本发明公开了一种提高断奶仔猪非特异性免疫功能的饲料添加剂、其制备方法和应用,该饲料添加剂为溶菌酶二聚体。该饲料添加剂在断奶仔猪饲料中添加量是0.09~1.00Kg/T。本发明使用溶菌酶二聚体饲料添加剂,完全可以替代饲料中的抗生素,通过完善及修复断奶仔猪非特异性免疫功能,改善绒毛与隐窝比值、维持消化道菌群平衡,从而有效降低断奶应激,避免断奶仔猪疾病发生,促进仔猪的生长性能充分发挥。本实施例方法简单,操作简便,条件温和;本实施例方法简单易行,成本低,适合推广使用。

背景技术
每当在设计猪饲料配方时,配方师一般都会考虑在配合饲料中添加饲料添加剂。常用的饲料添加剂分为营养性饲料添加剂,维生素类添加剂:维生素A、E、K、D、B等;微量元素类添加剂:铜、铁、锌、锰等;氨基酸类添加剂:赖氨酸、蛋氨酸等;为促进猪的对饲料转换功能的制剂:酶制剂、活菌剂、酸化剂等;为提高猪的抗病能力和猪的生产性能,在猪用配合饲料中添加药物添加如抗生素添加剂:土霉素、金霉素、喹乙醇等,激素类添加剂:生长激素、肾上腺皮质激等。
近年来,世界多国农业农村部明确规定药物饲料添加剂将全部退出饲料生产领域,并确定了养殖端减抗和限抗的时间表,其中中国国家农业农村部明确规定药物饲料添加剂将在 2020年全部退出。人们认识到长期低剂量或滥用抗生素必然会产生超级细菌而危害人类的生存。而高度集约化养殖面临着动物疾病增加和应激因子不断发生,造成养殖业很大损失。由此,迫使人们寻找替代抗生素物品。以保障人类健康、动物安全、稳定的生产。因此,如何开发一种药物饲料添加剂的新产品,成为亟待了解决的技术问题。

为了解决现有技术问题,本发明的目的在于克服已有技术存在的不足,提供一种提高断奶仔猪非特异性免疫功能的饲料添加剂、其制备方法和应用,本发明使用溶菌酶二聚体饲料添加剂,完全可以替代饲料中的抗生素,通过完善及修复断奶仔猪非特异性免疫功能,改善绒毛与隐窝比值、维持消化道菌群平衡,从而有效降低断奶应激,避免断奶仔猪疾病发生,促进仔猪的生长性能充分发挥。

