Technical Field
The present application relates to a cosmetic composition having collagen synthesis effect comprising a single lactic acid culture, and more particularly, to a cosmetic composition for promoting collagen synthesis and improving skin wrinkles comprising a lactobacillus plantarum CJ L P133 culture.
Background
The human skin protects the human body from various oxidants, air pollution, heavy metals, ultraviolet radiation (UV), etc., prevents bacteria, fungi, viruses, etc. from invading the skin, and has the function of preventing water loss as a protective barrier. The structure of the skin is roughly divided into three layers of epidermis, dermis and subcutaneous fat, and the dermis layer, which accounts for 90% of the skin, is composed of collagen, elastin, hyaluronic acid and glycoprotein. Collagen is the major component of skin, an important fibrin protein, which accounts for about 25% of the total weight of human protein. It constitutes most connective tissues and has important roles such as intercellular matrix function, structural support function, induction of cell division and differentiation, provision of skin tensile strength, and the like. It is known that aging, ultraviolet rays, harmful oxygen, etc. cause a decrease in collagen production and destroy the structure thereof, and thus it is closely related to the formation of skin wrinkles. In recent years, deteriorating environmental factors (e.g., fine dust, etc.) cause the breakdown of skin barrier, and thus skin aging is continuously promoted by the decrease in collagen synthesis and the increase in decomposition.
Therefore, it is very important in cosmetics to maximize the effects of each ingredient and minimize the side effects thereof to prevent skin aging. In this regard, there is a need to develop a cost-effective material that promotes collagen synthesis without side effects and an easy-to-use cosmetic comprising the same.
Lactic acid bacteria are bacteria that have a beneficial effect in the human body, producing lactic acid through the fermentation of carbohydrates or fibers. To date, about 400 species of lactic acid bacteria have been found, and about 18 species have been developed as commercial products.
Once lactic acid bacteria enter the human intestinal tract, they secrete specific substances through their own metabolic activities. Since these substances have beneficial effects on the human body, many studies have been made on lactic acid bacteria. The effects of lactic acid bacteria on antibacterial, antioxidant and immunological activities have been reported. In recent years, research has been focused on whitening and moisturizing effects of lactic acid bacteria, and the possibility as a cosmetic material has been increasing. Among lactic acid bacteria, many studies have been made on the action of lactobacillus, and enhancement of immunity, antibacterial action, skin whitening, improvement of atopic dermatitis, and the like have been reported.
Among them, the plant-derived lactic acid bacteria (PO L AB) are lactic acid bacteria isolated from fermented foods of plant materials (e.g., pickles, kimchi, etc.), and are different from dairy-derived lactic acid bacteria.
The lactic acid bacteria of plant origin not only include species at least 10 times more than animal lactic acid bacteria, but also have excellent adaptability to the external environment. In particular, the plant lactic acid bacteria isolated from kimchi having high salt content, low pH and high content of natural antibacterial substances can grow in extreme environments, and thus the plant-derived lactic acid bacteria have excellent productivity of various physiologically active substances.
Representative physiologically active substances may include antibacterial peptides, immunostimulants, gamma-aminobutyric acid (GABA), which exhibit various physiological functions including anti-inflammatory and immunosuppressive effects, and the like. In addition, lactic acid bacteria are heat stable peptides and produce bacteriocins as antimicrobial agents. Bacteriocins are natural antimicrobial proteins or proteinaceous substances produced by a variety of microorganisms, generally referring to substances having bactericidal mechanisms against bacterial strains that are morphologically and phylogenetically similar to the bacteriocin-producing microorganisms. Therefore, the development of cosmetic materials using lactic acid bacteria cultures or lactic acid bacteria fermented plant extracts and the like containing these substances is actively underway, and this progress is in line with the latest trend of being favorable for environmental protection and safety materials.
Nowadays, consumers expect not only general care effects in cosmetics but also various functions thereof (e.g., anti-aging, fine wrinkle improvement, anti-irritation, anti-inflammation, whitening effects, etc.).
In recent years, various plant extracts have been used for the purpose of these cosmetics, and there is an increasing desire for products based on a new concept that shows an improved range of performance characteristics by combining various characteristics.

In line with the environmental era, the inventors of the present application have made efforts to research the cosmetic effects of lactobacillus cultures derived from kimchi, which are safe to the skin, have no side effects and can be effectively used to prevent aging, improve skin elasticity and improve skin wrinkles through collagen biosynthesis, and in addition, have studied the possibility of their development in domestic industrialization.

