根据一个示例,可在体内使用(例如通过局部乳膏经皮肤放置、口服或注入血流等)具有特定光学特征(例如散射和/或吸收)的粒子。可定制粒子的光学特征以在存在生物标记物的情况下发生改变,并且可使用位于体外但能够成像到体内几厘米的光谱OCT在体内检测光谱特征的这种改变。这不依赖于大量纳米粒子的聚集来进行体内OCT检测。而是,单个共振粒子发生了修改或分解,这使得能够使用的粒子更少,并且使得能够进行多重检测。
根据一个示例,功能化粒子在该粒子的第一结构构造中具有第一光谱特征,并且在该粒子的第二结构构造中具有第二光谱特征。因此,粒子的结构构造的改变将导致其光学特性的改变,该改变可被追踪或确定。
图1是根据一个示例的直径为5μm的高介电(折射率,n=1.9)球的散射幅度的示意图。在图5的示例中,功能化粒子100包括涂覆有薄材料103(n=1.45)的介电芯101(n=1.9)。粒子100在该粒子的第一结构构造中具有第一光谱特征,在第一结构构造中粒子包括芯101和涂层103,并且粒子100在该粒子的第二结构构造中具有第二光谱特征,在第二结构构造中涂层103被降解或不存在而仅留下芯101。即,在图1的示例中,第一光学特征与粒子100的第一折射率有关,其中,n=1.45,而粒子的第二光学特征与粒子100的第二折射率有关,其中,n=1.9(即不存在涂层)。
因此,当涂层103被去除或以其他方式降解或不存在时,粒子100的散射光谱红移(red-shift)。散射光谱的该移动在图1中示出,其中,示出了在存在和不存在涂层的情况下粒子100的散射光谱。
在一个示例中,粒子100的涂层103或壳可被功能化以在存在例如生物标记物的另一种材料的情况下降解。因此,在存在这样的材料的情况下,涂层103将降解,从而导致粒子100的光学特性改变,并且因此随后的散射光谱将改变。
由于粒子的尺寸的改变很小,常规OCT方法将无法区分任何改变。但是,根据一个示例,通过用光谱OCT追踪粒子100的光谱,可通过更明显的光谱改变来检测小的结构改变,例如如图1所示。
根据一个示例,粒子芯和粒子涂层可由各种各样的天然和合成材料制成。以下是一些示例:
天然材料——壳聚糖、胶原蛋白、藻酸盐、右旋糖酐、明胶和白蛋白。
合成材料(性质上通常为聚合的)——聚苯乙烯、聚丙烯酰胺、聚甲基丙烯酸甲酯[PMMA]、聚乙二醇[PEG]、聚(乳酸)[PLA]、聚(乙醇酸)[PGA]、聚(丙交酯-共-乙醇酸)[PLGA]、聚(N-异丙基丙烯酰胺)[pNIPAM]、聚(ε-己内酯)[PCL]和聚(3-羟基丁酸酯-共-3-羟基戊酸酯)[PHBV]。
其他类型的材料(通常将其用作芯)——玻璃、二氧化钛、陶瓷、二氧化硅、磁性的和金属的(金、银)。
根据一个示例,可通过使用局部乳霜将功能化粒子注入皮肤中或放置在皮肤下,使得它们随后扩散浸泡在间隙液中的真皮层。在一个示例中,可将粒子的外壳层的性质设计或构造为响应于特定的生物标记物或生化提示而改变。该光学改变(即,提供从第一光谱特征到第二光谱特征的移动)可替代地由粒子的溶胀或降解引起,这可以是可逆的,从而改变粒子的折射率,或通过粒子中的化学改变来改变材料吸收光谱。也就是说,从粒子的第一结构构造到第二结构构造的改变(反之亦然)可通过涂层的去除或降解、或溶胀/去溶胀(deswelling)来实现。在一个示例中,可使用两种结构构造的组合。即,粒子可包括在存在第一材料的情况下降解的涂层和在存在第二材料的情况下溶胀的芯。
因此,当粒子与特定的生物分子(例如癌症标记物)接触时,外壳会降解,并且随后的光学性质改变可使用光谱OCT来远程检测。OCT系统可是台式机,也可是位于体外的集成可穿戴设备。
