Abstract
The invention discloses application of a composition of donkey-hide gelatin and new donkey-hide gelatin in preparation of an antioxidant drug, the experimental method of the invention adopts 24hpf (duct post fertilization) skin fluorescence transgenic zebra fish as an experimental animal, the drug and an embryo are incubated for 24h, and after the incubation is finished, the change of the number of skin fluorescence points is observed under a fluorescence microscope. Experiments prove that the donkey-hide gelatin and new donkey-hide gelatin composition has a reversing effect on reduction of skin fluorescent spots of the zebra fish induced by metronidazole, and both the donkey-hide gelatin and the new donkey-hide gelatin composition have antioxidant activity under the conditions of proper action concentration and proportion. Experiments show that the composition of the donkey-hide gelatin and the new donkey-hide gelatin can be used for preparing antioxidant medicaments; in particular to a medicine for preparing diseases related to cell oxidation caused by metronidazole.
Description
Technical Field

Background

Detailed Description
Example (b): evaluation experiment of antioxidant activity of donkey-hide gelatin and novel donkey-hide gelatin composition
The experimental method used in this example was:
the skin fluorescent zebra fish which develops for 24h is taken as an experimental animal, and the outer layer of the embryo of the zebra fish is stripped off by using 1mg/mL pronase. Randomly distributing embryos without the egg membranes, setting 2 compound holes for each group of experiments with 10 embryos, and averaging the experimental results after each group of experiments are repeated for three times. All of the above embryos were placed in a constant temperature incubator at 28.5 ℃, treated with each experimental solution in table 1 for 24 hours, anesthetized with ethyl m-aminobenzoate mesylate (MS222), and photographed by observation under a fluorescence microscope. The number of skin fluorescent spots of the zebra fish was counted using Imagepro-plus software, and the data were statistically analyzed using GraphPad software to evaluate the antioxidant activity of the samples.
The experimental apparatus used in this example includes: model SZX16 fluorescent microscope and DP2-BSW image acquisition system (Olympus, Japan); an inverted microscope model X51 (Olympus, japan); forma 3111 type water jacket CO2Incubators (Forma corporation, usa); zebra fish breeding equipment (Beijing Aisheng science and technology company).
The experimental drugs used in this example include: colla Corii Asini and new colla Corii Asini samples are provided by Fupai colla Corii Asini company; metronidazole and vitamin C were purchased from shanghai bio-chemical company; pronase and ethyl aminobenzoate mesylate were both obtained from sigma. Grinding a donkey-hide gelatin sample into powder, and dissolving the powder with sterile water; both metronidazole and vitamin C are dissolved in sterile water.
The experimental animals used in this example include: the skin fluorescent transgenic zebra fish and the AB line zebra fish are provided by a zebra fish drug screening platform of biological research institute of academy of sciences in Shandong province.
The specific implementation conditions are as follows:

As can be seen from the experimental results in table 1 and fig. 1, the zebra fish of the control group 1 and the zebra fish of the control group 2 are normally developed, but the number of fluorescent spots on the skin of the zebra fish of the control group 2 is obviously reduced compared with the control group 1, which obviously indicates that under the influence of metronidazole, the cells on the skin of the zebra fish are oxidized; the zebra fish in examples 1-7 developed normally, but the number of skin fluorescent spots was increased compared with the control group 2, indicating that the combination of donkey-hide gelatin and new donkey-hide gelatin has a significant antioxidant effect.
From the experimental data in table 1, in the concentration range of 10-100 μ g/mL, as the concentration of the donkey-hide gelatin and the new donkey-hide gelatin composition increases, the number of the fluorescent spots on the skin of the zebra fish also increases, and shows a good increasing relationship, as shown in fig. 1. The reversing effect on the reduction of the number of fluorescent spots on the skin of the zebra fish can be achieved when the mass ratio of the donkey-hide gelatin to the new donkey-hide gelatin is 1: 10-10: 1, and the reversing effect is better especially when the mass ratio of the donkey-hide gelatin to the new donkey-hide gelatin is 1: 1.
Compared with the comparative example 1, the comparative example 2 and the example 3, the reversing effect of the combination of the donkey-hide gelatin and the new donkey-hide gelatin on the reduction of the skin fluorescent spots of the zebra fish is far better than the reversing effect of the combination of the donkey-hide gelatin and the new donkey-hide gelatin on the reduction of the skin fluorescent spots of the zebra fish.
In conclusion, the combination of the donkey-hide gelatin and the new donkey-hide gelatin can be used for preparing antioxidant medicaments; in particular to a medicine for preparing diseases related to cell oxidation caused by metronidazole.
