(2) uniformly mixing 0.010g of silicon dioxide, 0.30g of isolated soy protein and 0.30g of resistant dextrin in a three-dimensional mixer, and then adding 1.34g of hydrolyzed yolk powder and 0.34g of whole milk powder to obtain first-grade premixed powder;
(3) adding 4.80g of milk mineral salt into the first-stage premixed powder, performing second-stage mixing, and uniformly mixing to obtain second-stage premixed powder which is named as mixed powder A;
(4) with vitamin D30.0056g of microcapsule powder (total amount is 1) and 1 st mixing with the same weight of the powder A; on the basis, adding the mixed powder A with the same weight for the 2 nd mixing; and so on. Mixing the two powders completely to obtain total mixed powder;
(5) taking 0.60g of sodium carboxymethylcellulose and 0.40g of methylcellulose as a total amount of 1, and taking materials with equal weight from the total mixed powder to mix with the mixed powder for the 1 st time; on the basis, adding the 2 nd mixing with the total mixed powder with the same weight; and so on. Mixing the two materials completely to obtain powder to be filled;
(6) and (3) putting the powder to be filled into a hard capsule machine, and filling No. 5 hard capsules with 0.08g per capsule to obtain the hard capsule health food.
Example 12
In this example, the health food for increasing bone density in examples 8, 10 and 11 is taken as an example to study the quality of the health food of the present invention.
In this example, 3 batches of health food preparations for increasing bone density were prepared in different formulations as described in examples 8, 10 and 11. The powder prepared in example 8 was used for organoleptic examination; each dosage form was randomly tested for functional active ingredient in 1 lot each.
(1) Sensory requirements
The sensory requirement detection results of the health food for increasing bone density by the powder of the invention are shown in table 1:

(2) Functional active ingredient

The results show that the health food prepared by the mixing preparation method of the invention with the equivalent incremental mode and the multiple amplification has stable quality, and the materials with low proportion in the prescription can be uniformly mixed with other materials, thereby ensuring the uniformity of the health food in the prescription and the efficacy of the health food.
Example 13
This example takes the health food for increasing bone density in example 8 as an example, and studies the effect of the health food of the present invention in promoting bone growth.
Test materials: SD rats with weaning period of about 4 weeks are selected, cleaned, female and 70-90g in body weight. The temperature of the breeding room is 20-25 ℃, and the relative humidity is 40-60%. The feed is composed of protein, fat and carbohydrate. The drinking water is deionized water. After the rats acclimatized, the rats were fasted for 12 hours, weighed, measured for body length, and randomly bred in 4 cages, 30 rats/cage.
Dose design: the feed of rats in the blank control group is not added with any other substances. Calcium carbonate control rats were fed 580 mg/kg. Referring to the pre-test dose, the lower and upper sample dose groups were 205.6, 308.5mg/kg, respectively, which is equivalent to 0.67, 1 times the recommended dose for human.
The test method comprises the following steps: the time and mode of feeding the rats are shown in table 3. Feed and deionized water served as the rat survival base. The body weight was weighed every 10 days, 10 rats in each group were sacrificed after measuring the body length, and the bones were examined.
Statistical treatment: all data are expressed as mean ± standard deviation. The differences among the groups are processed by SPSS software and single-factor variance analysis, wherein p is less than 0.05 and has statistical significance, and p is less than 0.01 and has significant statistical significance.