代替降解,可在这样的材料中涂覆粒子,该材料被功能化以在存在某生物标记物的情况下改变折射率。图2是根据一个示例的已经涂覆有100nm介电涂层的金纳米棒的散射光谱的示意图。随着介电涂层改变折射率,金纳米粒子的散射共振也改变。如果涂层被功能化以改变与例如周围的葡萄糖浓度成比例的折射率,则可将粒子注入血流中或放置在皮肤的真皮层中。然后可用光谱OCT检测金纳米棒的散射光谱,以确定体内血糖水平。由于来自粒子的总散射保持相对恒定,因此使用传统的OCT方法不会出现可观察到的改变。但是,如果使用光谱OCT,则可检测到峰值光学共振的这种移动。在图2的示例中,金纳米棒的直径为30nm,长度为175nm,并涂覆有100nm厚的介电材料。当介电涂层的介电常数从1.33变为1.45时,金纳米棒的散射光谱(图2的右侧)被描绘。
图3是描绘根据一个示例的功能化粒子的光谱特征的改变的示意图。图3a描绘了粒子301的光学特征,该粒子包括芯303和涂层305。在图3b中,粒子301以可预测的方式暴露于影响涂层305的生物或化学提示(chemical cue)。即,涂层305被构造为对生物或化学提示的存在做出反应。暴露于生物或化学提示的结果在图3c中示出,其中,涂层305被改变,从而更改了粒子的光谱。在图3c的示例中,描绘了对粒子涂层的一些潜在改变——可在存在生物或化学提示的情况下对涂层进行改变(310),从而导致粒子的折射率、吸收或偏振的变化;涂层可溶胀(311),或者反之可减小尺寸;或者涂层可降解(312)。降解可显著到涂层不再存在于粒子的芯上的程度。
图4是根据示例的方法的流程图。更具体地,图4是根据示例的用于检测功能化粒子的光谱特征的变化的方法的流程图。在框401中,提供具有第一光谱特征的粒子。第一光谱特征由粒子的第一结构构造定义。该第一结构构造可例如通过粒子的尺寸(例如处于溶胀或未溶胀状态)或通过如上所述的涂层来提供。在框403中,使用光谱光学相干断层扫描来检测第一光谱特征。这可在体内或体外进行。在框405中,追踪在存在识别、结合、亲和或改变粒子的材料的情况下从该粒子的第一光谱特征到第二光谱特征的变化。即,检测该粒子的光谱特征的变化。
图5是根据一个示例的方法的流程图。更具体地,图5是根据一个示例的用于辅助诊断或预后监测的方法的流程图。在框501中,提供具有第一光谱特征的粒子。第一光谱特征由粒子的第一结构构造定义。该第一结构构造可例如通过粒子的尺寸(例如处于溶胀或未溶胀状态)或通过如上所述的涂层来提供。在框503中,使用光谱光学相干断层扫描来检测第一光谱特征。这可在体内或体外进行。在框505中,追踪在存在识别、结合、亲和或改变粒子的材料的情况下从该粒子的第一光谱特征到第二光谱特征的变化。即,检测粒子的光谱特征的变化。
根据一个示例,可以采用多种方式制备被功能化以在存在特定刺激/生物标记物的情况下变化的粒子。例如,可将酶可裂解(enzyme-cleavable)的肽掺入聚合物粒子的主链中,以促进在特定酶的存在下的降解。
已知酶的表达和活性会随着癌症、疾病、炎症或病理失调而更改。因此,在一个示例中,粒子可被定制为具有被相关联的蛋白水解酶(例如基质金属蛋白酶,MMP)特别地识别的识别元素(例如肽链/接头(linker)、聚合物-肽缀合物)。酶与基于肽的识别元素的后续相互作用导致肽的裂解,进而导致整个粒子的降解,从而更改其光谱特征。
在另一个示例中,可将基于小分子的识别元素掺入粒子主链中以促进在生化物质存在下的溶胀/去溶胀。在该示例中,靶生化物质与识别元素的结合更改了聚合物粒子内的电荷分布或疏水/亲水相互作用,导致渗透溶胀/去溶胀。例如,葡萄糖结合至苯基硼酸衍生物,所述苯基硼酸衍生物共价结合至粒子的聚合物主链。
