背景技术
数据采集、存储和处理的不断发展使得医疗方法和治疗策略越来越为患者定制化。作为治疗前分期过程的一部分,获得关于原发肿瘤和潜在肿瘤的定位、组织密度、肿瘤范围(tumour extent)、灌注和/或代谢状态和分化程度的肿瘤相关信息。信息量使得可以基于跨学科共识为各患者开发指南导向的、个性化的治疗概念。对于具有治疗意图的主要手术治疗概念,在不损失术中检查和恶性组织划分(demarcation)的信息的情况下,不能应用分期过程的信息。术中获取有关侵袭性癌组织(invasive carcinoma tissue)的范围和定位的信息限于临床视觉和触诊发现,并且术中组织学控制限于软组织切除边缘(margin)的代表性区域。位点区域(loci-regional)复发和转移可能是侵袭性癌细胞的潜在残留物引起的,且代表了治愈性治疗概念中的限制因素。
特别地,恶性组织的不完全去除降低了肿瘤治疗概念的成功。例如,在头颈癌中,残留的癌组织(R1状态)导致局部复发100%的增加,在5年内90%的死亡风险。1,2

基于作为侵袭性癌细胞增殖、去分化和浸润(infiltrate)到间质组织中的上皮细胞经历分子和形态学的上皮-间质转变(epithelial-mesenchymal transition,EMT)相关的变化的假设,认为EMT-标记蛋白整合素αvβ6是用于侵袭性癌细胞的合适靶标。EMT是胚胎发育期间至关重要的生理过程。在纤维化组织重塑、伤口愈合、侵袭性癌生长和肿瘤转移过程中也观察到了相应的细胞变化。10在肿瘤学方面,如图1所示,整合素αvβ6的膜结合表达介导侵袭性癌细胞的致癌潜力,抑制凋亡并促进侵袭性增殖。αvβ6的表达在具有仅限于上皮细胞、发育中的肺组织和肾脏上皮细胞的高水平的胚胎发育期间开始。11,12在生理学方面,在分化的上皮细胞中没有构成型表达αvβ6。然而,在包括伤口愈合和致癌作用(carcinogenesis)的组织重塑方面,它再次被上调。13已经在几种不同类型的恶性肿瘤中描述了αvβ6表达的普遍性(prevalence)。αvβ6被称为“癌症整合素”。14在文献中已知αvβ6表达与侵袭性癌症和转移相关联。15-38

还存在关于使用另一种化合物的术中生物成像的出版物。40b然而,尽管作者描述了所述化合物表现出αvβ6特异性结合,但担心所述化合物也可能结合αvβ3,使得所述化合物对确实表达αvβ6的头颈癌肿瘤组织的术中生物成像的适用性仍然值得怀疑。
将染料偶联到肽配体上会涉及负面影响配体结合特性的风险。此外,染料可能会影响体内的生物利用度和分布。这些也是成功的术中生物成像的重要属性。
发明目的
鉴于上述困难,本发明的目的是提供适于术中生物成像的化合物。特别地,本发明的目的是提供含有肽配体部分和荧光染料部分的化合物,其表现出对于表达αvβ6的癌细胞的高亲和性和选择性、高的荧光强度的组织背景比,并且该化合物表现出高生物利用度,且不会在健康组织中积聚。根据本发明的进一步目的,提供了额外的表现出高生物稳定性和长荧光稳定性的此类化合物。

发明内容
本发明通过提供实现上述目的的化合物来解决上述问题,因此允许侵袭性癌细胞的术中分子生物成像,以实时获得关于侵袭性癌组织的位置和范围的直接视觉信息。术中癌组织的视觉检测和划分可以辅助手术治疗,以改进对切除边缘的侵袭性癌细胞的控制,并有助于具有治愈意图的手术治疗概念。

“前体”是指带有官能团、并在一个或多个化学反应步骤、通常是偶联步骤中产生相应部分的化合物。当然,考虑到预期的合成途径,前体也可以视情况而定而带有保护基。

选择性结合αvβ6的部分A
原则上,可以使用任何表现出与αvβ6的强的、选择性结合的部分。本发明具体地提供了化合物,其中部分A源自WO 2017/046416A的化合物18,其为环状九肽cyc(FRGDLAFp(NMe)K)。该肽化合物已在WO 2017/046416A中显示出表现出对αvβ6的高结合亲和性和选择性。在实施例部分中,将其称为化合物OM_1204。
