperature sterilization and the solid content during storage are improved.
Examples 6 to 8
On the basis of example 1, the influence of different treatment conditions on the solid content and sialic acid content was investigated.

As can be seen from table 2, the concentration of transglutaminase and pH at the time of acid shrinkage have a large influence on the content of solids after sterilization during the period when the content of solids has been stored, and when the concentration of transglutaminase is too low, the degree of cross-linking between molecules of bird's nest protein is low, and the content of solids at the later stage is low; when the pH value of the system is too low, the bird's nest silk structure shrinks too tightly, so that the good water holding performance of the bird's nest is lost, the smooth mouthfeel of the bird's nest is lost, the solid content of the bird's nest is reduced, and meanwhile, when the pH value is too low, sialic acid is hydrolyzed from the bird's nest protein molecules, so that the nutritional value of the bird's nest is reduced. Preferred conditions for internal crosslinking in the present invention: 2-3% of glutamine transaminase, 40-50 ℃ of treatment temperature and the condition of structure shrinkage: the concentration of the sulfuric acid solution is 0.05mol/L, the treatment temperature is 80 ℃, the time is 40min, and the purity and the yield of the obtained sialic acid product reach the best effect.
Examples 9 to 10
On the basis of example 1, the influence of different treatment conditions on the solid content and the sialic acid content was investigated, and the conditions and results of examples 9 to 10 are shown in table 2.

As can be seen from Table 2, after the bird nest silk is subjected to the glutamine transaminase and acid denaturation treatment, the bird nest silk and the acid transaminase act synergistically to enable the internal structure of the bird nest silk to be more compact, and the heat stability during high-temperature sterilization and the solid content during storage are improved. The inventor further researches and discovers that after the bird's nest silk is treated by the glutamine transaminase, the crosslinking degree of protein molecules in the bird's nest silk is improved, the pH value of a system is adjusted to 2, the internal structure of the bird's nest silk is further enabled to be more compact, the bird's nest silk and the protein molecules act synergistically, and the thermal stability of the bird's nest silk during high-temperature sterilization is improved. Meanwhile, when the pH value is adjusted to be acidic, a part of sialic acid is hydrolyzed from cubilose protein molecules, so that the nutritional value of cubilose is reduced.
Aiming at the problems that the cubilose shreds are easy to dissolve in the high-temperature sterilization process and the subsequent storage process and the like, the invention develops the convenient instant cubilose and the preparation method thereof based on the existing product form, the basic theory and a large amount of experimental research of the cubilose, so that the cubilose shreds are not dissolved in the high-temperature sterilization and later-period storage process, can still exist in a filiform form, and can ensure fine and smooth mouthfeel.
The invention provides a preparation method of convenient and instant cubilose, which is characterized in that cubilose filaments are treated by glutamine transaminase, so that the crosslinking degree of protein molecules in the cubilose filaments is improved, the pH value of a system is adjusted to 0-5, protein denaturation is further realized, the internal structure of the cubilose filaments is more compact, the cubilose filaments and the protein denaturation are synergistic, the thermal stability during high-temperature sterilization is improved, the cubilose filaments are not dissolved during high-temperature sterilization and later storage, and the problem of water melting of the cubilose filaments is avoided. After the treatment of glutamine transaminase, compared with the single acid denaturation treatment, the content of sialic acid is improved during the acid denaturation treatment, and the internal structure of the bird's nest silk is more compact and has inhibiting effect on the loss of sialic acid probably because the bird's nest silk is subjected to the glutamine transaminase and acid denaturation treatment. According to the invention, through glutamine transaminase treatment, the subsequent pH value of the system is adjusted, and corresponding parameters are optimized, so that the difficult problem of bird's nest filamentation water is avoided, the loss of sialic acid is reduced, and the nutritional value of the bird's nest is improved.
It should be noted that the above-mentioned embodiments are only for illustrating the technical solutions of the present invention and not for limiting, and although the present invention has been described in detail with reference to the preferred embodiments, it should be understood by those skilled in the art that modifications or equivalent substitutions may be made on the technical solutions of the present invention without departing from the spirit and scope of the technical solutions of the present invention, which should be covered by the claims of the present invention.
Claims (10)
1. A preparation method of convenient instant edible bird's nest is characterized by comprising the following steps: comprises the steps of (a) preparing a mixture of a plurality of raw materials,
soaking nidus Collocaliae raw material in normal temperature deionized water or purified water, removing nidus Collocaliae hair, cleaning for several times, taking out, draining to obtain pretreated nidus Collocaliae;
adding the drained bird's nest silk into a glutamine transaminase water solution for soaking, draining, soaking the drained bird's nest silk into water, adding acid to adjust the pH value of the system to 0-5, soaking for 20-600 min at 30-100 ℃, draining, cleaning with clear water at normal temperature, taking out the bird's nest silk and draining;
mixing the dried edible bird's nest shreds with water or sugar water, subpackaging, sealing, placing into a high-temperature pressure cooker, sterilizing at 121 deg.C for 20min, cooling, inspecting, and packaging to obtain the instant edible bird's nest.
2. The method for preparing instant edible bird's nest according to claim 1, which is characterized in that: the content of nitrite in the pretreated dry base of the cubilose is lower than 10 mg/kg.

The method for preparing instant edible bird's nest according to claim 1, which is characterized in that: the drained bird's nest silks are added into a glutamine transaminase aqueous solution to be soaked, wherein the mass concentration of glutamine transaminase in the glutamine transaminase aqueous solution is 2-3%.
4. The method for preparing instant edible bird's nest according to claim 3, characterized in that: the soaking temperature is 20-60 ℃, and the soaking treatment time is 6-8 h.

The method for preparing instant edible bird's nest according to claim 1, which is characterized in that: and adding acid to adjust the pH value of the system to 0-5, wherein the used acid is one or a combination of sulfuric acid, hydrochloric acid, phosphoric acid, nitric acid, citric acid, malic acid, acetic acid, lactic acid and oxalic acid.
6. The method for preparing instant edible bird's nest according to claim 1, which is characterized in that: and soaking at the temperature of 30-100 ℃ for 20-600 min, wherein the soaking temperature is 40-60 ℃, and the soaking time is 30-60 min.
