Description

Composition with sun-screening, whitening and anti-aging effects and application thereof
Technical Field
The invention belongs to the field of cosmetics, and relates to a composition with sun-screening, whitening and anti-aging effects and application thereof.
Background
In recent years, with the occurrence of partial holes in the ozone layer of the atmosphere, the ultraviolet ray filtering effect is reduced, and the radiation intensity of sunlight is increased. Although the sun ultraviolet is a necessary condition for all things to grow, excessive ultraviolet can damage the immune system of the human body, damage collagen and elastin in the dermis, reduce the elasticity of the skin, generate wrinkles and cause skin aging. Meanwhile, long-time and high-intensity ultraviolet irradiation can enable the skin to generate a large number of free radicals to induce peroxidation of cell membranes, so that melanocytes generate a large number of melanin, and then the melanin moves layer by layer through cell metabolism to the epidermis of the skin, and skin problems such as color spots, uneven skin color and the like seen by people are formed. Therefore, the importance of sun protection is an important way to maintain the health of skin.
Throughout the cosmetic market, most products contain sunscreens, which integrate the concept of sunscreen protection into the daily skin care life. However, most sunscreen products add a large amount of organic uv absorbers in the pursuit of high SPF sunscreen effect, which undoubtedly will greatly increase the possibility of skin irritation and irritation. Under the background that whitening and anti-aging become an industrial hotspot, natural plant extracts integrating a plurality of functions of sun-screening, whitening and anti-aging are developed and researched, and the natural plant extracts have good application prospects.

Effect example 1 tyrosinase Activity inhibition Rate test
Tyrosinase is a polyphenol oxidase, is a key enzyme and a rate-limiting enzyme in the process of forming skin melanin, the activity of the tyrosinase is in positive correlation with the melanin synthesis amount, and the activity change of the tyrosinase can cause pigmentation disorder dermatosis. The tyrosinase inhibitor can be used for preventing and treating pigmentation by inhibiting tyrosinase activity and preventing melanin formation.
Test samples: the compositions or natural substances in examples 1 to 4 and comparative examples 1 to 8 were each formulated into a solution of 0.1mg/mL using 0.2mol/L Phosphate Buffered Saline (PBS) having pH of 6.8 as a solvent, to obtain a test sample.
The test method comprises the following steps: to each of the above test samples (0.1 mg/mL) was added 1mL of L-tyrosine (0.3mol) and 1mL of Phosphate Buffer (PBS) having a pH of 6.8, and the mixture was preheated in a constant temperature water bath at 37 ℃ for 10min, then 0.1mL of an aqueous solution of tyrosinase (1.0mg/mL, mcllains buffer) was added, the mixture was sufficiently stirred, and after a reaction at 37 ℃ for 15 minutes, absorbance at 475nm was measured (D)1). In addition, the same reaction was carried out using the above phosphate buffer solution in place of the 0.1mg/mL solution prepared from the composition prepared in examples 1-4 and comparative examples 1-5, and the absorbance (D) was measured2). For comparison, the absorbance was measured without adding the sample solution and tyrosinase (D)3). The tyrosinase activity inhibition was calculated using the following formula:
tyrosinase activity inhibition ratio (%) - (D)2-D1)/(D2-D3)×100%;
The test results are shown in table 2.
TABLE 2 tyrosinase activity inhibition test results

Figure BDA0002386441110000061
And (4) test conclusion: in comparative examples 1 to 4, one of the four natural substances, namely the scutellaria root extract, rutin, curcumin and ferulic acid, is independently used, and as can be seen in comparison with example 1, the tyrosinase activity inhibition rate is remarkably lower than that of the composition provided by the invention when any one of the four natural substances is independently used; comparative examples 5 to 8 are compositions obtained by using any three of the four natural substances, and the same as other conditions of example 1, it can be seen that the clearance is improved compared with the clearance of one of the natural substances used alone, but the effect is different from that of the composition of the invention, which indicates that the composition of the invention can achieve a remarkable tyrosinase activity inhibition effect under the synergistic effect of the four extracts.
Effect example 2 melanin production inhibition test
The test method comprises the following steps: selecting the same generation of cells in logarithmic growth phase, blowing and beating the cells into single cell suspension after conventional pancreatin digestion treatment, counting and quantitatively inoculating the cells into a 96-well plate. After 24 hours, the supernatant was discarded, and the culture solutions containing examples 1-4 and arbutin, a positive control, 50. mu.g/ml, were added, respectively (the volume contents of the respective compositions were the same). At 37 deg.C, 5% by volume CO2Culturing under saturation and humidity conditions, adding RPMI1640 culture solution into a control hole, culturing for three days, replacing the culture solution once in the middle, then removing supernatant, counting cell density after conventional pancreatin digestion treatment, centrifuging to obtain cell precipitate, washing twice with PBS, centrifuging to remove supernatant, quantitatively adding 1N NaOH solution containing 10 vol% DMSO, carrying out water bath at 80 ℃ for 50 minutes, and then measuring the light absorption value at 450nm of an enzyme-labeling instrument. The inhibition of melanin production was calculated by the following formula:
the melanin production inhibition (%) was (test absorbance/control absorbance) × 100%;
the test results are shown in table 3.

And (4) test conclusion: the compositions of examples 1 to 3 all had the effect of inhibiting melanin production, and the composition of example 4 had the effect of inhibiting melanin production equivalent to arbutin, and was applied to whitening cosmetics.
Effect example 3 anti-wrinkle test
The free radicals can damage the skin from various aspects, so that the aging of the skin is accelerated, and therefore, whether the anti-aging product has an anti-aging effect can be judged by the effects of scavenging and inhibiting the free radicals.

The enzyme label plate is insulated for 30min at 30 ℃, and then the absorbance value of the sample at the wavelength of 517nm is measured (A)1) Simultaneously, the absorbance (A) of a blank sample (100. mu.L of absolute ethanol instead of DPPH) without DPPH free radical reagent was measured2) And the absorbance (A) of the sample (100. mu.L of absolute ethanol instead of the sample) with the DPPH radical reagent3). The DPPH radical clearance was calculated using the following formula:
