Abstract
本发明公开了一种体外诱导iPS细胞向乳腺方向分化的方法及其专用培养基。该体外诱导方法包括以下步骤:将iPS细胞消化为单细胞,以该单细胞作为种子细胞接入iPS细胞维持培养基,并获得高密度单层细胞;将单层iPS细胞在分化培养基I中进行培养,细胞每天换液;弃去培养基,更换为分化培养基II培养,细胞进行隔天换液;弃去培养基,更换为分化培养基III培养细胞,细胞进行隔天换液;弃去培养基,更换为分化培养基IV继续培养细胞。本发明可获取大量、单一、处在不同分化阶段的乳腺细胞,具有操作简单、成本低廉和分化效率高等优点,应用前景广阔。
Description
技术领域
本发明属于发育生物学和生物医学工程技术领域。具体地,涉及一种体外诱导iPS细胞向乳腺方向分化的方法及其专用培养基;更具体地,涉及一种基于体外定向诱导分化,把iPS细胞向乳腺细胞转变,获得大量、单一、处在不同分化阶段的乳腺细胞的方法及其专用培养基。
背景技术
干细胞因具有自我更新及多向分化潜能在生命科学、生物、医疗等各个领域备受关注。目前,全球范围内针对干细胞的研究层出不穷,从血液病到组织损伤修复到抗衰老,干细胞技术正逐步从临床基础研究过渡到临床应用阶段,为许多传统医学难以解决的重大临床问题带来了新希望。干细胞治疗已成为血液系统疾病、神经系统疾病、心血管及肝脏疾病等重大疾病的新的治疗选择。
乳腺癌是女性发病率最高的肿瘤类型。家族遗传性乳腺癌,如BRCA1突变导致的三阴性乳腺肿瘤,临床上转移、复发率高,是最难治疗的乳腺肿瘤类型之一。而BRCA1突变携带者一生罹患乳腺肿瘤的风险高达40%~80%,患病后,病人5年生存率仅为10%,因此,阐明相关突变诱发肿瘤发生、发展的过程,发现潜在治疗靶点,阻断肿瘤起始,对家族性乳腺肿瘤的防治意义重大。癌变发生过程的阐明依赖于疾病模型。然而,目前尚无研究体系可在人乳腺发育水平模拟遗传性肿瘤发生的过程,如源自病人肿瘤组织的细胞系无法重现癌变发生的进程、小鼠模型具种属缺陷等。
诱导多能干细胞应用于乳腺疾病研究中具有巨大的潜在优势。iPS细胞可从患者自身体细胞直接诱导而获得,保留了疾病基因组的遗传信息,具有分化为任意细胞的潜能,因而,可在体外模拟疾病的病理进程与表型。早在2013年,Soyombo研究组已将BRCA1突变家系病人的体细胞重编程为iPS细胞。2017年,Qu等实现了人iPS细胞以3D形式分化为类乳腺结构,然而,该体外诱导分化方法在组织工程,及疾病模型建立的应用中均存在一定的缺陷。该分化方法存在以下不足:1、体外诱导分化过程中未对细胞命运进行足够人为干预,导致分化效率低。尤其是在分化早期,细胞以EB球的形式随机分化,而诱导分化的培养基仅起到将随机分化而来的乳腺方向的细胞进行富集。在胚胎发育中,乳腺细胞来自表皮外胚层,由于诱导多能干细胞具有朝三个胚层分化的潜能,上述方法随机分化阶段,仅部分细胞向外胚层分化,且主要为神经外胚层,而非表皮外胚层细胞,因而,基于随机分化的乳腺细胞得率低。2、体外诱导分化过程中细胞始终以EB球,或包被了基质胶的悬浮球的形式培养,最终获得的类乳腺结构同样被包被在基质中,难于分离到足够的、处在特定分化阶段的乳腺上皮细胞。3、鉴于上述两个技术特点,该分化方法步骤繁琐,乳腺细胞得率低,难以获得大量、单一、处在不同分化阶段的乳腺细胞。
综上所述,由于缺乏iPS细胞诱导乳腺分化的有效方法,极大限制了干细胞在乳腺疾病领域的应用,开发新型iPS细胞乳腺诱导分化方法意义重大。
发明内容
本发明要解决的技术问题是克服上述现有技术的缺陷和不足,提供一种新的体外诱导iPS细胞向乳腺方向分化的方法。本发明所述诱导iPS细胞向乳腺方向分化的方法,可获得大量、单一、处在不同分化阶段的乳腺细胞,易于体外扩增,足量、质优,为乳腺干细胞在组织工程中的应用及疾病模型建立创造了条件。
