本发明涉及一种体外诊断检测试剂,具体涉及一种用于体外诊断的葡萄糖定量检测干片。
背景技术
所谓的“干片试剂”是传统的“湿化学“相对比较而言的。它是以被检测样品中的液体作为反应媒介,待测物直接与固定于载体上的试剂反应的一种方式,它与传统湿化学的最大区别就在于参与化学反应的媒介不同。随着生物化学中的酶的分离,提纯,储存等技术的发展,传感器,光度计,和电极技术的进步,以及计算机的普及,干化学也迅速发展起来。
与湿化学相比,干化学分析法有对应的检测仪器,操作简单,检验时间短,得到的结果准确等优点。检测的样本由最初的尿液到现在的全血、血清、血浆或其他液体,试剂干片的结构从定性到半定量的二层结构到能精确定量的多层膜结构,因此,干化学试纸法在临床检验的应用范围明显扩大,能进行的生化检验项目越来越多,美国奥森多公司推出的VITORS350,适用于急诊和常规生化的全自动分析系统,可以检测血清、血浆、全血、尿液及脑脊液等样本,检测项目包括肝功、肾功、心肌酶、血脂、蛋白、离子等组合项目或任意单项,真正实现结果的快速、准确、能充分满足临床准确、快速和灵活的诊断需求,但配套仪器过大,不能实现即时即地进行检测。
随着生物技术的进步,利用免疫渗透、免疫层析等技术研制出的含酶标记、荧光标记,尤其胶体金或硒标记抗体(或抗原)的免疫干片,可用于肌钙蛋白、特种蛋白、激素、某些治疗药物及病毒抗体或抗原等的分析测定,现在采用的荧光光度法的多层膜干片及其配套仪器也已出现,并被越来越多地使用,河南安图生物公司的阴道炎五联检测试剂,则是通过干化学酶法,检测特异性生化标记物,用于妇科疾病的诊断。经过20多年的发展,干化学分析技术已广泛应用于检验医学的各个方面,包括常规生化、内分泌激素、毒素药物浓度分析及特种蛋白等免疫检查。但荧光光度技术对仪器的要求比较高,在液相中,可以用来检测的分光光度计很贵,且结果不能长期保存;胶体金技术则面临产品质量相差较大,不能质控,不能确保质量,且存在后带现象,标本需稀释测试,只能进行定性,不能进行定量,故只作为初筛诊断试剂,用于疾病诊断的辅助治疗。
发明内容

为解决上述技术问题,本发明提供一种用于体外诊断的葡萄糖定量检测干片,包括带样品滴加孔的上外壳、扩散层、酶层、色素层、树脂层、支持层、带透光孔的下外壳,其中,上外壳与下外壳相互贴合,上外壳中心位置开有样品滴加孔,下外壳中心位置开有透光孔,扩散层、酶层、色素层、树脂层、支持层通过涂布工艺从上到下依次分层贴合在一起。
所述扩散层主要包括亲水性高分子材料以及白色反光材料组成,扩散层湿膜厚度为200~400μm,干膜厚度为60~300μm。
所述亲水性高分子材料为醋酸纤维素,所述白色反光材料为二氧化钛、硫酸钡、聚苯乙烯、微晶纤维素中的一种或多种组成的混合物。
所述酶层包括葡萄糖氧化酶、过氧化物酶、缓冲液以及亲水性聚合物。
所述色素层包括色素、缓冲液,亲水性聚合物,所述色素包括氢供体和耦合器的组合。
所述氢供体为4-氨基安替比林衍生物,耦合器为1-羟基萘衍生物。
所述亲水性聚合物包括凝胶、明胶衍生物、阿胶、聚乙烯醇、聚乙烯吡咯烷酮或聚丙烯酰胺,所述缓冲液选用磷酸缓冲液或柠檬酸缓冲液。
所述色素层及酶层湿膜厚度为50~300μm,干膜厚度为20~200μm。
所述的支持层为透明塑料,支持层的厚度为30~200μm。
所述树脂层是水性环氧树脂,所述树脂层湿膜厚度10~100μm,干膜厚度为2~50μm。
