技术领域
本发明涉及中药组合物技术领域,特别涉及一种激活酪氨酸酶活性的中医药组合物及其制备方法。
背景技术
近年来,随着人们工作生活节奏的加快,脑力劳动的过度负担,生活的烦恼等原因,致使少年白发、中年白发的现象越来越普遍、越来越严重,影响人们的整体形象;针对白发丛生的现象,现有技术中主要使用永久性染发剂进行处理,其染发原理是采用氨水使头发膨胀和软化,毛鳞片打开从而使染料中间体和偶合剂更好的扩散渗透到皮质层,染料中间体和偶合剂发生氧化反应、偶合和缩合反应形成较大的染料分子,染料大分子被封闭在头发纤维内,从而使头发染上颜色。采用染发剂处理虽然暂时解决了白发问题,但没有从根本改善,随着新陈代谢的进行,新生毛发生长处出来后依然是白色的,需要定期对头发进行复染。
但是现有技术中的染发剂其化学组分复杂,长时间使用化学药剂会由头皮深入皮肤内部,严重影响人类的健康,不仅无法从根源上解决白发问题,还可能对毛孔及皮肤造成永久性损害,从而导致秃头、脱发甚至引发癌变。
传统中医学认为白发的病因主要在肾,若肝肾两虚气血不足,全身的血液循环疲软,无力将营养物质输送到人体直立的最高处“头顶”,头上毛囊得不到滋养,渐渐萎缩,就会引起脱发,且酪氨酸是合成黑色素的主要物质,肾气虚阻碍了酪氨酸活性的激发,难以合成黑色素,从而产生白发。以目前市场上所使用的的生发及白发变黑发的产品无法从根本上清除人体内血液里的杂质及促进黑色素的正常合成,难以从根源上达到生发及白发转黑的目的。
因此,如何提供一种中药组合物使其发挥最佳的协同作用,有效激活自身酪氨酸酶活性,从根源上解决白发,是本领域技术人员亟待解决的问题。
发明内容
有鉴于此,本发明提供了一种有效激活酪氨酸酶活性的中药组合物,按一定比例配伍而成,通过补肝肾益精、清热除烦、养心安神,激发毛囊细胞中酪氨酸的生成,进而激活酪氨酸酶的活性,促进毛囊细胞黑色素的生成达到黑发的作用。
为了达到上述目的,本发明采用如下技术方案:
一种激活酪氨酸酶活性的中药组合物,包括以下重量份的组分:
酪氨酸酶10~30份、何首乌1~40份、当归1~40份、马鞭草1~40份、红花1~30份、苦参1~20份、人参1~20份、党参1~20份、沙参1~20份、丹参1~20份、黑灵芝1~40份、黑芝麻1~40份、鸡血藤1~30份、冬虫夏草1~10份、咖啡黄葵果1~20份、银杏叶1~20份、黄精1~20份。
优选的,包括以下重量份的组分:
酪氨酸酶20~25份、何首乌10~35份、当归20~30份、马鞭草25~35份、红花10~20份、苦参10~15份、人参5~15份、党参5~15份、沙参5~15份、丹参5~15份、黑灵芝10~35份、黑芝麻10~35份、鸡血藤15~25份、冬虫夏草4~8份、咖啡黄葵果9~15份、银杏叶8~14份、黄精10~15份。
优选的,包括以下重量份的组分:
酪氨酸酶23份、何首乌23份、当归25份、马鞭草22份、红花17份、苦参13份、人参12份、党参12份、沙参11份、丹参11份、黑灵芝28份、黑芝麻29份、鸡血藤19份、冬虫夏草7份、咖啡黄葵果13份、银杏叶11份、黄精13份。
本发明所提供的一种激活酪氨酸酶活性的中药组合物,还可以加入其它有效物质载体。
优选的,所述载体为海藻酸钠、羧甲基纤维素钠、低聚糖。
本发明还提供了一种激活酪氨酸酶活性的中药组合物的制备方法,包括以下步骤:
(1)将何首乌、当归、马鞭草、红花、苦参、人参、党参、沙参、丹参、黑灵芝、黑芝麻、鸡血藤、冬虫夏草、咖啡黄葵果、银杏叶、黄精混合后粉碎,得到混合料;
(2)向混合料中加入蒸馏水和无水乙醇的混合物中浸泡60-80min后,加热煮沸后保持沸腾120min-150min,过滤,得滤液一和药渣;
(3)将药渣中加入蒸馏水,煮沸后保持沸腾20-40min,过滤,得到滤液二;
(4)合并滤液一和滤液二,静置24-30h,取上清液用柠檬酸调节pH为6.5-6.8,加入酪氨酸酶混合均匀后即得中药组合物。
优选的,所述步骤(1)中的粉碎粒度为≥10目。
优选的,所述步骤(2)中加入的蒸馏水为混合药材总重量的5-7倍,无水乙醇为混合药材的1-3倍。
优选的,所述步骤(3)中加入的蒸馏水为混合药材总重量的3-5倍。
本发明的上述技术方案中,酪氨酸酶作为外加剂,有效弥补酪氨酸酶不足从而导致黑色素合成量不足的问题,从而有效提升黑色素的合成;
何首乌:性苦、甘、涩,味温。养血益肝,乌须发,祛风。《本草求真》谓其“滋水补肾,黑发轻身”。现代医学证实,何首乌可调节神经与内分泌功能和营养发根,可促使头发黑色素生成,使头发变黑;
当归:伞形科当归属植物,味甘、辛、苦,性温,归肝、心、脾经。《日华子本草》载其“治一切风,一切血,补一切劳,破恶血,养新血”,有效促进血液循环;
马鞭草:性凉,味微葳,有凉血、散瘀、通经、清热、解毒、止痒、驱虫、消胀的功效;
红花:活血化瘀、散湿去肿;
苦参:养肝胆气,安五脏,定志益精,利九窍;
人参:补气生津,其浸出液可被皮肤缓慢吸收且无不良刺激,能扩张皮肤毛细血管,促进皮肤血液循环,增加皮肤营养,调节皮肤的水油平衡;
党参:性甘,平,具有补中益气之功效;
沙参:具有养阴清热之功效;
丹参:有祛瘀、生新、活血、调经等效用,有效促进血液循环;
黑灵芝:具有益肾气通九窍之功效,有效补气且促进血液循环;
黑芝麻:含有大量的脂肪和蛋白质,还含有糖类、维生素A、维生素E、卵磷脂、钙、铁、铬等营养成分。有健胃、保肝、促进红细胞生长的作用,同时可以增加体内黑色素,有利于头发生长;
鸡血藤:补肝肾、益精壮阳,促进血液循环;
冬虫夏草:可调节免疫系统功能,促进酪氨酸的合成,有效激发酪氨酸酶活性,并且其还具有良好的补肾益气以及促进循环的作用;
咖啡黄葵果:咖啡黄葵的营养丰富,幼果中含有大量的粘滑汁液,具有特殊的香味,其汁液中混有果胶,牛乳聚糖及阿拉聚糖,具有滋补阴阳之功效,含有丰富的维生素和矿物质元素,促进红细胞生长,有效促进酪氨酸的合成,进而激活酪氨酸酶活性,从而促进黑色素合成;
银杏叶:含有天然活性黄酮及苦内酯等与人体健康有益的多种成份,具有扩张血管的作用,可有效促进头部血液循环,提升代谢进而促进黑色素合成;
黄精:补中益气,具有良好的促循环作用;
即本发明的激活酪氨酸酶活性的中药组合物主要采用纯中药加食材为原料,按一定比例配伍而成,通过补肝肾、益精血、促循环等作用,激发毛囊细胞中酪氨酸的生成,进而激活酪氨酸酶的活性,促进毛囊细胞黑色素的生成达到黑发的作用,能够治疗长期处于高压状态,精神高度紧张等引起的白发增多,具有很好的疗效,且能完全控制白发继续生成。

下面将结合本发明的实施例,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
