Exclusion criteria: a. pregnant or lactating women; b. those known to be allergic to the main raw materials of the product; c. systemic or topical glucocorticoid or immunosuppressive therapy, with a drug withdrawal time of less than 4 weeks; d. serious systemic diseases, such as liver and kidney diseases, blood system diseases, autoimmune diseases, malignant tumors, chronic serious infection, diabetes, psychosis, drug addiction, alcoholism, etc.
2) The implementation method comprises the following steps: all volunteers enrolled were randomly assigned to three parallel groups, test, control and blank, 30 persons each, using a computer generated random grouping table; the volunteers in the test group eat 3 candies obtained in the example 1 after half an hour every day according to the requirement; the volunteers in the control group eat the candies prepared in the comparative example 13 times a day after half an hour every day according to requirements; the volunteers in the blank group eat similar candies without functional components for half an hour every day according to the requirement, 3 candies are taken every day, and three groups of test clothes adopt a back-to-back mode; volunteers were asked to not change previous facial wash habits until the end of the 28-day trial cycle. And performing phenotype expression evaluation and skin barrier function related index determination on the 0 th day and the 28 th day, and recording evaluation detection results and adverse reactions or case specific situations. Volunteers were asked to be unable to use other skin barrier repair type products during the trial.
3) Evaluation of phenotypic performance: evaluating the severity of 4 physical signs of stabbing pain and flushing, dryness and pruritus, desquamation and erythema and erythra and acne, and scoring according to a 3-grade and 4-grade method, wherein each item is counted from 0 to 3, and 0 is zero, so that the severity cannot be seen after careful observation; 1, light, signs exist but need to be observed carefully; 2-medium, this sign is immediately visible; when it is heavy, 3, this sign is very obvious. The degree of itching and tingling was evaluated, 0 ═ none; 1 is light and does not need scratching; 2, scratching is required; when 3 is heavy, it affects sleep or life obviously. The symptom integral is the integral sum of stabbing pain and redness, dry itching, desquamation erythema, erythra and acne and pruritus signs, and the symptom integral reduction index (SSRI is the integral of symptoms before treatment-integral of symptoms after treatment)/integral of symptoms before treatment, the SSRI is less than 20%, the progress is that the SSRI is more than or equal to 20% and less than or equal to 60%, the effect is that the SSRI is more than or equal to 60% and less than or equal to 90%, and the cure is that the SSRI is more than 90%.
4) And (3) determination of related indexes of barrier function: the measurement of the relevant indexes of the barrier function needs to be carried out in the environment without direct sunlight and wind, at 22-24 ℃ and with the humidity of 40-60%, the volunteer is rested for 10-20 min, the water loss rate of the skin at the nasolabial sulcus of the patient through The Epidermis (TEWL) and the water content of the horny layer are measured by using a skin tester (produced by Courage-khazaka company, Germany), and each index is detected for 3 times and the average value is taken. Among them, the transepidermal water loss rate is also called transdermal water loss rate.
5) The statistical method comprises the following steps: SPSS statistical software is used for analyzing data, and when P is less than 0.05, the difference is considered to have statistical significance.
6) Trial results
6.1 volunteer Condition
The trial period amounted to 28 days. Evaluating 1 case of each of the volunteer test group and the control group on day 28, and 2 cases of the blank group; results analysis retained 29 people in the test group, 29 people in the control, and 28 people in the blank group. Average age of volunteers (31.62 ± 9.72) years (22-48 years). The average ages of volunteers in the test group, the control group and the blank group are compared and are (31.59 +/-8.66) years, (32.02 +/-7.64) years and (30.00 +/-9.82) years, and the difference among the three groups has no statistical significance. Comparing the symptom score, TEWL, stratum corneum water content and epidermal sebum content of three groups of patients entering the group at the baseline of evaluation on 0 day, wherein the differences of the three groups have no statistical significance (P is more than 0.05).
6.2 phenotypic Performance assessment
When evaluated on day 0, the symptom scores are respectively (2.75 +/-1.60), (2.77 +/-1.36) and (2.48 +/-1.21), and the differences among the three groups have no statistical significance; on the 28 th day evaluation, the symptom scores of the test group, the control group and the blank group are respectively (1.02 +/-0.96), (1.53 +/-1.05) and (1.92 +/-1.17), the test group is obviously superior to the blank group (P is less than 0.05), the control group has a trend superior to the blank group, but the difference has no statistical significance (P is more than 0.05).
6.3 Barrier function index
Transdermal water loss rate (TEWL): the TEWL [ (10.99. + -. 5.88) g/(m2h) ] of the test group was lower than that of the blank group [ (16.17. + -. 8.47) g/(m2h) and the control group [ (12.17. + -. 6.42) g/(m2h) when evaluated on day 28, and the difference was statistically significant (P < 0.05).
Water content of horny layer: the test group contained higher water content in the stratum corneum (40.22. + -. 11.66) than the blank (34.28. + -. 10.86) and the control (37.32. + -. 9.86) at day 28, the difference being statistically significant (P < 0.05).
As can be seen from the results of the symptom integral related to skin barrier damage, and the indexes of the transdermal water loss rate and the moisture content of the stratum corneum related to the skin barrier function, compared with the blank group, the indexes of the symptom integral related to skin barrier damage, the transdermal water loss rate, the moisture content of the stratum corneum and the like of the volunteers in the test group are significantly improved, which indicates that the candy prepared in example 1 can enhance the defense capability of the skin barrier.
While the volunteers in the control group improved the symptoms of integral, transdermal water loss rate, stratum corneum water content and the like related to the skin barrier damage, but the improvement effect was lower than that of the volunteers in the test group, which indicates that the candy prepared in example 1 has better skin barrier strengthening defense capability than the candy prepared in comparative example 1.
