Extract for treating gastric cancer and preparation method thereof
Technical Field
The invention belongs to the technical field of medicines, and particularly relates to an extract for treating gastric cancer and a preparation method thereof.
Background
Gastric cancer is a malignant tumor originated from gastric mucosa epithelium, and is clinically mainly manifested by epigastric distention and discomfort, dull pain, intolerance of food, progressive anemia and emaciation, mass in late epigastric period and persistent positive fecal occult blood. Pain and weight loss are the most common clinical symptoms of advanced gastric cancer. Patients often have more definite upper gastrointestinal symptoms, such as upper abdominal discomfort, fullness after eating, aggravation of upper abdominal pain along with the progress of the disease, appetite reduction and hypodynamia. It is also characterized by different tumor sites. Carcinoma of the gastric fundus of the cardia may have poststernal pain and progressive dysphagia; gastric cancer near the pylorus shows pyloric obstruction. The incidence of the gastric cancer is the first in various malignant tumors in China, and the gastric cancer has higher incidence and mortality. At present, stomach cancer tends to be younger due to changes in dietary structure, increased working pressure, infection with helicobacter pylori, and the like. Gastric cancer can occur in any part of the stomach, more than half of which occur in antrum, greater curvature, lesser curvature, anterior and posterior walls, etc. The treatment methods of gastric cancer include surgery, radiotherapy, chemotherapy, biotherapy and the like, and even if the methods are numerous, the treatment results are still unsatisfactory in general. The operation treatment has high risk, and simultaneously causes physical trauma to patients, and the patients need further treatment by combining chemotherapy or radiotherapy in the later period generally, and the chemotherapy or radiotherapy has great damage to normal cells of human bodies, great toxic and side effects and great physical damage. Biological antibody drugs are expensive to treat, and the treatment effect varies from person to person.
At present, drug therapy is still an important way for treating gastric cancer, but most of the existing drugs for treating gastric cancer are chemical drugs, are easy to generate drug resistance, and have large side effects and unobvious effects. As a preferred embodiment, the vinegar is selected from: one or more of Zhangjiajiuquxiangzhi vinegar, Shanxi mature vinegar, Zhenjiang aromatic vinegar, Fujia Yongchun mature vinegar, Lanzhonggning vinegar, Henan mature turtle special vinegar, Tianjin uniflow mature vinegar, Liyunggang dripping vinegar and Beijing Longmen rice vinegar. More preferably, the vinegar is Zhang Jiajiu Jiuquxiangzhi vinegar. The total flavone in the Zhang Jiajiu Jiuqu aromatic ester vinegar reaches 1800mg/100mL, and the content of lactic acid accounts for not less than 25% of the total acid; the soluble salt-free solid is not less than 25g/100mL and is far higher than the national standard 1.0g/100 mL; the content of the characteristic active peptide is not less than 4.5g/100 mL. In addition, the aspongopus japonicus ester vinegar also contains active substances such as polysaccharide, amino acid, ferulic acid, gamma-aminobutyric acid, ligustrazine, catechin, epicatechin and the like, and the active substances are high in abundance and high in medicinal value. The aspongopus fragrant ester vinegar contains rich active substances, has effects of sterilizing, removing toxic substances, preventing common cold, removing blood stasis, relieving swelling, astringing blood, stopping bleeding, expelling ascaris and killing trichomonad, and has remarkable therapeutic effect on digestive system diseases, especially gastric cancer.
In a preferred embodiment, the vinegar has a total acid concentration of 8 to 16g/100 ml. When the total acid concentration of the vinegar is 8-16g/100ml, the extraction efficiency of active substances in the traditional Chinese medicinal materials is optimal, and the active substances in the traditional Chinese medicinal materials can be extracted fully and efficiently. In some embodiments, the vinegar is prepared by solid state fermentation.
As a preferred embodiment, the preparation of the vinegar at least comprises: soaking the raw materials in water, steaming, and stewing to obtain raw materials to be fermented; and then, sequentially carrying out alcoholic fermentation, acetic fermentation, fumigating, vinegar pouring, vinegar freezing and aging on the raw materials to be fermented. Specifically, the preparation method of the vinegar comprises the following steps: a) adding water into the raw materials for soaking to ensure that the raw materials fully absorb water to obtain soaked grain raw materials; b) adding water accounting for 140-240% of the weight of the grain raw materials into the soaked grain raw materials, and stewing the materials for 15-80 min to obtain the raw materials to be fermented; c) adding famous wine fungus series yeast which accounts for 20-70% of the weight of the cereal raw material into the raw material to be fermented, uniformly mixing, adding water which accounts for 40-70% of the total weight of the cereal raw material, and fermenting for 15-730 days at 28-35 ℃ to obtain wine mash; d) adding grain processing peel substances accounting for 100-300% of the weight of the grain raw materials into the fermented glutinous rice as loose materials, uniformly mixing, adding vinegar fermented grains accounting for 5-15% of the weight of the grain raw materials and fermenting for 6-8 days for acetic fermentation for 8-360 days to obtain vinegar fermented grains; e) smoking 25-75% of the fermented vinegar residue obtained in the step d) at 60-80 ℃ for 4-8 days to obtain smoked fermented residue products; f) adding cold water and the light vinegar liquid for pouring vinegar into the rest fermented vinegar grains to increase the total weight to 2-3 times of the original weight, soaking for more than 12 hours, and pouring out the vinegar liquid to obtain a first vinegar liquid; heating the first vinegar liquid to 80-90 ℃, adding the smoked grain product, soaking for more than 10 hours, and leaching out the vinegar liquid again to obtain a second vinegar liquid; g) freezing the second vinegar liquid obtained in the step f) in a freezing environment to form vinegar blocks; then, placing the vinegar blocks in an environment with the temperature of 0-20 ℃ to slowly melt the vinegar blocks, and collecting flowing thick vinegar liquid; h) putting the concentrated vinegar liquid collected in the step g) into a brewing jar, and brewing in the open air for at least 12 months to obtain the edible vinegar. The contents of active substances such as flavone, polysaccharide, polyphenol, active peptide and the like in the vinegar prepared by the embodiment of the invention are high, so that on one hand, the vinegar is beneficial to exerting a better treatment effect; on the other hand, the method has better extraction effect on the traditional Chinese medicinal materials matched with the vinegar. Therefore, the content of each active substance in the prepared medicine for treating gastric cancer is higher, the synergistic coordination mechanism among the active substances is more complete, and the curative effect of the medicine is better.
