Description

Skin burn and scald wound repairing preparation and preparation method thereof
Technical Field
The invention belongs to the technical field of medical products and preparation processes thereof, and particularly relates to a skin burn and scald wound repairing preparation and a preparation method thereof.
Background
Burns and scalds are injuries caused by external factors such as boiling water (oil), fire, light, radiation or chemical substances, including skin mucosa injuries, serious injuries to subcutaneous tissues can occur, and the burns and scalds are one of the most common and extremely complex trauma diseases which occur in battlefields, fire fighting, mines, factories and various natural disasters and daily work and life. If not properly handled, adverse consequences can result.
The severity of burn and scald is mainly judged according to the position, area and depth of the burn and scald, and generally divided into three degrees according to the depth of the burn and scald:
first-degree burn and scald: only the epidermis layer is injured, and the injured skin is reddened and swollen, has strong pain feeling and does not have blisters;
second degree burn and scald: damage to the dermis, local red swelling, fever, severe pain and obvious blisters;
three-degree burn and scald: the whole skin including subcutaneous fat, bone and muscle are damaged, the skin is scorched and necrotic, and the pain is not severe because nerves are damaged.
At present, a lot of medicines for treating burns and scalds exist, and a lot of patents and documents exist, namely, medicines and treatment measures for treating burns and scalds mainly based on western medicine, wherein the western medicine treatment is to clean (namely debridement) wound surfaces, treat and disinfect necrotic muscle tissues of the wounds and then wrap the wounds. Western medicine does not advocate the application of drugs on the wound surface, mainly takes internal drugs (such as anti-inflammatory and analgesic drugs), and the method looks scientific, has not ideal actual effect and is expensive. While the theoretical literatures and patents of traditional Chinese medicine for external treatment of burns and scalds in China are disclosed and published in large quantities, new dosage forms are continuously appeared, such as: "Longhu Shaoshangshang oil"; concentrated suspension and liquid agent, such as Jinyusan solvent, Huzhang scald liquid, etc., ointment, Huanglian ointment; tincture named FUQIPING tincture; powder, ulcer powder; the film agent is a compound rhubarb burn medicine film. (Guoshi, et al, the progress of burn treatment with external traditional Chinese medicine preparation, vol. 13, 4, in 2004, 8 months in traditional Chinese medicine J for external treatment). The medicines have long healing time and high price in different degrees, and scars can be left after healing. For example, the Chinese patent application No. 95110884.0 discloses a Chinese medicine for treating burns and scalds, which is powder prepared from more than ten Chinese herbal medicines such as centipede, raw scorpion, sky-blueness, angelica dahurica, coptis root and the like, but the using procedure is complex, the best effect is difficult to achieve if the Chinese patent application No. 95110884.0 is not properly prepared for patients, the use is troublesome, the treatment effect is poor, and the treatment time is long, so that a preparation which can quickly relieve pain of the burn and scald wound surface, can diminish inflammation and inhibit bacteria, quickly and effectively repairs the skin of the wound surface, can heal the wound surface in a short time, does not leave scars after healing and has high.

The control group used the conditioning solution prepared in CN108403450A in example III, and the control group used only the surface stem cell activating factor.
Treating burns and scalds: each mouse was coated with 8% sodium sulfide depilatory, gently scraped off after several minutes, and wiped clean with clear water. Observing for several hours, and determining that the depilated part has no abnormal conditions such as red swelling, inflammation and damage. Adopts chloral hydrate for shallow anesthesia and is arranged on a healing operation table. The pot with water was placed on an electric furnace and a 20g weight was placed in the pot and heated to boiling for 10 min. Then the upper end of the weight is clamped by a signature, the weight is quickly placed on the skin of the mouse for 5s, and slight pressure is applied, so that the scald area is 2cm multiplied by 2 cm.
Administration treatment: spraying the test components with the sprays obtained in the corresponding examples 5-8 times per day, 3-5mL each time; spraying the repairing liquid obtained in the third embodiment to a control group, and spraying three times every day, wherein three times are sprayed each time; the control group was applied with surface stem cell activating factor 3-5 times daily, 1-2mL each time.
Wound surface observation: after the daily application of the medicine is finished, the skin wound of the mouse is observed, data are directly measured by adopting a ruler, and the area of the wound is calculated. The total observation period was 5 days.
The results of observation and calculation are as follows:
TABLE 1 treatment Change in scald wounds (/ cm) in subject mice2)
Figure BDA0002371994360000111
Figure BDA0002371994360000121
According to the data in the table, the prepared skin burn and scald wound spray preparation has obvious treatment effect on experimental subjects in each example, wherein the overall repair and healing effects of the examples 13, 15 and 16 are more effective, and the overall healing effect of the other examples tends to increase. The repair liquid of the control group has a certain repair effect, but the final observed effect is obviously better than the healing effect of the repair preparation spray. Therefore, the skin burn and scald wound repair preparation has an obvious healing effect, can achieve a high healing rate in a short time, and does not have any side effect. Meanwhile, the effect of wound surface repair by only using the surface stem cell activation factor is obviously weaker than that of the whole scheme.