根据一个示例,利用光谱OCT来识别和监测功能化粒子的独特光学特征使得能够进行体内生物传感和连续监测。如上所述,粒径、形状和成分决定了介电粒子的光学特性,其可通过改变表面特性来进一步调节。各种化学功能化可用于使表面对周围环境中的特定分析物做出响应,从而产生了用于远程生物传感的多功能体内传感器。
如本文所用,功能化粒子是指具有已经针对特定应用进行了功能化的表面的粒子或其他小的局部化物体。功能化包括粒子的表面改变,也就是说,粒子包括芯和涂层,并且如上所述,涂层(至少)可用于在特定生物标记物的存在下做出反应。在另一个示例中,如上所述,粒子的功能化可使得粒子整体上(与仅涂层相对)可在生物标记物的存在下做出反应。例如,如上所述,粒子可溶胀/去溶胀,并且该结构改变可限于涂层,或者可整体发生在粒子上,在这种情况下,粒子可例如由单一材料形成。

Claims (14)
1.一种功能化粒子,其中,所述粒子在所述粒子的第一结构构造中具有第一光谱特征,并且在所述粒子的第二结构构造中具有第二光谱特征。
2.根据权利要求1所述的功能化粒子,其中,所述粒子包括介电芯和围绕所述芯的介电涂层。
3.根据权利要求2所述的功能化粒子,其中,所述粒子的所述第一结构构造包括所述芯和涂层,并且所述粒子的所述第二结构构造包括无所述涂层的所述芯。
4.根据权利要求1所述的功能化粒子,其中,所述粒子的所述第一结构构造包括处于未溶胀或溶胀状态的所述粒子,并且所述粒子的所述第二结构构造包括处于对应的溶胀或未溶胀状态的所述粒子。
5.根据前述权利要求中任一项所述的功能化粒子,其中,所述粒子在所述第一和第二结构构造中具有各自不同的折射率、吸收或偏振。
6.根据前述权利要求中任一项所述的功能化粒子,其中,从所述第一光谱特征到所述第二光谱特征的变化源自所述第一结构构造和所述第二结构构造之间的在所述粒子的所述折射率、吸收和偏振中的一个或多个方面的改变。
7.根据前述权利要求中任一项所述的功能化粒子,其中,所述粒子包括涂覆有第二介电材料的介电粒子芯,所述第二介电材料被构造为在所述材料的存在下降解。
8.一种用于检测功能化粒子的光谱特征的改变的方法,所述方法包括:
提供具有第一光谱特征的粒子;
使用光谱光学相干断层扫描来检测所述第一光谱特征;以及
追踪在存在识别、结合、亲和或改变所述粒子的材料的情况下从所述粒子的所述第一光谱特征到第二光谱特征的变化。
9.根据权利要求8所述的方法,还包括:
检测所述粒子的折射率、吸收和偏振中的一个或多个的变化。
10.根据权利要求8或9所述的方法,其中,提供所述粒子包括:用被构造为在所述材料的存在下降解的第二介电材料涂覆介电粒子芯。
11.一种用于辅助诊断或预后监测的方法,所述方法包括:
提供具有第一光谱特征的粒子;
使用光谱光学相干断层扫描来检测对象中的所述第一光谱特征;以及
追踪在存在识别、结合、亲和或改变所述粒子的材料的情况下从所述粒子的所述第一光谱特征到第二光谱特征的变化。
12.一种功能化粒子用于诊断或预后应用的用途,其中,所述粒子在所述粒子的第一结构构造中具有第一光谱特征,并且在存在识别、结合、亲和或改变所述粒子的材料的情况下在所述粒子的第二结构构造中具有第二光谱特征。
13.根据权利要求12所述的用途,其中,所述材料是其存在会引起所述功能化粒子的折射率、吸收或偏振方面的变化的生物标记物、酶、蛋白质、碳水化合物、小分子药物或核酸。
14.一种用于诊断或预后应用的套件,包括:
功能化粒子,其中,所述粒子在所述粒子的第一结构构造中具有第一光谱特征,并且在所述粒子的第二结构构造中具有第二光谱特征;以及
光谱光学相干断层扫描设备,其被构造为追踪或检测从所述粒子的所述第一光谱特征到第二光谱特征的变化。