源自荧光染料的部分B

在本发明中,优选使用NHS-酯化合物作为原料。当与肽或接头的氨基偶联时,NHS-酯基将被所述氨基取代以产生酰胺基。
接头/间隔子
接头将肽部分A与染料部分B连接。使用具有合适长度的接头作为使染料部分对结合αvβ6的肽部分的干扰最小化的手段可能是有利的。以这种方式,肽结合的亲和性和选择性可以被保留,甚至可能增加。
原则上,在本发明的上下文中可以使用任何二价化学基团作为接头。合适的尤其是在WO 2017/046416 A中描述的接头。优选的是源自具有两个官能团的前体化合物的接头,该两个官能团对结合配偶体的官能团具有反应性。这样的官能团可以是(活化的或未活化的)羧酸基团、氨基、适合于点击化学偶联的官能团,诸如叠氮化物基团、乙炔基团和二苯基环辛炔(DBCO)基团。
例如,赖氨酸残基的ω-氨基有利地与羧基反应以产生酰胺基。
为了利用这些有利的偶联反应,优选采用在一端带有羧基而在另一端带有氨基的接头。可以在合成过程中根据需要适当地保护这些官能团。
在两个反应性基团之间,接头通常包含原子链。在接头的主链(backbone)中,该链的长度优选为1至10个原子。这些原子通常独立地选自C、O、N、S和P、优选具有C。这些原子的自由价当然被氢或非反应性取代基所饱和,该非反应性取代基不干扰肽部分与αvβ6靶受体的结合。更优选的是具有4-6个碳原子的亚烷基链的接头。优选的是源自具有5至7个碳原子的ω-氨基烷基羧酸的。最优选的是使用源自6-氨基己酸(AHX)的接头。
本发明的化合物及其制造方法
本发明的化合物通过偶联反应从WO 2017/046416 A的化合物18和荧光染料获得。任选且优选地,该偶联是通过间隔子/接头完成的。因此,本发明的化合物可以由以下通式表征:

肽部分中的偶联位置是赖氨酸残基的ω-氨基。通过引入脯氨酸侧链或相邻苯丙氨酸侧链的取代基的偶联原则上也是可能的,但是将需要更大的合成工作(syntheticeffort),并且至少由于这个原因,这种选择不是优选的。
如上所述,额外的接头/间隔子的使用并非是绝对必须的。这是因为800CW的反应性NHS-酯偶联基团通过五个碳原子的亚烷基链从发色团分离。该亚烷基链(与赖氨酸侧链一起)可以用作“内部”接头/间隔子,实现了肽部分A从染料部分B的所需空间分离。

可以使用合适的反应性基团之间的常规偶联反应来制备本发明的化合物。各个步骤的顺序不受限制。因此,它涉及以任何合理的顺序的以下步骤:
·提供肽部分前体;

·将肽部分前体的一个官能团与接头前体偶联;
·提供染料前体;
·将接头前体的另一个官能团与染料前体偶联。
依赖本领域公知的既定过程执行偶联反应。
根据一个方面,肽部分A和接头之间的偶联可以首先完成,然后将所得到的分子偶联至染料前体。根据另一方面,接头前体可以首先偶联至染料前体,然后将所得到的分子偶联至肽部分前体。根据再另一方面,接头前体与肽部分前体的偶联可以整集成到肽合成中。例如,接头可以与线性肽前体偶联。在该偶联步骤之后,肽被环化,然后与染料前体偶联。该最后的替代方案由下面的实施例1的过程来说明。
本发明化合物的用途
本发明的化合物可用于侵袭性癌组织的体内标记和检测。该特征使本发明的化合物可用于术中生物成像。
本发明化合物的具体用途还包括术前划分、术中划分和切除侧的控制,以控制残余的侵袭性αvβ6阳性癌细胞和切除侧的术后控制,这也可以用作肿瘤患者的后续护理的附加手段。所有可被认为是上述应用的αvβ6阳性恶性肿瘤总结于表1中,并以绿色标记。
在这种情况下,其他可能的用途是切除状态的控制和淋巴结的绘制。
术中生物成像是本领域已知的。最近已经综述了相关文献。41手术中生物成像可以如该综述文章和其中引用的文献中所述进行(当然,主要区别在于使用本发明的化合物代替这些文献中描述的化合物)。参考文献42和43还描述了用于术中生物成像的合适方法,其可以适用于本发明的化合物。
原则上,术中生物成像包括以下步骤/动作:
(a)化合物的系统给药。尽管其他给药形式本身不排除在外(只要实现所需的生物分布和稳定性),但是这通常通过静脉给药(i.v.administration)完成。
对待施用的制剂没有特别限制。对于静脉给药,可以使用典型的注射用溶液/悬浮液。特别合适的是基于具有5%二甲基亚砜(DMSO)的无菌磷酸盐缓冲盐水(PBS)的制剂。适当地调整总剂量和浓度,从而实现所需的TBR(荧光强度中的肿瘤背景比(tumor tobackground ratio))。还可以适当调整给药和手术之间的时间间隔,以实现期望的TBR。24小时的时间间隔可能会给出令人满意的结果。
(b)用能够通过染料部分诱导荧光发射的光照射具有疑似肿瘤细胞的组织。这有利地借助于激光来完成。文献42中描述了用于照射的适当设备,尤其是结合其“2.2成像系统”部分描述的第一照相机系统所描述的激光光源。
(c)检测荧光发射。再次,文献42中描述了合适的设备,尤其是该文献“2.2成像系统”部分所描述的第一照相机系统。
(d)手术切除显示荧光发射的组织、或被显示荧光发射的组织包围的组织。
本发明的化合物可用于与表达αvβ6的任何类型的癌症有关的术中生物成像和相关用途。这尤其包括结肠癌、胃癌(gastric carcinoma)、口腔鳞状细胞癌(OSCC)、胰腺导管腺癌、肠腺癌、头颈鳞状细胞癌(HNSCC)、侵袭性子宫内膜癌、基底细胞癌、乳腺癌、子宫内膜癌、胃癌(gastric cancer)、肝癌、非小细胞肺癌、肺癌脑转移、卵巢癌、胰腺癌和前列腺癌。
实施例
1、合成实施例-本发明化合物和对比化合物的合成
1.1带有接头的环肽的合成:cyc(FRGDLAFp(NMe)K(Ahx))作为本发明化合物的前体,且cyc(FRADLAFp(NMe)K(Ahx))作为对比化合物的前体。
遵循标准Fmoc策略46,在2-氯-三苯甲基聚苯乙烯(2-CTC)树脂上合成环肽,然后在溶液中环化。如别处47所述,在树脂上进行N-甲基化。简而言之,Fmoc保护的甘氨酸(或分别为丙氨酸)(1.2eq)用DIEA(2.5eq)在无水DCM(2mL)中固定在树脂(0.969mmol/g)上1小时。使用Fmoc-aa-OH(2eq)、HATU(2eq)和DIEA(5eq)在DMF中进行1小时来完成肽链延长。赖氨酸侧链的Dde保护用于具有Boc-Ahx-OH的线性肽的正交树脂上修饰。在DCM中用20%HFIP从树脂上裂解后,线性肽在DMF中使用DPPA(3eq)和NaHCO3(5eq)环化16h。随后将环状肽用TFA/DCM/TIPS/水(80:15:2.5:2.5%)去保护1小时。
1.2肽接头前体与染料前体的偶联
使用HPLC纯化的cyc(FRGDLAFp(NMe)K(Ahx))或cyc(FRADLAFp(NMe)K(Ahx))分别(1eq),青色素-5.5NHS酯(1eq)、或青色素-7.5NHS酯、或IRDye800CW NHS-酯和DIEA(3eq)在DMF中执行荧光标记1小时(通过HPLC-MS监测)。最终将偶联物通过半制备型HPLC纯化并冻干,分别得到目标化合物OM1231、FRX103、FRX109或FRX110。
化合物OM1231为对比化合物,其含有源自WO 2017/046416A的化合物18的肽部分,该肽部分通过AHX接头与源自青色素-5.5染料的染料部分偶联。
化合物FRX103为对应于OM1231的另一对比化合物,但是其中掺入了青色素-7.5染料代替青色素-5.5染料。

染料的性质影响整个肽-染料复合物与整合素的结合亲和性(表1)。本发明的化合物FRX110对整合素αvβ6表现出优异的结合亲和性和选择性,这使得该化合物特别适合于术中成像。
2、参考实施例1–癌细胞中αvβ6表达的体外评估
进行了侵袭性癌细胞的分子生物学和功能表征。鉴定了EMT标记蛋白整合素αvβ6,其被重新表达为细胞去分化的一部分,并在侵袭性癌细胞的间质边缘处的信号分子(TGF-β)的影响下重新表达。侵袭性癌组织的致癌功能和增殖前沿(proliferation front)的独特表达模式使整合素αvβ6成为检测侵袭性癌细胞并将其与健康组织中区分开来的靶点。
