Abstract
本发明公开了一种益生菌包埋颗粒,原料包括益生菌菌粉、浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉、多聚果糖、抗性糊精和磷脂。本发明还提供了制备所述包埋颗粒的方法,包括:(1)将所述益生菌的菌粉作为芯材,用浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉和水进行流化床喷雾包埋得蛋白胶体保护层,干燥后得包埋物A;(2)将包埋物A用多聚果糖、抗性糊精、磷脂和水进行流化床喷雾包埋得多糖保护层,干燥后得益生菌颗粒。本发明的优点在于,获得通过蛋白胶体保护层和多糖保护层保护的益生菌粉,使其具有高耐酸性、高流动性和高分散特性,可室温保藏,货架期稳定,并适用于多种产品的开发与使用。
Description
技术领域
本发明属于食品技术领域,具体为一种益生菌包埋颗粒及其制备方法。
背景技术
2001年FAO和WHO将益生菌定义为“活的微生物,摄入适量对宿主健康产生有益作用的活菌”,多年来研究也发现“活的”益生菌对人体健康并不仅限于肠道健康,还涉及免疫健康及脑健康等。因此益生菌产品中益生菌的稳定性至关重要,同时益生菌顺利到达肠道的能力也至关重要。影响益生菌在储存过程中稳定性的因素主要为产品的属性(水分活度、水分含量、含氧量、pH等)和产品储藏条件(温度、湿度、光照等),其中产品储藏条件我们能通过简单的技术手段加以控制,而其产品属性的控制则需要复杂的加工工艺实现。
通常情况下各种益生菌被加工为粉状产品,主要为降低水分活度对于益生菌的影响,降低其代谢活动,使益生菌处于休眠/或半休眠状态而延长益生菌的货架期。但是常规的益生菌粉剂多数仅为简单的冻干粉,需要冷藏或冷冻储藏以保持其活力,且这些产品的耐酸性主要靠益生菌本身的耐酸特性来实现,一些耐酸性不强的菌株很难通过胃环境实现大量活菌到达肠道。
目前涉及益生菌包埋的专利主要为:
CN 109464425 A公开了一种益生菌包埋颗粒及其制备方法,所述益生菌包埋颗粒包括益生菌核芯和所述益生菌核芯外的三个包埋层,所述包埋层从内到外依次为:淀粉保护层、薄膜包衣隔离层和肠溶包衣层。且所述方案包埋后活菌存活率为39~49%。
CN 107788534 A公开了一种可以避免肠道内多菌共生、益生菌遭受胃酸、胆盐攻击,提高多菌共生益生菌活菌数的通过包埋技术制备的多菌共生益生菌微胶囊。
上述发明涉及到益生菌包埋技术,但是CN 109464425 A益生菌包埋颗粒主要通过添加薄膜包衣及肠溶衣来解决益生菌耐受胃酸环境,到达肠道的实际问题,但是所用材料涉及化学试剂的使用,并不都是天然的食品原料实现的以上功效。CN 107788534 A则是利用β环糊精(β-cyclodextrin)作为载体,实现益生菌的简单包埋,很难实现益生菌在胃内的高效通过率。
发明内容
为解决现有技术中益生菌粉货架期短,耐酸性弱的问题,本发明提供了一种益生菌包埋颗粒及其制备方法。其实现的目的为,获得通过蛋白胶体保护层和多糖保护层保护的益生菌粉,使其具有高耐酸性、高流动性和高分散特性,可室温保藏,货架期稳定,并适用于多种产品的开发与使用。
为了实现上述目的,本发明提供以下技术方案:本发明提供的一种益生菌包埋颗粒,原料包括益生菌菌粉、浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉、多聚果糖、抗性糊精和磷脂,浓缩乳清蛋白、酪蛋白酸钠和圆苞车前子粉的总用量为益生菌粉的5%-15%,多聚果糖、抗性糊精和磷脂的总用量为益生菌粉的10%-20%,所述百分比为质量百分比;浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉的质量比为:0.2~0.4:0.1~0.3:0.01~0.03,多聚果糖、抗性糊精、磷脂的质量比为:0.1~0.5:0.1~0.5:0.002~0.005。
进一步的,所述益生菌菌粉包括动物双歧杆菌、乳双歧杆菌、短双歧杆菌、鼠李糖乳杆菌、干酪乳杆菌、嗜酸乳杆菌、副干酪乳杆菌、植物乳杆菌、发酵乳杆菌和肠膜明串珠菌中任意一种或一种以上。
进一步的,所述浓缩乳清蛋白为浓缩乳清蛋白WPC70、浓缩乳清蛋白WPC80、WPI中任意一种。
本发明还提供了制备所述益生菌包埋颗粒的方法,包括如下步骤:
(1)将所述益生菌的菌粉作为芯材,用浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉和水进行流化床喷雾包埋得蛋白胶体保护层,干燥后得包埋物A;
(2)将包埋物A用多聚果糖、抗性糊精、磷脂和水进行流化床喷雾包埋得多糖保护层,干燥后得益生菌颗粒。
进一步的,所述步骤(1)中浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉和水的质量比为:0.2~0.4:0.1~0.3:0.01~0.03:1。
进一步的,所述步骤(2)中多聚果糖、抗性糊精、磷脂和水的质量比为:0.1~0.5:0.1~0.5:0.002~0.005:1。
进一步的,所述步骤(1)中水为去离子水。
进一步的,所述步骤(1)中包埋物A干燥后的水分含量≤5%。
进一步的,所述步骤(1)和步骤(2)中所述的流化床喷雾干燥进风温度均为30-60℃,出风温度均为25-45℃。
进一步的,所述步骤(2)中干燥后益生菌颗粒的水分含量为1.5~2.5%。
进一步的,所述步骤(1)中将浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉和水均质后再进行流化床喷雾,所述均质为二级均质,一级均质压力10-20MPa,二级均质压力2-3MPa,均质温度45-55℃。
进一步的,所述步骤(2)中将多聚果糖、抗性糊精、磷脂和水均质后再进行流化床喷雾,所述均质为二级均质,一级均质压力18MPa,二级均质压力2-3MPa,均质温度55-60℃。
本发明采用上述技术方案,包括以下有益效果:本发明的益生菌颗粒实现了益生菌粉的高流动性,高分散特性和低吸湿性,所开发的产品可室温保藏,货架期稳定,耐酸性强,并适用于多种产品的开发与使用。本发明的两步流化喷雾干燥技术实现了益生菌的分层分功能包埋,乳清蛋白和圆苞车前子粉形成蛋白胶体包埋层在益生菌食用时可形成胶束有效提高益生菌的耐酸特性,而多聚果糖和抗性糊精形成的包埋层则提高了益生菌颗粒的流动性,降低了其吸湿性和结块性。

