本发明涉及一种piggyBac转座子系统介导的真核生物转基因细胞系及构建方法,先将过表达元件整合到piggyBac转座子的末端反向重复序列(inverted terminal repeat,ITR)之间,构建转基因过表达载体PB‑Vector,再将转基因过表达载体PB‑Vector和piggyBac transposase表达载体A3‑helper按照摩尔比1:1混合后转染真核生物,筛选获得真核生物转基因细胞系。

技术领域
本发明属于基因工程技术领域,涉及piggyBac转座子系统介导的真核生物转基因细胞系及构建方法。
背景技术
随着越来越多的生物的全基因组序列测序完成,功能基因组的研究成为生命科学的重要方向,与个体水平相比,细胞因为易于培养,成本较低,周期较短等优点而日益成为了功能基因组研究的重要对象。遗传操作技术是研究功能基因组的重要技术手段,转基因细胞系的构建对于研究基因功能和生物医药研究非常重要。
目前应用最广泛的构建转基因细胞系的方法是基于慢病毒的转基因技术,已经取得了巨大的成就。但是,慢病毒系统主要是作用于哺乳动物细胞,在其他动物细胞上(如昆虫)转基因效率很低;而且,慢病毒系统的承载能力有限,只有几kb,而且随着承载DNA片段的增长,转基因效率急剧下降。
因此,开发一种光谱高效的转基因细胞系构建方法非常不要。piggyBac转座子系统是来源于鳞翅目昆虫(如粉纹夜蛾)的一种Ⅱ型转座子,全长2476bp,包括两个末端反向重复序列(inverted terminal repeat,ITR)和一个编码转座酶的阅读框。piggyBac转座子采用“剪切-黏贴”模式发生转座,转座效率高。
目前,piggyBac转座子系统已经被证明可以在从昆虫到哺乳动物的许多物种中高效转座。piggyBac转座子系统具有强大的承载能力,已有报道显示最大可承载超过200kb的核苷酸,承载能力远远超过慢病毒系统。因此,开发piggyBac转座子系统介导的真核生物转基因细胞系的构建方法具有非常广阔的应用前景。
发明内容
有鉴于此,本发明的目的在于提供一种piggyBac转座子系统介导的真核生物转基因细胞系及构建方法。
为达到上述目的,本发明提供如下技术方案:
1、piggyBac转座子系统介导的真核生物转基因细胞系的构建方法,具体步骤如下:
(1)合成piggyBac转座子系统基础载体,命名为piggyBacModify,其核苷酸序列如SEQ ID NO.1所示;
(2)合成IE2启动子启动的筛选标记表达框,命名为PUC57-IE2-GST-Zeocin-ISCEIsiet-EGFP-Ser1PA,核苷酸序列如SEQ ID NO.2所示;然后将载体PUC57-IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA的表达框IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA连接到步骤(1)的piggyBacModify,构建成功piggyBac细胞转基因基础载体,命名为pB-Modified{IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA},其核苷酸序列如SEQ ID NO.3所示;
(3)将外源基因表达框连接到基础载体pB-Modified{IE2-GST-Zeocin-ISCEIsiet-EGFP-Ser1PA},即可构建成转基因过表达载体PB-Vector;
(4)将PB-Vector和核苷酸序列如SEQ ID NO.4所示的piggyBac转座酶表达载体A3-helper混合转染真核生物细胞,筛选,完成所述细胞系的构建。
作为优选的技术方案之一,步骤(1)中,piggyBacModify包括:
两个转座臂(包含piggyBac转座子的末端反向重复序列,inverted terminalrepeat,ITR),其核苷酸序列如SEQ ID NO.5和SEQ ID NO.6所示。
作为优选的技术方案之一,步骤(2)中,所述筛选标记表达框包括GST标签-Zeocin抗性基因表达蛋白-EGFP(绿色荧光蛋白),其核苷酸序列如SEQ ID NO.7所示。
作为优选的技术方案之一,步骤(2)中,载体PUC57-IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA中的表达框IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA用酶切连接的方式连接到piggyBacModify的AgeI/AscI位点。
作为优选的技术方案之一,步骤(3)中,利用酶切连接或者无缝克隆的方法将外源基因表达框连接至基础载体的AscI位点。
作为优选的技术方案之一,步骤(3)中,所述外源基因表达框的核苷酸序列具有以下规律:NNNNNNNNNN-NNNNNNNNNN。
作为优选的技术方案之一,步骤(4)中,PB-Vector和piggyBac转座酶表达载体A3-helper(核苷酸序列如SEQ ID NO.4)按照摩尔比1:1混合。
作为优选的技术方案之一,步骤(4)中,转染方法包括但不限于脂质体转染法、电穿孔转染法。
作为优选的技术方案之一,步骤(4)中,细胞完成转染后,用含有Zeocin的完全培养基筛选2个月,直至所有细胞都包含piggyBac转座子系统,即完成所述细胞系的构建。
2、利用上述方法构建的piggyBac转座子系统介导的真核生物转基因细胞系。
本发明的有益效果在于:
本发明提供了一种基于piggyBac转座子系统的真核生物转基因细胞系的构建方法,先将过表达元件整合到piggyBac转座子的末端反向重复序列(inverted terminalrepeat,ITR)之间,构建转基因过表达载体PB-Vector,再将转基因过表达载体PB-Vector和piggyBac transposase表达载体A3-helper(核苷酸序列如SEQ ID NO.4)按照摩尔比1:1混合后转染真核生物,筛选获得真核生物转基因细胞系。
本发明最大的优点在于:
1、piggyBac转座子系统在很多种生物中具有高效转座活性;
2、piggyBac转座子系统具有非常大的承载容量。
3、由于piggyBac转座子是以“剪切-黏贴”的方式来实现来实现转座,因此可以通过控制转座子浓度等方式来控制piggyBac转座子系统携带的外源基因整合到宿主细胞的拷贝数。
因此,piggyBac转座子系统可以在很多种物种实现高效转基因细胞系的构建,尤其是在构建全基因组敲除细胞库时,可以方便的控制每个细胞只整合一个外源sgRNA。

具体实施方式
下面将结合附图,对本发明的优选实施例进行详细的描述。
以下凡是未注明的具体实验方法,都按照公认的实验方法与条件实施,例如,按照试剂耗材厂商提供的说明书操作,或者按照经典实验书籍《分子克隆实验指南》(第三版,J.萨姆布鲁克等著))来完成实验。
实施例:
本实施例中所用到的家蚕胚胎细胞系(The Bombyx mori embryonic cell lineBmE)为生物实验中常用细胞系(PMID:17570024)。
构建piggyBac转座子介导的过表达绿色荧光蛋白(EGFP)的真核生物细胞系
1、设计并合成piggyBac转座子系统基础载体piggyBacModify。其核苷酸序列如SEQ ID NO.1所示,载体图谱如图1所示。
2、设计并合成IE2启动子启动的筛选标记表达框,包括GST标签-Zeocin抗性基因表达蛋白-EGFP(绿色荧光蛋白),命名为PUC57-IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA,核苷酸序列如SEQ ID NO.2所示。载体图谱如图2所示。
3、将载体piggyBacModify用AgeI和AscI双酶切,琼脂糖凝胶回收大小为4913bp的核酸作为骨架;将载体PUC57-IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA用AgeI和AscI双酶切,琼脂糖凝胶回收大小约为2900bp的核酸作为片段;将片段(IE2-GST-Zeocin-ISCEIsiet-EGFP-Ser1PA)和骨架(载体piggyBacModify)按照摩尔比1:1连接,转化后挑选正确的单克隆,成功构建pB-Modified{IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA}(载体图谱见图3)。
1)酶切条件为50μL体系,包含1μg的载体【piggyBacModify或PUC57-IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA】,5μL的CutSmart缓冲液,AgeI和AscI各1μL,用双蒸水补齐50μL,酶切条件为37℃过夜。
2)DNA连接酶为T4 DNA连接酶,连接总体系为50μL,其中骨架和片段按照摩尔比1:10添加,总质量为2μg,T4 DNA连接酶缓冲液5μL,T4 DNA连接酶1μL,用双蒸水补齐50μL,16℃连接4小时,T4 DNA连接酶采购自NEB公司。
3)转化所用的感受态为Trans1-T1感受态细胞,购买自全式金公司,按照公司说明书来执行转化实验。
4)正确单克隆的挑选。首先是挑选约24个单克隆,用含有氨苄青霉素的LB液体培养基在37℃、220rmp摇10小时,然后用菌液PCR的方式挑选滞后克隆,氨苄青霉素工作浓度为50μg/ml。然后将滞后克隆抽提质粒,酶切验证;最后将酶切验证正确的单克隆执行sanger测序,最终挑选到正确的单克隆。菌液PCR所用引物如下:
>pBacR-R,5’-GTACTGTCATCTGATGTACCAGG-3’,如SEQ ID NO.8所示;
>IE2-R,5’-TTCGAACAGATGCTGTTCAACT-3’,如SEQ ID NO.9所示;
PCR酶为普通rTaq酶,PCR反应条件为:94℃预变性4min;94℃变性30s,55℃退火30s,72℃延伸90s;35个循环;72℃延伸10min;12℃保存;阳性克隆的菌液PCR产物大小为1086bp。
4、构建SpCas9蛋白表达框的T载体PUC57-Hr3-Hsp70-Cas9-SV40(核苷酸序列如SEQ ID NO.10所示),该载体由pUC57-hA4-Cas9(核苷酸序列如SEQ ID NO.11所示,PMID:24671069)将启动子A4更换为Hsp70得到。
5、将载体pB-Modified{IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA}用AscI酶切,琼脂糖凝胶电泳回收大小为7899bp的核酸作为骨架,去磷酸化后备用;将载体PUC57-Hr3-Hsp70-Cas9-SV40的Hr3-Hsp70-Cas9-SV40片段用无缝克隆的方法连接到pB-Modified{IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA}上,构建成PB-Vector-SpCas9,核苷酸序列如SEQ ID NO.12。