2.1免疫组织化学分析
在从骨浸润的(bone-infiltrating)FFPE癌样品的4μm厚的切片(section)上执行整合素αvβ6的免疫组织化学染色。热固定后,将石蜡载玻片脱蜡以及再水化。在黑暗中用0.3%(v/v)MeOH/H2O2封闭内源性过氧化物酶20分钟。在用PBS(2%(w/v)牛血清白蛋白(BSA))脱去(unmasking)抗原并封闭非特异性位点后,将载玻片与针对人整合素αvβ6(百健艾迪(Biogen Idec),剑桥,MA,USA)的单克隆抗体(稀释1:2.000)一起孵育。49在孵育2小时并在PBS中洗涤步骤后,应用生物素化的次级兔抗小鼠抗体(稀释1:200;达科(Dako),格洛斯楚普(Glostrup),丹麦),然后用辣根过氧化物酶(HRP)-链霉亲和素复合物(达科,格洛斯楚普,丹麦)处理30分钟。为了可视化免疫复合物,在0.05M Tris-HCl缓冲液(pH 7.6)中含有0.0018%(v/v)的H2O2的0.05%(w/v)的二氨基联苯胺溶液(试剂盒5001;达科)在施用至切片。癌细胞膜的棕色染色表明整合素αvβ6呈阳性。在不存在初级抗的情况下的染色用作对照,并且是阴性的。
2.2整合素αvβ6表达的免疫组织化学评估
整合素αvβ6蛋白质表达通过免疫反应评分(IRS)在不同的目标区域(regions ofinterest,ROI)中进行半定量评估。在癌组织中,定义了三个ROI:侵袭前沿、从侵袭前沿(invasive front)到中心的过渡和癌的中心。浸润骨的癌细胞巢内的位置被细分为三个ROI:与基质组织接壤的区域、癌细胞巢的中心区域和癌组织内的细胞角蛋白沉积区域(角蛋白珠(keratine pearl))。
在细胞水平上的定位分为亚细胞ROI:细胞膜、胞质溶胶和细胞核。最后,评估健康的上皮和基质组织。
对于各ROI,应用IRS标准:A=αvβ6阳性细胞的百分比:0=否,1=<10%,2=10-50%,3=51-80%,4=>80%阳性细胞。B=染色强度:0=否,1=轻度,2=中度,3=强染色。通过A×B计算出各ROI的最终IRS评分,整合素αvβ6表达定义为:0-1=阴性,2-3=低,4-8=中,9-12=高。
使用配对样本符号测试来计算类别之间的差异,显着性水平为p≤0.001。
图2示出了该实验的结果:
A)αvβ6的表达定位在浸润性癌的侵袭前沿。在癌的中心区域没有检测到αvβ6。周围的基质显示出炎症反应,没有诱导αvβ6表达。
B)在松质骨中浸润性癌细胞的整合素αvβ6表达。上皮角蛋白珠不显示αvβ6表达。
C)特定αvβ6表达的图示,这限于癌细胞膜。
该图呈现了所研究的癌样本(n=55)的百分比部分(y轴),并说明了与定位相关的表达水平(x轴)。
总之,在癌侵袭区域中可以检测到最高的表达水平。癌细胞浸润间质组织的特征是高整合素αvβ6表达水平。
3、参考实施例2–体外结合癌细胞
3.1细胞系和培养条件
人OSCC细胞系HN从德国布伦瑞克的德国微生物和细胞培养物保藏中心(Collection of Microorganisms and Cell Cultures)(DSMZ号ACC 417(HN))购买。HN细胞系从软腭的侵袭性鳞状细胞癌的颈淋巴结转移而建立。转移发生在原发肿瘤治疗后7年,转移到肺和脑。49作为对照细胞,我们使用了低整合素αvβ6和高整合素αvβ3表达的人卵巢癌细胞系OVMZ6。50所有细胞均在杜尔贝科改良的依格尔培养基(Dulbecco′s ModifiedEagle′sMedium,DMEM)(西格玛-奥尔德里奇(Sigma-Aldrich),圣路易斯,密苏里州,美国)中培养,并补充了10%(v/v)胎牛血清(FCS)(Gibco,LifeTechnologiesTM,卡尔斯巴德,加利福尼亚州,美国)。
