本发明涉及生物医药技术领域,具体是一种快速判断样本离体时间试剂及方法。
背景技术
离体作为生物学用语在现代语文中常被使用,指的是为了各种研究目的而把生物体的一部分摘出和游离于生物体外的状态。与此相反,把处于“生物体内”的自然未动的状态则称为“生物体内”的“在体”。例如把摘出后的蛙心脏放在任氏液中观察其博动状况,把摘出的组织放在特定培养基上培养,即体外培养等。
发明内容
本发明提供快速判断样本离体时间试剂及方法,用以解决问题样本离体时间难以精确检测的弊端。
本发明的技术方案如下:
一种快速判断样本离体时间试剂,其特征在于其为包含硫化铵或氰化钾的溶液。
一种快速判断样本离体时间的方法,其特征包括如下步骤:取离体组织的抗凝血在小三角瓶中摇,如血很快变为鲜红,表明血中含有较多还原血红蛋白;如血不能变红,则可肯定血中仍有其他异常血红蛋白;
肉眼观察时高铁血红蛋白呈现巧克力样的棕褐色;硫化血红蛋白呈蓝褐色;血红蛋白M则因生成高铁血红蛋白而呈巧克力棕褐色;
以蒸馏水稀释血液10~20倍,加入硫化铵或1%氰化钾数滴,如为高铁血红蛋白则由棕褐色变为鲜红色;如时间过久,则变为蓝褐色,即高铁血红蛋白-氧合血红蛋白-硫化血红蛋白);如颜色不变鲜红色则为硫化血红蛋白。
还包括如下步骤:
分光镜下观察高铁血红蛋白之吸收光带在波长618~630nm处,硫化血红蛋白在波长607~620nm处;加入硫化铵或氰化钾溶液后高铁血红蛋白之吸收光带消失,而硫化血红蛋白因硫化血红蛋白一旦生成即不能再还原其所吸收光带不变;依据多波长吸光度校正的方法,即可以计算出硫化血红蛋白的含量,进而推算出样本离体时间。
有益效果:
本发明的快速判断样本离体时间试剂及方法检测手段流程可控,可实现精确检测,具有成本低,实施方便的优点。