1)酶切条件为50μL体系,包含1μg的载体pB-Modified{IE2-GST-Zeocin-ISCEIsiet-EGFP-Ser1PA},5μL的CutSmart缓冲液,AscI酶1μL,用双蒸水补齐50μL,酶切条件为37℃过夜。
2)无缝克隆试剂盒采购自NEB公司,按照说明书操作。
3)转化所用的感受态为Trans1-T1感受态细胞,购买自全式金公司,按照公司说明书来执行转化实验。
4)正确单克隆的挑选。首先是挑选约24个单克隆,用含有氨苄青霉素的LB液体培养基在37℃、220rmp摇10小时,然后用菌液电泳的方式挑选滞后克隆,氨苄青霉素工作浓度为50μg/ml。然后将滞后克隆抽提质粒,酶切验证;最后将酶切验证正确的单克隆执行sanger测序,最终挑选到正确的单克隆。
6、将载体PB-Vector-SpCas9和piggyBac transposase表达载体A3-helper(核苷酸序列如SEQ ID NO.4)按照摩尔比1:1共转染家蚕胚胎细胞系BmE,转染方式包括但不限于脂质体转染法、电穿孔转染法等;6-8小时后更换完全培养基,培养大约1个月时间,直至细胞状态恢复正常。然后更换含有Zeocin的完全培养基培养2个月,直至全部细胞都稳定整合绿色荧光蛋白(EGFP)表达框(表明全部细胞稳定整合了SpCas9基因)。完全培养基为包括体积浓度10%胎牛血清(fetal bovine serum,FBS,赛默飞世尔公司)和青霉素-链霉素(Penicillin-Streptomycin,20万单位/升,赛默飞世尔公司)的Grace’s培养基(赛默飞世尔公司),Zeocin的工作浓度为200μg/ml。用流式细胞仪或者梯度稀释法分选单克隆细胞,扩大培养,可以得到过表达SpCas9的真核生物细胞系。
最后说明的是,以上优选实施例仅用以说明本发明的技术方案而非限制,尽管通过上述优选实施例已经对本发明进行了详细的描述,但本领域技术人员应当理解,可以在形式上和细节上对其作出各种各样的改变,而不偏离本发明权利要求书所限定的范围。

Claims (10)

1.piggyBac转座子系统介导的真核生物转基因细胞系的构建方法,其特征在于,具体步骤如下:
(1)合成piggyBac转座子系统基础载体,命名为piggyBacModify,其核苷酸序列如SEQID NO.1所示;
(2)合成IE2启动子启动的筛选标记表达框,命名为PUC57-IE2-GST-Zeocin-ISCEIsiet-EGFP-Ser1PA,核苷酸序列如SEQ ID NO.2所示;然后将载体PUC57-IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA的表达框IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA连接到步骤(1)的piggyBacModify,构建成功piggyBac细胞转基因基础载体,命名为pB-Modified{IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA},其核苷酸序列如SEQ ID NO.3所示;
(3)将外源基因表达框连接到基础载体pB-Modified{IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA},即可构建成转基因过表达载体PB-Vector;
(4)将PB-Vector和核苷酸序列如SEQ ID NO.4所示的piggyBac转座酶表达载体A3-helper混合转染真核生物细胞,筛选,完成所述细胞系的构建。
2.根据权利要求1所述的构建方法,其特征在于,步骤(1)中,piggyBacModify包括:两个转座臂,其核苷酸序列如SEQ ID NO.5和SEQ ID NO.6所示。
3.根据权利要求1所述的构建方法,其特征在于,步骤(2)中,所述筛选标记表达框包括GST标签-Zeocin抗性基因表达蛋白-EGFP,其核苷酸序列如SEQ ID NO.7所示。
4.根据权利要求1所述的构建方法,其特征在于,步骤(2)中,载体PUC57-IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA中的表达框IE2-GST-Zeocin-ISCEI siet-EGFP-Ser1PA用酶切连接的方式连接到piggyBacModify的AgeI/AscI位点。
5.根据权利要求1所述的构建方法,其特征在于,步骤(3)中,利用酶切连接或者无缝克隆的方法将外源基因表达框连接至基础载体的AscI位点。
6.根据权利要求1所述的构建方法,其特征在于,步骤(3)中,所述外源基因表达框的核苷酸序列具有以下规律:NNNNNNNNNN-NNNNNNNNNN。
7.根据权利要求1所述的构建方法,其特征在于,步骤(4)中,PB-Vector和piggyBac转座酶表达载体A3-helper(核苷酸序列如SEQ ID NO.4)按照摩尔比1:1混合。
8.根据权利要求1所述的构建方法,其特征在于,步骤(4)中,转染方法包括但不限于脂质体转染法、电穿孔转染法。
9.根据权利要求1所述的构建方法,其特征在于,步骤(4)中,细胞完成转染后,用含有Zeocin的完全培养基筛选2个月,直至所有细胞都包含piggyBac转座子系统,即完成所述细胞系的构建。
10.利用权利要求1~9中任一项所述方法构建的piggyBac转座子系统介导的真核生物转基因细胞系。