本发明的另一目的是提供一种用于体外诱导iPS细胞定向分化为乳腺细胞的培养基。
本发明的再一目的是提供使用上述方法或上述培养基诱导得到的乳腺细胞。
本发明上述目的通过以下技术方案实现:
一种用于体外诱导iPS细胞定向分化为乳腺细胞的培养基,包括依次使用的以下五种培养基;其中,
iPS细胞维持培养基:包括mTeSR培养基和5~15 μM Y27632;
分化培养基I:包括E6培养基、5~15 ng/mL BMP4、5~15 μM SB431542和5~15 μMSU5402;
分化培养基II:包括E6培养基、5~15 ng/mL BMP4、5~15 μM SB431542;
分化培养基III:包括MammoCult™ Human Medium Kit培养基和1~10 ng/mL BMP4;
分化培养基IV:是MammoCult™ Human Medium Kit培养基。
本发明能够提供更适于乳腺细胞发育分化的微环境,有利于获得大量、单一、处在不同分化阶段的乳腺细胞,为人iPS细胞向乳腺细胞的定向分化提供了一种新的研究思路,为探讨人iPS细胞向乳腺细胞分化的调控机制提供了新的研究线索。

在本发明较佳的实施例中,iPS细胞维持培养基中,Y27632的浓度可以为5 μM、6 μM、7 μM、8 μM、9 μM、10 μM、11 μM、12 μM、13 μM、14 μM、15 μM等。
在本发明较佳的实施例中,分化培养基I中,BMP4的浓度可以为5 ng/mL、6 ng/mL、7 ng/mL、8 ng/mL、9 ng/mL、10 ng/mL、11 ng/mL、12 ng/mL、13 ng/mL、14 ng/mL、15 ng/mL等。

S1. 将iPS细胞消化为单细胞,以该单细胞作为种子细胞接入iPS细胞维持培养基中进行培养;
S2. 弃去步骤S1的培养基,更换为所述的分化培养基I培养细胞48 h;并每天换液;
S3. 弃去步骤S2的培养基,更换为所述的分化培养基II培养细胞,并隔天换液;
S4. 弃去步骤S3的培养基,更换为所述的分化培养基III培养细胞,并隔天换液;
S5. 弃去步骤S4的培养基,更换为所述的分化培养基IV继续培养细胞,并隔天换液。
本发明将iPS细胞以单层培养的方式获得表皮外胚层细胞后,直接更换培养基,诱导细胞向乳腺方向分化,通过该方法可获取大量、单一、处在不同分化阶段的乳腺细胞。

本发明提供了一种利用iPS细胞具有高度自我更新和多能性的生物学特性,将其体外定向诱导分化为乳腺细胞的方法。该方法通过利用专用培养基、控制培养时间点来控制细胞命运,先将iPS细胞分化为表皮外胚层,再进一步诱导表皮外胚层细胞向乳腺细胞分化,从而实现iPS细胞体外定向分化为乳腺细胞的目的。本发明提供的将iPS细胞高效诱导分化为乳腺细胞的方法可获取大量、单一、处在不同分化阶段的乳腺细胞,具有操作简单、成本低廉和分化效率高等优点,更适于疾病模型的建立,为干细胞移植、及乳腺癌等疾病研究奠定了基础。本发明将在基础及转化医学领域发挥重要作用,应用前景广阔。

2、体外诱导人iPS细胞定向分化为乳腺细胞的方法,具体包括以下步骤:
S1. 取汇合度达70%~80%的人iPS细胞,以accutase消化2~5 min,离心收集单细胞,用PBS清洗后,得到种子细胞;将种子细胞以350,000~400,000细胞/cm2的密度接种于Matregel包被30 min的培养板,将细胞接入iPS细胞维持培养基I中进行培养24 h;
S2. :取培养24 h的细胞,弃去步骤S1的培养基,用PBS清洗细胞1次,更换为上述分化培养基I培养48 h,每天换液;