3.2细胞整合素αvβ6的免疫细胞化学检测
HN和OVMZ6细胞在纤连蛋白包被的微室载玻片(ChamberSlideTM系统,西格玛-奥尔德里奇)上生长,并在室温(RT)下在2%(w/v)多聚甲醛(PFA)中固定15分钟,在PBS中洗涤一次,然后在磷酸盐缓冲液盐水(PBS)、2%(w/v)牛血清白蛋白(BSA)中在室温下封闭1小时。指向整合素αvβ61的单克隆抗体(1.6μg/mL)在1%(w/v)BSA中、在PBS中的细胞上于室温下孵育2小时,随后在室温下加入次级Alexa-488标记的山羊抗小鼠IgG(0.6g/mL)45分钟。将载玻片安装在PBS中,并通过蔡司(Zeiss)LSM 700(蔡司,耶拿(Jena),德国)来评估荧光强度。为了将荧光染色强度转换为辉标度(glow scale)的颜色,应用LSM扫描软件Zen(蔡司)提供的查找表(LUT)“橙色至白色”:低强度(红色)、中强度(黄色)和高强度(白色)。
该实验的结果如图3a所示。在该图中,左面板(panel)显示了在HN细胞(A,B)和OVMZ6细胞(E,F)中的整合素αvβ6表达。HN细胞的特征是高整合素αvβ6表达水平。在OVMZ6细胞中,几乎不存在整合素αvβ6。
右面板显示了HN细胞(C,D)中整合素αvβ3的低表达。相反,OVMZ6细胞显示了升高的整合素αvβ3(G,I)表达水平。
3.3整合素αvβ6靶向OM_1231偶联物的细胞结合实验
HN和OVMZ6细胞在纤连蛋白包被的微室载玻片上以、25×103/孔的密度培养24小时,然后在室温下在2%(w/v)PFA中固定15分钟,并在PBS中洗涤。溶于PBS、5%(v/v)二甲基亚砜(DMSO)中的Cy5.5偶联化合物OM_1231以10μM的最终浓度在室温下施用1小时,然后在PBS中洗涤3次。为了证明OM_1231对于整合素αvβ6的结合特异性,加入OM_1231(10μM)之前,将其未标记的类似物OM_1204在室温下以10倍摩尔过量在细胞上孵育1小时。作为对照肽的是用Cy5.5荧光染料(代码OM_1224)通过赖氨酸侧链NH2基团标记的非结合环(RβADfK)。在培养期后,将细胞在PBS中洗涤6次、并安装载玻片。如上所述,通过显微镜蔡司LSM 700(蔡司)检测荧光信号强度。通过向荧光图像应用LUT“蓝色到红色”来合并图像,低强度:蓝色;中强度:绿色;高强度:红色。
图3b示出了该实验的结果:
在图3b中,面板A-C:用Cy5.5标记的化合物OM_1231处理的HN细胞在细胞膜上显示出强荧光信号强度。
D-F:非结合Cy5.5标记的化合物(OM_1224)不产生荧光信号强度。
G-I:阻断研究:通过首先添加未标记的OM_1204,然后用其Cy5.5偶联类似物(OM_1231)孵育细胞进行的结合竞争实验导致荧光信号强度的强烈下降。
J-L:OM_1231并未识别OVMZ6细胞上的细胞整合素αvβ3。
M-O:整合素αvβ6阴性的上皮细胞未显示αvβ6选择性RGD肽的任何结合。来自39。
因此,体外分析证实了对比化合物OM_1231与αvβ6阳性癌细胞的特异性结合,该对比化合物OM_1231是一种功能化的αvβ6特异性RGD肽。
4、发明性实施例1–本发明化合物与癌细胞的体内结合
开发了一种原位HNSCC PDX(头颈部鳞状细胞癌患者衍生的异种移植)小鼠模型,该模型允许侵袭性、正交各向异性的人类癌组织的分析。将HNSCC组织样品移植到NOD/SCID小鼠的颈部区域。这提供了间质组织的环境中的上皮衍生的癌组织,以模拟侵袭性癌组织与间质之间的上皮-间质界面。
在HNSCC PDX小鼠模型中,整合素αvβ6特异性PET成像显示了整合素αvβ6作为侵袭性癌组织的敏感诊断标记。45
4.1具有人癌细胞的小鼠模型
在手术期间从增生性侵袭区域收集人癌症样本,并将该人癌症样本移植到含有1%青霉素/链霉素和2.5μg/ml两性霉素B的杜尔贝科改良的依格尔培养基(DMEM)中。将癌组织切成2×2×2mm的组织块,在切除后1-2小时将其移植到NOD/SCID小鼠的颈部肌肉组织中。