如下实施例的激活酪氨酸酶活性的中药组合物的制备方法,均采用以下步骤:
(1)将何首乌、当归、马鞭草、红花、苦参、人参、党参、沙参、丹参、黑灵芝、黑芝麻、鸡血藤、冬虫夏草、咖啡黄葵果、银杏叶、黄精混合后粉碎至粒度为≥10目,得到混合料;
(2)向混合料中加入混合药材总重量的5-7倍的蒸馏水和混合药材重量1-3倍的无水乙醇,浸泡60-80min后,加热煮沸后保持沸腾120min-150min,过滤,得滤液一和药渣;
(3)将药渣中加入混合药材总重量的3-5倍的蒸馏水,煮沸后保持沸腾20-40min,过滤,得到滤液二;
(4)合并滤液一和滤液二,静置24-30h,取上清液用柠檬酸调节pH为6.5-6.8,加入酪氨酸酶混合均匀后即得中药组合物。

酪氨酸酶10份、何首乌40份、当归1份、马鞭草40份、红花1份、苦参20份、人参1份、党参20份、沙参1份、丹参20份、黑灵芝1份、黑芝麻40份、鸡血藤1份、冬虫夏草10份、咖啡黄葵果1份、银杏叶20份、黄精1份。
实施例2
一种激活酪氨酸酶活性的中药组合物,包括以下重量份的组分:
酪氨酸酶30份、何首乌1份、当归40份、马鞭草1份、红花30份、苦参1份、人参20份、党参1份、沙参20份、丹参1份、黑灵芝40份、黑芝麻1份、鸡血藤30份、冬虫夏草1份、咖啡黄葵果20份、银杏叶1份、黄精20份。

一种激活酪氨酸酶活性的中药组合物,包括以下重量份的组分:
酪氨酸酶25份、何首乌10份、当归30份、马鞭草25份、红花20份、苦参10份、人参15份、党参5份、沙参15份、丹参5份、黑灵芝35份、黑芝麻10份、鸡血藤25份、冬虫夏草8份、咖啡黄葵果9份、银杏叶14份、黄精10份。
实施例5
一种激活酪氨酸酶活性的中药组合物,包括以下重量份的组分:
酪氨酸酶23份、何首乌23份、当归25份、马鞭草22份、红花17份、苦参13份、人参12份、党参12份、沙参11份、丹参11份、黑灵芝28份、黑芝麻29份、鸡血藤19份、冬虫夏草7份、咖啡黄葵果13份、银杏叶11份、黄精13份。
实验例
通过测定实施例1-5所制得的中药组合物对黑素瘤细胞增殖,酪氨酸酶活性以及黑色素合成的影响,评价其黑发养发功效。
实验材料:小鼠黑素瘤细胞;
细胞培养:用含10%胎牛血清的DMEM培养基37℃、5%CO2条件下常规培养,传代24小时后对细胞进行加药刺激,并根据实验要求加药处理适当的时间。
细胞增殖的测定:将传代细胞的浓度调至1×105个/mL加入96孔培养板,每孔0.1mL。孵育24小时,待细胞贴壁厚,吸去旧培养液,加入实施例1-5所制取的中药组合物,置于37℃、5%CO2条件下继续培养48小时,采用经典的MTT比色法测定细胞增值率,每一实施例设立3个复孔,取平均值,并设置培养基为空白对照。
细胞增殖率(%)=各浓度平均吸光度值/空白对照组平均吸光度值×100%;
酪氨酸酶活性测定:实施例1-5所制取的中药组合物作用细胞2天后,测定酪氨酸酶活性,每一实施例设立3个复孔,取平均值,并设置培养基为空白对照。
酪氨酸酶的激活率(%)=各实施例平均吸光度值/空白对照组平均吸光度值×100%;
黑色素含量测定:将配置的细胞悬液以1×105个/mL密度接种于6孔板,孵育1天后加入实施例1-5所制取的中药组合物继续培养2天,测定黑素含量;每一实施例设立3个复孔,取平均值,并设置培养基为空白对照。