S3. 弃去步骤S2的培养基,用PBS清洗细胞1~2次,重新更换为权利要求1或4所述的分化培养基II培养细胞,培养细胞9 d,并隔天换液;
S4. 弃去步骤S3的培养基,用PBS清洗细胞1~2次,更换为权利要求1或5所述的分化培养基III培养细胞,培养细胞6 d,并隔天换液;
S5. 弃去步骤S4的培养基,用PBS清洗细胞1~2次,更换为权利要求1所述的分化培养基IV继续培养细胞,培养细胞14 d,并隔天换液。
实施例2 体外诱导人iPS细胞定向分化为乳腺细胞
1、实施例中使用以下培养基:
iPS细胞维持培养基:mTeSR培养基和10 μM Y27632;
分化培养基I:E6培养基、10 ng/mL BMP4、10 μM SB431542和10 μM SU5402;
分化培养基II:E6培养基、10 ng/mL BMP4、10 μM SB431542;
分化培养基III:包括MammoCult™ Human Medium Kit培养基和10 ng/mL BMP4;
分化培养基IV:是MammoCult™ Human Medium Kit培养基。
2、体外诱导人iPS细胞定向分化为乳腺细胞的方法,具体包括以下步骤:
S1. 取汇合度达70%~80%的人iPS细胞,以accutase消化2~5 min,离心收集单细胞,用PBS清洗后,得到种子细胞;将种子细胞以350,000~400,000细胞/cm2的密度接种于Matregel包被30 min的培养板,将细胞接入iPS细胞维持培养基I中进行培养24 h;

S4. 弃去步骤S3的培养基,用PBS清洗细胞1~2次,更换为权利要求1或5所述的分化培养基III培养细胞,培养细胞4 d,并隔天换液;
S5. 弃去步骤S4的培养基,用PBS清洗细胞1~2次,更换为权利要求1所述的分化培养基IV继续培养细胞,培养细胞16 d,并隔天换液。
实施例3 体外诱导人iPS细胞定向分化为乳腺细胞
1、实施例中使用以下培养基:
分化培养基I:mTeSR培养基和15 μM Y27632;
分化培养基II:E6培养基、15 ng/mL BMP4、10 μM SB431542和10 μM SU5402;
分化培养基III:包括MammoCult™ Human Medium Kit培养基和10 ng/mL BMP4;
分化培养基IV:是MammoCult™ Human Medium Kit培养基。
2、体外诱导人iPS细胞定向分化为乳腺细胞的方法,具体包括以下步骤:
S1. 取汇合度达70%~80%的人iPS细胞,以accutase消化2~5 min,离心收集单细胞,用PBS清洗后,得到种子细胞;将种子细胞以350,000~400,000细胞/cm2的密度接种于Matregel包被30 min的培养板,将细胞接入iPS细胞维持培养基I中进行培养24 h;
S2. :取培养24 h的细胞,弃去步骤S1的培养基,用PBS清洗细胞1次,更换为上述分化培养基I培养48 h,每天换液;
S3. 弃去步骤S2的培养基,用PBS清洗细胞1~2次,重新更换为权利要求1或4所述的分化培养基II培养细胞,培养细胞10 d,并隔天换液;
S4. 弃去步骤S3的培养基,用PBS清洗细胞1~2次,更换为权利要求1或5所述的分化培养基III培养细胞,培养细胞4 d,并隔天换液;
S5. 弃去步骤S4的培养基,用PBS清洗细胞1~2次,更换为权利要求1所述的分化培养基IV继续培养细胞,培养细胞36 d,并隔天换液。
实施例4 iPS细胞体外乳腺诱导分化细胞鉴定
1、按照实施例1的体外诱导方法,先将人iPS细胞诱导分化为表皮外胚层,再将细胞向乳腺方向诱导分化。实验发现,分化第30天的细胞呈现乳腺上皮细胞样(见图1中A图、B图);同时发现,乳腺相关marker在分化过程中表达,并逐渐上调(见图1中C图)。