类似地,将健康的口腔上皮移植用于对照实验。
将动物放在专门的通风笼子中抚育小鼠(Tecniplast IVC)。根据欧盟指令(EUDirective)2010/63,鼠笼中的最大饲养密度(I型超长(Superlong),基础面积16×37cm)是基于动物重量。食物(丸化的10毫米圆形的高压灭菌的鼠食物大鼠/小鼠,特殊处理,住房饲料(housing feed)号1324SP,Fa,阿尔托明(Altromin))和水(酸化的饮用水(1N HCl,),每周更换一瓶)可随意接受动物。特殊的木质颗粒(Select Fine,Ssniff)用作垫料(litter),每周两次更换。筑巢材料以高压灭菌纸浆的形式提供,还有红色的聚碳酸酯鼠室(Bioscape)。房间专门设计为牲畜房(人员受限的出入控制,由培训过的动物护理技术人员进行护理,空气调节,12小时的明暗节律、每个具有微光(twilight)阶段)。动物的抚育按照EU指令2010/63中列出的条件进行。
手术过程:
固定小鼠并称重、以施用腹膜内麻醉。术前施用皮下镇痛剂(卡布洛芬(Rimadyl),4.0-5.0mg/kg以稀释1:10,10ml/kg体重),术后头三天每24小时也给予镇痛剂作为止疼药。
麻醉:
对于完全拮抗的麻醉,用27g插管(cannula)腹膜内给予咪达唑仑(midazolam)(5mg/kg)、美托尼定(medetonidine)(0.5mg/kg)和芬太尼(fentanyl)(0.05mg/kg)。大约每30分钟以起始剂量的1/3进行一次后续给药。通过皮下给药氟马西尼(flumazenil)(0.5mg/kg)、阿帕米唑(atipamezole)(2.5mg/kg)和纳洛酮(naloxone)(1.2mg/kg)对药物产生拮抗作用。
异种原位移植的手术过程:
在控制手术耐受性之后,将小鼠放在温暖的垫上并给予潘森(Bepanthen)眼药膏。仔细清洁脸颊区域和下颌角的手术区域,被毛(coat)在颈部处轻柔的剃掉大约7×7mm的区域。消毒皮肤,用解剖刀切断皮肤和皮下组织。切口的长度约为5mm。将组织制备到咬肌的表面。咬肌抻开(stumped open),将大约3×3×3mm的来自人类癌组织片植入到小鼠的颈肌中。癌组织是在手术中从人鳞状细胞癌的侵袭区域术中获得的,使用拉诺维酮(Braunovidone)防腐预处理并迅速提供以用于异种移植。植入癌组织后,检查手术部位并用维克里尔(Vicryl)6-0缝线缝合。
术后管理:
镇痛:麻醉时已经向小鼠皮下注射了0.05mg/kg剂量的丁丙诺啡(buprenorphine)。之后,以8小时的节律(rhythm)给予丁丙诺啡24小时。此外,术前准备的卡布洛芬每24小时给药一次。
4.2本发明化合物的给药和生物成像
如前所述,通过HNSCC PDX小鼠的尾静脉,将FRX110作为无菌PBS中的10-100μM溶液(200μl)缓慢给药,并在给定时间点进行成像。
使用750nm CW激光二极管(BWF2-750-0,B&W泰克,特拉华州,美国)、依靠42描述的设备执行生物成像,使用最大功率为300mW来激发荧光分子。由250W卤素灯(KL-2500LCD,肖特AG,美因茨(Mainz),德国)进行照明。在85mW/cm2处、在15厘米的工作距离下测量激光功率,根据美国国家标准协会(American National Standards Institute,ANSI)的标准规定,该激光功率低于最大允许照射量(exposure)。使用短通滤波器(E700SP,色彩技术(Chroma Technology),罗金厄姆(Rockingham),佛蒙特州,美国)消除了场照明NIRF信号的分量(component),以排除荧光检测场和场照明光路(F1)之间的干扰。