本发明带来的有益效果:本发明旨在提供一种用于体外诊断的干化学法多层膜试剂干片,该方法具有快速诊断、操作简单、节约成本等优势,能够提高检测的准确度、精密度和灵敏度,检测方法具有溯源性,试剂稳定,保存时间长。

具体实施方式
为使本发明的目的、技术方案和优点更加清楚,以下结合附图及具体实施例,对本发明作进一步地详细说明。
如图1所示,本发明提供一种用于体外诊断的葡萄糖定量检测干片,包括带样品滴加孔的上外壳1、扩散层2、酶层3、色素层4、树脂层5、支持层6、带透光孔的下外壳7。其中,上外壳1与下外壳7相互贴合。上外壳1中心位置开有用于样品滴加的样品滴加孔8,下外壳7中心位置开有透光孔9。扩散层2、酶层3、色素层4、树脂层5、支持层6通过涂布工艺分层贴合在一起。
本发明是由含干化学试剂的模块附着在坚固的塑料条上构成。血液中的各种化学检查内容与干化学试剂发生反应,使模块颜色发生变化,其颜色深浅或颜色变化的快慢与血尿液中相应化学成分的浓度成比例关系。
进一步来说,上述葡萄糖多层膜干片可用于定量测定血清、血浆、尿和脑脊髓液中的葡萄糖浓度。葡萄糖干片为涂覆在聚酯基材上的多层分析成分,样品通过上外壳样品滴加孔滴加到扩散层表面,通过扩散层均匀地分布到下面的试剂层。在葡萄糖氧化酶的催化下,样品中的葡萄糖被氧化产生过氧化氢和葡萄糖酸盐。随后过氧化物酶催化染剂前体进行氧化联结反应,生成一种染剂。该染剂的强度通过反射光测定,其强度与血尿液中葡萄糖的浓度成正比例关系。具体反应如下:
其中,扩散层主要包括亲水性高分子材料组成,如醋酸纤维素,以及白色反光材料,如二氧化钛、硫酸钡、聚苯乙烯、微晶纤维素、或以上反光材料的混合物;扩散层的主要作用包括均匀扩散样本,过滤样本中大分子物质和干扰物质,保护试剂层;扩散层湿膜厚度为200~400μm,干膜厚度为60~300μm,如在100~250μm内则效果更佳。所述试剂层则是待测物质发生化学反应的区域,与其他发明不同,本发明的试剂层包括色素层和酶层两层,色素层包括必要的色素、缓冲液,亲水性聚合物等必要生物制剂;酶层则包括葡萄糖氧化酶、过氧化物酶及缓冲液、亲水性聚合物等;色素包括氢供体和耦合器的组合,氢供体为4-氨基安替比林衍生物,如4-氨基安替比林,4-氨基-2-甲基-3-苯基-1-(2,4,6-三氯苯基)-3-吡唑-5-1等等;耦合器为1-羟基萘衍生物,如1,7-二羟基萘,钠或钾-羟基磺酸盐,等等;亲水性聚合物包括凝胶(如酸洗明胶,去离子胶)、明胶衍生物(如钛酸盐,羟甲基丙烯酸甲酯等等)、阿胶、聚乙烯醇、聚乙烯吡咯烷酮、聚丙烯酰胺等等,其中,明胶是首选。PH缓冲液的加入是为了保持氧化酶的最佳的PH值或附近,可选用磷酸缓冲液或柠檬酸缓冲液。色素层及酶层湿膜厚度50~300μm,干膜厚度为20~200μm,如在40~200μm内则效果更佳。所述的支持层可以由各种透明塑料,如聚乙烯PE、聚丙烯PP、聚酯甲基丙烯酸甲酯PMMA、聚苯乙烯PS、聚碳酸酯PC、聚对苯二甲酸乙二醇酯PET等,优选PET;支持层的厚度为30~200μm,50-150um为最佳;可见光范围的透光率85%以上,95%以上最佳,保证入射光源及反射光均可通过支持层。支持层与试剂层涂膜之间需借助具有粘附作用的的树脂层才能更好的附着在一起,进而降低对基材的材质和性能的要求,节约成本。树脂层湿膜厚度10~100μm,干膜厚度为2~50μm,如在10~20μm内则效果更佳。上外壳和下外壳组成的外壳层主要是保护试剂在运输,储存,测试的过程中由于各种力导致的形变,从而避免试剂的损坏,尤其是在上机测试中,无论是自动或半自动的将干片试剂送入检测机器中,干片不可避免的受到相应的力,试剂外壳可以避免试剂干片破损。上外壳和下外壳可采用塑料材质。