下面通过实施例的方式进一步说明本发明,但并不因此将本发明限制在所述的实施例范围之中。下列实施例中未注明具体条件的实验方法,按照常规方法和条件,或按照商品说明书选择。所用的原料或试剂都是市售可得。
实施例一:
本发明提供的一种益生菌包埋颗粒,原料包括益生菌菌粉、浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉、多聚果糖、抗性糊精和磷脂,浓缩乳清蛋白、酪蛋白酸钠和圆苞车前子粉的总用量为益生菌粉的5%,多聚果糖、抗性糊精和磷脂的总用量为益生菌粉的20%,所述百分比为质量百分比;浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉的质量比为:0.2:0.3:0.01,多聚果糖、抗性糊精、磷脂的质量比为:0.5:0.1:0.002。
所述益生菌菌粉包括动物双歧杆菌、乳双歧杆菌、短双歧杆菌、鼠李糖乳杆菌、干酪乳杆菌、嗜酸乳杆菌、副干酪乳杆菌、植物乳杆菌、发酵乳杆菌和肠膜明串珠菌按任意配比混合菌种;

所述益生菌粉为乳双歧杆菌B420菌粉,总活菌数为5×1010CFU/g。
所述乳清蛋白为浓缩乳清蛋白WPC80。

所述益生菌粉为鼠李糖乳杆菌LGG和副干酪乳杆菌LC2W按活菌数1:1制成的混合菌粉,总活菌数为3.8×1010CFU/g;
所述乳清蛋白为WPI;
所述益生菌颗粒制备方法包括如下步骤,(1)将所述益生菌的菌粉作为芯材,用浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉溶于水经均质后进行流化床喷雾包埋得蛋白胶体保护层,干燥后得包埋物A;

