As can be seen from figure 3, the industrial hemp extract is helpful for improving the expression level of AQP-3, Caspase-14 and F L G proteins related to the skin barrier, recovering the damage of external stimuli (such as ultraviolet light, air pollution and the like) to the skin barrier, enhancing the water locking, moisturizing and other performances of the skin barrier, repairing the damaged skin stratum corneum and having good recovery effect on sensitive skin.
(6) Influence of industrial hemp extract on TRPV family gene expression level
According to the existing report, TRPV4 protein forms a pentagonal structure together with β -catenin and cadherin in epidermal keratinocytes of mice, and the interaction plays an important role in connection of the keratinocytes and further plays an important role in the health condition of skin barriers.
HaCaT cells were first cultured in medium containing 0.04mM calcium (low calcium medium) withThen transferred to a medium added with 1.5mM CaCl2In the medium (high calcium medium), HaCaT cells are induced to differentiate by a high calcium switch. As can be seen from Table 3, the industrial cannabis extracts have inhibitory effects on the expression of TPRV4 by HaCaT cells stimulated by the external temperature environment, and are presumed to have a repairing effect on sensitive skin.
Test example 3 clinical efficacy verification of composition containing industrial hemp extract
The series of facial masks, sprays and lotions prepared in examples 4, 5 and 6 were administered to 30 sensitive skin patients (diagnosed and confirmed by clinical dermatologists) and self-evaluated for dimensions such as stinging, erythema, dry skin, desquamation, pain, tightness, pruritus and perm, with a score of 0-5 (0 points representing no stinging, no erythema, low dryness of skin, no desquamation, no pain, no tightness, no pruritus symptom and no perm, and gradually increasing in degree from 0 to 5 points). The sensitive skin patients self-score according to the scoring table before using the facial mask product, then use the facial mask product, respectively score 15min, 1 day, 3 days, 5 days, 15 days and 30 days after using the facial mask,
typical effects are shown in fig. 4, and according to results, the facial prickling, erythema, dry skin, desquamation, pain, tightness, pruritus and perm of sensitive skin patients are obviously improved after the industrial hemp extract facial mask is used.
In conclusion, the external composition disclosed by the invention can inhibit hyaluronidase, remove free radicals, improve the expression level of skin barrier-related proteins (AQP-3, F L G and Caspase-14), has a remarkable effect on treating skin sensitivity, is not limited by conventional sensitive skin treatment instruments and professional dermatologists, is convenient to use and high in safety, and has a wide application prospect.

Claims (12)

1. A composition for external use containing an extract of industrial hemp, characterized in that: the preparation is prepared by taking industrial hemp extract as an active ingredient and adding auxiliary materials acceptable in cosmetics; the industrial hemp extract is prepared from the following raw materials in a mass ratio of 5-7: 2-4: 1 industrial hemp leaf, industrial hemp flower and industrial hemp stalk, and extracting with ethanol solution.
2. The composition for external use according to claim 1, wherein: the industrial hemp extract is prepared from the following raw materials in a mass ratio of 6: 3: 1 industrial hemp leaf, industrial hemp flower and industrial hemp stalk, and extracting with ethanol solution.
3. The composition for external use according to claim 1, wherein: the concentration of the ethanol solution is 50-100%, v/v.
4. The composition for external use according to any one of claims 1 to 3, characterized in that: the preparation is water agent, emulsion, spray, cream, ointment, pellicle or gel.
5. The topical composition according to claim 4, characterized in that: the preparation is prepared from the following raw materials in percentage by weight:
3-10% of industrial hemp extract, 2-4% of glycerol, 5-7% of butanediol, 0.05-0.15% of preservative and the balance of deionized water;
or:
3-10% of industrial hemp extract, 13-16% of polyvinyl alcohol PVA, 2.8-4.0% of glycerol, 200.1-0.3% of Tween, 0.04-0.06% of EDTA-2Na, 0.5-1.5% of hyaluronic acid, 0.05-0.15% of preservative and the balance of deionized water;
or:
3-10% of industrial hemp extract, 3.0-4.0% of glycerol, 1-3% of 1, 2-pentanediol, 0.4-0.6% of glycerol stearate, 0.6-0.8% of alkyl glycoside SS, 200.6-0.8% of alkyl glycoside SSE, 1-3% of squalane, 0.5-1.5% of β -glucan, 0.4-0.6% of hyaluronic acid, 0.05-0.15% of preservative and the balance of deionized water.
6. The composition for external use according to claim 5, wherein: the preparation is prepared from the following raw materials in percentage by weight:
5% of industrial hemp extract, 3% of glycerol, 6% of butanediol, 0.1% of preservative and the balance of deionized water;
or:
5% of industrial hemp extract, 15% of polyvinyl alcohol PVA, 3.5% of glycerol, 200.2% of Tween, 0.05% of EDTA-2Na, 1% of hyaluronic acid, 0.1% of preservative and the balance of deionized water;
or:
5% of industrial hemp extract, 3.5% of glycerol, 2% of 1, 2-pentanediol, 0.5% of glycerol stearate, 0.7% of alkyl glycoside SS, 200.7% of alkyl glycoside SSE, 2% of squalane, β -glucan 1%, 0.5% of hyaluronic acid, 0.1% of preservative and the balance of deionized water.
7. A process for preparing the composition for external use according to claim 5 or 6, characterized in that; it comprises the following steps:
1) weighing raw and auxiliary materials according to the proportion;
2) mixing glycerol, butanediol and deionized water uniformly to obtain phase A; industrial hemp extract as phase B; preservative as phase C;
3) mixing the phase A and the phase B, adding the phase C, and mixing;
or:
1) weighing raw and auxiliary materials according to the proportion;
2) uniformly mixing polyvinyl alcohol (PVA), glycerol, tween 20, EDTA-2Na and deionized water to obtain a phase A; industrial hemp extract and hyaluronic acid as phase B; preservative as phase C;
3) mixing phase A and phase B, adding phase C, mixing, and soaking in facial mask paper for 5-10 min;
or:
1) weighing raw and auxiliary materials according to the proportion;
2) heating and uniformly mixing 1, 2-pentanediol, glycerol and deionized water to obtain phase A, heating and uniformly mixing glycerol stearate, alkyl glycoside SS, alkyl glycoside SSE-20 and squalane to obtain phase B, uniformly mixing industrial hemp extract, β -glucan and hyaluronic acid to obtain phase C, and using preservative as phase D;
3) heating and stirring phase A and phase B at 50-60 deg.C to obtain emulsion, cooling to 35-40 deg.C, sequentially adding phase C and phase D, and mixing.
8. A method of preparing an extract of industrial cannabis sativa according to any of claims 1 to 4, characterized in that it comprises the following steps:
weighing leaves, stems and flowers of cannabis sativa according to a ratio, adding 10 times (v/w, ml/g) of 50-100% ethanol aqueous solution, performing ultrasonic extraction for 1.5-2.0h for 2 times in total, combining the extracting solutions, and performing vacuum concentration at 60 ℃ until the relative density is 1.03 to obtain the cannabis sativa extract.
9. Use of a topical composition according to any one of claims 1 to 6 in the manufacture of a medicament for the treatment of skin sensitivity.
10. Use according to claim 9, characterized in that the skin sensitivity is skin prick, erythema, dryness, desquamation, pain, tightness, itching and/or perm.
11. The use according to claim 9, wherein the medicament is a medicament for inhibiting hyaluronidase, scavenging free radicals and/or increasing the amount of expression of skin barrier-related proteins.
12. Use according to claim 11, wherein said skin barrier associated protein is AQP-3, F L G and/or Caspase-14 protein.