本发明干片试剂制备方法如下:
1、将树脂溶液(直接购买)均匀涂覆在基材上,在45°条件下干燥10min,干燥后形成厚度均匀的树脂层。
2、在干燥的树脂层上继续涂覆以下色素层溶液,在45°条件下干燥30min,干燥后形成厚度均匀色素层。色素层溶液的配方如下:
3、在干燥后的色素层上涂覆以下酶层溶液,在45°条件下干燥30min,干燥后形成厚度均匀的酶层。酶层溶液的配方如下:
4、在干燥的酶层上继续涂覆以下扩散层溶液,在25°条件下干燥5h,干燥后即完成干粉干片的制作。扩散层溶液的配方如下:
5、将干粉干片裁剪成一定大小的尺寸,装在与机器配套的干片外壳中,即完成干片的制作。
该方法制作的干化学试剂性能优越、试剂稳定、储存时间长、方便携带,完全实现即时即地检测的目标。
干片检测操作流程如下:
1、干片按需要量取出,回温1h;
2、将干片装入检测机器,取10ul全血/血浆/尿液/脑脊液直接滴加在干片上外壳的样本滴加中,测试5分钟出结果。
结果判断:参照参考值范围,超出范围即为异常值。

Claims (10)
1.一种用于体外诊断的葡萄糖定量检测干片,其特征在于,包括带样品滴加孔的上外壳(1)、扩散层(2)、酶层(3)、色素层(4)、树脂层(5)、支持层(6)、带透光孔的下外壳(7),其中,上外壳(1)与下外壳(7)相互贴合,上外壳中心位置开有样品滴加孔(8),下外壳中心位置开有透光孔(9),扩散层(2)、酶层(3)、色素层(4)、树脂层(5)、支持层(6)通过涂布工艺从上到下依次分层贴合在一起。
2.如权利要求1所述的用于体外诊断的葡萄糖定量检测干片,其特征在于,所述扩散层(2)主要包括亲水性高分子材料以及白色反光材料组成,扩散层湿膜厚度为200~400μm,干膜厚度为60~300μm。
3.如权利要求2所述的用于体外诊断的葡萄糖定量检测干片,其特征在于,所述亲水性高分子材料为醋酸纤维素,所述白色反光材料为二氧化钛、硫酸钡、聚苯乙烯、微晶纤维素中的一种或多种组成的混合物。
4.如权利要求1所述的用于体外诊断的葡萄糖定量检测干片,其特征在于,所述酶层(3)包括葡萄糖氧化酶、过氧化物酶、缓冲液以及亲水性聚合物。
5.如权利要求1所述的用于体外诊断的葡萄糖定量检测干片,其特征在于,所述色素层(4)包括色素、缓冲液,亲水性聚合物,所述色素包括氢供体和耦合器的组合。
6.如权利要求5所述的用于体外诊断的葡萄糖定量检测干片,其特征在于,所述氢供体为4-氨基安替比林衍生物,耦合器为1-羟基萘衍生物。
7.如权利要求4或5所述的用于体外诊断的葡萄糖定量检测干片,其特征在于,所述亲水性聚合物包括凝胶、明胶衍生物、阿胶、聚乙烯醇、聚乙烯吡咯烷酮或聚丙烯酰胺,所述缓冲液可选用磷酸缓冲液或柠檬酸缓冲液。
8.如权利要求1所述的用于体外诊断的葡萄糖定量检测干片,其特征在于,所述色素层及酶层湿膜厚度为50~300μm,干膜厚度为20~200μm。
9.如权利要求1所述的用于体外诊断的葡萄糖定量检测干片,其特征在于,所述的支持层(6)为透明塑料,支持层的厚度为30~200μm。
10.如权利要求1所述的用于体外诊断的葡萄糖定量检测干片,其特征在于,所述树脂层(5)是水性环氧树脂,所述树脂层湿膜厚度10~100μm,干膜厚度为2~50μm。
