本发明涉及2‑巯基吡啶‑N‑氧化物三价钴盐配合物的抗菌应用。本发明公开2‑巯基吡啶‑N‑氧化物三价钴盐配合物对铜绿假单胞菌、大肠杆菌和枯草芽孢杆菌均具有优异的抗菌活性。该抗菌剂制备工艺简单,反应条件温和,产率高,具有重大应用价值。

技术领域
本发明属于生物领域,特别涉及2-巯基吡啶-N-氧化物三价钴盐配合物的抗菌应用。
背景技术
随着人民生活水平的不断提高,人们对健康、卫生的生活环境也有了更高的要求,抗菌材料的需求越来越大。抗菌材料的核心是抗菌剂,一种具有抑菌杀菌性能的物质。抗菌剂可分成无机抗菌剂和有机抗菌剂。以金属银离子为代表的无机抗菌剂,抗菌杀菌能力强,但具有选择性差、容易变质的缺点。以抗生素为代表的有机抗菌剂,具有杀菌选择性好的优点,但大量使用后,细菌容易产生耐药性。为此,我们考虑制备无机-有机复合抗菌剂,将无机抗菌剂和有机抗菌剂的优点结合起来。
2-巯基吡啶-N-氧化物钠盐本身就是一种高效、安全、广谱杀菌剂,在农业上用于防治苹果疮痂病和卷叶病,在食品上对酵母菌和霉菌有抑制作用,在医学上用于治疗脂溢性皮炎、花斑癣等体表性疾病。而钴元素是人体内重要的微量元素,例如它是维生素B12组成部分,能促使血红蛋白的合成等。将2-巯基吡啶 -N-氧化物和钴金属配位结合起来,该配合物对人体和环境都是友好的。
尽管2-巯基吡啶-N-氧化物和钴金属离子的配位反应已有报道,但其在抗菌材料方面的应用研究未见报道。值得说明的是,钴有二价和三价两种价态,本发明公开的是三价钴盐配合物。该抗菌剂配合物制备工艺简单,反应条件温和,产率高,具有重大应用价值。

本发明提供了2-巯基吡啶-N-氧化物三价钴盐配合物的抗菌应用。
具体技术方案如下:
2-巯基吡啶-N-氧化物三价钴盐配合物的抗菌应用,其中2-巯基吡啶-N-氧化物三价钴盐配合物具有如下分子结构:
所述抗菌对象包括铜绿假单胞菌、大肠杆菌和枯草芽孢杆菌;2-巯基吡啶-N-氧化物三价钴盐配合物能高效抑制铜绿假单胞菌、大肠杆菌和枯草芽孢杆菌的生长。
所述2-巯基吡啶-N-氧化物三价钴盐配合物浓度优选为0.3~1.36mg/mL。

具体实施方式
以下将结合具体实施实施例对本发明作进一步描述,但本发明的实施方式不限于此。
下列实施例中未注明具体条件的实验方法,通常按照常规条件。所用试剂均为市售普通试剂,2-巯基吡啶-N-氧化物三价钴盐配合物溶液配置所用溶剂均为二甲亚砜(DMSO)。
本发明中涉及的2-巯基吡啶-N-氧化物三价钴盐配合物可以参考并改进文献方法进行合成(D.X.West, C.A.Brown,J.P.Jasinski,J.M.Jasinski,R.M.Heathwaite,D.G.Fortier,R.J.Staples,and R.J.Butcher,Crystal structures of the cobalt(III),nickel(II),copper(II),and zinc(II)complexes of 2-thio-6-picoline N-oxide,Journal of Chemical Crystallography,1998,28(12):853-860.),具体过程可参见实施例,其合成反应方程如下所示:
实施例1:
2-巯基吡啶-N-氧化物三价钴盐配合物的合成:
称取0.586g的六水合氯化钴于烧杯中,加适量水溶解,得到桃红色溶液。然后取1.85mL的2-巯基吡啶-1-氧化钠水溶液(质量百分浓度40%)加入上述溶液,立即产生黄色沉淀,离心,分走水相,沉淀用四氢呋喃溶解,得墨绿色溶液,将溶液转移到试管里敞开口静置,一周内会慢慢析出墨绿色三价钴抑菌剂晶体,收集,干燥备用。
实施例2:
2-巯基吡啶-N-氧化物三价钴盐配合物抑制铜绿假单胞菌生长实验:
采用双层平板法。在一个经过高温灭菌处理的培养皿上倒入10mL加热融化的LB固体培养基,冷却后,将直径为8mm的牛津杯用火灼烧底部后固定在凝固的固体培养基上,每个培养皿均匀放置三个牛津杯。再在该平板中倒入10mL融化的固体培养基,凝固后,将牛津杯取出,制成有三个直径为7mm的圆孔的双层平板。用移液枪取200μL的培养好的铜绿假单胞菌菌液到每个平板中,用涂布棒涂布均匀(灼烧后冷却使用)。在平板圆孔中加入配置好的浓度为1.36mg/mL的2-巯基吡啶-N-氧化物三价钴盐配合物的DMSO溶液100μL,冰箱冷藏2h后37℃培养箱培养24h,观察抑菌圈大小,测得抑菌圈半径为13mm。
实施例3:
2-巯基吡啶-N-氧化物三价钴盐配合物抑制大肠杆菌生长实验:
具体实施过程跟实施例2相似,只是用培养好的大肠杆菌菌液代替实施例2中的铜绿假单胞菌菌液,测得抑菌圈半径为15mm。
实施例4:
2-巯基吡啶-N-氧化物三价钴盐配合物抑制枯草芽孢杆菌生长实验:
具体实施过程跟实施例2相似,只是用培养好的枯草芽孢杆菌菌液代替实施例2中的铜绿假单胞菌菌液,测得抑菌圈半径为15mm。
为了比较不同金属离子对配合物抗菌性能的影响,我们还合成了2-巯基吡啶-N-氧化物镍和银的配合物,在同等条件下比较他们的抗菌性能。
对比例1:
2-巯基吡啶-N-氧化物镍配合物抑制铜绿假单胞菌生长实验:
具体实施过程跟实施例2相似,只是用镍配合物代替实施例2中的三价钴配合物,测得抑菌圈半径为 11mm。
对比例2:
2-巯基吡啶-N-氧化物银配合物抑制铜绿假单胞菌生长实验:
具体实施过程跟实施例2相似,只是用银配合物代替实施例2中的三价钴配合物,测得抑菌圈半径为 8mm。
实施例2-4和对比例1-2的主要结果列于表中。

注:配合物浓度为1.36mg/mL
由表可见,2-巯基吡啶-N-氧化物三价钴盐配合物抗菌性能最好;金属离子对配合物性能影响很大,说明配合物的抗菌性能是2-巯基吡啶-N-氧化物配体和金属离子协同作用结果,并不是简单的叠加,因为单独钴离子、镍离子是没有抗菌活性的。