And (3) test results:
(1) the low and high dose groups of rats extended bones, inhibited osteoclast differentiation and promoted osteogenic growth compared to the blank control group in all periods of day 10, day 20 and day 30. The bone elongation of the rats in the high dose group was increased by 15% on day 30, and the difference was statistically significant (p < 0.01).
(2) The cell number proliferation rate of osteoblasts was increased to 105% for the low dose group rats on day 10, day 20, and day 30, compared to calcium carbonate control group rats, for which the difference was statistically significant (p < 0.05); the bone elongation is improved by 3.5 percent, and the difference has statistical significance (p is less than 0.05); the activity of bone alkaline phosphatase in serum is improved by 5%, and the difference has statistical significance (p is less than 0.05).
(3) The cell number proliferation rate of the osteoblasts of the rats of the high dose group increased to 160% over the entire period of days 10, 20 and 30, compared to the calcium carbonate control group rats on day 30, the difference being statistically significant (p < 0.01); the bone elongation is improved by 15 percent, and the difference has statistical significance (p is less than 0.01); the activity of bone alkaline phosphatase in serum is improved by 16%, and the difference has statistical significance (p is less than 0.01). Calcification of the bone joints was promoted as observed by electron microscopy.
(4) The rats orally take the sample of example 8, which is equivalent to 0.67 and 1 time of the dose recommended by human bodies, and the final data shows that both dose groups can promote the growth of the bones of the rats, and compared with the low dose group, the effect of the high dose group is more obvious. Indicating that the test result is positive.
And (4) test conclusion:
according to the test results, the novel composition for increasing bone density has the effect of promoting bone growth.
Example 14
This example takes the health food for increasing bone density in example 8 as an example, and studies the effect of the health food of the present invention in promoting bone growth. Experiments are designed according to the scheme of 'testing method for increasing bone mineral density' in 'health food testing and evaluation technical Specification' (2003 edition).
Test materials: SPF grade SD male healthy rats weighing 60-75g were provided by southern university of medical laboratory animal center. The laboratory is a shielding environment, the temperature is 20-26 ℃, and the relative humidity is 40-70%. The pellet feed and the basic feed powder are provided by the experimental animal center of Guangdong province. The animals were randomly fed in 5 dose groups of 10 animals/group.
The test dose is as follows: the low, medium and high sample dosage groups are respectively set as 0.25, 0.50 and 1.50g/kg BW, which are 5, 10 and 30 times of the recommended dosage of human body, and a low calcium control group (150mg/100g feed) and a calcium carbonate control group (0.58g/kg BW, calcium carbonate control group with the same level as the high dosage calcium) are additionally set.
The test method comprises the following steps: the experimental animals were fed daily ad libitum with sufficient feed and water for each mouse, and with deionized water to avoid calcium gain from the water for 3 months.
Observation indexes are as follows: general conditions (general physiological signs, appearance, behavior, stool and urine, fur, etc. in rats); femur weight (animals sacrificed after feeding for 3 months, right femur was dissected off, baked in 105 ℃ oven to constant weight, bone dry weight was weighed); bone density (measured using a dual-energy X-ray bone densitometer for the center and distal end of the left femur); bone calcium content (measured by inductively coupled plasma spectrometer).
Statistical treatment: using SPSS software to process the anova, firstly carrying out anova test according to an anova program, wherein the anova is uniform, calculating an F value, and the F value is less than F0.05, and concluding: the difference between the average numbers of all groups is not significant; f value is more than or equal to F0.05, and statistics is carried out by a pairwise comparison method of mean values between a plurality of experimental groups and a control group; carrying out appropriate variable conversion on the data which are not normal or uneven in variance, and counting by using the converted data after the requirements of normal or uniform variance are met; if the variable still does not reach the goal of being normal or uniform in variance after conversion, the statistics is carried out by using the rank sum test.
The test results are shown in table 4:
(1) general conditions are as follows: the physiological signs, behaviors, stool and urine, fur and the like of the rats are not abnormal generally.
(2) Left femoral bone density: compared with the low-calcium control group, the bone density of the distal end and the bone density of the midpoint of the left femur of each dose group of rats are increased, and the difference has statistical significance (P is less than 0.05); and compared with the calcium carbonate control group, the high-dose group has no statistical significance on the difference between the bone density at the far end of the left femur and the bone density at the midpoint (P is more than 0.05). The test sample is proved to have positive bone density test result.
(3) Calcium content of right femur: compared with the low-calcium control group, the weight of the right femur of each dose group of rats is increased, and the difference is statistically significant (P is less than 0.05); and the weight difference of the right femur in the high dose group compared with the calcium carbonate control group is not statistically significant (P > 0.05). Compared with the low-calcium control group, the calcium content of the right femur in the medium and high dose groups is increased, and the difference has statistical significance (P is less than 0.05); and compared with the calcium carbonate control group, the calcium content difference of the right femur has no statistical significance (P is more than 0.05). The test result of the bone calcium content of the test sample is positive.

The composition for increasing bone density according to claim 1, wherein the following raw materials are included per 100 formulation units: 4.32-1.00 kg of milk mineral salt, 1.20-275.60 g of hydrolyzed yolk powder and vitamin D30.005-1.16 g of micro-capsule powder and 0.15-383.30 g of whole milk powder.
3. Use of the bone density increasing composition according to claim 1 or 2 for the preparation of a health food for increasing bone density.
4. A health food for increasing bone density, comprising the composition for increasing bone density according to claim 1 and an auxiliary material acceptable in the field of health foods.
5. The health food for increasing bone density according to claim 4, wherein the auxiliary materials acceptable in the field of health foods comprise solid preparation auxiliary materials and/or adhesives.
6. The health food for increasing bone density according to claim 5, comprising the following raw materials per 100 formulation units: 4.32-1.00 kg of milk mineral salt, 1.20-275.60 g of hydrolyzed yolk powder and vitamin D30.005 g-1 g of microcapsule powder.16g, 0.15-383.30 g of whole milk powder, 1.52-23.08 g of solid preparation auxiliary material and/or 0.02-2.00 g of adhesive.
7. The health food for increasing bone density according to claim 5 or 6, wherein the solid preparation excipient is at least one selected from the group consisting of soybean protein isolate, maltodextrin, corn starch and resistant dextrin; the adhesive is selected from at least one of sodium carboxymethylcellulose, hydroxypropyl cellulose, hydroxypropyl methyl cellulose, starch slurry, purified water, methyl cellulose, ethyl cellulose, hydroxypropyl methyl cellulose, polyvinylpyrrolidone and sucrose.
8. The health food for increasing bone density according to any one of claims 4 to 6, wherein the health food is in the form of powder, tablet or capsule.
9. The method for preparing a health food for increasing bone density according to any one of claims 4 to 8, wherein the raw materials are mixed in an equivalent incremental multiple amplification manner.
10. The manufacturing method according to claim 9, wherein the raw materials are mixed using at least one of a three-dimensional mixer, a double cone mixer, a V-type mixer, a two-dimensional mixer, a spray dryer, a fluidized bed spray dryer, a powder filling and sealing machine, a tablet press, a coating machine, a granulation machine, a hard capsule machine, and a soft capsule machine.