一种提高断奶仔猪非特异性免疫功能的饲料添加剂,将蛋清溶菌酶通过生物交联方法,生成稳定的溶菌酶多聚体,作为断奶仔猪的饲料添加剂。
作为本发明优选的技术方案,所述溶菌酶多聚体为溶菌酶二聚体、溶菌酶三聚体、溶菌酶四聚体、溶菌酶五聚体中的至少一种溶菌酶多聚物。溶菌酶二聚体是非特异性免疫因子之一,它具有抗菌消炎、抗病毒作、提高动物非特异性免疫能力等作用。用于解决断奶仔猪因断奶应激而造成的抗病力低下,腹泻频发,死亡率增加。通过修复完善非特异性免疫功能,阻止病原菌入侵,促进免疫细胞吞噬活性,清除入侵的病原菌。进而确保断奶仔猪健康生长。
作为本发明优选的技术方案,当所述溶菌酶多聚体主要包括溶菌酶二聚体、溶菌酶三聚体、溶菌酶四聚体、溶菌酶五聚体时,以溶菌酶多聚体总量为100%,并按照质量百分比计算,其中,溶菌酶二聚体含量≥35wt.%,包括溶菌酶三聚体、溶菌酶四聚体、溶菌酶五聚体的溶菌酶多聚体总含量≥15wt.%,溶菌酶单体≤50wt.%。
作为本发明优选的技术方案,将蛋清溶菌酶溶解于无金属离子液体中,投入溶菌酶交联剂,混合形成反应物体系溶液;控制温度65℃,反应物体系溶液酸碱度pH为7.2,进行反应,在180分钟后,生成溶菌酶多聚体产物;随后再向产物混合液中投入溶菌酶交联稳定剂,在 62℃低温负压烘干,喷雾制粒,形成微囊颗粒。
作为本发明优选的技术方案,提高断奶仔猪非特异性免疫功能的饲料添加剂,其分子量为28800,包含258个氨基酸,并携带大量正电荷,碱性蛋白;酶活≥500u/mg。
作为本发明优选的技术方案,提高断奶仔猪非特异性免疫功能的饲料添加剂为微黄或黄色粉末,其中含有水分≤10wt.%。
一种本发明提高断奶仔猪非特异性免疫功能的饲料添加剂的制备方法,步骤如下:
⑴将计量好的溶菌酶二聚体投入混合机中;
⑵再加入计量好的脱脂米糠粉投入混合机中;
⑶将上述步骤⑴和⑵中加入的原料进行混合,混合时间至少7s;
⑷将在所述步骤⑶中混合好的成品进行计量打包,得到溶菌酶二聚体材料,其中溶菌酶二聚体≥3wt.%,酶活性≥50u/mg。
一种本发明提高断奶仔猪非特异性免疫功能的饲料添加剂的应用,所述饲料添加剂在断奶仔猪饲料中添加使用量为0.09~1.00Kg/T。
本发明与现有技术相比较,具有如下显而易见的突出实质性特点和显著优点:
1.本发明使用溶菌酶二聚体饲料添加剂,完全可以替代饲料中的抗生素,通过完善及修复断奶仔猪非特异性免疫功能,改善绒毛与隐窝比值、维持消化道菌群平衡,从而有效降低断奶应激,避免断奶仔猪疾病发生,促进仔猪的生长性能充分发挥;
2.本发明方法简单,操作简便,条件温和,成本低,适合推广使用。
具体实施方式
以下结合具体的实施例子对上述方案做进一步说明,本发明的优选实施例详述如下:
实施例一:
在本实施例中,一种提高断奶仔猪非特异性免疫功能的饲料添加剂,将蛋清溶菌酶通过生物交联方法,生成稳定的溶菌酶二聚体,作为断奶仔猪的饲料添加剂。溶菌酶二聚体是非特异性免疫因子之一,它具有抗菌消炎、抗病毒作、提高动物非特异性免疫能力等作用。用于解决断奶仔猪因断奶应激而造成的抗病力低下,腹泻频发,死亡率增加。通过修复完善非特异性免疫功能,阻止病原菌入侵,促进免疫细胞吞噬活性,清除入侵的病原菌。进而确保断奶仔猪健康生长。
在本实施例中,将蛋清溶菌酶溶解于无金属离子液体中,投入溶菌酶交联剂,混合形成反应物体系溶液;控制温度65℃,反应物体系溶液酸碱度pH为7.2,进行反应,在180分钟后,生成溶菌酶多聚体产物;随后再向产物混合液中投入溶菌酶交联稳定剂,在62℃低温负压烘干,喷雾制粒,形成微囊颗粒,其分子量为28800,包含258个氨基酸,并携带大量正电荷,碱性蛋白;酶活≥500u/mg。微囊颗粒为微黄或黄色粉末颗粒,溶菌酶活性检测SOP,溶菌酶二聚体SOP,其中含有水分≤10wt.%。卫生指标达标。
实验测试分析:
为了验证本实施例饲料添加剂对断奶仔猪非特异性免疫功能实际使用效果,选择了川农雅安实验猪场进行了饲喂试验。共选用252头仔猪进行试验。
对照组饲喂添加抗生素添加剂20mg/kg硫酸抗敌素、50mg/kg吉它霉素的饲料。试验组饲喂按添加本发明的溶菌酶二聚体饲料添加剂的饲料。空白组饲喂无添加抗生素添加剂也没有添加溶菌酶二聚体添加剂的饲料。本实施例的饲料添加剂实施如下:
一、28日龄-56日龄的仔猪阶段对腹泻率的影响参加表1。

在表1中,a,b同行数据标注不同字母的表示差异显著,p<0.05,相同或无字母表示差异不显著,p>0.05:结果表示为平局值。由表1可知,本实施例添加溶菌酶二聚体90mg/kg, 可有效控制腹泻,可替代抗生素。
二、28日龄-56日龄的仔猪阶段对中性粒细胞吞噬活性的影响参见表2。

试验结果表明,在表2中,与对照组、空白组相比,60mg/kg、90mg/kg溶菌酶二聚体组仔猪腹腔巨噬细胞吞噬细胞吞噬率和吞噬指数均显著提高,p<0.01,从而提高断奶仔猪的非特异性免疫能力。由表2可知,本实施例添加溶菌酶二聚体能有效提高断奶仔猪的非特异性免疫能力。
三、28日龄-56日龄的仔猪阶段对断奶仔猪生产性能的影响参见表3。