Detailed Description
In order to accomplish the above objects, one aspect of the present application provides a cosmetic composition for improving skin wrinkles, comprising a lactobacillus plantarum culture, from which bacterial bodies are removed.
Hereinafter, the cosmetic composition for improving skin wrinkles, comprising lactobacillus plantarum culture of the present application, is explained in detail.
As used herein, the term "lactobacillus plantarum" refers to a microorganism of the genus lactobacillus, which is a gram-negative bacterium capable of producing lactic acid. It is found in fermented foods such as sauerkraut, pickles, sauerkraut, etc.
The lactobacillus plantarum strain may specifically be lactobacillus plantarum CJ L P133 strain, and it may be the strain deposited under accession number KCTC 11403 BP.
Specifically, lactobacillus plantarum CJ L P133 strain was deposited at 16.10.2008 under the budapest treaty at the korean typical culture collection (KCTC) of the international depository organization korean bioscience and biotechnology institute (KRIBB), and assigned registration number KCTC 11403 BP.
As used herein, the term "culture" refers to a medium comprising a byproduct produced by culturing a strain of a microorganism in the medium, wherein the strain is allowed to take up and metabolize nutrients contained in the medium. The term "culture" includes all preparations which can be formed using a culture medium (e.g., a diluent or a concentrate of the culture medium, a dried product obtained by drying the culture medium, a crude or purified product of the culture medium, a mixture thereof, and the like).
Specifically, the culture may be a culture obtained by culturing a lactobacillus plantarum strain, or may be a culture obtained by culturing a strain from which bacterial bodies are removed after the culture. That is, a culture medium from which the bacterial body has been removed can be used as the culture.
More specifically, the culture may be obtained by a method comprising: (a) culturing a lactobacillus plantarum strain by inoculating a culture medium; (b) centrifuging the resulting product of step (a); (c) recovering the supernatant from the resulting product of step (b).
In step (a), the medium may be any medium used in the art as long as the medium can culture the strain and recover the culture after the culture, specifically a liquid medium, and more specifically an MRS liquid medium.
In order to obtain a pure isolate, step (a) may further comprise the step of plating the lactobacillus plantarum strain on a solid medium and culturing and collecting those colonies which are confirmed to be pure isolates, prior to step (a).
In step (b), centrifugation can be performed using a high speed centrifuge at 1 to 7 ℃, specifically 2 to 6 ℃, and more specifically 3 to 5 ℃ at 1,500 to 3,500rpm, specifically 2,000 to 3,000rpm, and more specifically 2200 to 2800rpm for 10 to 30 minutes, specifically 15 to 25 minutes, and more specifically 17 to 23 minutes.
Step (c) may further comprise the step of filtering the supernatant. In addition, the step (c) may further comprise the step of pulverizing the filtered supernatant using a lyophilizer.
As used herein, the term "wrinkles" refers to fine lines resulting from skin aging, and wrinkles may be caused by genes, a decrease in the level of collagen present in the dermis of the skin, the external environment, and the like.
In the present application, the composition can improve skin wrinkles by a method of increasing the level of collagen synthesis, thereby preventing a decrease in collagen.
Collagen is an important fibrin protein, which accounts for about 25% of the total human protein by weight. It occupies most connective tissue and has the effects of intercellular matrix, structural support, induction of cell division and differentiation, providing tensile strength to the skin, etc.
As used herein, the term "ameliorating" refers to suppressing or inhibiting the symptoms of skin wrinkles, or alleviating the symptoms of skin wrinkles that have occurred on the skin using the compositions of the present application.
In the present application, the cosmetic composition may be formulated in the form of a solution, a topical ointment, cream, foam, nutritional emollient, soft emollient, filler (pack), soft water, milky lotion, makeup base, essence, soap, liquid cleanser, bath agent, sunscreen, suntan oil, suspension, emulsion, paste, gel, lotion, powder, soap, surfactant-containing cleanser, oil, foundation, emulsion foundation, wax foundation, patch, and spray, but the formulation is not limited thereto.
In the present application, the cosmetic composition may further comprise at least one cosmetically acceptable carrier for incorporation into the cosmetics of ordinary skin. As conventional components of the cosmetically acceptable carrier, for example, oil, water, surfactant, humectant, lower alcohol, thickener, chelating agent, pigment, preservative, perfume, etc. may be appropriately mixed, but the components are not limited thereto.
The cosmetically acceptable carrier incorporated into the cosmetic composition of the present application may vary depending on the formulation of the cosmetic composition.
When the formulation of the present application is an ointment, a paste, an emulsifiable concentrate or a gel, animal oil, vegetable oil, wax, paraffin, starch, tragacanth, a cellulose derivative, polyethylene glycol, silicone, bentonite, silica, talc, zinc oxide, or the like can be used as a carrier component, but the carrier component is not limited thereto. These carrier components may be used alone or in combination of two or more.
When the formulation of the present application is a powder or a spray, lactose, talc, silica, aluminum hydroxide, calcium silicate, polyamide powder, etc. may be used as a carrier component, and particularly, in the case of a spray, the formulation may further include a propellant such as chlorofluorocarbon, propane/butane and dimethyl ether, but the carrier component is not limited thereto. These carrier components may be used alone or in combination of two or more.
When the formulation of the present application is a solution or an emulsion, a solvent, a solubilizing agent or an emulsifier (e.g., water, ethanol, isopropanol, ethyl carbonate, ethyl acetate, benzyl alcohol, benzyl benzoate, propylene glycol, 1, 3-butylene glycol, etc.) may be used as a carrier component, and in particular, cottonseed oil, peanut oil, corn seed oil, olive oil, castor oil and sesame oil, aliphatic esters of glycerin, and fatty acid esters of polyethylene glycol or sorbitan may be used, but the carrier component is not limited thereto. These carrier components may be used alone or in combination of two or more.
When the formulation of the present application is a suspension, a liquid diluent (e.g., water, ethanol or propylene glycol), a suspending agent (e.g., ethoxylated isostearyl alcohol, polyoxyethylene sorbitol ester and polyoxyethylene sorbitan ester), microcrystalline cellulose, aluminum metahydroxide, bentonite, agar, tragacanth, and the like may be used as a carrier component, but the carrier component is not limited thereto. These carrier components may be used alone or in combination of two or more.
When the formulation of the present application is a soap, an alkali metal salt of a fatty acid salt, a half ester salt of a fatty acid, a fatty acid protein hydrolysate, isethionate, a lanolin derivative, a fatty alcohol, a vegetable oil, glycerin, a sugar, or the like may be used as the carrier component, but the carrier component is not limited thereto. These carrier components may be used alone or in combination of two or more.
Another aspect of the present application provides a method for preventing or improving skin wrinkles, comprising administering to a subject a lactobacillus plantarum culture in which bacterial bodies are removed.
The method of improving skin wrinkles may be a method comprising applying to a subject in need of improving skin wrinkles a cosmetic composition comprising a lactobacillus plantarum culture and an additional cosmetically acceptable carrier. The cosmetically acceptable carrier is the same as described above.
The subject may or may not include a human. Specifically, the subject may be a mammal including a cow, a pig, a sheep, a chicken, a dog, a human, and the like; birds; and the like, and can be any subject including, but not limited to, those who can ameliorate skin wrinkles by administering the above compositions.
Another aspect of the present application provides use of a composition for preparing a cosmetic for improving skin wrinkles, comprising a lactobacillus plantarum culture, from which bacterial bodies of lactobacillus plantarum have been removed.
Examples
Hereinafter, the present application will be described in detail by way of exemplary embodiments. However, it will be apparent to those skilled in the art to which the present application pertains that these exemplary embodiments are for illustrative purposes only, and are not intended to limit the scope of the present application.
Example 1 preparation of Lactobacillus plantarum CJ L P133 culture
Lactobacillus plantarum CJ L P133 strain deposited under accession number KCTC 11403BP was plated on solid MRS medium (BD Difco, Lactobacillus MRS agar) and cultured at 37 ℃ for about 18 hours after confirmation, pure isolated colonies were collected and inoculated into liquid MRS medium (BD Difco, Lactobacillus broth MRS) using platinum ring and cultured at 37 ℃ for about 18 hours after completion of culture, culture had 1.0 × 109CFU/m L concentration to separate bacterial bodies from the culture, the culture was centrifuged at 4 ℃ and 2500rpm for 20 minutes using a high speed centrifuge (L ABOGENE 1248R.) after filtering the supernatant of the culture, the filtered supernatant was pulverized using a lyophilizer (HETO P L90000). The culture thus prepared was named "Lactobacillus plantarum CJ L P133 culture".
Example 2 confirmation of collagen synthesizing ability of Lactobacillus plantarum CJ L P133 culture
To confirm the collagen synthesis effect of the lactobacillus plantarum CJ L P133 culture prepared in example 1, the amount of collagen expression in skin fibroblasts was measured.