Specifically, in the step S20, the Chinese medicinal material components are pretreated, crushed, and sieved with a 10-80 mesh sieve to obtain powdery Chinese medicinal material components. The pretreated Chinese medicinal material components are crushed, which is favorable for the extraction of active substances in the subsequent Chinese medicinal material components. On one hand, if the crushing granularity is too large, the extraction of active substances is slow, insufficient extraction is easy to occur, raw materials are wasted, and the process cost is increased. On the other hand, the small crushing particle size destroys the crystal phase structure of active substances in the traditional Chinese medicinal materials matched with vinegar, damages the efficacy of the active substances and increases the separation difficulty.
As a preferred embodiment, the method of the pretreatment is: cleaning the traditional Chinese medicine components, and drying for 68-80 hours at 45-65 ℃. The method comprises the steps of cleaning the Chinese medicinal material components with purified water, removing impurities such as silt and deteriorated medicinal materials in the Chinese medicinal material components, drying the cleaned Chinese medicinal material components to obtain cleaned dry Chinese medicinal material components, and carrying out follow-up pharmacy. The pretreated traditional Chinese medicine components can not introduce unnecessary impurity components to the subsequent pharmacy process, and the pollution of the medicines at the source is avoided.
In a preferred embodiment, the pulverization is performed by a pulverizer or a pulverizer. In some embodiments, the traditional Chinese medicine components are crushed by a high-speed crusher to enable the particle size of the traditional Chinese medicine components to reach 10-80 meshes.
Specifically, in step S30, vinegar is used to extract the powdered Chinese medicinal components to obtain an extract. In a preferred embodiment, the powdered Chinese medicinal materials are extracted 1 to 3 times with vinegar of 7 to 10 times volume, respectively, and the extracts are combined. The vinegar is used as an extraction solvent to extract powdery traditional Chinese medicinal material components for multiple times, on one hand, the vinegar greatly improves the dissolution of active substances of the traditional Chinese medicinal material components such as periplaneta americana, dandelion, pseudo-ginseng, ginseng and the like, and the active substances in the traditional Chinese medicinal material components are completely dissolved out through multiple extraction, so that the utilization rate of the traditional Chinese medicinal material components is improved, and the content of the active substances in the medicine is improved. On the other hand, solved the many-sided problem that exists of traditional soaking with white spirit, if: the dissolution efficiency of the soaking active substances is low, the dissolution speed is slow, and many patients can not drink alcohol and even have alcohol allergy. Through multiple extraction, active substances in medicinal materials matched with vinegar are fully dissolved out, the medicine is utilized at the maximum efficiency, and the content of the active substances in the medicine is increased, so that the curative effect of the medicine is improved. In some embodiments, a first extraction of powdered herbal ingredients is performed with 7 volumes of vinegar; then, vinegar with 8 times of volume is adopted to carry out secondary extraction on the Chinese medicinal material components after the primary extraction; and extracting the Chinese medicinal material components after the second extraction for the third time by using vinegar with 9 times of volume, and finally combining the extracted products of the three times to obtain the extracting solution.
As a preferred embodiment, the method of extracting includes: extracting for 1-5 hours at the temperature of 20-80 ℃ and the rotating speed of 50-80 r/min, and filtering to obtain filtrate, namely the extracting solution. According to the embodiment of the invention, low-temperature extraction is adopted, so that the damage of high-temperature decoction to effective components is effectively avoided, the active substances in the extract for treating gastric cancer are kept in the optimal active states, and the optimal drug properties of the active substances are favorably exerted.
Specifically, in step S40, the extract is subjected to concentration, filtration, and sterilization in this order to obtain an extract for treating gastric cancer. The excessive water in the extracting solution is removed through concentration treatment, and the effective active substances in the medicine are concentrated, thereby being beneficial to taking, storing and transporting the extracting solution. And then filtering to remove impurity particulate matters in the concentrated medicine. Through sterilization treatment, impurity bacteria and other substances which are inevitably introduced in the preparation process of the extracting solution are removed, and the extracting solution is purified, so that better drug effect is brought into play.
As a preferred embodiment, the method of the concentration process includes: freezing the extracting solution into solid blocks at the temperature of-25 to-18 ℃; and (3) placing the solid block of the extracting solution at room temperature, slowly melting the solid block, collecting the molten liquid, and stopping collecting when the solid block becomes white. According to the embodiment of the invention, freezing concentration treatment is adopted, water molecules in the extracting solution are solidified into ice crystals by utilizing the solid-liquid phase equilibrium relation of the extracting solution below a freezing point, and ice is removed by a mechanical means, so that the solvent water in the solution is reduced, the concentration of the solution is improved, and the solution is concentrated. The concentration method does not affect the activity of active substances in the extract. When the freezing temperature is too high, the extract is not fully frozen, the melting speed of the extract block is not balanced, the concentration effect is not good, and the water content in the concentrated solution is high.