从上述测试结果可看出实施例1-5所制备的中药组合物对小鼠黑色瘤细胞有促进作用;实施例1-5所制备的中药组合物可明显促进小鼠黑色素瘤细胞酪氨酸酶的活性;实施例1-5所制备的中药组合物可以明显促进黑色素含量的增多。
本说明书中各个实施例采用递进的方式描述,每个实施例重点说明的都是与其他实施例的不同之处,各个实施例之间相同相似部分互相参见即可。对于实施例公开的装置而言,由于其与实施例公开的方法相对应,所以描述的比较简单,相关之处参见方法部分说明即可。

Claims (8)
1.一种激活酪氨酸酶活性的中药组合物,其特征在于,包括以下重量份的组分:
酪氨酸酶10~30份、何首乌1~40份、当归1~40份、马鞭草1~40份、红花1~30份、苦参1~20份、人参1~20份、党参1~20份、沙参1~20份、丹参1~20份、黑灵芝1~40份、黑芝麻1~40份、鸡血藤1~30份、冬虫夏草1~10份、咖啡黄葵果1~20份、银杏叶1~20份、黄精1~20份。
2.根据权利要求1所述一种激活酪氨酸酶活性的中药组合物,其特征在于,包括以下重量份的组分:
酪氨酸酶20~25份、何首乌10~35份、当归20~30份、马鞭草25~35份、红花10~20份、苦参10~15份、人参5~15份、党参5~15份、沙参5~15份、丹参5~15份、黑灵芝10~35份、黑芝麻10~35份、鸡血藤15~25份、冬虫夏草4~8份、咖啡黄葵果9~15份、银杏叶8~14份、黄精10~15份。
3.根据权利要求1所述一种激活酪氨酸酶活性的中药组合物,其特征在于,包括以下重量份的组分:
酪氨酸酶23份、何首乌23份、当归25份、马鞭草22份、红花17份、苦参13份、人参12份、党参12份、沙参11份、丹参11份、黑灵芝28份、黑芝麻29份、鸡血藤19份、冬虫夏草7份、咖啡黄葵果13份、银杏叶11份、黄精13份。
4.一种激活酪氨酸酶活性的中药组合物,其特征在于,由有效剂量的权利要求1~3中任一项所述的中药组合物及药剂学所用载体组成。
5.一种激活酪氨酸酶活性的中药组合物的制备方法,其特征在于,包括以下步骤:
(1)将何首乌、当归、马鞭草、红花、苦参、人参、党参、沙参、丹参、黑灵芝、黑芝麻、鸡血藤、冬虫夏草、咖啡黄葵果、银杏叶、黄精混合后粉碎,得到混合料;
(2)向混合料中加入蒸馏水和无水乙醇的混合物中浸泡60-80min后,加热煮沸后保持沸腾120min-150min,过滤,得滤液一和药渣;
(3)将药渣中加入蒸馏水,煮沸后保持沸腾20-40min,过滤,得到滤液二;
(4)合并滤液一和滤液二,静置24-30h,取上清液用柠檬酸调节pH为6.5-6.8,加入酪氨酸酶混合均匀后即得中药组合物。
6.根据权利要求5所述的一种激活酪氨酸酶活性的中药组合物的制备方法,其特征在于,所述步骤(1)中的粉碎粒度为≥10目。
7.根据权利要求5所述的一种激活酪氨酸酶活性的中药组合物的制备方法,其特征在于,所述步骤(2)中加入的蒸馏水为混合药材总重量的5-7倍,无水乙醇为混合药材的1-3倍。
8.根据权利要求5所述的一种激活酪氨酸酶活性的中药组合物的制备方法,其特征在于,所述步骤(3)中加入的蒸馏水为混合药材总重量的3-5倍。