试验结果表明,由表3可知,本实施例添加溶菌酶二聚体,断奶仔猪生产性能优于空白组,与抗生素组持平。
本实施例提高断奶仔猪非特异性免疫功能的饲料添加剂及其使用方法。该饲料添加剂为溶菌酶二聚体。该饲料添加剂在断奶仔猪饲料中添加量是0.03~1.00Kg/T。本实施例使用溶菌酶二聚体饲料添加剂,完全能替代饲料中的抗生素,通过完善及修复断奶仔猪非特异性免疫功能,改善绒毛与隐窝比值、维持消化道菌群平衡,从而有效降低断奶应激,避免断奶仔猪疾病发生,促进仔猪的生长性能充分发挥。
实施例二:
本实施例与实施例一基本相同,特别之处在于:
在本实施例中,一种提高断奶仔猪非特异性免疫功能的饲料添加剂的制备方法,步骤如下:
⑴将计量好的溶菌酶二聚体投入混合机中;
⑵再加入计量好的脱脂米糠粉投入混合机中;
⑶将上述步骤⑴和⑵中加入的原料进行混合,混合7s;
⑷将在所述步骤⑶中混合好的成品进行计量打包,得到溶菌酶二聚体材料,其中溶菌酶二聚体≥3wt.%,酶活性≥50u/mg。
本实施例方法简单,操作简便,条件温和,成本低,适合推广使用。
上面对本发明实施例进行了说明,但本发明不限于上述实施例,还可以根据本发明的发明创造的目的做出多种变化,凡依据本发明技术方案的精神实质和原理下做的改变、修饰、替代、组合或简化,均应为等效的置换方式,只要符合本发明的发明目的,只要不背离本发明提高断奶仔猪非特异性免疫功能的饲料添加剂、其制备方法和应用的技术原理和发明构思,都属于本发明的保护范围。
Claims (8)
1.一种提高断奶仔猪非特异性免疫功能的饲料添加剂,其特征在于:将蛋清溶菌酶通过生物交联方法,生成稳定的溶菌酶多聚体,作为断奶仔猪的饲料添加剂。
2.根据权利要求1所述提高断奶仔猪非特异性免疫功能的饲料添加剂,其特征在于:所述溶菌酶多聚体为溶菌酶二聚体、溶菌酶三聚体、溶菌酶四聚体、溶菌酶五聚体中的至少一种溶菌酶多聚物。
3.根据权利要求2所述提高断奶仔猪非特异性免疫功能的饲料添加剂,其特征在于:当所述溶菌酶多聚体主要包括溶菌酶二聚体、溶菌酶三聚体、溶菌酶四聚体、溶菌酶五聚体时,以溶菌酶多聚体总量为100%,并按照质量百分比计算,其中,溶菌酶二聚体含量≥35wt.%,包括溶菌酶三聚体、溶菌酶四聚体、溶菌酶五聚体的溶菌酶多聚体总含量≥15wt.%,溶菌酶单体≤50wt.%。
4.根据权利要求1所述提高断奶仔猪非特异性免疫功能的饲料添加剂,其特征在于:将蛋清溶菌酶溶解于无金属离子液体中,投入溶菌酶交联剂,混合形成反应物体系溶液;控制温度65℃,反应物体系溶液酸碱度pH为7.2,进行反应,在180分钟后,生成溶菌酶多聚体产物;随后再向产物混合液中投入溶菌酶交联稳定剂,在62℃低温负压烘干,喷雾制粒,形成微囊颗粒。

⑴将计量好的溶菌酶二聚体投入混合机中;
⑵再加入计量好的脱脂米糠粉投入混合机中;
⑶将上述步骤⑴和⑵中加入的原料进行混合,混合时间至少7s;
⑷将在所述步骤⑶中混合好的成品进行计量打包,得到溶菌酶二聚体材料,其中溶菌酶二聚体≥3wt.%,酶活性≥50u/mg。
8.一种权利要求1所述提高断奶仔猪非特异性免疫功能的饲料添加剂的应用,其特征在于:所述饲料添加剂在断奶仔猪饲料中添加使用量为0.09~1.00Kg/T。