本发明公开的一种益生菌包埋颗粒,原材料包括益生菌粉,蛋白胶束层,多糖保护层。
所述益生菌粉为动物双歧杆菌Bb-12、乳双歧杆菌B420按活菌数1:1制成的混合菌粉,总活菌数为2.8×1010CFU/g;
所述的蛋白胶束层指由浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉溶于水经均质后喷雾包埋制成的包埋层。
所述的多糖保护层由多聚果糖、抗性糊精和磷脂溶于水后经喷雾包埋制成的包埋层。
所述益生菌颗粒制备方法包括如下步骤,(1)将所述益生菌的菌粉作为芯材,用浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉溶于水经均质后进行流化床喷雾包埋得蛋白胶体保护层,干燥后得包埋物A;

以下通过列举部分与未经包覆的益生菌颗粒效果对比试验例,用以说明本发明技术方案的技术效果:
效果试验例1:
对实施例1-4的益生菌颗粒进行铝箔袋包装,同时也对未经造粒的对应于实施例1-4的益生菌粉进行铝箔袋包装,并进行加速试验。加速条件:温度37℃,相对湿度75%,存储时间3个月,分别于加速起始0月,加速1个月,加速2个月和加速3个月进行菌粉活菌数的检测。并计算存活率,存活率指存储一段时间后活菌数的检测值与起始值之比,用百分比表示;在进行益生菌活菌数检测前,先进性粉体流动性和分散性的观察。观察方法:轻轻取出(不进行挤压或强烈震荡)益生菌粉或包埋颗粒,打开封口将益生菌至于白色瓷盘内,观察其有无团块,轻轻晃动瓷盘观察其分散性和流动性。益生菌存活率具体结果见表1。

加速试验益生菌存活情况见表1,由表1的结果可知,益生菌经过包埋益生菌存活率得到了极大的提高,加速条件下,至加速3个月不同益生菌的存活率比未加工前提高了12-15%,所有益生菌包埋颗粒的益生菌存活率都高于57%,甚至达到75%;常温条件下,益生菌包埋颗粒的益生菌存活率在78-88%,远高于对应未经包埋的益生菌粉的存活率(52-75%)。说明该加工工艺的益生菌颗粒稳定性极好,货架期稳定。
另外,通过跟踪观察的益生菌粉或颗粒的分散性及流动性结果也显示,常温下所有益生菌包埋颗粒及所有对应未经包埋的益生菌粉都无团块出现,分散性和流动性都较好。但是在加速条件下,实施例3的原菌粉在加速2个月时有小团块出现,分散性有所降低;实施例4的原菌粉在加速3个月时有小团块出现,分散性有所降低;其他益生菌颗粒及益生菌粉都无出现明显的分散性或流动性下降。以上说明,益生菌包埋颗粒,不但提高了益生菌粉的稳定性,而且提高了其流动性和分散性。
效果试验例2:
对实施例1-4的益生菌颗粒,及未经造粒的对应于实施例1-4的益生菌粉进行吸湿性试验。试验条件:将试验样品5g放置于直径为9cm的培养皿内,盖盖放置于温度30℃,相对湿度50%,恒温恒湿培养箱内12h,分别于放之前记录培养皿、样品质量和放置12h后称量其质量。计算吸湿率,吸湿率为样品增重的质量/样品的原质量,用百分比表示,具体结果见表。

由表2的吸湿性试验结果可知,益生菌经过包埋,益生菌颗粒的吸湿性急剧下降,益生菌颗粒的吸湿率13-20%远低于对比益生菌原粉的吸湿率25-37%,特别是实施例1的益生菌包埋颗粒吸湿率仅为原菌粉吸湿率的38%,说明该加工工艺的益生菌包埋颗粒吸湿率极低,易于工业加工生产。
效果试验例3:
对实施例1-4的益生菌颗粒,及未经造粒的对应于实施例1-4的益生菌粉进行耐酸性试验。试验方法:用盐酸,加水稀释配制pH值为2的盐酸溶液,并加入1%的胃蛋白酶,混匀,即得模拟胃酸。然后将益生菌包埋颗粒或益生菌粉加入到(活菌数接种量3×107CFU/ml,对数值为7.5),模拟胃酸里,37℃水浴处理2h,进行活菌数的检测。活菌数检测结果以对数值表示,结果以数量级的下降并计算存活率,存活率指经过模拟胃酸处理后活菌数的检测值与起始值之比,用百分比表示,具体结果见表3。