From the above results, it was confirmed that the culture of lactobacillus plantarum CJ L P133 of the present application can increase the synthesis of collagen, which is known to have a major role in the skin aging mechanism, in which wrinkles are formed by decreasing substrate proteins constituting the skin, or increasing enzymes decomposing the substrate proteins, and thus the culture is effective for improving skin wrinkles.

1. A cosmetic composition for improving skin wrinkles, comprising a Lactobacillus plantarum culture from which Lactobacillus plantarum bacterial bodies have been removed.

wherein the Lactobacillus plantarum strain is the Lactobacillus plantarum CJ L P133 strain deposited under accession number KCTC 11403 BP.
3. The cosmetic composition according to claim 1,
wherein the culture is obtained by a method comprising:
(a) culturing the lactobacillus plantarum strain by inoculating a culture medium;
(b) centrifuging the resulting product of step (a); and
(c) recovering the supernatant from the resulting product of step (b).
4. A method for preventing or ameliorating skin wrinkles comprising administering to a subject a lactobacillus plantarum culture from which the bacterial bodies of the lactobacillus plantarum have been removed.
5. Use of a composition comprising a culture of lactobacillus plantarum from which the bacterial bodies of said lactobacillus plantarum have been removed, for the preparation of said cosmetic product intended for improving skin wrinkles.