磨砂玻璃扩散器(DG10-220,索尔实验室(Thorlabs),牛顿(Newton),新泽西州,美国)用于实现来自两个光源(D)的均匀照明。光学信号通过电动变焦/聚焦透镜(CVO GAZ11569M,Goyo光学公司,朝霞市(Asaka),埼玉县(Saitma),日本)解析,并通过分色镜(700DCXXR,AHF分析技术(Analysentechnik)AG,图宾根,德国)(DM)在两个通道中进行频谱解析。光谱范围内的第一通道范围在720至850nm内,通过带有NIRF发射滤光片(ET810/90,色彩技术)(F2)的NIRF消色差双合透镜对(achromatic doublet pair)(MAP10100100-B,索尔实验室(Thorlabs))(RL1)进行过滤,并通过iXon电子倍增电荷耦合器件(EMCCD,DV897DCS-BV,安道尔科技公司,贝尔法斯特,北爱尔兰)记录。光谱范围为450-700nm的第二通道穿过一对可见的消色差双合透镜(MAP10100100-A,索尔实验室)(RL2),该消色差双合透镜通过短通滤光片(ET700SP-2P,色彩技术)(F3)、并来自12位彩色电荷耦合器件(CCD)相机(pixelfly qe,PCOAG,凯尔海姆,德国)过滤。
相机系统[基于EMCCD检测(Luca R,安道尔科技公司(Andor Technology))。该相机具有荧光滤光片(D850/40m,色彩技术)(F4),并使用镜头(变焦7000微距镜头,纳维塔尔(Navitar),纽约,美国)。相机的捕获和控制是通过索利斯(Solis)软件(Solis I,安道尔科技公司)和我们小组开发的基于GPU的C++软件完成的。所有数据处理均在MATLAB(迈斯沃克(MathWorks)公司,马萨诸塞州,美国)中实现。
4.3结果
该实验的结果如图4a所示:用FRX110对NOD/SCID小鼠颈部区域的增殖性侵袭性HNSCC组织生物成像。人HNSCC异种移植(p.TX)后,从第1到第12周执行成像,以对癌增殖进行成像。放大的图片显示了第5周(p.TX)增殖的侵袭性癌组织的生物成像,具有高的组织背景(TBR)比。图4显示了FRX110与侵袭性生长癌异种移植物的特异性结合,能够术中分子生物成像和侵袭性癌组织划分成为可能。
4.4补充实验
在通过FRX110排除非特异性信号后,在HNSCC PDX模型中使用FRX110对整合素αvβ6的分子生物成像(=MBI)进行测试。以下结果按时间顺序显示。在我们的初步研究中,4-6小时后发现未结合的NIRF示踪剂的排泄,没有非特异性积累。为了针对整合素αvβ6的MBI设置具有NIRF信号/背景信号之间的最佳比率的最佳时间,将时间窗延长设定为24h p.i.。表2显示了测试系列的概述。

人癌组织异种移植后,在一周后(时间W1)首先检测HNSCC PDX模型中FRX110的积累和生物分布。
为了进行生物分布分析,在通过尾静脉的αvβ6特异性NIRF示踪剂(FRX110)注射后(p.i.)0、4、6和24h执行分子生物成像(MBI)。
6小时后,示踪剂显示出肾脏清除,并在24小时后排除(请参见图5b和下面的项目5.2下的相应文本)。
在随后的几周中,增殖癌组织中αvβ6表达的MBI在移植后第4、5、8、10和12周(p.Tex)、在24小时注射后(p.i.)。
在两个目标区域(ROI)中进行平均荧光强度/像素的测量。ROI1是异种移植的定位。ROI2是对侧的参考,具有可比较的组织灌注。
每只动物都以4个不同的通道表示:彩色光=临床情况;荧光=黑色/白色荧光信号,融合=彩色光和荧光通道的叠加;在荧光强度标度中,显示了荧光强度/像素的分布。
平均荧光强度/像素的商ROI1/ROI2作为肿瘤组织背景比(TBR)给出。TBR>1对应于背景信号上方的荧光信号,并且相对于ROI2通过FRX110在癌移植区域(ROI1)中的积累。
平均荧光强度/像素在每个时间点以图形方式显示。在测试系列的时间进程中,将TBR以数值形式以及图表形式被制成表。