益生菌颗粒的耐酸性结果表明,经过包埋的益生菌其耐酸性都比未包埋的对应菌株耐酸性高,经过包埋的益生菌经过模拟胃酸2h处理后,实施例1-4的益生菌包埋颗粒活菌数减少到约5lg(cfu/ml),而未经包埋的益生菌粉活菌数都低于对应实施例活菌数约1个数量级。说明经过本发明的益生菌包埋颗粒,具有良好的耐酸性,经过包埋的益生菌穿过胃肠道的生存能力得到了提高。
效果试验例4:
用实施例1,实施例2的益生菌颗粒添加到调制乳粉中,分别制成调制乳粉1和调制乳粉2。考察益生菌在乳粉中的分散性及稳定性。分散性考察方法:将益生菌包埋颗粒按5×106CFU/g通过简单的干混添加到调制乳粉内,并分装200g/袋,通过抽样检测(抽样点包装前段、包装中段,包装后段各3个样)益生菌在不同包装内的活菌数来评估益生菌包埋颗粒在奶粉中的分散性。稳定性检测:通过将乳粉置于加速条件下观察,加速条件37℃,3个月,然后测其活菌数。具体结果见表4。

将益生菌包埋颗粒应用于调制乳粉,结果表明,益生菌包埋颗粒可以很好地分散于乳粉内,并具有良好的稳定性。益生菌包埋颗粒应用于乳粉内的均匀度分析显示,两种益生菌包埋颗粒的相对标准偏差都小于等于6%。并且含益生菌包埋颗粒的乳粉,其益生菌存活量较高,经过3个月的加速试验,益生菌存活保持数量级不变,且存活率高于50%。说明本发明的益生菌包埋颗粒,适用于调制乳粉,并表现为良好的分散性和稳定性。

1.一种益生菌包埋颗粒,其特征在于,原料由益生菌菌粉、浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉、多聚果糖、抗性糊精和磷脂组成,浓缩乳清蛋白、酪蛋白酸钠和圆苞车前子粉的总用量为益生菌粉的5%-15%,多聚果糖、抗性糊精和磷脂的总用量为益生菌粉的10%-20%,所述百分比为质量百分比;浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉的质量比为:0.2~0.4:0.1~0.3:0.01~0.03,多聚果糖、抗性糊精、磷脂的质量比为:0.1~0.5:0.1~0.5:0.002~0.005;
所述的益生菌包埋颗粒的制备方法包括如下步骤:
(1)将所述益生菌的菌粉作为芯材,用浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉和水进行流化床喷雾包埋得蛋白胶体保护层,干燥后得包埋物A;所述包埋物A干燥后的水分含量≤5%;

所述步骤(2)中将多聚果糖、抗性糊精、磷脂和水均质后再进行流化床喷雾,所述均质为二级均质,一级均质压力18MPa,二级均质压力2-3MPa,均质温度55-60℃。
2.根据权利要求1所述的益生菌包埋颗粒,其特征在于,所述益生菌菌粉为动物双歧杆菌、乳双歧杆菌、短双歧杆菌、鼠李糖乳杆菌、干酪乳杆菌、嗜酸乳杆菌、副干酪乳杆菌、植物乳杆菌、发酵乳杆菌和肠膜明串珠菌中任意一种或一种以上。
3.根据权利要求1所述的益生菌包埋颗粒,其特征在于,所述浓缩乳清蛋白为浓缩乳清蛋白WPC70、浓缩乳清蛋白WPC80、WPI中任意一种。
4.根据权利要求1所述的益生菌包埋颗粒,其特征在于,所述步骤(1)中浓缩乳清蛋白、酪蛋白酸钠、圆苞车前子粉和水的质量比为:0.2~0.4:0.1~0.3:0.01~0.03:1。
5.根据权利要求1所述的益生菌包埋颗粒,其特征在于,所述步骤(2)中多聚果糖、抗性糊精、磷脂和水的质量比为:0.1~0.5:0.1~0.5:0.002~0.005:1。
6.根据权利要求1所述的益生菌包埋颗粒,其特征在于,所述步骤(1)中水为去离子水。
7.根据权利要求1所述的益生菌包埋颗粒,其特征在于,所述步骤(1)和步骤(2)中所述的流化床喷雾干燥进风温度均为30-60℃,出风温度均为25-45℃。