图1中的B图为乳腺分化中细胞形态图。由图1中B图可知,细胞以单层培养的方式诱导分化,在分化第25天,单层细胞上出现成团生长细胞,该细胞形态上类似于分化的乳腺腺泡。在图1中C图乳腺分化方法优化中,乳腺基底方向marker随分化过程逐渐上调。
2、对分化第30天的细胞进行流式分选,发现诱导分化的细胞主要分为EPCAM+CD10-,EPCAM+ CD10+,EPCAM- CD10+三群(图2中A图)。
RNA-seq实验对上述三群细胞进行鉴定,结果显示EPCAM+ CD10-,EPCAM+ CD10+具有与成体乳腺细胞类似的基因表达谱,而EPCAM- CD10+则疑似神经细胞(图2中B图、图3中C图)。
随后,对EPCAM+ CD10-,EPCAM+ CD10+两群细胞进行了功能鉴定,实验结果显示,分化的EPCAM+ CD10+乳腺细胞具有重建小鼠乳腺结构的能力(图3中D图),提示该细胞为乳腺干细胞(MaSC)。
综上结果可知,利用本发明的专用培养基,将iPS细胞以单层培养的方式获得表皮外胚层细胞后,直接更换培养基,可高效诱导细胞向乳腺方向分化,而且通过该方法可获取大量、单一、处在不同分化阶段的乳腺细胞,易于体外扩增,足量、质优,具有操作简单、成本低廉和分化效率高等优点,更适于疾病模型的建立,为干细胞移植、及乳腺癌等疾病研究奠定了基础。
Claims (10)
1.一种用于体外诱导iPS细胞定向分化为乳腺细胞的培养基,其特征在于,包括依次使用的以下五种培养基;其中,
iPS细胞维持培养基:包括mTeSR培养基和5~15 μM Y27632;
分化培养基I:包括E6培养基、5~15 ng/mL BMP4、5~15 μM SB431542和5~15 μMSU5402;
分化培养基II:包括E6培养基、5~15 ng/mL BMP4、5~15 μM SB431542;
分化培养基III:包括MammoCult™ Human Medium Kit培养基和1~10 ng/mL BMP4;
分化培养基IV:是MammoCult™ Human Medium Kit培养基。
2.体外诱导iPS细胞定向分化为乳腺细胞的方法,其特征在于,包括以下步骤:
S1. 将iPS细胞消化为单细胞,以该单细胞作为种子细胞接入权利要求1所述的iPS细胞维持培养基;
S2. 弃去步骤S1的培养基,更换为权利要求1所述的分化培养基I培养48 h;细胞进行每天换液;
S3. 弃去步骤S2的培养基,重新更换为权利要求1所述的分化培养基II培养细胞,并隔天换液;
S4. 弃去步骤S3的培养基,更换为权利要求1所述的分化培养基III培养细胞,并隔天换液;
S5. 弃去步骤S4的培养基,更换为权利要求1所述的分化培养基IV继续培养细胞,并隔天换液。
3.根据权利要求2所述的方法,其特征在于,步骤S3的培养时间为8~12 d。
4.根据权利要求3所述的方法,其特征在于,步骤S3的培养时间为9~11 d。
5.根据权利要求2所述的方法,其特征在于,步骤S4的培养时间为1~10 d。
6.根据权利要求5所述的方法,其特征在于,步骤S4的培养时间为4~6 d。
7.根据权利要求2所述的方法,其特征在于,步骤S5的培养时间为5~45 d。
8.根据权利要求7所述的方法,其特征在于,步骤S5的培养时间为9~34 d。
9.根据权利要求2所述的方法,其特征在于,步骤S1中,将种子细胞以350,000~400,000细胞/cm2的密度接种于Matregel包被的培养板,培养24 h获得高密度单细胞层。
10.使用权利要求1所述培养基或权利要求2~9任一所述方法诱导得到的乳腺细胞。