图4b总结了来自表2中所示测试系列的MBI数据。
该实施例的实验表明,本发明的化合物在表现出高荧光强度的肿瘤细胞中选择性地积累,以产生高的肿瘤背景比。
5、参考实施例–生物分布
5.1比较化合物在生物体内的生物分布
将比较化合物FRX103以10μmol的浓度静脉内注射到HNSCC PDX小鼠中。依靠荧光强度测量,在所示的时间段内测定了FRX103在生物体中的分布和积累。这些实验的结果在注射后(p.i.)长达24小时的时间段内显示在图5a中。
FRX103在癌组织区域显示出恒定的积累。在注射后24h后,癌组织区域中的特异性信号与背景信号之间的比率最佳。但是,FRX103在24小时后未显示出任何排除,但在肝脏和胰腺中出现了不希望的残留积累。
在经受健康上皮移植的对照动物(对照TX)中,未能观察到移植区域中的非特异性积累。但是,与HNSCC PDX组类似,所有动物(n=7)在24小时后在肝脏和胰腺中均显示出持续的信号。
这些观察表明,所测试的FRX103化合物的生物分布和排除是不令人满意的。
5.2本发明化合物在生物体内的生物分布
使用本发明的化合物FRX110重复上述实验。该实验的结果如图5b所示。从该图可以得出,注射后0-24h的时间范围内的生物分布并未显示出非特异性的积累或结合(TBR1.0)。未结合的FRX110在24小时内通过肾脏系统排除。与FRX103相比,已证明FRX110已是更适合生物成像应用的化合物。
5.3本发明化合物与健康细胞的体内结合
用假手术(sham operations)和具有健康对照组织的移植来执行对照实验,用FRX110和FRX109作为对照示踪剂对其成像。
在对照TX模型和对照动物中测试生物分布,以进行进一步研究。
在体内模型中使用对照示踪剂FRX109研究了结合的特异性。用人癌组织的假移植(假TX)(无组织移植)在小鼠对FRX109进行可视化。在未操作的对照小鼠、假TX小鼠和已经移植了人健康上皮的小鼠(对照TX)中对FRX110进行可视化。
表3给出了对照实验的概述,以便能够排除NIRF示踪剂的非特异性结合。

图5c显示了FRX110和FRX109在注射后(p.i.)6h表现出良好的肾脏清除率,没有非特异性示踪剂增强、和低的组织背景(TBR)比。
6、发明性实施例2–本发明化合物在癌细胞的术中检测中的体内用途
在HNSCC PDX小鼠模型中,执行了术中分子生物成像辅助的癌切除术。图6示出了切除的癌的组织学/免疫组织学表现。根据本发明的术中生物成像提供了关于恶性组织的定位、范围和癌生物学特征的信息,该信息在术中实时可用。
在临床前HNSCC PDX小鼠模型中进一步证明,生物成像辅助的癌切除术允许在术中检测侵袭性癌组织并控制切除状态。

(i)提供带有部分A的接头;
(ii)提供含有部分B的荧光染料;
(iii)使所述荧光染料与所述带有部分A的接头反应,使得所述接头和源自所述荧光染料的部分B之间形成共价键;
或者
(i’)提供接头,所述接头带有如权利要求1所限定的源自荧光染料的部分B;
(ii’)提供包含部分A的化合物;
(iii’)使包含部分A的化合物与带有所述部分的接头反应,使得所述接头和所述部分A之间形成共价键。
6.根据权利要求1-4中任一项所述的化合物,其用在治疗癌症的可互操作的手术方法中。
7.根据权利要求6所述的用途的化合物,其中,所述方法包括以下步骤:
(a)向患者系统性的施用根据权利要求1-4中任一项所述的化合物;
(b)用能够通过染料部分诱导荧光发射的光照射具有疑似肿瘤细胞的组织;
(c)检测荧光发射;
(d)手术切除显示荧光发射的组织、或被显示荧光发射的组织包围的组织。
8.根据权利要求6或7所述的用途的化合物,其中所述癌症选自:结肠癌、胃癌、口腔鳞状细胞癌(OSCC)、胰腺导管腺癌、肠腺癌、头颈鳞状细胞癌(HNSCC)、侵袭性子宫内膜癌、基底细胞癌、乳腺癌、子宫内膜癌、胃癌、肝癌、非小细胞肺癌、肺癌脑转移、卵巢癌、胰腺癌和前列腺癌。
